A bispecific antibody (BsAb) is an artificial protein containing two kinds of specific antigen binding sites. BsAb can connect target cells to functional cells or molecules, and thus stimulate a directed immune response. Last several decades a wide variety of bsAb formats and production technologies have been developed. BsAbs are constructed either chemically or biologically, exploiting techniques like cell fusion and recombinant DNA technologies. There are over 100 different formats of bsAb so far developed, but they could be classified into the two main categories such as Fc-based (with a Fc region) bsAbs and fragment-based (without a Fc region) bsAbs. BsAb has a broad application prospect in tumor immunotherapy and drug delivery. Here, we present a brief introduction to the structure of antibody, pharmacological mechanisms of antibodies and the trend in the production technologies of therapeutic antibodies. In addition, we address a review on the current status of various bsAb format development and their production technologies together with global situation in the clinical studies of bsAb.
Polyclonal antibodies against human DNA-dependent RNA polymerase II (HPP II) were generated from chicken egg yolk after immunization with RNA polymerase II as an antigen. The antibodies from egg yolk (IgY) were purified and characterized. IgY showed a specificity against DNA-dependent RNA polymerase II, and was a polyclonal antibody against 12 subunits of polymerase II. An amount of 0.35 mg of IgY was obtained freman HPP II-Sepharose affinity column using 10 eggs from a chicken immunized against RNA polymerase II as an antigen. These antibodies can be used for isolating the genes for RNA polymerase II components, and for in vitro transcription assays using HP-RNA polymerase II.

In a previous study, we obtained polyclonal antibodies against the organophosphorus insecticide fenitrothion and developed an enzyme-linked immunosorbent assay (ELISA) for this pesticide. Using these antibodies and an enzyme tracer, a direct competitive ELISA method specific for fenitrothion using a dipstick format was developed. Dipstick ELISA using antibodies to fenitrothion immobilized on an Immunodyne membrane allowed the quick visual detection of fenitrothion at concentrations above $10\;{\mu}g/L$. The $IC_{50}$ value of dipstick ELISA using reflectance detection was $27\;{\mu}g/L$ with a detection limit of $2\;{\mu}g/L$. The recovery of fenitrothion from spiked lettuce and rice samples using the dipstick ELISA ranged from 87-107%.
High titer rabbit polyclonal antibodies (pAbs) which show a specificity for saikosaponin a (SSA), have been generated. The immunogen used was a conjugate of SSA linked through its glucose moiety to bovine serum albumin by periodate oxidation method. The antibody titers obtained from two rabbits, innoculated with the immunogen, reached a plateau after the fourth and third booster injection, respectively. The specificity of the pAbs was determined by hapten inhibition assays using several SSA-like structures. SSA competitively inhibited the binding of the rabbit anti-SSA pAbs to SSA-ovalbumin on solid phase, a coated antigen on the well. The antibodies showed high specificity to SSA, exhibiting no significant cross-reactivity with any of SSA analogues tested.
Transcription of mitochondrial DNA in the yeast S. cerevisiae depends on recognition of a consensus nonanucleotide promoter sequence by mitochondrial RNA polymerase specificity factor, which is a 43 kDa polypeptide encoded by the nuclear MTF1 gene. Mtf1p has only limited amino acid sequence homology to bacterial sigma factors, but functions in many ways like sigma in that it is required for promoter recognition and initiation of transcription. To analyze the corebinding region of Mtf1p, monoclonal antibodies to this protein were prepared. Recombinant Mtf1p overproduced in E. coli was purified to near homogeneity and used to raise monoclonal antibodies (mAbs). From fused cells screened for Mtf1p mAbs by immunodot blot analysis, 19 positive clones were initially isolated. Further analysis of positive clones by Western blotting resulted in 4 mAbs of Mtf1p.
This survey was carried out to investigate the seroprevalence of antibodies to Neospora (N.) caninum in dairy cattle raised in Kangwon province in Korea. A total of 349 sera collected from dairy cattle were tested for N. caninum antibodies using an indirect immunofluorescent antibody test (IFAT). Ninety six (27.5%) dairy cattle were positive by IFAT. Seroprevalence of cows was 28.9% (91/315) and seroprevalence of boars was 14.7% (5/34). The seroprevalences of cows were increased according to the ages from 19.6% in cows less than 2 year-old to 50.0% in cows more than 7 year-old. Among the seroprevalences of cattle according to the raised areas, five counties or cities, Wheongsung was 34.6% (27/78), Wonju was 29.3% (46/157), Hongcheon was 28.9% (13/45), Chuncheon was 15.0% (3/20) and Cheolwon was 13.3% (2/15). It was proved that dairy cattle raised in Kangwon provinces exposed extensively and seriously to N. caninum.
From among the group of patients diagnosed clinically to have Guillain-Barre syndrome(GBS), subgroups with pure motor involvement have been identified. Some of such patients appear to have an axonal neuropathy by eletrophysiology. Such cases have been termed acute motor axonal neuropathy(AMAN). Many of these patients are found clinically to have normal sensation and to have electrodiagnostic patterns consistent with selective degeneration of motor axons. A serological survey showed some of individuals with AMAN had evidence of antecedent Campylobacter jejuni(CJ) infection. And AMAN has an association with the presence of anti-ganglioside antibodies. This article reviewed briefly the AMAN and their relationship to CJ infection and anti-ganglioside antibodies.
A serological survey for antibodies to Leptospira (L.) interrogans in dairy and Korean native cattle in Gyeongbuk area was performed using 6 different living antigens, which were L. icterohaemorrhagiae(RGA), L. canicola(Hond Utrecht IV), L. autumnalis(Akiyami A), L. australis(Ballico), L. pomona(Pomona) and L. hebdomadis(Hebdomadis), by microscopic agglutination test. In the microscopic agglutination test, greater than partial agglutination at a serum dilution of 1:300 or over was recorded as positive. In the dairy cattle(Holstein), four(4.7%) of 86 sera from 13 dairy farms were positive for L. canicola and L. pomona antibodies. In the Korean native cattle, four(3.2%) of 124 sera from the slaughter house in Taegu city were positive for L. canicola, L. icterohaemorrhagiae and L. pomona antibodies. Among the positive sera, L, canicola was dominated in this experiment.
