Alginate의 면역조절작용 (Immunomodulating Activity of Alginate)

  • Alginates are polysaccharides with gel-forming properties composed of 1,4-linked $\beta$-D-mannuronic acid (M), $\alpha$-L-guluronic acid (G), and alternating (MG) blocks. The M-and the MG-blocks, but not the G-blocks, have been known to be the active components of the alginates in experimental models. In this study, we have examined the ability of high M-alginate to activate immune cells. Alginate induced the macrophage anti-viral activity and the lymphocyte blastogenesis, and enhanced cytotoxicity of natural killer cell. In addition, alginates stimulated the macrophages to induce the production of $H_{2}O_{2}$, whereas alginates had no effect on NO production and suppressed the production of TNF-$\alpha$. These findings suggest that high M-alginate may be modulating various elements of the host immune response.

  • Although most enterovirus infections are not serious enough to be life threatening, several enteroviruses such as enterovirus 71 are responsible for severe, potentially life-threatening disease. The epidemic patterns of enteroviruses occur regularly during the year, but they may change due to environmental shifts induced by climate change due to global warming. Therefore, enterovirus epidemiological studies should be performed continuously as a basis for anti-viral studies. A great number of synthesized antiviral compounds that work against enteroviruses have been developed but only a few have demonstrated effectiveness in vivo. No proven effective antiviral agents are available for enterovirus disease therapy. The development of a new antiviral drug is a difficult task due to poor selective toxicity and cost. To overcome these limitations, one approach is to accelerate the availability of other existing antiviral drugs approved for antiviral effect against enteroviruses, and the other way is to screen traditional medicinal plants.

  • In this paper, we study a within-host mathematical model of HCV infection and carry out mathematical analysis of the global dynamics and bifurcations of the model in different parameter regimes. We explore the effect of reverse transcriptase inhibitors (RTI) on spontaneous HCV clearance. The model can produce all clinically observed patient profiles for realistic parameter values; it can also be used to estimate the efficacy and/or duration of treatment that will ensure permanent cure for a particular patient. From the results of the model, we infer possible measures that could be implemented in order to reduce the number of infected individuals.

  • To isolate Lactic acid bacteria for animals, we have screened from Kim-chi, swine intestine, swine feces, and dairy products by random selection and anti-viral, antipathogenic bacteria test. Among them, CLP-1 shown that inhibitory effect against rotavirus, porcine epidemic diarrhea (PED) virus, Salmonella sp, and E.coli. By examining biological property, API-ZYM and identified Lactobacillus plantarum by 16S rDNAgene sequence. CLP-1 determined resistance to low pH and bile salt. Futhermore, the cell body of CLP-1 adhered to the intestinal epithelium tissue of swine and Caco-2 cell. CLP-1 was examined on cell immune system modulating activity in vitro. The whole cell and cell culture supernatant was increasing of interferon-${\beta}$ activity. And then, CLP-1 increased prevention effect by Salmonella enteritidis infection in SPF chickens. And we determined similar result in pigs.

해양생물 추출물의 대장암세포주에 대한 항암 작용 검색 (Anti-cancer Effect of Marine Resources Against Human Colorectal Cancer Cells)

  • 해양생물자원은 차세대 바이오산업의 중요한 자원으로 관심이 증대되고 있다. 다양한 해양생물자원이 생물학적 활성을 가지고 있을 것으로 기대됨에도 불구하고, 지금까지는 시료 채취의 어려움 등으로 개발에 제한이 있었다. 해면동물, 해조류, 산호 등의 대부분의 해양생물자원들이 험난한 환경에서 살아남기 위해 특수한 대사산물을 만들어 낼 것으로 여겨지고 있어 이를 활용하기 위한 노력들이 기울여지고 있다. 많은 종류의 해양생물자원 중에서 바이오산업에 활용할 수 있는 항암, 항균, 항바이러스 및 항암 작용 등과 같은 생물학적 활성을 가진 물질을 선별하는 것이 시급한 실정이다. 본 연구에서는 항암 작용을 갖는 해양생물자원을 도출하기 위하여 사람유래 대장암 세포주에서 세포독성시험을 실시하였다. 해양생물자원은 2013년 3월 마크로네시아에서 채취한 샘플들로 메탄올로 추출한 물질을 사용하였다. 해양생물자원의 세포독성시험을 실시하여 20개의 시료 중에서 3개의 시료에서 농도의존적인 세포 생존을 억제하는 것을 확인하였다. 검색된 시료들 중 2종만이 동정되어, 해면동물 Hyrtios sp.임이 밝혀졌다. 한종은 아직 밝혀지지 않은 상태로 추가적인 연구를 통해 동정이 필요하였다. Hyrtios sp. 추출물(1304KO-327과 1304KO-329)의 HCT116세포 증식억제작용은 이전 연구에서 보고된 RKO에서의 작용과 일치함을 알 수 있었다. 대장암세포주의 생존 억제 활성을 밝혀낸 3종의 도출 물질은 앞으로의 지속적인 연구를 통해 추출물 중의 항암 활성 물질을 규명함으로써 바이오산업의 새로운 개발 자원으로 활용될 것이 기대된다.

