천금연수단(千金延壽丹)이 노화 유발된 철쥐의 항산화효능에 미치는 영향 (An anti-Dxidative Effect of Cheongeumyeonsudan of Rats Induced Aging by D-Galactose)

  • Objectives : The purpose of this study is to find out the effect of antioxidation related to aging of Cheongeumyeonsudan which is written on Dongui-bogam experimentally. Method : 14 weeks aged SD albino rats were separated into uncontrolled group, controlled group and CGY group. As controlled and CGY groups were induced aging by subcutaneous injection of D-galactose, at the same time we administered the extract of Cheongeumyeonsudan to CGY group for 6 weeks. After then we drew blood from each group, and took measurements; the activity of SOD, GSH-px, catalase in erythrocytes, TBARS value, concentration of total lipid, tryglycende, total cholesterol, HDL-cholesterol in blood plasma. Results : The activities of SOD, GSH-px in erythrocytes were significantly increased in the CGY group compared with control group. The activity of catalase showed a tendency to increase, but it was nor remarkable. The concentration of total lipid, the values of TBARS and total cholesterol was significantly decreased in the CGY group compared with control group, and the concentration of plasma HDL-cholesterol was not remarkable. The concentration of tryglycende in plasma showed a tendency to decreased. Conclusions : it is suggested that Cheongeumyeonsudan decreased the activities of free radical, the concentration of lipid in plasma and generate enzyme which form lipid peroxide.

약용식물 추출물에 대한 항산화성과 알코올 탈수소효소 저해성 연구 (Activities of Antioxidation and Alcohol Dehydrogenase Inhibition of Methanol Extracts from Some Medicinal Herbs)

  • 32종의 생약재를 메탄올로 추출한 후 진공 건조한 후 각추출물의 생리활성 물질 및 알코올 대사와 관련된 효소활성을 측정하였다. DPPH법으로 free radical 소거능을 측정한 결과 정향, 녹차, 목단, 적양은 90% 이상의 소거활성을 보이고, 부위별로 살펴보면 줄기껍질> 잎 > 열매 > 뿌리 순으로 활성이 나타났다. 아질산염 소거능은 천초, 적양, 녹차, 지구자목, 팔각향 등이 우수한 소거활성을 보였고, 이 또한 부위별로는 줄기껍질이나 잎에서의 활성이 더 우수한 것으로 나타나 여러 생약재 중 줄기껍질이나 잎에 존재하는 페놀성 화합물이나 flavonoids류가 이와 같은 기능성에 영향을 줄 것으로 간주된다. ADH 활성 측정은 일반적으로 숙취현상에 효과가 있다고 알려져 있는 생약재 13종을 사용하여 알코올 탈수소효소 저해를 측정한 결과 13종의 생약재 모두 90% 이상의 강한 저해효과를 보였다. 그러므로 free radical 소거능이나 아질산염 소거능이 우수한 몇몇 생약재, 즉 녹차, 적양 등은 알코올의 대사와 관련하여 알코올의 대사과정 중 ADH의 활성을 저해하므로써 간 보호 효과와 숙취현상 억제에 효과적이라 사료된다.

청기화담환(淸氣化痰丸) 추출물이 비만쥐의 지질강하, 항산화효과 및 전염증성 Cytokines 생산에 미치는 영향 (Effects of Cheunggihwadamhwan Extract on Lowering Lipid, Antioxidation and Production of Proinflammatory Cytokines in Rats Fed on High Fat Diet)

