Cerulein 유도 급성췌장염 마우스모델에서 자가분해 조절과 항산화 활성에 미치는 [6]-gingerol의 영향 ([6]-Gingerol Attenuates Autophagy and Increases Activities of Antioxidative Defense Enzymes in Mice with Cerulein-induced Acute Pancreatitis)

  • 열대아시아 원산의 다년생 초본 생강의 주성분인 [6]-gingerol은 항산화 및 항염증 등의 특성이 잘 알려져 있지만 cerulein 유도 급성췌장염에서의 자가분해 관련 유전자 발현 조절과 항산화 효소 활성에 대한 연구는 거의 없다. 본 연구에서는 cerulein 유도 급성췌장염 동물모델에서 [6]-gingerold의 자가분해 조절과 항산화 작용을 조사하였다. 급성췌장염 유발 전 4일 동안 [6]-gingerol (0.1 mg/20 g mouse/day)을 경구투여 한 후 $50{\mu}g/kg$ cerulein을 복강주사로 급성 췌장염을 유도하였다. 그 결과 혈중 ${\alpha}$-amyase 활성, 자가분해 표적 유전자(Beclin-1 및 cleaved LC3-II)의 발현, 지질과산화는 [6]-gingerol 투여군에서 유의적으로 감소하였으며, 항산화지표 효소인 SOD와 GSH-Px 활성은 [6]-gingerol 투여군에서 유의적으로 증가하였다. 이상의 결과들은 천연식물소재 생강의 유효성분 중 하나인 [6]-gingerol이 cerulein 유도 급성 췌장염에서 자가분해 조절과 감소된 항산화효소 활성을 강화하는 효과를 나타내므로 생강이 급성췌장염의 예방과 치료에 대한 기능성 식품소재로 그 활용이 매우 높을 것으로 사료된다.

톳(Hijikia fusiforme) 추출물이 고지혈증 횐쥐의 지질대사 및 간 효소활성에 미치는 영향 (Effects of Hijikia fusiforme Extracts on Lipid Metabolism and Liver Antioxidative Enzyme Activities in Triton-Induced Hyperlipidemic Rats)

  • 본 연구는 용매별 톳 추출물이 고지혈증을 유발한 횐쥐의 혈청과 간의 지질 및 항 산화 효소활성을 측정하기 위하여 체중이 평균 100 g 되는 Sprague-Dawley계 수컷 횐쥐를 7마리씩 7군으로 나누어 1군은 정상군, 2군 이하 7군까지는 용매별 톳 추출물을 체중 kg당 0.5 mg의 용량을 복강으로 일주일간 주사한 다음 마지막 날 체중 kg당 triton 0.5 mg을 복강으로 투여하여 고지혈증을 유발하여 나타난 결과는 다음과 같다. 혈청 및 간 중 유리 콜레스테롤함량은 핵산층 투여군이 대조군에 비해 유의적으로 낮게 나타난 반면 HDL-콜레스테롤 함량은 대조군에 비해 에탄올층과 핵산층 투여군이 유의적으로높았다. 혈청 및 간의 총 콜레스테롤 함량의 경우 대조군에 비해 수층 투여군이 유의적으로 낮게 나타났으며, 특히 간 은 에탄올층 투여군과 에틸아세테이트층 투여군이 대조군에 비해 낮게 나타났다. 수층 투여군의 혈청 중 중성지방함량은 대조군에 비하여 에탄올층 투여군과 핵산층 투여군이 유의하게 낮게 나타났다. 간 중 TBARS 함량은 대조군에 비해 수용성, 비수용성 톳 분회물 투여군의 TBARS함량이 모두 낮게 나타났다. 간 중 SOD의 활성도는 대조군에 대해 톳 수층 투여군이 낮은 경향이었고, 비수용성 대조군에 비해 핵산층 투여군이 유의적으로 낮은 경향을 보였다. 간의 catalase 활성도의 경우 정상군에 비해 대조군과 분획물 투여군이 높은 경향을 보였으나 에틸아세테이트층 투여군은 정상군 수준 이하로 떨어졌다. 이러한 결과로 볼 때 톨은 생체내 항 고지혈증, 항 고콜레스테롤 및 항 산화효과가 있음을 알 수 있었다.

