Glucocorticoid Regulation of Gene Expression in Hippocampal CA3 and Dentate Gyrus (글루코코티코이드 호르몬에 의한 뇌해마의 CA와 Dentate Gyrus 부분의 유전자 발현 변화)

  • Glucocorticoids (GCs) alter metabolism, synaptogenesis, apoptosis, neurogenesis, and dendritic morphology in the hippocampus. To better understand how glucocorticoids regulate these aspects of hippocampal biology, we studied gene expression patterns in the CA3 (Hippocampal pyramidal cell field CA3) and dentate gyrus (DG). Litter-matched Lewis inbred rats treated for 20 days with either 9.5 mg per day sustained-release corticosterone or placebo pellets were compared with high-density oligonucleotide microarray analysis (Rat Neurobiology U34 Arrays, Affymetrix). In placebo-treated rats, 32 genes were expressed at greater levels in CA3 than DG, whereas 3 genes were expressed at great levels in DC than CA3. Regional differences were also apparent in corticosterone-induced changes in the hippocampal transcriptome. Six genes in CA3 and 41 genes in DC were differentially regulated by corticosterone. As per the glucocorticoid effects on gene transcription in the brain, forty three of these genes were upregulated, and 4 genes were downregulated. Genes differentially expressed in hippocampus included those for 13 neurotransmitter proteins, 5 ion channel related proteins, 4 transcription factors, 3 neurotrophic factors, 1 cytokine, 1 apoptosis related protein, and 5 genes involved in synaptogenesis. Interestingly, GCs can have suppressive effects on brain BDNF mRNA transcription, one of the neurotrophic factors. These results indicate the diversity of targets affected by chronic exposure to corticosterone and highlight important regional differences in hippocampal neurobiology.

Effects of Soil Water Potential and Nitrogen Fertilization on Characteristics of Photosynthesis and Chlorophyll Fluorescence Induction in Schisandra chinensis Baillon

  • Management of soil water and fertilization is known to primarily affect physiological properties and yield in plant. The effect of soil water potential and nitrogen application on characteristics of photosynthesis and chlorophyll fluorescence in Schisandra chinensis Baillon was investigated on a sandy loam soil. Net photosyntheis rate and transpiration rate increased as a photon flux density and was highest at -50kPa of soil water potential. Light compensation point ($1.5{\mu}molm^{-1}s^{-1}$) and dark respiration ($0.13{\mu}molCO_2m^{-1}s^{-1}$) was lowest at -50 kPa but maximum photosynthesis rate ($13.10{\mu}molCO_2m^{-1}s^{-1}$) and net apparent quantum yield ($0.083{\mu}molCO_2m^{-1}s^{-1}$) was highest at -50 kPa. As results of chlorophyll fluorescence by OJIP analysis, maximum quantum yield (Fv/Fm) of photosystem II (PSII) and PIabs was higher in treatments of -50 kPa and -60 kPa respectively, which reflects the relative reduction state of PSII. But the relative activities per reaction center such as ABS/RC and DIo/RC were low with decreasing soil water potential. Net photosyntheis rate and transpiration rate were highest at treatment of soil testing 1.0 times ($92kgha^{-1}$). Application of nitrogen resulted in high Fv/Fm, $PI_{abs}$ and low ABS/RC, DIo/RC. This result implies that -50 kPa of soil water potential and nitrogen fertilizer may improve the efficiency of photosynthesis through controlling a photosystem in Schisandra chinensis Baillon.

Scientific Comparison Study on the Joseon Dynasty Palace Roof Tiles and Modern Handmade Roof Tiles

