  • In the present study, anti-inflammatory activity of amygdalin isolated from persicae Semen have been evaluated on lipopolysaccharide (LPS)-induced release of nitric oxide (NO), prostaglandin $E_2\;(PGE_2)$ and tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$) by the macrophage RAW 264.7 cells. Amygdalin significantly inhibited generation of NO and $TNF-{\alpha}$ on LPS-stimulated RAW264.7 cells in a concentration-dependent manner. Consistent with these observations, the expression of inducible NO synthase (iNOS) enzyme was also inhibited by amygdalin in a concentration-dependent manner. However, amygdalin did not show any influence on the synthesis of $PGE_2$ and the expression of COX-2. Thus, this study suggests that amygdalin-mediated inhibition of iNOS expression, and $TNF-{\alpha}$ release may be one of the mechanisms responsible for the anti-inflammatory effects of Persicae Semen.

캐롭 추출물을 활용한 경옥고의 항산화, 항염증 효과 증진 (Improvement of Antioxidant and Anti-inflammatory Effects of Gyeongokgo Using Carop Extract)

  • Objectives: Gyeongokgo is a traditional medicine typically used for individuals with wasting disorders and decreased immunity. Carop is well-known for its exceptional antioxidant effects. The objective of this study was to determine whether the antioxidant and anti-inflammatory properties of Gyeongokgo may be enhanced by the addition of carob. Methods: Cytotoxicity was confirmed using an MTT assay. Free radical scavenging activity ability was evaluated using DPPH assay. The amount of produced nitrogen oxide, TNF-α, and IL-6 were measured using an ELISA reader. Results: The Gyeongokgo with carob showed no cytotoxicity and a synergistic effect on antioxidant activity, when compared with the Gyeongokgo without carob. In addition, the inhibitory effect on nitric oxide production was higher for the Gyeongokgo with carob when compared with the Gyeongokgo without carob. The anti-inflammatory response of the Gyeongokgo with carob was possibly through the suppression of the production of TNF-α and IL-6. Conclusion: The addition of carob to Gyeongokgo may be an effective measure to enhance the antioxidant and anti-inflammatory effects.

수종의 생약제제가 human fetal osteoblasts의 염기성 인산분해 효소 활성에 미치는 영향 (Effects of Several Herbal Medicines on Alkaline Phosphatase Activity in Human Fetal Osteoblasts)

  • Several growth factors and polypeptides are not commonly yet used for regenerators of bone tissue or alveolar bone because of the insufficiency of studies on their side effects, genetic engineering for mass production and stability for clinical application. Recently, many herbal medicines, which have advantage of less side effects and possibility of long-term use, have been studied for their capacity and effects of anti-bacterial, antiinflammatory and regenerative potential of periodontal tissues. Morindae Radix, Cibotium Barometz (L.), Albizziae Cortex, Cistandhis Herba have been traditionally used as medicines for treatment of bone disease in Eastern medicine. The objective of the present study is to examine the ability of alkaline phosphatase (ALP) activity of human fetal osteoblast (hFOB1) when several natural medicines were supplemented. hFOB1 were cultured with Dulbecuo's Modified Eagle's Medium Nutrient Mixture F-12 HAM ( DMEM/F-12 1:1 Mixture, Sigma, USA) and negative control, dexamethasone (positive control), and each natural medicines for 3 days. And then ALP activity was measured by spectrophotometer for enzyme activity and Alizarin red S staining for morphometry. Among the natural medicines of this study, Morindae Radix, Cibotium Barometz (L.) and Cistanchis Herba induced higher activity of ALP synthesis than negative controls in all experimental group. Albizziae Cortex showed mild increases than negative control group. According to measurement of positively stained area, all of the natural medicines of this study increased compared to negative control. Especially, Cibotium Barometz (L.) and Cistanchis Herba showed statistical significance compared to negative control (p<0.05). These results indicate that Morindae Radix, Cibotium Barometz (L.), Albizziae Cortex, Cistandhis Herba have an inducing ability of ALP synthesis on osteoblast.

