붕어 고음추출물의 생리활성 (Biological Activities of Extracts from Crucian Carp)

  • The pharmacological effects of hydrocooked extracts of crucian carp have been investigated. All of the hydrocooked extracts showed the contractile effect on the isolated rat duodenum and decreased to a certain extent the normal blood pressure in anesthetized rat. The hydrocooked extracts also exhibited a dose-dependent relaxation on the isolated rat aorta precontracted with 50 mM KCl. All of these except for RM 60 fraction haying cytotoxic effect against MCF-7 human breast cancer cell showed neither antibacterial activity nor antitumor activity. In addition, we found that although body weight gain in the hydrocooked extracts treated group were all reduced significantly compared to control group, there were no significant changes in the hematological parameters, except that the levels of high-density lipoprotein was significantly increased. These results suggest that the hydrocooked extracts of crucian carp may contain a variety of bioactive materials.

저령(Grifola umbellata)의 균핵에서 추출한 조다당류의 면역활성 및 항암 효과 (Immuno-modulatory and Antitumor Effect of Crude Polysaccharides Extracted from Sclerotium of Grifola umbellata)

  • 저령의 균핵으로부터 중성염용액, 열수 및 메탄올 추출물을 분리하였다. 세포독성 실험 결과, 중성염용액 추출물은 $0{\sim}2,000\;{\mu}g/ml$의 농도에서 NIH3T3, Sarcoma 180 및 MCF-7에 대한 세포독성이 없었으나, 메탄올 추출물에서는 $1,000\;{\mu}g/ml$ 이상의 농도에서는 독성을 나타내었다. Sarcoma 180 복수암에 대한 항암 효과는 중성염용액 추출물을 투여한 실험군에서 66.74%의 높은 생명 연장 효과를 나타내었으며, 암세포의 생 세포 수 또한 54.2% 감소시키는 효과를 나타내었다. 중성염용액 추출물은 대조군에 비해 보체 대체 경로에서의 항보체 활성을 $85.05{\sim}88.73%$, B 임파구의 alkaline phosphatase 활성을 6배 이상 증가시킴으로써 면역 활성 효과를 향상시켰다. 또한 중성염용액 추출물을 50 mg/kg body weight의 농도로 마우스 복강에 투여하였을 때 대조군에 비하여 복강세포수가 1.7배 증가하였으며, 혈액 내 백혈구 수 또한 3.6배의 증가를 나타내었다. 간, 비장 및 흉선 등의 면역 관련 장기의 체중에 대한 중량을 측정한 결과, 중성염용액 추출물 투여군은 대조군에 비해 증가된 수치를 보였으며, 혈액생화학적 검사를 시행한 결과, 대조군과 유사한 경향을 나타내었다. 중성염용액 추출물의 총 다당류와 단백질의 함양은 각각 98.25%와 1.44%로 측정되었다. 따라서 저령균핵의 중성염용액 추출물의 항암 효과가 암세포에 대한 직접적인 세포독성에 의한 것이 아니고 면역활성에 의한 것으로 판단된다.

Antitumor effect of Careya arborea against Ehrlich ascites carcinoma with reference to lipid peroxidation and enzymatic and non enzymatic antioxidant system in Swiss albino mice

  • The methanol extract of stem barks of Careya arborea Roxb. (MECA) (Family- Myrtaceae) was evaluated for antitumor activity and antioxidant status against Ehrlich's Ascites Carcinoma (EAC) bearing Swiss albino mice. After 24 h of tumor inoculation the MECA was administered at the doses of 50, 100 and 200 mg/kg body weight/mice/day for 14 days. After the last dose and 18 h fasting mice were sacrificed. The effect of MECA on the growth of transplantable murine tumor, life span of EAC bearing hosts, hematological profiles, serum and liver biochemical parameters were estimated. The MECA showed significant (P < 0.01) decrease in ascites volume, packed cell volume and viable cell count and prolonged the life span of EAC tumor bearing mice. Hematological profiles reverted to more or less normal levels in extract treated mice. The MECA also produced protective effect by decreasing the activity of serum enzymes, bilirubin and increase the protein and uric acid levels. MECA significantly (P < 0.05) decreased the levels of lipid peroxidation, while significantly (P < 0.05) increased the levels of glutathione content, vitamin C, vitamin E, superoxide dismutase and catalase CAT. The results indicate that MECA exhibited significant antitumor and antioxidant activity in EAC bearing mice.

