A Study on the Cultivation of Enteromorpha prolifera(Muller) J.Agardh, Chlorophyta in Korea (녹조식물 가시파래, Enteromorpha prolifera의 양식연구)

  • This study aimed to develop the methods of growing Enteromorpha prolifera natural seedlings in its natural habitat and artificial indoor seedlings by inducing spore release. Likewise, the study examined the possibility of mass production by developing cultivation techniques with cultivating examination. The natural seedling of E.prolifera thrived in a sea area composed of sand and mud, which Is its natural habitat. Growing of this alga on the seedling frame 20 cm high from the bottom at the intertidal zone in summer and 40 cm high in fall was found to be very effective. However, enabling the best attachment rate for artificial indoor seedling requires inducing spore release after drying the mature thalli in a dark place fur about 12∼24 hours and setting seedling nets in a dark water tank (spore solution) for 24 hours. Breeding E.prolifera in a pole-system farm is best done in shallow sea areas with mud or mud and sand geological feature. However, floating-system lam is better for deep-sea areas with fast current. Ideal farming places are sea areas with plenty of nutritional salt and safe places that protect the lam facilities against billows. Furthermore, an exposure method on seawater surface to produce larger output should be used.

  • Experiments were carried out to asses the impotance of environmental conditions upon the growth rate, length compositions seedling and spore liberation of a red alga Glacilaric verrucosa. The alga grown in water of low salinity mixed with fresh water shows better growth rate, lager length composition and excellent seedling, whereas the one vegetated in an area of tidal exposure shows a somewhat retarded growth rate and poor seedlings. The rate of growth Increases from August to October and begins to diminish from December continuing to diminish through February. Therefore the spore settling time extends from May to lune. Although the sun-drying method is the most eacient way of inducing spore liberation, further investigations are necessary in oiler to use this method for the application to artificial spore settling.

  • Topics discussed in this paper are engineering design of artificial reefs, design considerations of compressive strength and pH, materials of silica fume concrete and volcanic stones for reef construction, and field monitoring of artificial reefs. Six artificial reefs were deployed offshore at Biyangdo in July 2006, and succession patterns on the reefs were investigated bimonthly from July 2006 to October 2007. The spore-bag method, which has been widely used in Korea for the attachment of seaweed on artificial reefs, was applied to the developed reefs using the kelp Ecklonia cava, which is the dominant species in Jeju, and recruitment and growth patterns were observed.

Studies on the Ginseng Tea using Spore-forming Lactic Acid Bacteria (유포자성 유산균을 이용한 인삼차 개발에 관한 연구)

  • In order to develop ginseng tea powder with spore forming lactic acid bacteria Lactobacillus sporogenes was used. In the jar fermentor experiment under optimal culture conditions, the number of spore of L. sporogenes reached about $20{\times}10^8\;CFU/mL$ and sporulation rate was 97%. Granulated ginseng tea was made from glucose 7 kg, lactose 2 kg, ginseng extract 1 kg and spores 5 g $(5200{\times}10^8\;CFU/g)$. In the treatment of artificial gastric juice (pH 3.0) for 4 h and artificial bile for 8 h, the survival rate of spores in the granulated ginseng tea was 55.4% and 90.0% respectively. The spores survived 77.6% after incubation for 20 min in boiled water. Its storage stability was about 75% for 12 months at room temperature.

A study on the pot cultures of arbuscular mycorrhizal fungi in Korea (Arbuscular 내생균근 균의 포트배양에 관한 연구)

  • Four plant (Sorghum bicolor, Cassia mimosoides var. nomame, Sesamum indicum and Glycine soja) were cultivated at the pots including the soils containing arbuscular mycorrhizal fungi and were also investigated with the colonizations and productions of arbuscular mycorrhizal fungi. Whereas the colonizations of arbuscular mycorrhizal fungi continuosly increased on the roots until 50 days, the productions of arbuscular mycorrhizal fungal spores were fluctuated with the terms of 30 days after inoculated. This indicated that the colonizations on the roots were not correlated with productions of arbuscular mycorrhizal fungal spores. Also, the various soils collected were applied to this technique by using pot cultures. Out of 82 various soils collected, the spore productions of arbuscular mycorrhiaze were observed only from 42 soils. The spores cultured under artificial conditions were identified to 15 species with four genera. The spore productions of arbuscular mycorrhizal fungi using this technique would be considered to be related to the soil pH: The spore productions were found in the low pH for the species of Acaulospora and Glomus, the those near pH 7.6 for the species of some Glomus, Scutellospora and Gigaspora.