RAW 264.7 세포에서 Carpinus pubescens Burkill 추출물의 항산화 및 항염증 활성 (Anti-Oxidative and Anti-Inflammatory Activities of Carpinus pubescens Burkill Extract in RAW 264.7 Cells)

  • 본 연구에서는 C. pubescens Burkill 에탄올 추출물(CPEE)의 항산화능과 항염증 생리활성을 DPPH 라디칼 소거능, ROS 생성 억제능, NO 소거 활성 등을 통해 분석하였다. 먼저 CPEE의 항산화능을 DPPH 라디칼 소거능으로 분석한 결과 강한 소거능을 보였으며, 좀 더 자세한 항산화능 작용기작을 알아보기 위해 ROS 생성 억제능으로 분석한 결과 농도 의존적으로 강한 ROS 소거능을 보였다. 뿐만 아니라 대표적인 항산화 효소인 HO-1 및 그 전사 인자인 Nrf2의 단백질 발현에 미치는 영향을 분석한 결과 CPEE에 의해 HO-1 및 Nrf2의 발현이 증가됨을 보였다. 한편 CPEE가 LPS에 의해 유도된 NO 생성에 미치는 영향을 분석한 결과 농도의 존적인 NO 생성 저해능을 보였으며 이는 NO 생성 단백질인 iNOS의 발현 저해에서 기인함을 확인하였다. 이러한 결과를 통해 CPEE의 높은 항산화능과 항염증 활성을 확인하였으며 향후 잠재적인 기능성 소재로서 유용하게 활용될 수 있을 것으로 판단된다.

위염 및 위궤양에 미치는 Propolis 추출물의 영향 (Effects of Propolis Extract on Antigastritic and Antiulcer)

  • 벌집으로부터 채취한 수지상의 물질인 propolis는 다양한 생화학적 성분을 함유하며 기원전 300년부터 사용되어 온 전통약물이다. 최근 항균, 항 바이러스, 항진균, 국소마취, 면역 활성, 항염, 항산화 작용등의 생물학적 활성이 보고된 바 있다. 본 연구에서는 위염과 위궤양에 미치는 propolis추출물과 분획물의 약조학적 효과를 평가하고자 에탄올 추출물을 hexane, toluene, ethyl acetate로 분획하여 항위염 및 항위궤양 활성을 조사하였다. Propolis 에탄을 추출물은 염산·에탄을 위손상, Shay의 위액 분비 실험에서 용량 의존적인 보호효과를 나타내었고 hexnae과 toluene 분획물은 염산·에탄을 위손상, aspirin에 의한 위손상, Shay의 위액 분비 실험에서 위손상과 위액분비를 감소시켰다. propolis 추출물과 분획물의 위염 및 위궤양에 대한 보호효과는 H+/K+ATPase activity의 억제에 의한 위액분비의 감소에 의한 것으로 생각된다.

황금(黃芩)과 구성 화합물의 약리작용에 대한 고찰 (Review of Pharmacological Effects of Scutellaria Baicalensis and Its Bioactive Compounds)

  • Objective : Scutellaria baicalensis is one of the most popular and multi-purpose herb in traditional medicine. It is also useful for its practicability to cultivate in Korea. The purpose of this study is to contribute to researches and applications of scutellaria baicalensis by analyzing and reviewing international researches on the compositions and the effects of scutellaria baicalensis. Methods : This study analyzed 146 articles from PubMed by searching with the keyword "Scutellaria baicalensis", "Huang quin", "Baical Skullcap", "Huang qin", "baical skullcap root", "ogon", "Hwanggeum" and "Hwangkeum", published within the last 10 years(from 2000 to 2009). We reviewed the 146 articles on Scutellaria baicalensis and its active constituents in terms of 'Active constituents', 'Experimental studies', 'Clinical studies', 'Drug interaction', 'Side Effects/Toxicity' and 'Pharmacokinetics'. Results : The active constituents of Scutellaria baicalensis are flavonoids such as baicalein, baicalin, wogonin and oroxylin-A. It is reported that scutellaria baicalensis and its active compounds have antiinflammatory activity, antitumor activity, antioxidant activity, antiviral and antibiotic activity, neuroprotective effects, hepatoprotective effects and cardiovascular effect. Conclusions : This study is aimed to summarize the results obtained within the last 10 years and to contribute to following researches and applications of Scutellaria baicalensis.