민긴뿌리버섯(Oudemansiella radicata)의 자실체로부터 추출한 조다당류의 항암 및 면역 활성 효과에 관한 연구 (Studies on Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Fruiting Body of Oudemansiella radicata)

  • 민긴뿌리버섯은 송이버섯과에 속하는 버섯으로 예로부터 식용은 물론 항암, 고혈압 및 진균감염증의 치료에 널리 이용해온 식의약용 버섯이다. 민긴뿌리버섯의 자실체로부터 중성염용액, 열수 및 메탄올을 이용하여 조다당류를 추출하여 Sarcoma 180에 접종된 ICR mice에 주사하여 수명연장 및 항암효과를 조사하였다. 세포독성 실험결과, 각각의 세포는 $10{\sim}1000\;{\mu}g/ml$ 추출물 농도에서 70% 내외의 생존율을 보여 세포독성을 나타내지 않았다. 각각의 조다당류가 투여된 실험군이 대조군에 비해 평균수명이 각각 $42.9{\sim}66.7%$ 연장되었다. 중성염용액 추출물은 B 임파구의 alkaline phosphatase 활성을 대조군과 LPS군에 비해 약 $1.4{\sim}3$배 내외의 증가율을 보였다. 총 복강 세포수도 대조군에 비하여 최고 3.5배 정도 증가하였으며, 혈액 중 백혈구의 수도 대조군에 비하여 약 2.5배 증가하였다. 그리고 면역에 관련된 장기인 간, 비장 및 흉선의 체중이 대조군에 비하여 증가된 것을 확인하였다.

삼색도장버섯(Daedaleopsis tricolor)에서 추출한 조다당류의 면역 활성 및 항암 효과 (The Immuno-Modulatory and Antitumor Effects of Crude Polysaccharides Extracted from Daedaleopsis tricolor)

  • 삼색도장버섯으로부터 중성염용액, 열수, 및 메탄올 추출물을 분리하였다. 세포독성 실험 결과, 열수 추출물은 $0{\sim}2,000\;{\mu}g/ml$의 농도에서 암세포에 대한 세포독성이 없었으나, 중성염용액 추출물과 메탄올 추출물에서는 약간의 독성을 나타내었다. Sarcoma 180 복수암에 대한 항암 효과는 중성염용액 추출물을 투여한 실험군에서 77.4%의 높은 생명 연장 효과를 나타내었다. 삼색도장버섯의 중성염용액 추출물과 열수 추출물은 대조군에 비해 비장세포를 $1.7{\sim}2.4$배 증가시켰고, B 임파구의 alkaline phosphatase 활성을 $2.2{\sim}8.7$배 증가시킴으로써 비장세포의 증식능과 B 임파구의 면역 활성 효과를 증가시켰다. 대식세포주 RAW 264.7에 $50;{\mu}g/ml$의 농도로 중성염용액 추출물을 처리하였을 경우, 양성 대조군이 $79\;{\mu}M$의 nitric oxide(NO)를 발생시킨 것에 비해 다소 높은 $90\;{\mu}M$의 NO를 발생되었다. 또한 중성염용액 추출물을 50 mg/kg body weight의 농도로 마우스 복강에 투여하였을 시 대조군에 비하여 복강세포수가 10배 증가하였으며 혈액 내 백혈수 또한 2배 증가를 나타내었다. 따라서 삼색도장버섯의 중성염추출물은 항암 효과와 숙주의 면역을 활성화시키는 것으로 사료된다.

  • Background: $\beta$-1,3-glucans are well known to enhance the immune reactions, resulting in antitumor, antibacterial, antiviral, anticoagulatory and wound healing activities. $\beta$-1, 3-glucans have various activities depending on molecular weight, degree of branching, conformation, water-solubility and intermolecular association. However, the $\beta$-1,3-glucan linked backbone structure is essential and $\beta$-D-glucopyranosyl units are required for immuno-potentiating activities. Result: In this study, we tested the immunophamacological activities of soluble $\beta$-1,3-glucans and confirmed the following activities: (1) $IFN-{\gamma}$ production in PBMCs in the presence or the absence of PHA, LPS, or IL-18; (2) induction of various cytokines in the spleen and thymus; (3) adjuvant effect on the antibody production; (4) nitrogen oxide synthesis in macrophages; (5) the cytotoxic and antitumor effects on cell lines and ICR mice. Conclusion: These results strongly suggested that $\beta$-1,3-glucans possessed various immuno-pharmacological activities.

