곽향의 성분 분석 및 주요 성분들의 in silico 항당뇨 타겟 단백질 탐색 (Analysis of Chemical Constituents of Agastachis Herba and in silico Investigation on Antidiabetic Target Proteins of its Major Compounds)

  • 곽향은 식욕부진, 메스꺼움 등의 치료에 사용될 뿐만 아니라 최근에는 항당뇨 효능도 알려졌다. 본 연구에서는 곽향의 항산화력과 주요 성분들을 조사한 다음, in-silico 방법론을 적용하여 타겟 단백질들을 예측하였다. 먼저 메탄올 추출물의 DPPH와 ABTS 라디칼 소거능의 EC50 값은 각각 78.6 ㎍/mL과 31.0 ㎍/mL이었다. 이것은 ascorbic acid의 값(9.9 ㎍/mL, 5.2 ㎍/mL)과 비교할 때 항산화력이 뛰어나다고 할 수 있다. HPLC-PDA-MS/MS를 이용하여 성분들을 정성 분석한 결과, 추출물의 주요 화합물로 rosmarinic acid, tilianin, agastachoside 그리고 acacetin을 확인하였다. 이들 성분들의 항당뇨 작용 타겟 단백질을 DIA-DB 서버를 사용하여 구조 유사도와 inverse doking 방법론을 적용하여 예측하였다. 본 가상 탐색 결과, 주요 타겟 단백질들은 PPAR-γ, DPP IV, glucokinase, α-glucosidase, SGLT2, aldose reductase, corticosteroid 11-beta-dehydrogenase로 예측되었다. 그리고 이들 단백질들 중 일부는 이미 실험적으로 검증된 타겟 단백질이었다. 따라서 in silico 검색 방법이 유효하다고 생각할 수 있다. 마지막으로 활성성분들의 최적의 추출 조건을 결정하기 위하여 여러 추출 용매들로 곽향을 추출하였다. 여러 유기 용매들 중에는 메탄올이 그리고 에탄올-물 혼합용매에서는 80% 에탄올이 가장 효과적인 추출 용매로 확인되었다.

발효기간에 따른 돌산갓김치의 glucosinolates 함량변화와 항산화, 항고혈압 및 항당뇨활성과의 상관관계 (Changes in Glucosinolate Content of Dolsan Leaf Mustard Kimchi during Fermentation and Correlation with Antioxidant, Antihypertensive, and Antidiabetic Activities)

  • 돌산갓김치의 조추출물을 이용하여 glucosinolates함량, 항산화활성, 항고혈압 및 항당뇨활성을 측정하였다. Glucosinolates 함량은 DLMK의 잎과 줄기에서 6.41, 7.92 mg/g으로 발효기간 중 낮은 함량을 나타냈다. Total polyphenol과 total flavonoid함량은 줄기보다 잎에서 발효기간 동안 2배 이상의 함량을 나타냈다. ABTS라디컬 소거활성과 EDA은 발효 14일째 대조군과 유사한 활성을 나타냈고, FRAP는 발효 14일째 잎에서 대조군보다 높은 함량을 나타냈다. 그리고 항고혈압 활성(ACE 저해활성)은 잎에서 대조군인 0.01% captopril과 비슷하거나 높은 저해활성을 나타냈으며, 항당뇨활성(${\alpha}$-glucosidase 저해활성)은 대조군인 0.05% acarbose보다 잎, 줄기에서 높은 저해활성을 나타냈다. 또한 glucosinolates 함량과 ABTS 및 EDA, ACE 저해활성 및 ${\alpha}$-glucosidase 저해활성의 상관관계는 줄기 추출물 보다 잎 추출물이 높은 양의 상관관계를 나타냈다. 발효기간 중 glucosinolates는 sinigrin, glucobrassicin, glucotropaeolin, progoitrin 검출되었다. 이 결과를 통해 돌산 갓김치 추출물은 항산화, 항고혈압 및 항당뇨활성에 효과가 높은 것으로 나타냈기에 기능성 식품으로서 가치가 높을 것으로 기대된다.

