Antimutagenic Compounds Indentified from Chloroform Fraction of Persimmon Leaves

  • Methanol extract of dried persimmon leaves was refractionated using hexane, chloroform, ethylacetate, n-butanol aqueous fractions. Among these chloroform fraction showed the highest inhibition rate on the mutagenicities of aflatoxin B₁(AFB₁) and 3,2' -dimethyl-4-amino-bipheny1(DMAB) in Salmonella typhimurium TA 100. Chloroform fraction was further fractionated into eight fractions by silica gel column chromatography and thin layer chromatography(TLC). The fraction 5 on TLC exhibited the highest antimutagenic activities in AFB₁and DMAB. 2,4-Decadienal, dihydro-4-methyl-2(3H)-furanone, hexanoic acid 1,4-bis(1-methy1 ethy1)benzene, heptanoic acid, phenol, octanoic acid, nonanoic acid and benzoic acid were tentatively identified from this antimutagenic fraction by GC-MS.

  • The extracts of aerial parts of Artemisia princeps var. orientalis, A. argyi, A. capillaris, A. rubripes and A. laciniata were investigated on the antimutagenic effect againt aflatoxin $B_1$ $(AFB_1)$. The methanolic extracts of 5 Artemisia species reduced the mutagenicity of $AFB_1$. And also 4,5-O-dicaffeoylquinic acid isolated from A. princeps var. orientalis revealed the antimutagenic activity.

  • The methanol extract of aerial part of Angelica keiskei Koidzumi exhibited a strong antimutagenic activity against aflatoxin $B_1\;(AFB_1)$. N-methyl-N'-nitro-N-nitrosoguanidine and 4-nitroquinoline-1-oxide in the Ames test with Salmonella typhimurium TA100. Cynaroside, isolated front ethylacetate fraction of the methanol extract Over silica gel, inhibited the mutagenicity of $AFB_1$ with an inhibition value of 96% at 1.0 mg/plate concentration and 87% at 0.5 mg/plate concentration. Other compounds, hyperoside, sucrose and luteolin-7-rutinoside, isolated from ethylactate or n-butanol fraction, also showed antimutagenic effect.

민간 생약재의 아플라톡신 $B_1$에 대한 항돌연변이 효과 (Antimutagenic Effects of Traditional Herbal Drugs on the Aflatoxin $B_1$)

  • The antimutagenic effects of 46 kinds of medicinal plants that have been used as traditional folk antitumor agents in Korea were studied by using Ames mutagenicity test. Most of the methanolic extracts from the plants which were used in this experiment showed strong antimutagenic activity toward aflatoxin B1(AFB1) in Salmonella typhimrium TA100 and TA98. However, N-methyl-N'-nitro-N-nitrosoguanidine(MNNG) induced mutagenicity was not blocked by adding the methanolic extracts of the plants except persimmon leaves (Diospyros kaki Thunberg)and Elaeagnus umbellata.

별불가사리의 항돌연변이 활성 (Antimutagenic Activity of Asterina pectinifera)

  • The antimutagenic activities of the total extract and several fractions of starfish, Asterina pectinifera (Asteriidae), were investigated in vitro by SOS chromotest with Escherichia coli PQ37 and Ames test with Salmonella typhimurium TA100. When various fractions was tested, the chloroform and butanol fractions showed low induction factors, which means both fractions increased antigenotoxicity against the base substitution mutagen MNNG. Even though higher antigenotoxic effect of the chloroform fraction, no effective result of Ames test was found in revertant formation of S. typhimurium TA100. The most effective antigenotoxic and antimutagenic fraction was a butanol one: i.e., When 0.5 mg/tube of butanol fraction was applied, the induction factor was 0.68. As the concentration of the fraction was increased the formation of revertants of S. typhimurium TA100 by about 81%. There was no cytotoxic effect of butanol fraction against S.typhimurium TA100. This result might be useful for further study to search a possible anticancer agent from the starfish, Asterina pectinifera.

  • The nutritional evaluation, and antimutagenic and anticancer effects of Kimchi were reviewed. Kimchi contains high levels of vitamins including vitamin C, $\beta$-carotene, vitamin B complex, niacin, and of minerals such as calcium, potassium, iron and phosphorous, etc. Kimchi is a low energy food, byt contains high quantities oforganic acids, dietary fiber and lactic acid bacteria in addition to the vitamins and minerals. Thus Kimchi could be developed as a protective food as it contains the various regulatory nutrients. The levels of NO3, NO2 and nitrosamines in Kimchi ingredients and Kimchi during theripening are not significant. However, high level of NaCl that could be used when prepared Kimchi in the warm region can be a problem, since high concentraitons of NaCl(>9.5%) in Kimchi could be comutagenic to themutagen of MNNG. The methanol extract of Kimchi, red pepper powder, garlic and lactic bacteria revealed antimutagenic or anticancer activities. The Kimchi extract also inhibited the growth of various human cancer cells.