Screening for in vitro Cytotoxic Activity of Seaweed, Sargassum sp. Against Hep-2 and MCF-7 Cancer Cell Lines

  • Discovery of anticancer drugs that kill or disable tumor cells in the presence of normal cells without undue toxicity is a potential challenge for therapeutic care. Several papers in the literature have emphasized the potential implications of marine products such as seaweeds which exhibit antitumor activity. Study attempts to screen the antitumor effect of Sargassum sp, against chosen cell lines such as MCF-7 (Breast cancer) and Hep-2 (Liver Cancer). Ethanol extract of Sargassum sp. was concentrated using a Soxhlet apparatus and dissolved in DMSO. In vitro cytotoxic activity of Sargassum sp at various concentrations ($100{\mu}g/ml-300{\mu}g/ml$) screened for antitumor effect against the chosen cell lines using MTT assay (3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide, a yellow tetrazole). The study documented that the percentage of cell viability has been reduced with increased concentration, as evidenced by cell death. Sargassum sp extract shows potential cytotoxic activity ($P{\leq}0.05$) with $IC_{50}$ of $200{\mu}g/ml$ and $250{\mu}g/ml$ against Hep-2 and MCF-7 cell lines respectively. The ethanol fraction of Sargassum sp induced cell shrinkage, cell membrane blebbing and formation of apoptotic bodies with evidence of bioactive components as profound influencing factors for anti-tumor effects. Further research need to be explored for the successful application of Sargassum sp as a potent therapeutic tool against cancer.

  • Alcohol extracts of six plant namely Coix agrestis L$_{OUREIRO}$, Chloranthus Japonicus (Siebi) N$_{AKAI}$, Ajuga Spectabilis N$_{AKAI}$, Juglans mandsuria Max., Cirsium Macki var Ussuriense $K_{ITA}$ and Sasamorpha Purpurascense (Hack) N$_{AKAI}$ were subjected to the screening tests for anti-tumor activities against SN-36 Leukemia, Sarcoma 180 and Ehrlich ascites carcinoma. Of the six extracts, Ajuga Spectabilis N$_{AKAI}$ and Coix agrestis L$_{OUREIRO}$ were found to be potential in survival effect of tumor transplanted mice.

  • DA-125, a new anthracycline antitumor antibiotic, was administered at dose levels of 0, 0.2, 0.6 and 1.8 mg/kg/day intravenously to pregnant New Zealand White rabbits from day 6 through 18 of gestation. The does were subjected to the caesarean section on day 28 of gestation. Effects of test agent on general toxicity of does and embryonic development of F1 fetuses were examined. At 1.8 mg/kg, the organ weight for ovary of does was significantly decreased. The decrease in the number of corpus lutea, implantations and litter size, and the increase in the rate of resorptions were also observed. In addition, various types of external, visceral and skeletal malformations occurred in fetuses at an incidence of 7.7, 7.7 and 20.6%, respectively. The results show that the no effect dose levels (NOELs) of DA-125 are 0.6 mg/kg/day for does and F1 fetuses.

전갈(全蝎)이 노화(老化)에 따른 2단계(段階) 발암화(發癌化) 과정(過程)에 미치는 영향(影響) (Effects of Buthus martensi Karsch on tumor promotion in two-stage carcinogenesis in mice)

  • To clarifiy the activating effects of Buthus martensi Karsch(BMK) on tumor promotion in two-stage carcinogenesis in mice was investigated. In vivo system, BMK was seen to gave an inhibitory activity on TPA-induced mouse ear edema. In addition, the BMK was proved to have antitumor-promoting activity in two-stage mouse skin carcinogenesis induced by DMBA and two-stage mouse lung carcinogenesis induced by 4-NQO as a initiator plus TPA and glycerol as a promoter. Moreover, BMK significantly exhibited an cytolytic effect in HepG2 cells and showed significant antitumor activity against Sarcoma-180 bearing mice by oral administration. These results suggest that BMK could be effective in adjuvant chemotherapy for human cancer.