혈관내피세포에서 우방자(牛蒡子) 에탄올 추출물의 항염증 효과 (Anti-Inflammatory Effect of Ethanol Extract from the Seeds of Arctium Lappa L. in Vascular Endothelial Cells)

  • Objectives: The seeds from Arctium lappa have been considered for its various pharmacological properties, which include anti-carcinogenic, anti-inflammatory, anti-diabetic, and anti-viral activities. Methods: In the present study, we investigated the anti-inflammatory effect of the ethanol extract from the seeds of Arctium lappa L (EAL) on cytokine-induced vascular inflammation in human umbilical vein endothelial cells (HUVEC). Results: Pretreatment with EAL significantly decreased tumor necrosis factor alpha ($TNF-{\alpha}$)-induced cell adhesion molecules expression such as intercellular adhesion molecule-1 (ICAM-1), vascular cell adhesion molecule-1 (VCAM-1), and endothelial-selectin (E-selectin) in a dose-dependent manner. Cell adhesion assay showed that pretreatment with EAL suppressed HUVEC-monocyte adhesion by $TNF-{\alpha}$ over $1{\mu}g/ml$ concentration. We investigated the involvement of nuclear transcription factor kappa-B ($NF-{\kappa}B$) in $TNF-{\alpha}$-induced vascular inflammation. $NF-{\kappa}B$ p65 nuclear expression was induced by $TNF-{\alpha}$, however, pretreatment with EAL was attenuated that nuclear translocation. In cytoplasm, EAL was also attenuated $TNF-{\alpha}$-induced decrease of inhibitor of ${\kappa}B-{\alpha}$ ($I{\kappa}B-{\alpha}$) expression. Moreover, EAL significantly decreased $TNF-{\alpha}$-induced production of intracellular reactive oxygen species (ROS). Conclusions: Taken together, our findings suggest that seeds of Arctium lappa L could be a therapeutic herb for prevention of cardiovascular diseases throughout the inhibition of vascular endothelial inflammation.

Thioacetamide 유발 간손상모델에서 통규활혈탕의 간보호효과 (Protective Effect of Tongyuhwalhyeol-tang on Liver Injury in Thioacetamide-induced Rat)

  • Objectives : Liver disease is an inflammatory reaction caused by oxidative stress, viral, alcohol, and drug properties. Inflammatory reaction causes hepatitis and chronic hepatitis is persistent, it progresses to liver fibrogenesis and liver cancer. The aim of this study was to confirm the hepatoprotective effect of Tongyuhwalhyeol-tang(Tongqiaohuoxue Decoction) (TH) and Gamtongyuhawlhyeol-tang(GTH) in TAA-induced liver injury animal model. Methods : The antioxidant activities were evaluated through in vitro experiments, such as 1, 1-diphenyl-2-picrylhydrazyl (DPPH) and 2, 2'-azinobis-3-ethyl-benzothiazoline-6-sulfonic acid (ABTS) radical scavenging assays, total polyphenol and total flavonoid content measurement. To confirm the liver protective effect, induced by Thioacetamide (TAA) for 3 days injection at 200 mg/kg rats. TH and GTH were treated 3 days at 200 mg/kg/day. The changes of reactive oxygen species (ROS), peroxynitrite ($ONOO^-$), alanine aminotransferanse (ALT) and aspartate aminotransferase (AST) in serum were analyzed after experiment. Also, expression of anti-inflammation, anti-oxidant related proteins were investigated by western blot analysis. Results : TH was inhibited the antioxidant activities. In the TAA-induced rat, TH decreased ROS, $ONOO^-$, ALT, AST level in serum. Inflammation related protein expressions increased in TAA-induced rat compared to normal rat. However, TH group inhibited the down expression of these proteins. Also, anti-oxidant related protein expressions increased in TH group compared TAA-induced rat. Conclusion : Therefor, these results suggested that TH provided hepatoprotective effects on the hepatic injury leading to the reduction of inflammatory response. In addition, the effect of TH was superior to that of GTH.

