Effect of Anti-Skin Wrinkle and Antioxidant of Agastache rugosa Kentz through Fermentation Process of the lactic acid (유산균 발효 배초향 추출물의 항산화 활성과 주름개선 효능)

  • This study was to investigate anti-skin wrinkle effect of Agastache rugosa Kentz extracts by extraction processes. In the comparison of the effect of the solvent extraction, the extracts by 70% ethanol (EE) showed better biological activities that those by hot water. Therefore, further fermented Agastache rugosa was applied to 70% ethanol extraction process (FEE). FEE showed higher DPPH scavenging activity of 62.98% than EE's 62.71% at $1.0mg/m{\ell}$, but there was no significant. Elastase inhibition was measured 23.0% from FEE at $1.0mg/m{\ell}$. Cytotoxicity showed the highest 16.26% from FEE, this value is safe in the cell experiment. Collagen production showed $113.1ng/m{\ell}$ from FEE, on the other hand EE was measured $77.4ng/m{\ell}$ in adding $1.0mg/m{\ell}$. MMP-1 production was observed $1398pg/m{\ell}$ from FEE and EE was measured $1632pg/m{\ell}$. These results were found the highest antioxidant and anti-wrinkle effect. As a result, it was also confirmed that anti-skin wrinkle activities of the Agastache rugosa Kentz extract was correlated with anti-oxidant activities.

A Verification of Cosmetic Effect about Anti-oxidant and Anti-wrinkle of 11 Native Plants (자생식물 11종의 항산화 및 항주름에 관한 향장효능 검증)

  • To check the possibility of using natural extracts for cosmetic substances, we tested the bioactive effects of 11 crude extracts originating from native plants grown in the Republic of Korea on anti-oxidant and anti-wrinkle activities. When DPPH scavenging activities were measured, the crude extracts of Vitex rotundifolia, Scirpus triangulatus, S. wichurae, S. tabernaemontani, and Aeschynomene indica assays had more than 70% of DPPH scavenging activities at 100 μg/ml concentration. The data from the hydrogen peroxide scavenging assay revealed that the crude extracts from the 10 species, except for Eleocharis mamillata var. cyclocarpa, could scavenge more than 90% at 1,000 μg/ml concentration. The crude extract of V. rotundifolia had the highest xanthine oxidase scavenging activity at a 1,000 μg/ml concentration. Based on the four anti-oxidant assays, the crude extracts of V. rotundifolia, S. triangulatus, S. wichurae, S. tabernaemontani, and A. indica were shown to have excellent anti-oxidant capacities overall. In addition to the anti-oxidant capacities, the crude extract from S. wichurae exhibited high collagenase inhibitory activity, therefore, it could be a promising candidate for a cosmetic ingredient with anti-oxidant and anti-wrinkle qualities. The results of this study indicate that many wild plants native to the Korea Peninsula can be used as excellent natural substances for cosmetics based on their bioactive effects.

Pilot Study on Anti-wrinkle Effect of Herbal Cosmetic Containing the Extracts of Douchi (fermented Glycine max Merr.) (두시 추출물을 함유한 한방화장품의 주름개선 효과에 대한 예비 임상연구)

  • From March 1, 2017 to May 31, 2017, subjects were allowed to apply a wrinkle-improving herbal cosmetic product containing the extract of Douchi for 8 weeks. The effectiveness of herbal cosmetics was evaluated through wrinkle improvement efficacy parameter analysis, subjective wrinkle improvement through questionnaires, and adverse reaction. The herbal cosmetic products containing the extract of Douchi showed a significant decrease in the wrinkle improvement efficacy parameter analysis result. The results of the subjective satisfaction analysis after use were also evaluated positively. We could confirm the possibility of wrinkle improvement effect of Douchi extract, and it will be possible to contribute to the expansion of korean medicine field through more studies in the future.