  • The purpose of this study is to scientifically analyze physicochemical characteristics of the roof tiles used for palaces in the Joseon Dynasty which stored in Changdeokgung material storage and Seooreung Jaesil and the modern handmade ones which made by N company to understand the differences between their manufacturing techniques. Through chromaticity, cross-sectional observation, component analysis, and crystal structure analysis, it was possible to confirm the physicochemical properties and fired properties of the roof tile. Roof tiles from the Joseon Dynasty have a wider colorimetric range and higher apparent porosity and water absorption, on average, than the modern roof tiles. The cross section of the Joseon Dynasty roof tiles shows that most clay minerals have not been vitrified, remaining in the form of atypical particles, while the modern roof tiles have denser clay materials. X-ray diffraction analysis identified low-temperature minerals such as micas in Joseon roof tiles but no peak of these minerals was observed in the modern roof tiles, implying that the modern ones are fired at higher temperature than the Joseon ones. Therefore, the modern roof tiles are fired at higher temperature and have higher density than the Joseon ones due to the use of pugmills. The general content of main ingredients was similar between the two. Additionally, the principal component analysis of trace elements in the Joseon roof tiles showed that most samples were from similar areas. It seems that the Joseon roof tiles were manufactured using soils supplied from a specific region at the same timeframe and their consistency in the content of principal components implies that they also have similar mix proportions of clay.

Physicochemical Properties of Extruded Defatted Hemp Seed and Its Energy Bar Manufacturing (압출성형 삼종실의 이화학적 특성과 에너지바의 제조)

  • This study was to develop high-nutritious energy bar from extruded hemp obtained by extrusion process. Mixture of rice flour and defatted hemp was extruded at a barrel temperature of 110 and 130$^{\circ}C$, and moisture content of 20 and 25%. Properties of extrudates such as bulk density, expansion index, breaking strength, apparent elastic modulus, water absorption index (WAI), water solubility index (WSI) have been analyzed. The antioxidant potential was determined by the DPPH-radical scavenging assay. The expansion index was the highest in rice-hemp extrudate at 130$^{\circ}C$ barrel temperature and 20% moisture content among the other hemp-added extrudates. The WAI was increased with increase in moisture content, while the WSI was increased with increase in barrel temperature. The peak viscosity of rice extrudate had higher valule than those of extrudate added with hemp. DPPH scavenging activity of rice-hemp extrudate at 130$^{\circ}C$ barrel temperature and 20% moisture content showed the highest value. Sensory properties, moisture content and color were assessed for quality of energy bar. The color values of the energy bar indicated decreasing L (lightness) and b (yellowness), and increasing a (redness) after 30 days storage at ambient condition. The highest overall acceptable was the energy bar added with rice-hemp extrudate at 130$^{\circ}C$ barrel temperature and 20% moisture content.

Study on Optimum Mixture of Industrial By-Products for Lightweight Foamed Filler Production by Mixture Experimental Design (혼합물 실험계획법에 의한 경량기포 충전재 제조를 위한 산업부산물의 최적 배합 검토)

  • This research studied production of lightweight filling production for sink hole restoration utilizing various industrial by-products(2kinds of fly ash, petro-cokes CFBC ash, blast furnace slag fine particle). For this purpose, the mixed raw material properties(compressive strength) behaviors according to the blending ratio of industrial by-products were examined by applying the experimental design method and statistical analysis was performed using the commercial program MINITAB. Compressive strengths of industrial by-products were strongly dependent on blast furnace slag powder. Compressive strength(3days aging) was 3~11MPa depending on the amount of blast furnace slag powder used. The use of CFBC fly ash was evaluated to have the least effect on compressive strength. In addition, the compressive strength and the coefficient of permeability were measured by preparing foamed concrete for the experimental batch 1 condition in the mixture experimental design. In this case, the bulk density is 0.9 to 1.0, the apparent porosity is 30 to 50%, the compressive strength(3days old) is 1 to 2MPa, and the permeability coefficient is $10^{-2}$ to $10^{-3}cm/sec$.

Effects of Die Temperature and Moisture Content on the Quality Characteristics of Extruded Rice with Mealworm (사출구 온도와 수분함량이 갈색거저리(Mealworm) 첨가 압출성형 백미의 품질 특성에 미치는 영향)

  • This study aims to make extruded rice snack with high quality in texture and nutrition by adding mealworm. Addition of the mealworm has the merit to fill in high-quality protein and unsaturated fatty acids which are insufficient in rice. Thus, the physicochemical properties were investigated through the process of extrusion cooking. As the extrusion process varied, the die temperatures were set to $120^{\circ}C$ and $130^{\circ}C$. Also, the moisture contents were adjusted to 30% and 35%. The specific length, the expansion ratio, and the water absorption index increased as the added content of mealworm became higher. On the contrary, the density, the breaking strength, the apparent elastic modulus, and the water solubility index decreased. As mealworm and moisture content increased, DPPH radical scavenging activity significantly increased but the rancidity decreased. As a result, the addition of mealworm to the extruded rice snack was effective in improving texture, nutrition, and antioxidation.