Anti-tumor Constituents from Isodon xerophilus

  • Results of chemistry and biological activity of many years indicate that plants belonging to the Isodon genus are rich in ent-kaurane diterpenoids, which have been revealed to possess biological activities such as antitumor, antibacterial and antiinflammatory effects. In continuation of our research on diterpenoids in medicinal plants of this genus, the acetone extract from the leaves of I. xerophilus, which is a plant native to Yunnan province of China, showed potent antitumor activity against K562. After partition, the most active EtOAc part was studied. Four new diterpenoids named xerophilusin A(l), B(2), C(3), D(4), and eight known compounds including macrocalin B(5) and rabdorosthomin A(6) were isolated, whose structures were elucidated through a series of one- and two-dimensional NMR techniques(DEPT, COSY, HMQC, HMBC and ROESY experiments). Among them, compound 1, 2 and 5 had two unique epoxy units formed by two ether bridges from C-20 to C-7, C-14. Up to now, there are four compounds having such an peculiar structure besides these three compounds. Compound 3 and 4 were two of the few examples possessing $1{\beta}$ substitutes. All the diterpenoid compounds were subjected to the antitumor screening. It is interesting that only xerophilusin A(l), B(2) and macrocalin(5) exhibited significant antitumor activity against K562 by the method of MTT($IC_{50}$ were listed in Table 1.). The results inspired us to infer that the unique ether bridges from C-20 to C-7, C-14 possibly played an important role in the antitumor activity.

Anti-inflammatory Action of Phenolic Compounds from Gastrodia elata Root

  • Previous screening of the pharmacological action of Gastrodia elata (GE) root (Orchidaceae) showed that methanol (MeOH) extracts have significant anti-inflammatory properties. The antiinflammatory agents of GE, however, remain unclear. In this experiment, MeOH extracts of GE were fractionated with organic solvents for the anti-inflammatory activity-guided separation of GE. Eight phenolic compounds from the ether (EtOEt) and ethyl acetate (EtOAc) fractions were isolated by column chromatography: 4-hydroxybenzaldehyde (I), 4-hydroxybenzyl alcohol (II), benzyl alcohol (III), bis-(4-hydroxyphenyl) methane (IV), 4(4'-hydroxybenzyloxy)benzyl-methylether (V), 4-hydroxy-3-methoxybenzyl alcohol (VI), 4-hydroxy-3-methoxybenzaldehyde (VII), and 4-hydroxy-3-methoxybenzoic acid (VIII). To investigate the anti-inflammatory and anti-oxidant activity of these compounds, their effects on carrageenan-induced paw edema, arachidonic acid (AA)-induced ear edema and analgesic activity in acetic acid (HAc)-induced writhing response were carried out in vivo; cyclooxygenase (COX) activity, reactive oxygen species (ROS) generation in rat basophilic leukemia (RBL 2H3) cells and 1,1-diphenyl-2-picryl-hydroazyl (DPPH) scavenging activity were determined in vitro. These phenolic compounds not only had anti-inflammatory and analgesic properties in vivo, but also inhibited COX activity and silica-induced ROS generation in a dose-dependent manner. Among these phenolic compounds, compound VII was the most potent anti-inflammatory and analgesic. Compound VII significantly inhibited silica-induced ROS generation and compound VI significantly increased DPPH radical scavenging activity. Compounds I, II and III significantly inhibited the activity of COX-I and II. These results indicate that phenolic compounds of GE are anti-inflammatory, which may be related to inhibition of COX activity and to anti-oxidant activity. Consideration of the structure-activity relationship of the phenolic derivatives from GE on the anti-inflammatory action revealed that both C-4 hydroxy and C-3 methoxy radicals of benzyl aldehyde play an important role in anti-inflammatory activities.