조록나무 잎 에틸 아세테이트 분획물의 아데노바이러스 36에 대한 항바이러스 효과 (Antiviral Effects of Ethyl Acetate Fraction of Distylium racemosum Leaf Extract on Adenovirus 36)

  • 비만의 원인은 유전적, 대사적 및 환경적 요인 등 다양한 가운데 특정 세균 및 바이러스 감염에 의해 비만이 유도되는 것이 확인되며 이러한 비만은 감염성 비만이라고 한다. 또한 감염성 비만을 유발하는 병원체 가운데 아데노바이러스 36이 밝혀졌다. 현재 비만 예방 및 치료제 개발에 대한 연구가 활발히 진행되는 가운데 감염성 비만의 예방 및 치료제 개발에 대한 연구는 불충불한 실정이다. 이와 관련하여 본 연구에서는 제주에 자생하는 조록나무 잎을 이용하여 분획물을 제조하고, 비만을 유도하는 사람아데노바이러스 36 증식에 미치는 영향을 평가하고자 한다. A549세포에 사람아데노바이러스 36을 감염시키고, 조록나무 잎 에틸 아세테이트 분획물을 처리하여 바이러스 역가를 측정하였다. 그 결과, 조록나무 잎 에틸 아세테이트 분획물을 처리한군에서 농도 의존적으로 바이러스 증식 억제능을 확인하였다. 또한 조록나무 에틸 아세테이트 분획물의 물질 분리를 한 결과, quercitrin이 확인되었으며, 표준물질 quercitrin의 사람아데노바이러스 36에 대한 증식 억제능을 확인하였다. 이러한 결과 조록나무 잎 에틸 아세테이트 분획물은 감염성 비만 치료제 개발을 위한 효과적인 후보 물질로 사료되며, 본 연구는 감염성 비만 치료제 개발을 위한 기초 자료로 유용할 것으로 보인다.

Comparative Analysis of the Complete Genome of Lactobacillus plantarum GB-LP2 and Potential Candidate Genes for Host Immune System Enhancement

  • Acute respiratory virus infectious diseases are a growing health problem, particularly among children and the elderly. Much effort has been made to develop probiotics that prevent influenza virus infections by enhancing innate immunity in the respiratory tract until vaccines are available. Lactobacillus plantarum GB-LP2, isolated from a traditional Korean fermented vegetable, has exhibited preventive effects on influenza virus infection in mice. To identify the molecular basis of this strain, we conducted a whole-genome assembly study. The single circular DNA chromosome of 3,284,304 bp was completely assembled and 3,250 protein-encoding genes were predicted. Evolutionarily accelerated genes related to the phenotypic trait of anti-infective activities for influenza virus were identified. These genes encode three integral membrane proteins, a teichoic acid export ATP-binding protein and a glucosamine - fructose-6-phosphate aminotransferase involved in host innate immunity, the nonspecific DNA-binding protein Dps, which protects bacteria from oxidative damage, and the response regulator of the three-component quorum-sensing regulatory system, which is related to the capacity of adhesion to the surface of the respiratory tract and competition with pathogens. This is the first study to identify the genetic backgrounds of the antiviral activity in L. plantarum strains. These findings provide insight into the anti-infective activities of L. plantarum and the development of preventive probiotics.

Antiviral Activity of Glycyrrhetinic Acid on MA-104 Cell Infection of the K-21 Korea Human Rotavirus Isolate

  • The mutability and frequency of genetic reassortment characteristic of rotavirus and resultant antigenic changes make the rotavirus formidable challenges for control efforts such as the vaccine development. An alternative approach to overcome these difficulties in development of the rotavirus vaccine is to develop effective inhibitors of the virus infection. As an effort to achieve this, effects of glycyrrhetinic acid (GA), which is an active component of glycyrrhizin, on MA-14 cell infection were examined by employing the human rotavirus isolated from Korea, K-21. The data obtained showed that MA-104 cell infection of the K-21 rotavirus was greatly influenced by the presence of both $18{\alpha}-Ga\;and\;18{\beta}-GA$. Both types of GA have inhibited more than 60% of the rotaviral infection at the concentration of 7.68mM. This inhibition effect became much more evident at the higher concentrations of GA. However, the type of GA did not make much differences on the inhibition effect of the drug. Although GA has to be used in high concentrations to exhibit anti-viral activity and to be virostatic, a long history of safe and high dose usage of licoriece in clinical settings in the Far East makes the GA as an attractive inhibitor of the rotaviral infection.
