Effective Application of CF11 Cellulose for Detection of Apple scar skin viroid in Apple

  • The low virus titer in woody plant tissues and the presence of inhibitor compounds such as polyphenols, tannins and polysaccharides are common difficulties that compromise purification of plant viroids from their woody hosts. A simple, reliable method of RNA isolation using CF11 cellulose column on a microcentrifuge tube scale for detecting Apple scar skin viroid (ASSVd) in apple was developed. Total RNA extracted from leaf, woody bark and the fruit skin was used for reverse transcription. RT-PCR products could be detected from RNA prepared from dormant woody bark, fruit skin and fresh leaves with both the CF11 cellulose column method and NucliSens extractor in February, August and November. Meanwhile, with the RNeasy kit RT-PCR, products were detected only in leaves and not from bark or fruit skin. The PCR product, about 330 base pairs, was analyzed by agarose gel electrophoresis. The CF11 cellulose column method was effective for detecting ASSVd. The method enabled the processing of a large numbers of samples of dormant woody bark, leaf and fruit skin of apple.

Effect of Calcium on the Apple Varieties Resistance to Alternaria Leaf Spot and Mycelial Growth of Alternaria mali Roberts (Ca물질(物質)이 사과나무 반점낙엽병(斑點落葉病) 발생(發生) 및 병원균(病原菌)의 균사생육(菌絲生育)에 미치는 영향(影響))

  • This studies was carried out to find the effect of inorganic matters of Apple Leaves on the occurrence of Alternaria leaf spot and calicium compounds on mycelial growth of Alternaria mali. Diseased leaf rate of Alternaria leaf spot of susceptible apple varieties was 84.8%, moderate resistant 8.1% and resistant 0.3% respectively and in No. of lesion per leaf, the tendency was same. CaO contents of apple leaves was contained much in the resistant varieties than susceptible one. It was estimated that calcium is one of factors on resistance to apple leaf spot by the results of relations among CaO contents and diseased leaf rate from July to August and No. of lesion per leaf was correlated significantly as r=-0.551, r=-0.585, -0.485 but in T-N, $P_2O_5,\;K_2O,\;MgO,\;Na_2O$, there was no difference among varieties. Then, growth of Alternaria mali was possible in the media of which pH ranged up to $11{\sim}13$, and calcium has effect of inhibition on growth of Alternaria mali.

Etiology of Apple Leaf Spot Caused by Colletotrichum spp. in China

  • Glomerella leaf spot pathogens can infect apple leaves, causing extensive necrosis and premature defoliation, as well as necrotic spots on fruit. In recent years, the disease has been reported with increasing frequency in China, and appears to be spreading rapidly in some apple-producing areas. In this study, fungal isolates from diseased apples leaves collected in Henan and Shaanxi provinces were analyzed based on morphology, cultural characters, pathogenicity and molecular phylogenetics. It was found that Glomerella leaf spot of apple was caused by two pathogens, Colletotrichum fructicola and C. aenigma. Pathogenicity tests showed that C. fructicola and C. aenigma could infect apple leaves of cultivar Golden Delicious, as well as Gala, Qinguan, Pink Lady, Pacific Rose, Golden Century and Honeycrisp, all of which include Golden Delicious in their parentage. In wound inoculation experiments, C. fructicola and C. aenigma were pathogenic to fruit of Gala, Qinguan, Golden Delicious, Pacific Rose, Starkrimson and Fuji. With non-wounded fruit, C. fructicola was pathogenic to Gala and Golden Delicious, and C. aenigma was pathogenic to Gala. It is concluded that the two pathogens could be differentiated according to pathogenicity to leaves and fruits of different apple cultivars.

  • The Near Infrared Reflectance Spectroscopy(NIR) was used to evaluate nutritional diagnosis for rapid leaf analysis method, 177 'Fuji' apple and 130 'Campbell Early' grape leaves were measured by Near Infrared reflectance spectra in the NIR region(1,100~2.500nm). Total nitrogen content was measured by kjelldhal distillation, after salycilic acid-sulfuric acid digestion. An empirical equation to predict total nitrogen content from its spectral signature was developed by adapting the Near Infrared Reflectance Spectroscopy analysis(NIRa) technique and the results were apple-0.965(R). 0.086(SEC), grape-0.926(R), 0.152(SEC). Standard Error of Prediction(SEP) of NIRa for predicting the total nitrogen of apple and grape leaves was 0.360 and 0.210, respectively. It was concluded that Near infrared reflectance spectroscopy analysis is promising method for rapid analysis of apple and grape leaves.

  • A highly facile and eco-friendly green synthesis of Annona squamosa (custard apple) leaf extract reduced graphene oxide (CRG) nanosheets was achieved by the reduction of graphene oxide (GO). The as-prepared CRG was characterized with X-ray diffraction (XRD), transmission electron microscope (TEM), Fourier-transform infrared spectroscopy (FT-IR), ultraviolet-visible (UV-Vis), X-ray photoelectron spectroscopy (XPS) and Raman spectroscopic techniques. Removal of oxygen containing moieties from the GO was confirmed by UV-Vis, FT-IR and XPS spectroscopic data. The XRD and Raman data further confirmed the formation of the CRG. TEM images showed the sheet structure of the synthesized CRG. These results show that the phytochemicals present in custard apple leaf extract act as excellent reducing agents. The CRG showed good dispersion in water.