Physicochemical properties of the materials used for the production of celadon maebyeong inlaid with cloud-and-crane designs and changes in their morphological properties by production stage (청자상감운학문매병 제작 재료의 물리화학적 특성 및 제작 단계별 형상학적 특성 변화)

  • In order to investigate the diverse physicochemical changes that occurred in traditional Korean pottery during its production, including before and after firing, this study produced six replicas of a celadon maebyeong inlaid with cloud-and-crane designs, respectively corresponding to the process of shaping, carving, inlaying designs, first firing, glazing and second firing, respectively. It then conducted a scientific study of these six replicas and analyzed their images through high-resolution three-dimensional transmission imaging. The materials used for the replicas show different mineral phases and even colors depending on the components of each material. For example, black inlay with a high content of iron oxide (Fe2O3) shows dark colors and white inlay with a high alumina (Al2O3) content appears white. Physicochemical properties such as chromaticity and magnetic susceptibility and major components of the replicas were confirmed by the differences in the density in the computed tomography (CT) images. The characteristics of fired products such as fine structure, absorption ratio, apparent porosity, and other characteristics of the major mineral components were identified by the presence of pores and the formation of cracks inside the replicas in the image analysis.

Effects of Psyllium Husk Content on the Physical Properties of Extruded Rice Flour (차전자피 함량에 따른 쌀 압출성형물의 물리적 특성)

  • This study was performed to determine the effect of psyllium husk addition on the physical properties of rice extrudates. Rice-based formulations mixed with psyllium husk (0, 7, 14 and 21%) were extruded at a die temperature of 140℃, screw speed of 200 rpm, and moisture content of 20%. As the content of psyllium husk increased, expansion ratio decreased, while piece density and specific length increased. Apparent elastic modulus, breaking strength, adhesiveness, and hardness augmented with an elevation in psyllium husk content. Lightness declined as psyllium husk content furthered, while redness, yellowness, and color difference intensified. Water soluble index and water absorption index increased with an increased amount of psyllium husk. In conclusion, the addition of psyllium affected the expansion of extruded rice snack possessing hard texture, small cells, and sticky texture due to higher water absorption during hydration.

Influence of Ga Content on the Ionic Conductivity of Li1+XGaXTi2-X(PO4)3 Solid-State Electrolyte Synthesized by the Sol-Gel Method

  • In this study, NASICON-type Li1+XGaXTi2-X(PO4)3 (x = 0.1, 0.3 and 0.4) solid-state electrolytes for all-solid-state batteries were synthesized through the sol-gel method. In addition, the influence on the ion conductivity of solid-state electrolytes when partially substituted for Ti4+ (0.61Å) site to Ga3+ (0.62Å) of trivalent cations was investigated. The obtained precursor was heat treated at 450 ℃, and a single crystalline phase of Li1+XGaXTi2-X(PO4)3 systems was obtained at a calcination temperature above 650 ℃. Additionally, the calcinated powders were pelletized and sintered at temperatures from 800 ℃ to 1,000 ℃ at 100 ℃ intervals. The synthesized powder and sintered bodies of Li1+XGaXTi2-X(PO4)3 were characterized using TG-DTA, XRD, XPS and FE-SEM. The ionic conduction properties as solid-state electrolytes were investigated by AC impedance. As a result, Li1+XGaXTi2-X(PO4)3 was successfully produced in all cases. However, a GaPO4 impurity was formed due to the high sintering temperatures and high Ga content. The crystallinity of Li1+XGaXTi2-X(PO4)3 increased with the sintering temperature as evidenced by FE-SEM observations, which demonstrated that the edges of the larger cube-shaped grains become sharper with increases in the sintering temperature. In samples with high sintering temperatures at 1,000 ℃ and high Ga content above 0.3, coarsening of grains occurred. This resulted in the formation of many grain boundaries, leading to low sinterability. These two factors, the impurity and grain boundary, have an enormous impact on the properties of Li1+XGaXTi2-X(PO4)3. The Li1.3Ga0.3Ti1.7(PO4)3 pellet sintered at 900 ℃ was denser than those sintered at other conditions, showing the highest total ion conductivity of 7.66 × 10-5 S/cm at room temperature. The total activation energy of Li-ion transport for the Li1.3Ga0.3Ti1.7(PO4)3 solid-state electrolyte was estimated to be as low as 0.36 eV. Although the Li1+XGaXTi2-X(PO4)3 sintered at 1,000 ℃ had a relatively high apparent density, it had less total ionic conductivity due to an increase in the grain-boundary resistance with coarse grains.