Modelling Algae Transport in Coastal Areas with Marine Afforestation (바다숲 조성해역의 해조류 포자 확산모델링)

  • To arrange effectively artificial reefs for marine afforestation, tidal currents were analyzed by numerical experiments, and particle tracking based upon tidal currents were carried out to clarify the path of algae spore. The experiments were carried out by EFDC (Environmental Fluid Dynamics Code), and water column was vertically divided 10 layers. Tidal current patterns showed to be affected by main current at outside of study area, and circle currents of two were observed from analysis of residual currents. Particle tracking were experimented for 15 days at 2 installation places in which artificial reefs for marine afforestation would be deployed. According to the results of particle tracking experiment, particle movement at St.1 showed belt type along coastal line, and St.2 showed ellipse type at 300~500 m distant from coastal line. It suggest that artificial reefs for marine afforestation should be installed belt zone at station of St.1 and ellipse zone at St.2. Modelling algae transport was also tested to account for local dispersion of algae spore due to the suspended materials.

Isolation and Characterization of a Protease-Producing Bacterium, Bacillus amyloliquefaciens P27 from Meju as a Probiotic Starter for Fermented Meat Products

  • This study was performed to select protease-producing Bacillus sp. as a potential probiotic starter for fermented meat products. In order to isolate protease-producing bacterium from meju, measured the diameter of the clear zone on agar plate (TSA, 1% (w/v) skim milk) and analyzed for intracellular protease activity, then 10 Bacillus-like strains were isolated. Three Bacillus-like strains (P19, P27, and P33) among 10 strains were able to tolerate in acidic condition (TSB, pH 2.5, 2 h incubation). These 3 strains were showed antimicrobial activity against food-borne pathogenic bacteria. These vegetative cells of 3 strains were showed a survival rate of 0.04% to 0.08% under the artificial gastric acidic condition (TSB, pH 2.5 with 1% (w/v) pepsin), but spore-forming cells were 56.29% to 84.77%. Vegetative cells of 3 strains were the least bile-resistant, while spore-forming cells of 3 strains showed higher survival rate more than 76% under artificial bile condition (TSB, 0.1% (w/v) oxgall bile). In these strains, P27 strain was finally selected as a good probiotic strain. P27 strain was tentatively identified as Bacillus amyloliquefaciens by API CHB kit and 16S rDNA sequence analysis. The results of this study suggest that B. amyloliquefaciens P27 can be used as a potential probiotic starter for fermented meat product.

Ocurrence of Clubroot Caused by Plasmodiophora brassicae on Kohlrabi in Korea (Plasmodiophora brassicae에 의한 콜라비 뿌리혹병 발생)

  • From 2016 to 2018, approximately 15% of kohlrabi were observed displaying significant clubroot symptoms in farmer's fields in Jeju, Korea. The initial infection appeared as hypertrophy of root hairs, and as the disease progressed, galls formation occurred on the main roots, finally disease progress resulted in yellowing and wilting of leaves. Pathogenicity was proven by artificial inoculation of plants with resting spore suspension, fulfilling Koch's postulates. The resting spore is one-celled, spherical and subspherical, colorless, and $3-5{\mu}m$ in diameter. On the basis of the morphological characteristics and phylogenetic analyses of internal transcribed spacer rDNA, the causal agent was identified as Plasmodiophora brassicae. To our knowledge, this is the first report on the occurrence of P. brassicae on kohlrabi in Korea.

  • This experiment was carried out to know the effects of artificial apawning of Armillaria matsutake in the Pinus densiflora stands. The results obtained were as followed: 1. The inoculation experiment resulted in only the development of hypha in the spore-spread plot and there appeared 2 fruit bodies in the spore injection plot and 4 fruit bodies in the hypha-transplanted plot. 2. The spore flushing plot method was considered a recommendable one in view of labor problem. 3. It was seemed that Armillaria matsutake was not a commensal fungus but a parasitic ectotrophic mycorhiza to pine trees since pine trees in the Armillaria matsutake-appearing plot grew remarkably slow comparing with the non-appearing plot. 4. Armillaria matsutake appearance only in 20 to 60 year-old pine trees seems to have a correlation with a special matter. Therefore the experimenter thinks that Armillaria matsutake spores must be inoculared only with 20 to 60 year-old pine trees.