모래지엽(毛棶枝葉)(Cornus walteri Wanger) 추출물이 비만 쥐의 지질강하, 항산화효과 및 전염증성 cytokines 농도에 미치는 영향 (Effects of Stem Bark Extracts of Cornus walteri Wanger on the Lipid Lowering, Anti-oxidative Activity and Concentration of Proinflammatory Cytokines in Rat Fed High Fat Diet)

  • Objective : This study was designed to examine the effects of stem bark extracts of Cornus walteri Wanger on the lipid lowering, anti-oxidative activity and concentration of proinflammatory cytokines in hyperlipidemic rat. Methods : Male rats weighing $195.21{\pm}5.85g$ fed high fat diet for 8 weeks and 40 rats(above 400 g) were divided into 4 groups. Each groups were divided into a control group and 3 experimental groups. We fed a control group of rats a basal diet and administered normal saline(100 mg/kg, 1 time/1 day) for 4 weeks. And we fed each experimental group of rats Basal diet and administered an extract of Cornus walteri Wanger(100 mg/kg, 200 mg/kg, 300 mg/kg, 1 time/1 day) for 4 weeks. At the end of the experiment, the rats were sacrificed to determine their chemical composition. We measured lipid of plasma and liver, concentration of proinflammatory cytokines, anti-oxidative activity and gene expression. Results : 1. Concentration of plasma free fatty acid, LDL-cholesterol showed a tendency to decrease in Cornus walteri Wanger ext. groups. Concentration of plasma triglyceride, total cholesterol showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. HDL-cholesterol showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group. 2. Concentration of liver total cholesterol showed a tendence to decrease in Cornus walteri Wanger ext. groups. Concentration of triglyceride liver showed a significantly decrement in all Cornus walteri Wanger ext. group than that of control group. 3. Concentration of plasma and liver TBARS showed a tendence to decrease in Cornus walteri Wanger ext. groups. The values of GSH-Px, SOD and CAT activity showed a significantly increment in the 300 mg/kg Cornus walteri Wanger ext. group than that of control group. 4. The values of plasma AST and ALT activity showed no significantly different in all treatment groups. 5. Concentration of plasma $IL-1{\beta}$, IL-6 and $TNF-{\alpha}$ showed a tendency to decrease in the Cornus walteri Wanger ext. groups. However the concentration of IL-10 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly increment than that of control group. Concentration of liver $IL-1{\beta}$, $TNF-{\alpha}$ and IL-10 showed no significantly difference in all treatment groups. However concentration of IL-6 in the 200 and 300 mg/kg Cornus walteri Wanger ext. groups showed a significantly decrement than that of control group. 6. In the analysis of RT-PCR, gene expression of $TNF-{\alpha}$, Apo-B, Apo-E and leptin in the Cornus walteri Wanger ext. groups showed a lower expression than that of control group. 7. The ratio of $TNF-{\alpha}$, Apo-E and leptin expression per $\beta$-actin expression in the 200 and 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. The ratio of Apo-B expression per $\beta$-actin expression in the 300 mg/kg Cornus walteri Wanger ext. showed a significantly decrement than that of control group. Conclusions : According to above results, in lowering lipid effect, antioxidative activity and antiinflammatory effect, the Cornus walteri Wanger ext. gives positive effect.

Suppressive Effects of an Ishige okamurae extract on 3T3-L1 Preadipocyte Differentiation

  • The biological activity of tissue specific stem cell is under the control of their specific microenvironment and the exogenous chemicals derived from digestive tract can be one of the constructing factors of that. It is suggested that the extract of brown algae Ishige okamurae has antioxidant-, apoptosis induction-, and antiinflammatory-effects. On the other hand, a few studies have shown that antioxidant assist inhibition of accumulation of fat. So we studied the effect of the extract of I. okamura on the cellular activity and differentiation of 3T3-L1 preadipocyte to adipose cell. The viability of cell was analyzed using 3-[4,5-dimethylthiazo-2-yl]-2,5-diphenyl tetrazolium bromide (MTT) assay. Adipogenesis of 3T3-L1 cell was analyzed after induction in the induction medium containing the I. okamurae extract. The cellular activity was high compared with the vehicle and 0.05 mM caffeine in all groups of I. okamurae extract treated cells. The extract of I. okamura inhibited accumulation of lipids in 10 and $50{\mu}g/ml$. The expression of the marker genes for adipocyte differentiation coincided with cytochemical results. These results suggest that the extract of I. okamurae increases the cellular viability of adipose precursor cells. On the other hand, it suppresses the differentiation of preadipocyte to adipocyte and accumulation of lipids in concentration-dependent manners. It may be possible that the major component of the extract can be applied in the control of adipose tissuegenesis.