  • 본 연구에서는 스트렙토조토신(Streptozotocin,STZ)으로 유발된 당뇨쥐에게 맥문동(Liriope platyphylla,Lp)에탄올 추출물(수율:30.7%)을 1,000mg/kg용량으로 7일간 경구 투여 후 혈청속의 혈당함량과 당대사와 관련된 몇 몇 효소와 지질대사와 관련된 물질을 측정한 결과 혈당과 triglyceride(TG), total cholesterol의 농도는 STZ대조군과 비교하여 맥문동 투여군에서 유의적인 감소를 나타내었고, glucose-6-pase(g-6-pase)활성은 STZ 대조군과 비교하여 유의적인 감소를, glucokinase(gk)활성은 증가를 나타내었다. 또한 hepatic glycogen 함량은 STZ대조군과 비교하여 유의적인 증가를 나타내었다. 따라서 본 실험 결과 맥문동 에탄올 추출물을 스트렙토조토신으로 유발된 당뇨쥐에 있어서 항 당뇨 성분을 함유하고 있음을 알 수 있었다.

Effects of Fermented Red Ginseng Extracts on Hyperglycemia in Streptozotocin-induced Diabetic Rats

  • Fermented red ginseng (FRG) was prepared by inoculating 0.1% Lactobacillus fermentum NUC-C1 and fermenting them at $40^{\circ}C$ for 12 hours. The ginsenoside contents of FRG were increased compared with those of red ginseng (RG). Moreover, the levels of the ginsenosides Rg2, Rg3, and Rh2 in FRG increased significantly. In an oral glucose tolerance test (OGTT), blood glucose levels were lower in animals fed with RG and FRG extracts than in normal controls. In particular, FRG extracts in OGTT were superior to RG extracts. The antidiabetic effects of FRG in streptozotocin (STZ)-induced diabetic rats were investigated. Rats were divided into four groups: normal control, diabetes mellitus (DM), FRG administered at 100 mg/kg, and FRG administered at 200 mg/kg groups. FRG extracts were orally administered to each treatment group for 3 weeks, and blood glucose, insulin, and lipid levels of each group were determined. Orally administered FRG extracts significantly reduced blood glucose levels and increased plasma insulin levels in diabetic rats. Additionally, the activities of disaccharidases, including sucrase, lactase, and maltase, were decreased significantly in the FRG groups. FRG groups also had reduced triglyceride and total cholesterol levels, compared with the DM group. These results suggest that FRG may have antidiabetic effects in STZ-induced diabetic rats.

서귀포 백년초의 기능성 원료 제조 특성 (Characteristics of Opuntia monacantha Haw. for the Functional Raw Material Production)

  • 백년초(Opuntia) 손바닥 선인장은 우리나라 남쪽 지방인 거제도, 제주도, 남해안 등지에서 자생하며 매년 4~5월경에 꽃이 피고 11~12월경에 자주색 열매가 익는다. 백년초는 예로부터 민간요법에 흔히 사용되었으며 혈액순환을 원활하게 하고 해독, 진통, 항산화 작용 등에 큰 효과가 있다. 본 연구에서는 서귀포 백년초의 항산화성 기능, 항당뇨, 항균활성 효능을 입증하여 기능성 원료로서의 가치를 파악하고자 하였다. 연구 결과 백년초 내 항산화성을 갖는 물질인 택시폴린을 추출하여 강력한 항산화 효과를 확인하였고 항당뇨성 기능 실험에서 시판되고 있는 당뇨치료제와 비교하였을 때 높은 수준의 항당뇨 기능을 확인하였다. 또한 항균 효과 측정 결과 대장균과 살모넬라균 등에서 강한 항균활성을 보여 서귀포 백년초의 기능성 원료로서의 우수한 가치를 입증하였다.