Oxidative Stress in Ovariectomy Menopause and Role of Chondroitin Sulfate

  • Oxidative stress due to reactive oxygen species (ROS) can cause oxidative damage to cells. Cells have a number of defense mechanisms to protect themselves from the toxicity of ROS. Mitochondria are especially important in the oxidative stress as ROS have been found to be constantly generated as an endogen threat. Mitochondrial defense depends mainly on super-oxide dismutase (SOD) and glutathione peroxidase (GPx), whereas microsomal defense depends on catalase (CAT), which is an enzyme abundant in microsomes. SOD removes superoxide anions by converting them to $H_2O$$_2$, which can be rapidly converted to water by CAT and GPx. Also, GPx converts hydroperoxide (ROOH) into oxidized-glutathione (GSSG). Ovariectomized (OVX) rats are used as an oxidative stress model. An ovariectomy increased the levels of MDA, one of the end-products in the lipid peroxidative process, and decreased levels of the antioxidative enzymes; SOD, CAT and GPx. However, Chondroitin sulfate (CS) decreased the levels of MDA, but increased the levels of SOD, CAT and GPx in a dose-depen-dent manner. Moreover, inflammation and cirrhosis of liver tissue in CS- treated rats were sig-nificantly decreased. These results suggest that CS might be a potential candidate as an anti oxidative reagent.

Inhibitory Effect of Ligularia fischeri var. spiciformis and Its Active Component, 3,4-Dicaffeoylquinic Acid on the Hepatic Lipid Peroxidation in Acetaminophen-Treated Rat

  • To find the action mechanism of the MeOH extract (LFS) of Ligularia fischeri var. spiciformis herbs (Compositae) and its active component, 3,4-dicaffeoylquinic acid (DCQA) on antihepatotoxicity, the effect was investigated on hepatic lipid perxodation and drug-metabolizing enzyme activities in acetaminophen-treated rat. Pretreatment with 250 mg/kg LFS (p.o.) and 10 mg/kg DCQA (p.o.) significantly decreased hepatic lipid peroxidation caused by acetaminophen injection. Further, LFS and DCQA inhibited hepatic microsomal enzyme activation such as hepatic P-450 cytochrome $b_5$, aniline hydroxylase and aminopyrine N-demethylase, suggesting that the two substances might effectively prevent the metabolic activation or scavenge electrophilic intermediates capable of causing hepatotoxicity. Both LFS and DCQA increased hepatic glutathione content and glutathione reductase activity, indicating that both resultantly prevented hepatotoxicity via antioxidative mechanism. Therefore, it was found that LFS had antihepatotoxicity based on the antioxidative action of DCQA.

홍삼 추출물의 제조에서 단백질 분해효소의 활용 (Potential of proteolytic enzyme treatment for production of Korean red ginseng extract)

  • 본 연구에서는 홍삼 추출물 제조과정에서 추출 수율을 향상시킬 수 있는 효소를 선별하고 최적의 반응조건을 조사하였다. 상업용 단백질 분해효소 중 Alcalase가 단백질과 탄수화물 수율 향상에 효과적이었으며, 효소 사용량은 홍삼 중량의 2%, 반응 시간은 1.5시간이 적당하였다. 최적의 반응조건으로 홍삼을 Alcalase로 처리한 결과 대조구보다 고형분 수율은 45.1%에서 71.1%로 50% 이상, 총페놀 함량은 0.44%에서 0.80%로 80% 이상 증가하였으며, 항산화 활성은 대조군과 매우 유사하였다. 또한 진세노사이드 함량은 대조구의 1.48 mg/g에서 1.98 mg/g으로 30% 이상 증가하였다.

겨울철 저온스트레스에 의한 문주란 잎의 항산화효소 활성과 O-J-I-P 곡선 패턴 (Antioxidative Enzyme Activities and O-J-I-P Transients of Crinum asiaticum var. japonicum $B_{AK}$. Leaves under Winter Stress)

  • 본 연구는 환경요인의 변화, 특히 겨울철 저온이 식물체에 미치는 영향을 정량적으로 밝히기 위해, 저온감수성 식물인 문주란을 대상으로 겨울철 일주기에 있어서 항산화효소 활성과 O-J-I-P곡선의 변화를 조사하였다. 겨울철의 superoxide dismutase와 peroxidase의 활성은 여름철에 비해 다소 증가하였다. 특히, peroxidase는 겨울철 새벽과 밤에 높은 활성을 보였으며 초겨울에만 특이적으로 검출되는 isoenzyme들로 관찰할 수 있었다. 또한 겨울철 문주란 잎의 O-J-I-P곡선에서 J, I, P-단계의 형광세기가 여름철에 비해 현저하게 감소하였다. 그리고, O-J-I-P곡선으로부터 산출된 주요 형광변수들 중에 Fm과 $\Phi_{po}$는 겨울철 저온에 노출되는 기간이 길어짐에 따라 각각 $30\%$$50\%$로 감소하였으며, ABS/RC는 초겨울보다는 늦은 겨울에 2배정도 증가하였다.

