황기 에탄올 추출물의 항 당뇨 효과 (Antidiabetic Effect of Ethanol Extract on Astragali Radix)

  • Streptozotocin(STZ)을 45mg/kg.b.w의 용량으로 흰쥐의 미정맥에 투여 한 후 당뇨병이 유발된 당뇨 흰쥐에게 1일 1회 7일간 1,000mg/kg의 용량으로 황기에탄올 추출물을 투여 후 glucose함량과 당대사에 관여하는 효소인 glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dehydrogenase(G-6-PDH, glucokinase(GK)활성과 glycogen 함량, triglyceride(TG), total cholesterol등의 지질대사에 관여하는 물질들을 측정한 결과 황기 에탄올 추출물 투여군이 glucose, TG, total cholesterol등의 함량과 G-6-Pase 활성의 유의적인 감소를 나타내었으며 glycogen 함량과 G-6-PDH, GK의 활성이 유의적인 증가를 나타내었다. 이와 같이 황기 에탄올 추출물이 항당뇨 개선효과를 갖는 유효성분을 함유하고 있음을 알 수 있었다.

Antidiabetic Activity of an Ayurvedic Formulation Chaturmukha Rasa: A Mechanism Based Study

  • Objectives: The objective of this study was to evaluate antidiabetic activity of Chaturmukha rasa based on streptozotocin induced diabetes model, alpha amylase inhibitory activity, alpha Glucosidase inhibitory activity and inhibition of sucrase. Methods: Chaturmukha rasa was prepared as per Ayurvedic formulary. Antidiabetic activity was measured in experimentally streptozotocin induced rats. The dose was taken as 45 mg/kg, i.p. The antidiabetic activity of Chaturmukha rasa was compared Triphala Kwatha, a marketed formulation. Further In vitro $\acute{\alpha}$- Amylase Inhibitory Assay, In vitro salivary amylase Inhibitory Assay, In vitro ${\alpha}-Glucosidase$ Inhibitory Assay and In vitro Sucrase Inhibitory Assay was performed with respect to Chaturmukha rasa. The IC50 value was calculated for all the above activity. Results: Streptozotocin with Acarbose showed significant decrease in blood glucose level whereas streptozotocin with Triphala kwatha showed more decrease in blood glucose level than Streptozotocin with Acarbose. The combination of Streptozotocin + Triphala kwatha + Chaturmukha rasa showed a significant decrease in blood glucose level on 21st day. In vitro $\acute{\alpha}$- Amylase Inhibitory Assay the Chaturmukha rasa showed IC50 value $495.94{\mu}l$ when compared with Acarbose $427.33{\mu}l$, respectively. In the ${\alpha}-Glucosidase$ Inhibitory Assay Chaturmukha rasa showed IC50 value $70.93{\mu}l$ when compared with Acarbose $102.28{\mu}l$, respectively. In vitro Sucrase Inhibitory Assay Chaturmukha rasa showed IC50 value $415.4{\mu}l$ when compared with Acarbose $371.43{\mu}l$, respectively. Conclusion: This study supports that Chaturmukha rasa may inhibit diabetes by inhibition of salivary amylase or alpha Glucosidase or sucrase. This may be the mechanism by which Chaturmukha rasa inhibits diabetes. Further this study supports the usage of Chaturmukha rasa for the management of diabetes.

Momordica charantia Protects against Cytokine-induced Apoptosis in Pancreatic $\beta$-Cells

  • The unripe fruit of Momordica charantia (MC) has been shown to possess antidiabetic activity. However, the mechanism of its antidiabetic action has not been fully understood. In this study, the effects of the aqueous ethanolic extract of MC (AEE-MC) were evaluated on the apoptosis in pancreatic $\beta$-cells treated with a combination of the cytokines, interleukin (IL)-$1{\beta}$, tumor necrosis factor (TNF)-$\alpha$, and interferon (IFN)-$\gamma$. In MIN6N8 cells, the inhibitory effect of AEE-MC was significantly observed at 2 to 50 ${\mu}g/mL$: a 26.2 to 55.6% decrease of cytoplasmic DNA fragments quantified by an immunoassay. The molecular mechanisms, by which AEE-MC inhibited $\beta$-cell apoptosis, appeared to involve the inhibition on the expression of p21, Bax, and Bad, the up-regulation of Bcl-2 and Bcl-$X_L$, and the inhibition on the cleavage of caspase-9, -7, and -3 and poly (ADP-ribose) polymerase. This study suggests that MC may inhibit cytokine-induced apoptosis in $\beta$-cells and, thus, may contribute via this action to the antidiabetic influence in diabetes.