구기자 분획물이 Streptozotocin으로 유발 된 당뇨 흰쥐에 대한 항당뇨 및 항산화작용에 미치는 효과 (Antidiabetic and Antioxidative Effect of Lycii fructus in Streptozotocin-Induced Diabetic Rats)

  • This study was carried out to investigate the antidiabetic and antioxidative effect of Lycii fructus in the Streptozotocin(STZ)-induced diabetic rats. The effective fractions were prepared as a form of organic solvents of $CH_{3}(CH_{2})_{4}CH_{3}$ $CHCI_{3}$, EtOAc, BuOH and $H_{2}O$ fractions prepared from the EtOH extract of Lycii fructus and The diabetes were induced by an tail-intravenous injection of STZ with a dose of 45 mg/kg dissolved in citrate buffer. The various fractions of Lycii fructus were orally administrated once a day for 7 days. The contents of serum glucose, and triglyceride in the $CHCI_{3}$ fraction and hepatic lipid peroxidation in the EtOAc, BuOH and $H_{2}O$ fractions treated rats were significantly decreased when compared to those of the STZ-control group In addition, an activity of hepatic GST in the BuOH fraction treated rats was significantly increased compared to that of the STZ-control group. whereas, activities of hepatic catalase, GSH-Px in the BuOH fraction treated rats were significantly decreased compared to those of the STZ-control group. Meanwhile, The content of hepatic glycogen and avtivity of hepatic glucokinase in $CHCI_{3}$ fraction treated rats were significantly increased, but activity of glucose-6-pase was significantly decreased in the $CHCI_{3}$ fraction treated rats. In conclusion, these results indicated that the BuOH fraction of Lycii fructus was effective for the antioxidation, and also the $CHCI_{3}$ fraction of Lycii fructus was effective for the antidiabetes in the STZ-induced diabetic rats.

  • Kinsenoside is a principle bioactive compound of Anoectochilus formosanus. It exhibits various pharmacological effects such as antihyperglycemic, antioxidant, anti-inflammatory, immunostimulating, and hepatoprotective activities and has recently been developed as an antidiabetic drug candidate. In this study, as part of an in vitro pharmacokinetic study, the stability of kinsenoside in rat and human liver microsomes was evaluated. Kinsenoside was found to have good metabolic stability in both rat and human liver microsomes. These results will provide useful information for further in vivo pharmacokinetic and metabolism studies.

Antioxidant and Antidiabetic Activities of Eucommia ulmoides Bark

  • Eucommia ulmoides bark extracts by cold water, boiling water, 100% EtOH, 70% EtOH, 100% MeOH, 70% MeOH and $CHCl_3$ were assayed for their medicinal effects. The antioxidant activity of the extracts ranged from $IC_{50}$ 125.2 to $IC_{50}\;872.7{\mu}g/ml$ in the 1,1-diphenyl-2-picrylhydrazyl (DDPH) free radical-scavenging assay, and cold water extracts had the highest antioxidant activity. $CHCl_3$ extracts had the highest inhibitory effect on angiotensin I-converting enzyme (ACE) giving inhibition of up to 56.4% at a concentration of 1 mg/ml. Extracts in 100% EtOH had the greatest inhibitory effect on $\acute{a}-amylase$ activity ($IC_{50}=174.6{\mu}g/ml$), and 70% MeOH extracts had the greatest inhibitory effect on ${\alpha}-glucosidase$ activity ($IC_{50}=14.0{\mu}g/ml$). Taken together, these results provided the in vitro evidence on the ACE, amylase and glucosidase inhibitory actions of E. ulmoides bark that form the pharmacological basis for its antihypertensive and antidiabetic action.

db/db 마우스에서 상엽 에탄올가용분획의 항당뇨활성 (Antidiabetic Activity of Mori Folium Ethanol Soluble Fraction in db/db mice)

  • Antidiabetic activity of Mori folium ethanol soluble fraction (MFESF) was examined in db/db mice, which is a spontaneously hyperglycemic, hyperinsulinemic and obese animal model . 500 and 1000mg/kg dose for MFFSF (designated by SY 500 and SY 1000, respectively) and 5mg/kg dose for acarbose were administered for 6 weeks. Body weight gain, fasting and non-fasting serum glucose, glycated hemoglobin and triglyceride were all reduced dose dependently when compared between db/db control group and MFESF treated group. At 11th and 13th week after birth, MFESF increased an insulin secretion which may result in lowering serum glucose level. Total activities of sucrase and maltase in SY 500 treated group were decreased when compared to db/db control. On the other hand, those in SY 1000 and acarbose treated groups were increased. This result may suggest that proteins for sucrase and maltase were compensatorily induced due to significant inhibition of glycosidase-catalyzed reaction at doses administered in this study.