오공(蜈蚣)의 항산화효과에 관한 연구 (Antioxidative Effects of Scolopendra subspinipes)

  • Objective: The purpose of this study was to evaluate the antioxidative effects of the extract of Scolopendra subspinipes which has been used mainly for detoxication in the oriental medicine and reported to have sedative action, antiinflammatory effect, antihypertensive property and immunity enhancing activity. Method: Inhibitory activities on oxygen radical generating enzymes (aldehyde oxidase and xanthine oxidase) and increasing activities on oxygen radical scavenging enzymes (superoxide dismutase, glutathione peroxidase, glutathione-S-transferase) were investigated. Furthermore, the content of glutathione in the mouse brain, DPPH radical scavenging activity and also anti-lipid peroxidative effects in vivo and in vitro were estimated. Results: The extract showed weak inhibitory effects on the activities of aldehyde oxidase and xanthine oxidase which are oxygen radical generating enzymes. The extract inhibited lipid peroxidation with 26.1% against control group at 500 mg/kg in vivo and with 11.2% against control group at 10 mg/kg in vitro in a dose-dependent manner, which means this drug may protect radical-induced cell damages. The extract showed dose-dependently the scavenging effect on DPPH radical with 24.8% activity at 10 mg/ml in vitro. The extract enhanced the activities of superoxide dismutase, glutathione peroxidase and glutathione-S-transferase, which are oxygen radical scavenging enzymes, with 28.9%, 22.3% and 23.1%, respectively at 500mg/kg in vivo. Finally, this extract strongly increased the glutathione content in the mouse barin. Conclusion: Above results indicated that Scolopendra subspinipes can be useful for the protection or treatment of some diseases caused by reactive oxygen species.

감태 효소 가소분해물을 이용한 고품질 간고등어의 개발 및 특성 (Development and Characterization of High Quality Salted Mackerel Using Enzyme Hydrolysates of Ecklonia cava)

  • This study was conducted to develop and characterize of a high quality salted mackerel using enzymatic extracts of Ecklonia cava (EEC). In this study, potential antioxidative properties of EEC were evaluated by DPPH free radical scavenging activity, hydrogen peroxide scavenging activity, peroxide value, and fatty acid composition, and the antimicrobial properties were also measured by analysis for volatile basic nitrogen, pH, viable cells, Eschericia coli and biogenic amine. Compared to EEC-untreated salted mackerel, the salted mackerel with EEC was superior in antioxidative properties, while was negligible in the difference of antimicrobial properties. These results suggested that the high quality salted mackerel with antioxidative activity could be developed by treatment of EEC.

식이 철 수준과 커피 섭취가 흰쥐의 산화스트레스와 항산화효소 활성에 미치는 영향 (Effect of Dietary Iron and Coffee Intake on Oxidative Stress and Antioxidative Enzyme Activities of Rats)

  • Iron deficiency is a severe nutritional problem in the world. Coffee intake of the people is increasing every year and it can increase the loss of several essential body minerals including iron. Either iron deficiency or coffee intake may increase the oxidative stress of the body. However, the effect of iron deficiency and/or coffee intake on peroxidation have not been studied much. Therefore, the aim of this study was to investigate the effect of coffee intake on oxidative stress and antioxidative enzyme activities of iron-deficient rats. Forty-eight male rats of Sprague-Dawley strain were divided into two groups by dietary iron levels. Iron deficient group were fed 5 ppm iron diet and iron-sufficient group were fed 50 ppm iron diet. Each iron group were divided into three sub-groups by coffee levels (0%, 1%, 4%) included in the experimental diet. The experimental diets were fed for 4 weeks. The hemoglobin level was significantly low in iron deficient group and the level was exacerbated by high coffee intake. The malondialdehyde concentration of the plasma and liver were not affected by iron or coffee level in this study. However, plasma aspartate aminotransferase and alanine aminotransferase, the indicator of the liver damage, were increased by high coffee intake. The erythrocyte and liver superoxide dismutase (SOD) activities were elevated in iron deficient groups. Coffee intake increased erythrocyte SOD activity in iron sufficient groups. Glutathione peroxidase and catalase activities were not influenced much by either iron or coffee intake. In conclusion, high coffee intake in iron deficiency may not only increase the anemia symptoms, but also may increase the oxidative stress of the body.(Korean J Nutrition 35(9) : 919~925, 2002)