Sasa borealis extract exerts an antidiabetic effect via activation of the AMP-activated protein kinase

  • Leaf of Sasa borealis, a species of bamboo, has been reported to exhibit anti-hyperglycemic effect. However, its antidiabetic mechanism is not fully understood. In this study, we examined whether an extract of S. borealis activates AMP-activated protein kinase (AMPK) and exerts anti-hyperglycemic effects. Treatment with the S. borealis extract increased insulin signaling and phosphorylation of AMPK and stimulated the expression of its downstream targets, including $PPAR{\alpha}$, ACO, and CPT-1 in C2C12 cells and $PPAR{\alpha}$ in HepG2 cells. However, inhibition of AMPK activation attenuated insulin signaling and prevented the stimulation of AMPK target genes. The S. borealis extract increased glucose uptake in C2C12 cells and suppressed expression of the gluconeogenic gene, PEPCK in HepG2 cells. The extract significantly reduced blood glucose and triglyceride levels in STZ-induced diabetic mice. The extract enhanced AMPK phosphorylation and increased Glut-4 expression in the skeletal muscle of the mice. These findings demonstrated that the S. borealis extract exerts its anti-hyperglycemic effect through activation of AMPK and enhancement of insulin signaling.

인동초(Lonicerae Japonica Thunb) 추출물의 혈당강하 효과 (Antidiabetic Effect of Lonicerae Japonica Thunb Extract)

  • 본 실험은 문헌상으로 혈당강하효과가 알려져 있는 인동초를 에탄올로 추출하여 그 추출물을 streptozotocin(STZ)으로 당뇨를 유발시킨 흰쥐에게 투여 후 혈당강하작용을 확인하였다. 또한, 그 유효 성분의 추적을 위하여 여러 용매(hexane, chloroform, ethylacetate, butanol, water)로 계통분획하여 그 추출물의 항 당뇨효능을 분석한 결과 hexane, water층의 분획물이 당뇨유발 흰쥐에서 glycogen 함량증가와 glucose-6-phosphatase(G-6-Pase), glucose-6-phosphate dedehydrogenase (G-6-PDH)등의 당대사효소 활성에 효과가 있었다.

Streptozotocin으로 유발된 당뇨쥐에서 청국장 분말의 항당뇨 및 항산화 효과 (Antidiabetic and Antioxidant Effects of Chunggugjang Powder in Streptozotocin-Induced Diabetic Rats)

  • The purpose of this study was to investigate the effects of dietary chungguajang powder on blood glucose level and hepatic activities of antioxidant enzymes in normal and streptozotocin (STZ)-induced diabetic rats. Sprague-Dawley male ratt ($200{\sim}220g$) of six groups including normal group fed normal diet (N), diabetic group fed normal diet (C), diabetic groups fed chunggugjang powder diet (DC-1%, DC-5%, DC-10%, DC-20%) were used for the experiments. Diabetes was induced by single intraperitoneal injection of streptozotocin as a dose of 70 mg/kg body weight. After 3 weeks the animals were sacrificed and hepatic activities of antioxidant enzymes, serum level of glucose and organ weight were evaluated. Food and water intakes were higher in diabetic groups than normal group. Body weight gain and food efficiency ratio were lower in diabetic groups. However, they were higher in chunggugjang diet groups (DC) than normal diet group (C). The serum glucose levels were significantly lower (p<0.05) in the diabetic groups fed chunggugjang diet (DC-10%, DC-20%) than diabetic group fed normal diet (C). Hepatic activities of superoxide dismutase, catalase, glutathione peroxidase and glutathione S-transferase were lower in diabetic groups than normal group and they were significantly lower in diabetic groups fed chunggugjang diet (DC-20%) compared to diabetic rats fed normal diet (p<0.05). In conclusion, these results indicated that chunggugjang powder would have antidiabetic and antioxidant effects in STZ-induced diabetic rats.