Production System of Virus-free Apple Plants Using Heat Treatment and Shoot Tip Culture (열처리와 경정배양을 이용한 바이러스 무병 사과 생산 시스템)

  • In worldwide, viral diseases of apple plants has caused the serious problems like reduced production and malformation of fruits. Also, the damages of apple plants by virus and/or viroid infection (Apple chlorotic leaf spot virus, Apple stem grooving virus, Apple mosaic virus, and Apple scar skin viroid) were reported in Korea. However there is few report about the protection approach against the infection by apple viruses. Therefore, this paper introduced the experimental protocol for the development of virus-free apple cultivars (Danhong, Hongan, Saenara, Summerdream). Apple plants were treated at $37^{\circ}C$ for 4 weeks and shoot tips were cultured in vitro. After heat treatment, the detection of apple viruses was performed by RT-PCR using virusspecific detection primers in new apple cultivars. With the heat treatments followed by in vitro shoot tip culture, the proportion of virus-free stocks of 'Danhong', 'Hongan', 'Saenara', and 'Summerdream' was 28%, 16%, 12%, and 12%, respectively. Taken together, this approach can be a good tool for production of virus-free apple stocks.

Combining ex vitro thermotherapy with shoot-tip grafting for elimination of virus from potted apple plants (기외 열처리와 경정접목을 이용한 사과 폿트묘에서의 바이러스 제거)

  • Apples are the most grown fruit crops in the fruit industry of Korea. However, virus or viroid infection such as apple mosaic virus (ApMV), apple stem grooving capillovirus (ASGV), apple stem pitting virus (ASPV), apple chlorotic leaf spot virus (ACLSV), apple scar skin viroid (ASSVd) causes fruit yield reduction and poor fruit quality. Therefore, in this study, we examined to established an efficient virus-free system to eliminate the most infected ASGV virus in domestic apple orchard. We investigated that the shoot growth rate and the virus removal rate in ASGV infected potted apples that were treated with heat treatment in a growth chamber (constant temperature/humidity device) maintained at 36℃, 38℃ and 40℃ for 4 weeks. Here we found that the shoot growth rate was the highest in the heat treatment group (36℃) and the virus was removed in the middle and top of the shoot but not in the bottom. The virus was did not removed in the 38℃ and 40℃ heat treatment group in all section of shoots, and the heat treatment group (40℃) died after 4 weeks of heat treatment without growth of shoots. We performed in vivo shoot-tip grafting using the shoot-tip of potted apple heat-treated at 36 ℃, and we also investigated the viability and virus removal rate, which showed 94% viability and 20% virus removal rate. Collectively, our results suggest that it would be possible to produce the virus-free apple plants through heat treatment and shoot-tip grafting.

Leaf Thinning and Fruit Quality of 'Hongro'/M.9 Apple Trees by Foliar Application of Cu-EDTA and Fe-EDTA

  • Hand leaf thinning for improvement of fruit coloration and quality is one of the most labor-consuming works in Korean apple cultivation. The study was aimed at investigating whether foliar application of two chelate compounds (Cu-EDTA, Fe-EDTA) could defoliate effectively 'Hongro'/M.9 apple leaves and enhance fruit coloration and quality. At 30 days before harvest, foliar spray with two chelate compounds defoliated significantly the apple leaves. Chelate treatments did not affect the leaf chlorophyll content. High concentration of chelates decreased drastically photosynthetic rate of true leaves, and then it stayed in low levels, whereas low levels of chelates reduced sharply the photosynthetic rate but it was gradually restored over time. Cu-EDTA regardless of its concentration triggered higher defoliation compared to Fe-EDTA. Consequently, higher defoliation improved light interception of fruits and accumulated more anthocyanin. Particularly, Cu-EDTA could target mainly fruit cluster leaves which affect directly light interception of the fruits, i.e. more selective defoliants compared to Fe-EDTA. However, 3% Cu-EDTA rather defoliated excessively, accompanying with reduction of fruit weight and soluble solids. Therefore, our results suggest that 1% Cu-EDTA which defoliates properly the fruit cluster leaves could improve coloration and quality of 'Hongro'/M.9 apple fruits.

Effect of Calcium Solution Spray on Fruit or Leaf on Calcium Accumulation into Apple Fruit (사과나무 과실과 잎에 살포된 칼슘의 과실로의 축적)

  • This research was conducted to investigate the effects of calcium solution spray on the accumulation of calcium into apple fruit. $^{45}_{CaCl_2}$ applied to fruit with different growth stages showed that more $^{45}Ca$ was penetrated into fruits when applied in the late stage than early stage. Slight radioactivity was detected only in pedicel except leaf when $^{45}Ca$ was treated on the leaves proximate to the fruit. When the Ca was treated on fruit surface only, calcium contents of fruit was increased.

Occurrence Status of Five Apple Virus and Viroid in Korea (국내 주요지역의 사과 바이러스 및 바이로이드 5종의 발생 현황)

  • The investigation of the infection rate of domestic apple orchards by four types of apple viruses (Apple chlorotic leaf spot virus [ACLSV], Apple stem pitting virus [ASPV], Apple stem grooving virus [ASGV], Apple mosaic virus [ApMV]) and one type of viroid (Apple scar skin viroid, ASSVd) found that most apple trees were infected with viruses and viroid at the rate of 97.3%. By region, the infection rate in Jeongseon stood at 98.8%, Danyang at 100%, Yesan at 100%, Jangsu at 89.1%, and Muju at 98.1%. By each virus and viroid, the infection rate of ASGV was the highest at 93.4%, followed by ASPV at 85.7%, ACLSV at 59.0%, ASSVd at 6.7%, and ApMV at the lowest 3.6%. In addition, 84.8% of the cases were infected with two or more types of viruses and viroid, nearly seven times the single type infection rate of 12.4%, and the cases infected with three viruses, ASPV, ACLSV, and ASGV accounted for 56.2%, more than the half the total number of trees investigated.
