Aqueous extract of Morus alba L. blocked the toxic effect of paraquat on E. coli growth. The active components in the extract may be capable of crossing the cell membranes and protect against superoxide toxicity in E. coli, The extract inhibited $FeSO_4/H_2O_2$ induced lipid peroxidation in rat liver homogenate and protected against t-butyl hydroperoxide caused Ac2F cell damage. Moreover, the extract showed inhibitory effect on phospholipase $A_2$ activity in a dose dependent manner. Antiinflammatory effect of the extract was further investigated using the carrageenin-induced oedema model. A single adminstration of the extract (3g/kg body, p.o.) was more effective than indomethacin. These results suggest that the isolation and identification of the active components would have significant therapeutic application to inflammation associated with oxygen radicals.
Inflammation is an innate immune response that protects the body from pathogens, toxins, and other dangers and is initiated by recognizing pathogen-associated molecular patterns or danger-associated molecular patterns by pattern-recognition receptors expressing on or in immune cells. Intracellular pattern-recognition receptors, including nucleotide-binding oligomerization domain-like receptors (NLRs), absent in melanoma 2, and cysteine aspartate-specific protease (caspase)-4/5/11 recognize various pathogen-associated molecular patterns and danger-associated molecular patterns and assemble protein complexes called "inflammasomes." These complexes induce inflammatory responses by activating a downstream effector, caspase-1, leading to gasdermin D-mediated pyroptosis and the secretion of proinflammatory cytokines, such as interleukin $(IL)-1{\beta}$ and IL-18. Ginsenosides are natural steroid glycosides and triterpene saponins found exclusively in the plant genus Panax. Various ginsenosides have been identified, and their abilities to regulate inflammatory responses have been evaluated. These studies have suggested a link between ginsenosides and inflammasome activation in inflammatory responses. Some types of ginsenosides, including Rh1, Rg3, Rb1, compound K, chikusetsu saponin IVa, Rg5, and Rg1, have been clearly demonstrated to inhibit inflammatory responses by suppressing the activation of various inflammasomes, including the NLRP3, NLRP1, and absent in melanoma 2 inflammasomes. Ginsenosides have also been shown to inhibit caspase-1 and to decrease the expression of $IL-1{\beta}$ and IL-18. Given this body of evidence, the functional relationship between ginsenosides and inflammasome activation provides new insight into the understanding of the molecular mechanisms of ginsenoside-mediated antiinflammatory actions. This relationship also has applications regarding the development of antiinflammatory remedies by ginsenoside-mediated targeting of inflammasomes, which could be used to prevent and treat inflammatory diseases.
As a part of ongoing research to elucidate and characterize antiinflammatory nutraceuticals, the crude extracts from Atriplex gmelinii C. A. Mey. and their solvent-partitioned fractions were tested for their antiinflammatory potential in lipopolysaccharide (LPS)-stimulated RAW 264.7 mouse macrophages. The crude extracts of A. gmelinii C. A. Mey. were fractioned according to polarity with n-hexane, 85% aqueous methanol (85% aq. MeOH), n-butanol, and $H_2O$. Their antiinflammatory activities were investigated in LPS-induced inflammation in mouse macrophages by measuring nitric oxide (NO) generation and mRNA expression of inflammation mediators, namely, inducible nitric oxide synthase (iNOS), cyclooxygenase-2 (COX-2), interleukin-$1{\beta}$ ($IL-1{\beta}$), and IL-6. As a result, we confirmed that the crude extracts of A. gmelinii C. A. Mey. inhibited LPS-stimulated NO production and mRNA expression of iNOS and COX-2 as important inflammatory factors. The inhibition of NO production through the downregulation of important inflammatory factors such as iNOS, COX-2, $IL-1{\beta}$, and IL-6 was found by treatment with all solvent-partitioned fractions. Among all tested fractions, 85% aq. MeOH showed the strongest antiinflammatory response. Based on the current results, A. gmelinii C. A. Mey. was suggested to possess natural antiinflammatory components, indicating that it could be used as a valuable source of antiinflammatory substances.