A Clinical Research about Herbal Cosmetics Containing Momordica charntia L. Extracts on the Anti-wrinkle and Whitening Effects (여주 추출물을 함유한 한방화장품의 주름 및 미백 개선에 관한 임상적 연구)

  • Objective : The purpose of this clinical research was to investigate the effects of herbal cosmetics containing Momordica charntia L. extracts on the anti-wrinkle and whitening effects. Methods : A total of 25 subjects who visited Daegu Hanny Oriental Medical Center from November 1st, 2010 to December 31th, 2010 were included in this study. In this study, we observed change of R1, R2, R3, R4, R5 for using products, change of average melanin content and pigmental area, analysis results of subjective wrinkle improvement, Analysis results of subjective whitening improvement. Statistical analysis was performed by using paired t-test and wilcoxon signed ranks test. Statistical significance was achieved if the probability was less than 5%(p<0.05) Results : Statistically, herbal cosmetics containing Momordica charntia L. extracts showed siginificant effect on change of R1, R2, R3, R4, R5 for using products, change of average melanin content and pigmental area, analysis results of subjective wrinkle improvement, Analysis results of subjective whitening improvement(p<0.05). And satisfaction after using herbal cosmetics was near good. Conclusions : Considering the above results, we have concluded that herbal cosmetics containing Momordica charntia L. extracts have the remarkable effects on the anti-wrinkle and whitening effects.

Development of Vaccinium uliginosum L. extracts for whitening & anti-wrinkle functional food

  • This study is peformed to investigate the effect of water extract from Vaccinium uliginosum L., on melanin production in B 16 melanoma cells, procollagen production and matrix metalloproteinase-1(MMP-1) inhibition in human fibroblast cells. One hundred grams of the Vaccinium uliginosum L. was extracted with 2000 mL of water($90^{\circ}C$, 16h, 2times). The water extracts were lyophilized and stored at $4^{\circ}C$ until used. Dry weight yields of extracts of Vaccinium uliginosum L. were $3\%$(w/w). Extracts from Vaccinium uliginosum L. showed scavenger activities on DPPH radical, superoxide anion radical, hydroxyl radical, hydrogen peroxide and singlet oxygen radical. And these substances inhibited release of cyiokines from human keratinocyte after UV B exposure. Therefore we confirmed that extracts from Vaccinium uliginosum L. had antioxidative effect. These substances inhibited purified tyrosinase activity and melanogenesis in B 16 melanoma cells treated/untreated IL-$1{\alpha}$. Moreover this extract stimulated procollagen production and inhibited MMP-1 production in human fibroblast cells treated/untreated IL-$1{\beta}$. Therefore we confirmed that extracts from Vaccinium uliginosum L. had whitening effect. And these substances decreased degree of wrinkle in hairless mouse skin that induced by UV B irradiation. Therefore we confirmed that extracts from Vaccinium uliginosum L. had anti-wrinkle effect. From the above results, it is possible that Vaccinium uliginosum L. may be developed to be an anti-melanogenesis agent and anti-wrinkle agent.

Skin Volume Augmentation and Anti-wrinkle Effects of Tribulus terrestris Fruit Extract (질려자 추출물의 피부 볼륨 증진 및 주름개선 효과)

  • Aging of the face is mainly related to the features that are sagging or loss of elasticity of the skin by reducing the volume around the eyes or cheek. Intrinsic aging can be seen to cause thinner dermis, reduction of extracellular matrix and subcutaneous fat. This study was carried out to investigate the skin volume augmentation and anti-wrinkle effects of Tribulus terrestris fruit extract. Skin anti-aging effect of Tribulus terrestris fruit extract was evaluated by using lipid accumulation, expressin of type I procollagen and elastin in preadipocytes and human dermal fibroblasts. Tribulus terrestris fruit extract augmented preadipocytes differentiation about 56% at 100 µg/mL. The type I procollagen and elastin were increased about 35% and 25% by treatment 20% Tribulus terrestris fruit extract, respectively. The clinical study also showed that skin sagging, skin elasticity, and dermal density improved without adverse effect following 4 week application of cream containing 2% Tribulus terrestris fruit extract. We suggest that Tribulus terrestris fruit extract can have the good possibility as skin volume augmenting, skin elasticity and wrinkle improving agent.

