Cultivation of Poria cocos using plastic bag method I-effect of temperature and number of plastic bag layers (비닐봉지 재배에 의한 복령 생산 I-환경온도 및 비닐 겹수의 영향)

  • Poria cocos is an edible and pharmaceutical mushroom with a long history of medicinal use in Korea. For the last 30 years, the domestic cultivated supply of Poria cocos has been unable to meet consumer demand, so Poria cocos is collected in mountainous areas and also imported from China. Thus, to increase the supply of Poria cocos, many artificial cultivation methods have been studied. In this study, Poria cocos is cultivated under different environmental conditions using plastic bags and the results compared. When cultivating Poria cocos at different temperatures (20, 25, 30 and $35^{\circ}C$) and using different numbers of plastic bag layers (1, 2), the most efficient cultivation conditions were a temperature of $25-30^{\circ}C$ and 2 plastic bag layers. The fastest growth was at $25-30^{\circ}C$, and the Poria cocos exhibited no weight change when cultivated using layers of plastic bags (1, 2).

Reuse of Holdfasts in Hizikia Cultivation (양식 톳 포복지의 재활용에 관한 연구)

  • In theprevious Hizikia cultivation, holdfasts were threw into the sea after harvesting in May; the young thalli(5$\~$10cm in length) of Hizikia are annually collected from natural bed by seed collectors for the cultivation, resulting in a ruined natural populations. Therfore, the reuse method of holdfasts by regeneration capability of Hizikia fusiformis, was investigated. The effects of emergence on the growth of regenerated thalli from holdfasts over 6 months of outdoor culture from May to November, 1995. The vegetative growth from the holdfasts was good under the emergence of 3hrs/day on the air than 0, 1 and 2hr/day. The regeneration of holdfasts was determined by measuring total length, number of stipe and weight. The growth was facilitated under the exposure condition of 1$\~$3 hrs/day on the air. Outdoor cultivation for the comparsion of to artifical natural seeds were conducted from December 1995 to May 1996. There was no significant differences(0.05< P) between the two kinds of seeds. Therefore, artificial seed maybe used as a replacement for the natural seed in Hizikia cultivation. From the results, an useful method was established to obtain young fronds for the cultivation using the reuse method of holdfast, to conserve the natural population of Hizikia.

The Artificial Cultivation of Oudemansiella mucida on the Oak Sawdust Medium

  • To produce fruiting bodies of Oudemansiella mucida, porcelain fungus, on the oak sawdust medium, additives suitable for the mycelial growth and fruiting body formation were screened. In general, the mycelial growth of the three strains of O. mucida used in this study have been good on oak sawdust mixed rice bran of $20{\sim}30%$. The mycelia incubated in potato dextrose broth for 7 days were inoculated on oak sawdust medium supplemented with various ratios of rice bran and incubated for 30 days at $25^{\circ}C$ in the dark condition until the mycelia of O. mucida fully colonized the media from top to bottom. Then, top surface of the media in the bottles were horizontally scratched with a spatula and filled with tap water for 3 hours. To induce the primordial formation of O. mucida, the bottles were transferred to the mushroom cultivating room under 12 hrs of light (350 lux) and dark condition with relative humidity of 95% at $17^{\circ}C$. The primordia of O. mucida were formed on the surface of oak sawdust media after 7 days of incubation. The mature fruiting bodies were observed 5 days after primordial formation. The fruiting bodies O. mucida were formed on oak sawdust medium mixed with 5 to 30% rice bran. However, abundant fruiting-bodies of O. mucida were produced in oak sawdust medium supplemented with 20% rice bran. This is the first report associated with an artificial fruiting body production of O. mucida in Korea.

The Change in the Properties of Seeding Pad by the Oil Palm EFB Fiber (오일팜 EFB 섬유 적용에 따른 육묘패드 특성변화)

  • The crop production technologies keep in improving and the cultivation becomes more standardization owing to the significant developments of various agricultural materials. The artificial soil and base system for root could be one of the major technologies for the modern cultivation especially for controlled horticulture. Although the perlite, cocopeat, and peat moss are the major components of the artificial soil and are broadly used for various application, there is a great need for the new alternative materials for overcoming the low nutrition and the possible shortage of raw materials. In this study, the application of oil palm EFB fiber as an alternative materials for artificial soil especially for the seeding pad components was evaluated. The changes in the structural properties and the functional properties such as moisture holding properties were compared by laboratory produced seeding pads with different mixture of oil palm EFB fiber. The addition of fibrillated EFB fiber resulted in the significant increase in durability of the seeding pad, which showed the possible application of EFB fiber to the seeding pad instead of the wood fiber (UBKP). The moisture holding properties and the germination condition characteristics of the EFB fiber showed the slight less than those of the cocopeat, which require more sophisticated study for improving the functional properties of seeding pad made of the EFB fiber.