  • Objectives The purpose of this study was to investigate the effects of Cheunggihwadamhwan (CGHDH) extract on lowering lipid, antioxidation and production of proinflammatory cytokines in rats fed on high fat diet. Methods 40 Male Sprague-Dawley rats were fed on high fat diet for 8 weeks and 32 rats (above 400 g) were randomly divided into 4 groups (8 mice in each group) : control group, 100 mg/Kg CGHDH group, 200 mg/Kg CGHDH group, 300 mg/Kg CGHDH group. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And We fed each experimental group of rats basal diet and administered an extract of Cheunggihwadamhwan extracts (100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid in plasma and liver, concentration of proinflammatory cytokines, antioxidative activity and gene expression. The gene expression level was investigated by the way of reverse transcription-polymerase chain reaction (RT-PCR). Results 1. Concentration of plasma FFA, plasma TG, plasma total cholesterol and plasma LDL-cholesterol showed a significant decrement in Cheunggihwadamhwan groups. However, concentration of plasma HDL-cholesterol showed a significant increment in 200, 300 mg/kg Cheunggihwadamhwan groups. 2. Concentration of liver total cholesterol and liver TG showed a significant decrement in Cheunggihwadamhwan groups. 3. Concentration of plasma TBARS showed a significant decrement in all Cheunggihwadamhwan groups. Concentration of liver TBARS showed a significant decrement in 200, 300 mg/kg Cheunggihwadamhwan groups. Concentration of liver GSH-Px, SOD and CAT showed a tendency to decrease in all Cheunggihwadamhwan groups. 4. Concentration of plasma IL-$1{\beta}$, plasma IL-6, TNF-$\alpha$ and NO, showed a tendency to decrease in all Cheunggihwadamhwan groups. Concentration of plasma IL-10 showed a tendency to increase in all Cheunggihwadamhwan groups. 5. In the analysis of reverse transcription-polymerase chain reaction (RT-PCR), the gene expression of Apo-B and Apo-E in the Cheunggihwadamhwan groups showed a low expression than that of control group. The ratio of Apo-B expression per $\beta$-actin expression in the showed a significant decrement in all Cheunggihwadamhwan groups. The ratio of Apo-E expression per $\beta$-actin expression in the showed a significant decrement in 300 mg/kg Cheunggihwadamhwan groups. Conclusions According to this study, the extract of Cheunggihwadamhwan showed a positive effect of lowering lipid, antioxidation and a control of producing proinflammatory cytokines.

Study on Physiologically Active Compounds and Antioxidant Activity of Korean Yam (Dioscorea batatas DECNE.)

  • The bioactive compound and antioxidant property of Korean yam (Dioscorea batatas DECNE.) were studied using in vitro methods. Yam available in Korea was analyzed for lycopene, chlorophyll a, b, tannin, phytic acid and total saponin contents. 70% Methanol, 70% ethanol and chloroform-methanol mixture (CM, 2:1, v/v) were used to extract yam. Then the antioxidant activity evaluated through ferrous ion chelating activity, ${\beta}$-carotene bleaching method, lipid peroxidation inhibition and nitric oxide (NO) radical scavenging activity. 70% Methanol extract showed the highest ferrous ion chelating activity and NO radical scavenging activity. And CM extract was the most effective in inhibition of linoleic acid peroxidation evaluated by ${\beta}$-carotene bleaching assay and lipid peroxidation inhibition assay. Based on the results obtained, yam is a potential active ingredient that could be applied in antioxidation as well as bio-health functional food to take a good part in prevention of human diseases and aging.

금앵자(金櫻子) 추출물(抽出物)이 음경해선체(陰莖海線體)의Nitric oxide synthase 활성(活性) 및 항산화효과(抗酸化效果)에 미치는 영향(影響) (Effect of the Rosae laevigatae Fructus extract on the nitric oxide synthase activity and antioxidan action in Rat's corpus cavernosum penis)

  • Rosae laevigatae Fructus extract (RLF) was tested for the effects on the urethral nitric oxide synthase (NOS) activity and Antioxidation in streptozotocin (STZ) induced diabetic rats. RLF was treated firstly into samples, and then STZ induced diabetic rats were set with them. In vitro, the urethral NOS activity was not noted but the type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide were decreased in the level of Dose of extract prepared from RLF. In vivo, after the extract was administered to the animal model for fifteen days, the urethral NOS activity increased in STZ induced diabetic rats to the level of normal rats. The content of urethral nitrite and glutathione followed by RLF pre-medicating administration, increased as highly as normal group in compare with the group treated with STZ. The type O activity and type conversion ratio of xanthine oxidase and the level of urethral lipid peroxide followed by RLF pre-medicating administration, decreased as lowly as normal group in compare with the group treated with STZ. In conclusion, the extract of RLF will be able to restore erectile dysfunction of STZ induced diabetic rats.