  • Anti-wrinkle effect of N-Acetyl-D-glucosamine (NAG) was evaluated by collagen synthesis and proliferation of normal human fibroblast. NAG was obtained by purifying deacetylated chitin which can be derived from chitin-rich crab shell. We studied in in-vitro cultures of human normal fibroblast, whether synthesis of collagens and fibroblast growth activation in these cells can be enhanced in the presence of NAG. It 야d not show any adverse effects in human shin irritation patch test. In in-vivo mouse test, it showed anti-wrinkle effect in hairless mouse (6W/F). From the HPLC analysis, the stability of NAG in the cosmetics product could be maintained for a long time. These results demonstrated that NAS can be useful anti-wrinkle cosmetic ingredient.

Development of Anti-Wrinkle Agent from Nelumbo nucifera Root Extract (연근 추출물에서 주름개선 소재의 개발)

  • In this research, root extracts of Nelumbo nucifera was tested to see the possibility for functional cosmetic agent. 70-100% ethanol was used as solvent and nuciferin was confirmed as active component. To test cosmetic effect of root extracts of Nelumbo nucifera, safety effect(MTT assay), anti-wrinkle effect(elastase inhibition assay) and antioxidation effect(DPPH free radical scavenging assay) were measured. When 100% ethanol was used as extracting solvent, cell viability was over 80% at $100{\mu}g/ml$, which indicated that root extract of Nelumbo nucifera was suitable for cosmetic agent. Root extract of Nelumbo nucifera showed 40~50% elastase inhibition at $100{\mu}g/ml$ so that it had good anti-wrinkle characteristics. 50% antioxidation capacity($FSC_{50}$) was $5.0{\sim}38{\mu}g/ml$ and root extract of Nelumbo nucifera showed excellent antioxidation effect. From the research, root extracts of Nelumbo nucifera showed strong possibility for anti-wrinkle functional cosmetic agent.

Anti-oxidation and Anti-aging Effect of Mixed Extract from Korean Medicinal Herbs (한약재 복합 추출물이 항산화 및 항노화에 미치는 영향)

  • Objectives : The present study was designed to investigate effects of mixed extract from korean medicinal herbs (MIX) on oxidation/reduction reaction-related and aging-related enzyme in vitro. Methods : We performed MTT assay, collagenase inhibition assay, elastase inhibition assay, tyrosinase inhibition assay, DPPH free radical scavenging assay, SOD-like activity and xanthine oxidase inhibition assay. Results : Recently, many studies have reported that elastin is also involved in inhibiting or repairing wrinkle formation, although collagen is a major factor in the skin wrinkle formation. The MIX showed 97% inhibition of collagenase activity, and 64% inhibition of elastase activity at 1 mg/ml concentration of MIX, next only to positive control, which indicate good efficacy for anti-wrinkle ingredient. Also it's treatment showed 34% inhibition of tyrosinase activity, to relate whitening effect, at the same dose of MIX. Antioxidant activities were determined by DPPH radical scavenging, xanthine oxidase (XO) inhibiting activity and SOD-like activity. Also these scavenging, XO-inhibiting and SOD-like activities were measured in 91%, 80%, and 63% inhibition, respectively, at a treated dose of 1 mg/ml, compare to control. Conclusions : These results suggest that possibility of mixed korean medicinal herbs as a functional ingredient for anti-wrinkle and whitening, anti-oxidation and anti-aging cosmetic formula.

Anti-wrinkle Activity of Lindera obtusiloba Extract (생강나무 추출물의 광노화에 의한 주름형성 억제 효과)

  • Chronic exposure of solar ultraviolet (UV) light to human skin results in photoaging with wrinkle formation. This study was performed to investigate anti-wrinkle effects of Lindera obtusiloba extract (LO) on UVB-induced wrinkle formation. We first measured cell proliferation and type I pN collagen synthesis activities in normal human dermal fibroblasts. Cell proliferation and type I pN collagen synthesis were increased by 33.8 % and 91.8 %, respectively, compared with no treatment control. SKH-1 hairless mice were topically applied 5 % LO solution for 10 weeks with UVB irradiation three times a week. After 10 weeks, a visual assessment and replica assay were performed on each mouse. According to visual assessment of close-up photos and skin replica, application of 5 % LO solution inhibited UV-induced wrinkle formation in mouse skin as compared to the vehicle-applied control mice. These results indicated that LO could protect skin wrinkle formation caused by chronic photo-irradiation in hairless mice.