  • Recently the artificial intelligence green house system, which collects automatically the informations of plants cultivation circumstances and controls the growing circumstances, is studied using temperature, humidity and illuminance sensors. In this paper, the inference for plants cultivation of optimum circumstance conditions is simulated on the internet bases by predicting the temperature, humidity and illuminance. On the IOT circumstances, the plant cultivation conditions of temperature, humidity and illuminance, using Arduino sensor, are transmitted to the manager on realtime and if the optimum condition of temperature and humidity for plant cultivation is not equal to the values, the system transmits automatically the SMS warning messages on realtime. Although the sudden climite conditions(snow, rain, hot weather) are occurred, the optimum condition of plant cultivation can be controlled. In this paper, using Fuzzy rules and WEKA TOOL, although the same flora temperature zone is used, the simulation is produced for the optimum value of temperature, humidity and illuminance for the zone.

Physical Characteristics of Korean Red Pines According to Provinces (Goseong, Hongcheon and Bonghwa-gun) (한국산 소나무의 지역(고성, 홍천 및 봉화군)에 따른 물리적 특성)

  • Physical characteristics of Korean red pine (Pinus densiflora) were investigated with different cultivation locations in Taebaek Mountains as Goseong-gun, Hongcheon-gun and Bonghwa-gun as experimental sites. Moisture content based on air-dried or green (artificial) wood was no significant differences with different cultivation places. Specific gravities of both sapwood and heartwood of red pine from Bonghwa-gun were higher than those from other two sites (Hongcheon-gun or Goseong-gun). Specific gravity of heartwood of red pine from Goseong-gun was higher than it from Hongcheon-gun, but this trend was opposite in case of sapwood. Higher specific gravity of red pine heartwood from Goseong-gun maybe resulted in higher strength than those of other sites. In shrinkage ratio, there was no significant difference among different cultivation places in radial directions, but red pine from Bonghwa-gun was higher than those of other sites in tangential directions. Hygroscopicity was no significant differences with different cultivation places. These results will be helpful information for efficient use of pinewood and good quality of pinewood production for genetical breeding improved.

Development of Carrot Medium Suitable for Conidia Production of Venturia nashicola

  • The causal fungus of pear scab, Venturia nashicola, grows slowly and rarely produces conidia on artificial media in the laboratory, but it produced conidia on the Cheongah medium containing Cheongah powder. V. nashicola grew too slow to produce conidia until 15 days after cultivation but produced conidia with $4{\time}10^4$ conidia/plate 30 days after cultivation on the Cheongah medium containing 1% Cheongah powder. V. nashicola showed a peak production of conidia with $4.5{\times}10^5$ conidia/plate 60 days after cultivation on the carrot medium containing 2% carrot powder, one of the constituents of Cheongah powder. The carrot medium is considered to be the best medium to obtain conidia of V. nashicola in the laboratory until now. This is the first report on the development of a suitable medium for conidia production of V. nashicola, as far as we know.

  • Shaking cultivation was carried out in 250 mL Erlenmeyel' flasks containing 60 mL of culture solution for 7 days. Artificial seawater was used as a basal culture medium. The cultivation was performed under the conditions that the temperature ranged from $4^{\circ}C$ to $32^{\circ}C$, the agitation speed 50 rpm to 200 rpm, and the sugar concentration 5 g/L to 35 g/L. The medium condition was cultured that sugar and nitrogen ratio 1.25 to 10. The biomass. the lipid content in biomass, and the DBA content of lipid were investigated according to the cultivation conditions. The lipid in biomass was distributed between 10 and 349'0, the DHA in lipid 34% to 43% of the lipid. and the biomass concentration 0.425 g/L to 4.50 g/L.

Cultural Characteristics of Phellinus baumii Grown in Bottle (장수상황버섯(Phellinus baumii)의 병재배 특성)

  • For artificial cultivation of Phellinus baumii, we have conducted a study on cultural characteristics and condition of fruitbody formation in sawdust cultivation. Mycelial density and incubation rate were higher in oak sawdust+rice bran (4 : 1) medium than oak sawdust 100% medium. However, primordia and fruitbody formation were higher in oak sawdust 100% medium than rice bran added medium. Bigger bottle capacity (1,100 ml) resulted in a little higher yield and more biological efficiency than 850 ml bottle.

  • We aim at providing consumers and farmers with various parts of mushrooms through development of new mushroom cultivation. For the mass artificial cultivation, we elucidated suitable environment conditions of Neolentinus lepideus. The results were summarized as follows. For the efficient cultivation of N. lepideus, temperature of mycelial growth and development of the fruit body were suitable $20{\sim}23^{\circ}C$ and $20^{\circ}C$, respectively.