The Variations of Stratospheric Ozone over the Korean Peninsula 1985~2009 (한반도 상공의 오존층 변화 1985~2009)

  • The climatology in stratospheric ozone over the Korean Peninsula, presented in previous studies (e.g., Cho et al., 2003; Kim et al., 2005), is updated by using daily and monthly data from satellite and ground-based data through December 2009. In addition, long-term satellite data [Total Ozone Mapping Spectrometer (TOMS), Ozone Monitoring Instrument (OMI), 1979~2009] have been also analyzed in order to deduce the spatial distributions and temporal variations of the global total ozone. The global average of total ozone (1979~2009) is 298 DU which shows a minimum of about 244 DU in equatorial latitudes and increases poleward in both hemispheres to a maximum of about 391 DU in Okhotsk region. The recent period, from 2006 to 2009, shows reduction in total ozone by 6% relative to the values for the pre-1980s (1979~1982). The long-term trends were estimated by using a multiple linear regression model (e.g., WMO, 1999; Cho et al., 2003) including explanatory variables for the seasonal variation, Quasi-Biennial Oscillation (QBO) and solar cycle over three different time intervals: a whole interval from 1979 to 2009, the former interval from 1979 to 1992, and the later interval from 1993 to 2009 with a turnaround point of deep minimum in 1993 is related to the effect of Mt. Pinatubo eruption. The global trend shows -0.93% $decade^{-1}$ for the whole interval, whereas the former and the later interval trends amount to -2.59% $decade^{-1}$ and +0.95% $decade^{-1}$, respectively. Therefore, the long-term total ozone variations indicate that there are positive trends showing a recovery sign of the ozone layer in both North/South hemispheres since around 1993. Annual mean total ozone (1985~2009) is distributed from 298 DU for Jeju ($33.52^{\circ}N$) to 352 DU for Unggi ($42.32^{\circ}N$) in almost zonally symmetric pattern over the Korean Peninsula, with the latitudinal gradient of 6 DU $degree^{-1}$. It is apparent that seasonal variability of total ozone increases from Jeju toward Unggi. The annual mean total ozone for Seoul shows 323 DU, with the maximum of 359 DU in March and the minimum of 291 DU in October. It is found that the day to day variability in total ozone exhibits annual mean of 5.7% in increase and -5.2% in decrease. The variability as large as 38.4% in increase and 30.3% in decrease has been observed, respectively. The long-term trend analysis (e.g., WMO, 1999) of monthly total ozone data (1985~2009) merged by satellite and ground-based measurements over the Korean Peninsula shows increase of 1.27% $decade^{-1}$ to 0.80% $decade^{-1}$ from Jeju to Unggi, respectively, showing systematic decrease of the trend magnitude with latitude. This study also presents a new analysis of ozone density and trends in the vertical distribution of ozone for Seoul with data up to the end of 2009. The mean vertical distributions of ozone show that the maximum value of the ozone density is 16.5 DU $km^{-1}$ in the middle stratospheric layer between 24 km and 28 km. About 90.0% and 71.5% of total ozone are found in the troposphere and in the stratosphere between 15 and 33 km, respectively. The trend analysis reconfirms the previous results of significant positive ozone trend, of up to 5% $decade^{-1}$, in the troposphere and the lower stratosphere (0~24 km), with negative trend, of up to -5% $decade^{-1}$, in the stratosphere (24~38 km). In addition, the Umkehr data show a positive trend of about 3% $decade^{-1}$ in the upper stratosphere (38~48 km).