매실추출물이 활성산소종 소거효과와 B16F1 세포에서 멜라닌 생성에 미치는 영향 (Effect of Fruit Extract of Prunus mume on the Scavenging Activity of Reactive Oxygen Species and Melanin Production in B16F1 Cells)

  • 매실나무(Carthamus tinctorius L.)는 한국, 일본과 중국에서 전통적으로 약효가 있는 음식으로 사용되어 왔다. 특히 과일은 위염증 및 위궤양에 도움이 되는 생물학적 효과가 있다고 보고되었다. 그러나 피부미백과 관련된 효능에 대한 연구는 이직까지 미진하다. 따라서, 본 연구에서는 항산화 및 피부미백에 대한 매실추출물의 효능이 조사되었다. MTT assay를 이용한 세포생존에 대한 결과로 매실추출물은 0.1% 이하의 농도에서 세포독성이 없는 것으로 나타났다. 다음에 매실추출물의 환원력 뿐만 아니라 DPPH radical, hydrogen peroxide 및 superoxide에 대한 직접적인 소거효과가 in vitro에서 평가되었다. 그것은 이상의 활성산소종에 대한 소거효과를 발휘하게 하는 우수한 환원력을 가지고 있다. 더욱이 산화적 스트레스와 연관된 genomic DNA 손상에 대한 보호효과가 농도에 따라 증가하는 것이 관찰 되었다. 뿐만 아니라 L-dopa에 의하여 유발되는 멜라닌 생성에 대한 억제효과를 나타내었다. 그것은 또한 Western blot 분석에서 항산화와 괸련된 NRF-2, SOD-1 및 SOD-2 발현수준을 감소시킨다는 것이 발견되었다. 이러한 결과들은 매실추출물이 항산화에 의한 멜라닌생성의 억제를 통하여 피부 미백효과를 발휘할 수 있다는 것을 암시한다.

홍경천 추출물의 생리활성 (Biological Activity of the Fractions Extracted from Rhodiola dumulosa)

  • 홍경천(Rhodiola dumulosa)을 기능성 식품소재로 개발하기 위하여 홍경천(뿌리)을 80% 메탄올로 열수 추출하여 용매 분획한 후 이들에 대한 항산화, 암세포 성장 억제 및 대식세포의 면역활성 등과 같은 기능성을 조사하였다. 홍경천 메탄올 추출물의 헥산, 에틸 아세테이트, 부탄올, 메탄올 및 물 분획물의 수소공여능은 100 ${\mu}g/mL$의 농도에서 각각 28.30, 53.21, 35.48, 42.64 및 21.14% 로서 에틸아세테이트 분획물이 가장 높았으며, 이는 합성 항산화제인 BHT와 비슷한 항산화 효과를 나타내었다. A549 및 SW480 와 같은 암세포에 용매 분획물 1, 10 및 100 ${\mu}g/mL$의 농도로 48시간 처리한 결과 에틸아세테이트 분획물이 다른 분획물에 비하여 암세포주의 성장을 강하게 억제하였으며, 그 활성은 농도 의존적으로 나타났다. 또한 100 ${\mu}g/mL$의 농도로 48시간 동안 에틸아세테이트 분획물을 암세포에 처리시 처리구에서의 암세포는 뚜렷한 세포수의 감소와 함께 심한 형태학적 변화가 관찰되었다. 대식세포주(RAW 264.7)에 1, 10 및 50 ${\mu}g/mL$의 농도로 용매 분획물을 처리한 후 NO의 생성량을 측정한 결과 10 μg/mL 이상의 농도에서 메탄올 분획물에서 NO 생성을 강하게 유도하였다. 따라서 본 결과는 홍경천의 기능성 식품소재로의 활용 가능성을 시사한다.