Experimental studies were made with Melilotus officinalis extract which was extracted from flowers and leaves of Melilotus officinalis Dsr. (Leguminosae). In this paper, acute toxicity, analgesic action, prolongation of hypnosis time by induced pentobarbital-Nain mice, antiinflammatory effect in rats and effects on isolated intestines of mice and rats were studied, The result was as follows; 1. Very low toxicity in mice. 2. Analgesic action was recognized markedly in mice. 3. Prolongation of hypnosis time induced by pentobarbital-Na in mice was shown. 4. Relaxing action was shown on the isolated ileum in mice and antagonistic action was seen on $BaCl_2-induced$ contraction of the ileum that the relaxing effect of the intestinal smooth muscle was recognized. 5.Antiinflammatory effect was shown markedly in mice. 6.Hypotensive and vaso-dilating actions due to the vascular smooth muscle relaxation were noted in rabbits.
The biological activities of fruits of Poncirus trifoliata have been studied. 70% Ethanol extract of Ponciri Fructus was fractionated with ether, ethyl acetate and n-butanol by turns. The ether fraction showed antibacterial activity on Staphylococcus aureus and inhibited the contractability of the isolated mice ileum. The ethyl acetate and n-butanol fractions showed the anti-edematous effect on the carrageenin-induced edema of rat hindpaw. From the n-butanol fraction, one of the flavonoid glycoside was isolated and identified as poncirin. Ponciri Fructus contained about 6% poncirin which was 5,7-dihydroxy-4´-methoxy flavanone rhamnoglucoside and poncirin showed antiinflammatory activities. Poncirin was considered as the major effective component of Ponciri Fructus.
A new natural product, 3,4,8-trihydroxybenzopyranopyran-6,9-dione was isolated from the aqueous ethanolic gall extract of Tamarix aphylla (Tamaricaceae) along with the known phenolics, monodecarboxyellagic acid and brevifolin carboxylic acid as well. The structures have been established by ESI-MS, $^1H$ and $^{13}C$ NMR spectral analysis. Antiinflammatory, antipyretic and ulcerogenic activities determination for both plant (Tamarix aphylla and Phragmites australis) were carried out on aq. ethanolic of extracts.
Recently it has been demonstrated that selective cyclooxygenase-2 (COX-2) inhibitors retain the antiinflammatory effect but with markedly reduced GI toxicity compared to non selective inhibitors such as traditional NSAIDs. As a consequence, intense efforts have been made to develop selective COX-2 inhibtors during the last decade. Two compounds in this class. celecoxib and rofecoxib. are already in the market and are proved as potent and selective COX-2 inhibitors with much better gastric tolerance. However. there are still strong domands for a COX-2 inhibitor with improved efficacy and safety profiles. Here we report the synthesis and biological profiles of 1.5- and 4.5-disubstituted imidazole analogues as structural equivalents of cefecoxib and refecoxib. The imidazole analogues are overlapped well whth the 3D srructures of celecoxib and rofecoxib.

3-Substituted 5-aminoisoxazole-4-carboxylates were prepared by the reaction of corresponding bormoaldoximes with cyanoacetate. The 3-trifluoromethylisoxazole derivatives were acylated to amides with various aminopyridine derivatives to afford diamides. The ester group was hydrolyzed and decarboxylated easily to give 3-trifluoromethyl-5-aminoisoxazole. The aminoisoxazole was also converted to amides. 5-Amino-3-trifluoromethylisoxazole-5-one-4-carboxylate was prepared by the reaction of trifluoroacetoaldoximoyl bromide and malonate. 5-Amino-3-methylisoxazole-5-one-4-acetate was prepared by the reaction of hydroxylamine and acetylmalonate. The synthesized compounds were tested for antiinflammatory activities.
The alcoholic extract of Momordica dioica roots significantly reduced $CCl_4$ induced hepatotoxicity in rats upon oral administration (200 mg/kg), as judged from the serum enzyme levels [serum glutamate oxaloacetate transaminases (SGOT), serum glutamate pyruvate transminases (SGPT), and alkaline phosphatase (ALP)]. These results were comparable with sylimarin (1.25 mg/kg, p.o.). The alcoholic extract inhibited the formation of oxyten derived free radicals (ODFR) in vitro with $4000\;{\mu}g/ml$ ascorbic system. The alcoholic extract also significantly reduced carrageenan-induced paw edema when administered orally (200 mg/kg) and the activity was comparable with ibuprofen(200 mg/kg, p.o.).