Effect of Berberis tinctoria leaf (Berberidaceae) extract on antidiabetic, antihyperlipidemic and antioxidant status in streptozotocin induced diabetes in rats

  • The present study was carried out to investigate the antidiabetic and antioxidant effect of methanolic extract of Berberis tinctoria leaves (MEBT), in streptozotocin induced diabetic rats. Oral administration of MEBT extract (150 mg/kg and 300 mg/kg) for a period of 14 days. Blood glucose levels, body weight food and liquid intake were measured on every $5^{th}$ day over a period of 14 days. In diabetic rats, MEBT at the dose of 150 mg/kg and 300 mg/kg body weight resulted in significant reduction in blood glucose levels. The study was further investigated to determine antioxidant and antihyperlipidemic potential of MEBT in streptozotocin (STZ) induced diabetic rats. These results suggest that the MEBT possess antidiabetic activity and is able to ameliorate biochemical damages in STZ induced diabetic rats and the results were found to be in a dose dependent manner.

가시오가피 추출물의 항당뇨 활성 및 GLUT4 유전자 발현에 미치는 영향 (Effects of Antidiabetic and GLUT4 gene Expression of Acanthopanax senticosus Extracts)

  • Antidiabetic effects of an aqueous and solvent extract prepared from the root, stem and fruit parts of Acanthopanax senticosus, were investigated in experimental Streptozotocin (STZ)-induced diabetic rats model. The n-butanol and water extracts of A. senticosus were orally administrated once a day for 6 days. The n-butanol extracts of fruit (FB) showed highest efficiency than other groups (water extracts of stem, root and fruit; butanol extracts of stem, root) on serum glucose values in the STZ-induced diabetic rats. We have studied gene expression of glucose transporter genes in C2C12 skeletal muscle cell line during differentiation treated by the n-butanol and water extracts of A. senticosus, SW, RW, FW, SB, RB and FB. The GLUT4 gene was high expressed by FB treatment. These findings suggest that FB of A. senticosus have GLUT4 gene expression activity for glucose homeostasis and may have beneficial effects on blood glucose lowering in the diabetic patients.

Antidiabetic Stilbene and Anthraquinone Derivatives from Rheum undulatum

  • The antidiabetic-activity-guided fractionation and isolation of the $80\%$ EtOH extracts obtained from cultivated Korean Rhubarb rhizomes (Rheum undulatum, Polygonaceae) led to the isolation and characterization of one stilbene, desoxyrhapontigenin(1) and two anthraquinones, emodin (2) and chrysophanol (3). Their structures were established by chemical and spectroscopic methods. Compounds 1, 2, and 3 inhibited postprandial hyperglycemia by 35.8, 29.5, $42.3\%$, respectively.

항당뇨약 Rosiglitazone의 혈관 수축성에 대한 이중성 조절 (Biphasic Effects of Rosiglitazone on Agonist-induced Regulation of Vascular Contractility)

  • Rosiglitazone ($Avandia^{(R)}$) represents a new class of antidiabetic drugs which are $PPAR{\gamma}$ agonists. The present study was undertaken to determine whether the new antidiabetic rosiglitazone influences on the agonist-induced regulation of vascular smooth muscle contraction as an antihypertensive and, if so, to investigate the related mechanism. Endothelium-denuded arterial rings from male Sprague-Dawley rats were used and isometric contractions were recorded using a computerized data acquisition system. Rosiglitazone decreased Rho-kinase activating agonist (NaF or thromboxane $A_2$ mimetic)-induced contraction but not depolarization- or phorbol ester-induced contraction. Surprisingly, it slightly potentiated the latter contraction possibly opening a voltage-dependent calcium channel by its chemical structure on 50 mM KCI- or $1{\mu}M$ phorbol 12,13-dibutyrate-induced vasoconstriction. In conclusion, this study provides the evidence and possible related mechanism concerning the biphasic effect of an antidiabetic rosiglitazone as a possible antihypertensive on the agonistinduced contraction in rat aortic rings regardless of endothelial function.