재래식 메주 및 된장 중의 항산화성 물질에 관한 연구 4. 페놀화합물의 함량과 항산화력 (Antioxidative Materials in Domestic Meju and Doenjang 4. Separation of Phenolic Compounds and Their Antioxidative Activity)

  • In order to investigate the antioxidant activity of phenolic compounds contained in domestic Meju and Doenjang , the methanolic extract from defatted Meju and Doenjang was fractionated into the phenolic acid and the isoflavone fractions by alumina column and polyamide-6 column chromatography, respectively. Both phenolic acid and isoflavone fractions exhibited an identical antioxidative effect against the oxidation of linoleic acid. GC analysis revealed that the phenolic acid fractio contained vanillic , chlorogenic ,p-coumalic , ferulic, and caffeine acid. The content of caffic acid was greater than 70% in the phenolic acid fraction and the content of vanillic and chlorogenic acids were disappeared during fermentation, and thus, the facts are thought to be reasons for the low antioxidant activity of the phenolic aicd fraction from Doenjang. Similarly, the isoflavone fraction cotained daidzin, glycitin-65-0 glucosider and genistin and their aglycons such as daidzein, glyciten and genistein. The content of daidzin and genistin in Meju dramatically decreased at the early stage of fermentation ,whereas the content of daidzein an dgenistein rather increased ; however, these change in isoflavone contents did not affect the antioxidant activity of isoflavone fractions.

Characterization of Grape Seed Oil

  • Grape seed oil was characterized to assess the usefulness in the food industry. Among the various oils, the initial antioxidant activity was the highest for grape seed oil. Heating the oil at $180^{\sim}C$ for 20 min retained 86% of the initial activity. Grape seed and sesame oils showed a low peroxide value, about 2, implying a less oxidative reaction. The oxidation of grape seed oil was increased to a less extent by heat-treatment than other oils. Light exposure for 1 month resulted in a slight decrease in the antioxidant activity of grape seed oil, maintaining 96% of the initial activity. Other oils were all light-susceptible and the activities decreased significantly. The peroxide values of all the oils increased by light exposure, but the extent of oxidation was still the least for grape seed oil. The addition of grape seed oil to perilla oil was very effective, in that the peroxide value was 5-times decreased by 1 : 5 composition of grape seed oil versus perilla oil. These results indicate that grape seed oil can be used as a good cooking oil or an additive for other oils.

락토바실러스 람노서스 유래 엑소좀의 항염증, 항산화 효능 및 피부 장벽 개선에 관한 연구 (A Study on Anti-Inflammatory, Antioxidant Activity and Skin Barrier Improvement of Lactobacillus rhamnosus-derived Exosomes)

  • 본 연구에서는 락토바실러스 람노서스(Lactobacillus rhamnosus) J2K-821 균주로부터 한외여과를 통해 엑소좀을 분리하고, 피부에 대한 여러 효능을 평가했다. 나노입자추적분석 결과 3.22 × 108 particles/mL 농도의 크기가 50 ~ 200 nm인 락토바실러스 람노서스 유래 엑소좀(LRDEs)을 확인하였다. 염증 완화 효과를 검증하기 위해 LRDEs를 lipopolysaccharide (LPS)로 염증반응을 유도시킨 RAW 264.7 대식세포에서의 nitric oxide (NO) 생성 저해능을 확인한 결과, 농도 의존적으로 NO 생성을 저해하였다. 그리고 항산화 및 피부 장벽 개선 효능을 평가하기 위해 2,2-diphenyl-1-picrylhydrazyl (DPPH) 라디칼 소거 활성, hyaluronidase 억제 활성을 확인한 결과, 농도 의존적으로 항산화 및 hyaluronidase 억제 활성이 증가하는 것을 확인하였다. 이를 통하여 LRDEs는 항염증, 항산화 및 피부 장벽 개선에 효과적인 천연 화장품 소재로 사용 가능할 것으로 사료된다.