  • 동치미에서 분리한 유산균인 Leu. mesenteroides subsp.cremoris DLAB19 균주의 aflatoxin Bl에 대한 항 돌연변이원성 물질 생산을 위한 최적 조건을 조사한 결과, 탄소원으로 glucose를 첨가시 가장 높은 항 돌연변이 효과를 나타내었으며, 질소원으로서는 yeast extract 첨가시 항 돌연변이 효과가 우수하였다. 탄소원으로 glucose의 농도을 2$\%$ 첨가시aflatoxin Bl에 대한 항 돌연변이 효과가 가장 우수하였으며,질소원으로서 yeast extract 1$\%$ 첨가시 가장 우수한 항 돌연변이 효과를 나타내었다. 항 돌연변이원성 물질 생산을 위한 최적 배양 조건은 초기 pH, 배양 온도, 진탕 속도가 각각 7.0, 30$^{\circ}C$ 및 150 rpm이었다. 상기의 최적 조건에서 36시간 배양시 가장 높은 항 돌연변이 효과를 나타내었는데 S. enterica serovar Typhimurim TA100과 TA98을 이용한 경우항 돌연변이 효과가 87.11$\%$와 75.04$\%$이었다.

양파 열수추출물의 항산화능 및 인공소화후의 항돌연변이 효과 (Antioxidant and Antimutagenic Activities of Hot Water Extract from White and Yellow Onions after Simulated Gastric Digestion)

  • 양파 열수 추출물의 항산화능 및 인공소화 전과후의 항돌연변이 효과를 BHT와 ascorbic acid와 비교하여 조사하였다. 총 페놀 및 플라보노이드의 함량은 노란색 양파 추출물이 횐색 양파 추출물보다 많았으며, 양파 추출물의 과산화수소 소거능은 농도가 증가함에 따라 비례적으로 증가하였다. $\beta-carotene-linoleate\;system$에서 항산화능과 reducing power는 증가되었으나, BHT와 ascorbic acid보다는 낮았다. 양파열수 추출물의 인공소화 후에 IQ와 MNNG로 처리된 Salmonella typhimurium TA80과 TA100에 대하여 항돌연변이 효과가 나타났으며, 노란색 양파가 횐색 양파보다 높게 나타났다. 결론적으로 양파 추출물을 소화시키면 항산화 및 항돌연변이의 효과는 양파의 페놀과 플라보노이드의 성분 변화에 의한 것으로 판단된다.

표고버섯 에탄올 추출물의 산화적 스트레스 억제 효과와 항돌연변이 효과 (Antioxidative Stress and Antimutagenic Effects of Lentinus edodes Ethanol Extracts)

  • The antioxidative effect and antimutagenic capacity in ethanol extracts of Lentinus edodes were studied for suggestion of prevention and dietetic treatment of chronic diseases and development of antioxidative and antimutagenic functional food by employing biological and biochemical assay. The $IC_{50}$ of MDA with BSA conjugation reaction, lipid peroxidation and scavenging effect on DPPH radical in ethanol extracts of Lentinus edodes showed 74.58 mg/assay, 5.747 mg/assay and 0.939 mg/assay respectively. So, the most effective antioxidative capacity in ethanol extracts of Lentinus edodes was the scavenging effect on DPPH radical, among the method used this study. The indirect and direct antimutagenic effects of ethanol extracts of Lentinus edodes were examined by Ames test using Salmonella typimurium TA98 and TA100. The inhibition rates on indirect mutagenicity mediated by 2-anthramine showed 91.67% in the Salmonella typimurium TA98 and 96.60% in the Salmonella typimurium TA100. The inhibitory effect on direct mutagenicity mediated by sodium azide in Salmonella typimurium TA100 was 22.83%. and mediated by 2-nitrofluorene in Salmonella typimurium TA98 was 5.34%. This data indicates that ethanol extracts of Lentinus edodes have more effective effects on indirect mutagenicity than direct mutagenicity. From this result, it believed to have a possible antioxidative and antimutagenic capacities, and taken for the candidate of prevention and dietetic treatment of chronic diseases and development of antioxidative and antimutagenic functional food.

소금의 종류에 따른 염장 고등어의 항돌연변이효과 및 암세포 성장억제효과 (Antimutagenic and Anticancer Effects of Salted Mackerel with Various Kinds of Salts)

  • Raw meat of mackerel (Scomber japonicus) was salted by refined, sun-dried, bamboo, and KC1-added bamboo salts. Antimutagenic activity on N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in Ames test and growth inhibitory effects of AGS human gastric and HT-29 human colon adenocarcinoma cells were investigated using methanol extracts of the salted mackerels. Mackerel salted sun-dried, bamboo, and KC1-added bamboo salts used increased the antimutagenic activities against MNNG, however, the sample treated with refined salt reduced the antimutagenic activity. Inhibitory effects of the salted-mackerels on the growth of human cancer cells were increased as dose dependent pattern. Mackerel salted with refined salt activated the growth of AGS human gastric adenocarcinoma cells, but mackerel salted with sun-dried, bamboo, and KC1-added bamboo salts kept or increased anticancer effect compared to the raw mackerel. Mackerel salted with KC1-added bamboo salt led to the highest antimutagenic and anticancer activities. These results suggest that antimutagenic and anticancer effects of mackerel during manufacturing of the salted-mackerel could be enhanced by using different kind of salts such as bamboo, or KC1-added bamboo salts.
