Background: Lidocaine spray is a local anesthetic that improves random-pattern skin flap survival. The fractional ablative carbon dioxide laser (FxCL) produces vertical microchannels that delivers topically applied drugs to the skin. In this study, we hypothesized that FxCL therapy would enhance the lidocaine effect to improve random-pattern skin flap survival in rats. Methods: McFarlane random-pattern skin flaps were elevated in 48 rats, which were divided into four groups according to treatment: FxCL+lidocaine, FxCL, lidocaine, and nontreatment (control). On postoperative day 7, necrotic flap areas, the number of capillary vessels, and neutrophil count were evaluated. Anti-rat vascular endothelial growth factor (VEGF) and CD31 antibody activity were also evaluated by immunohistochemical staining. Results: Flap survival rate was $53.41%{\pm}5.43%$, $58.16%{\pm}4.80%$, $57.08%{\pm}5.91%$, and $69.08%{\pm}3.20%$ in the control, lidocaine, FxCL, and FxCL+lidocaine groups, respectively. Mean neutrophil count in the intermediate zone excluding the necrotic tissue was $41.70{\pm}8.40$, $35.43{\pm}6.41$, $37.23{\pm}7.15$, and $27.20{\pm}4.24cells/field$ in the control, lidocaine, FxCL, and FxCL+lidocaine groups, respectively. Anti-rat VEGF and CD31 antibody activity were the highest in the FxCL+lidocaine group. Conclusion: FxCL with lidocaine had a positive effect on random-pattern skin flap survival in rats. Thus, FxCL with lidocaine spray should be considered as a new treatment option to improve flap viability.
목적: 혈관내피성장인자(VEGF)와 그 수용체는 종양의 성장과 전이에 매우 중요한 역할을 한다 3개의 수용체가 알려져 있는데 그 중에서도 VEGFR2 (Flk-1/KDR)가 종양 angiogenesis에 매우 밀접하게 관련한 것으로 알려져 있다. 본 연구에서는 마우스 Flk-1 항체로 알려진 DC101에 I-131을 표지하여 매우 공격적인 종양으로 알려진 흑색 종의 치료 정도를 확인해 보고자 하였다. 방법: 하이브리도마 세포에서 Flk-1 항체인 DC101을 분리하여 western blot, ELISA, maldi-tof 방법을 이용하여 순도를 확인하고 항체 에 chloramin T를 이용하여 I-131을 표지하였다. 누드마우스에 B16F10세포를 주사하여 흑색종 모델을 만들고 평균 $200-250\;mm^3$으로 키워 $^{131}I$-DC101을 주사하여 영상과 시간별 장기섭취율(%ID/g)을 비교하고, 종양내 Flk-1 발현을 확인하기 위하여 면역염색 등을 시행하였다. 흑색종 동물모델을 5개 군으로 나누어 각각 치료를 시행하였다. 1군은 PBS만을, 2군은 $^{131}I$-DC101 $50\;{\mu}g(200\;{\mu}Ci)$을, 3군은 DC101 $50\;{\mu}g$을, 4군은 $^{131}I$-DC101 $30\;{\mu}g(200\;{\mu}Ci)$을, 5군은 $^{131}I$-DC101 $15\;{\mu}g(200\;{\mu}Ci)$을 각각 매 3일 ${\sim}$ 4일마다 주사하고 전체 5회를 주사하였고 종양볼륨을 측정하였다. 결과: $^{131}I$-DC101을 흑색종 모델에 정맥주사하고 78시간까지 영상을 얻은 결과 시간에 따라 종양섭취가 증가 하는 영상을 보였다. 시간대별 장기섭취를 정확히 확인하기 위하여 1시간, 6시간, 24시간, 48시간, 72시간 장기섭취율을 비교한 결과 시간에 따라 혈액내 방사능치가 서서히 감소하였고 다른 장기의 섭취도 시간에 따라 감소하였고 종양의 섭취는 48시간까지 증가하였다가 그 이후는 감소하였다. 흑색종 동물모델에 $^{131}I$-DC101 치료를 시행한 결과 3번째 주사까지는 각군간의 유의한 차이를 보이지 않다가 4번째 주사를 시행한 때부터 1군과 2군, 또는 1군과 4군간의 유의한 차이를 보이기 시작했다. 또한 5번째 주사 이후에는 5군에서도 유의한 차이를 보여 I-131 에 의한 효과가 뚜렷해짐을 확인하였다. 결론 마우스 Flk-1 항체로 알려진 DC101을 흑색종 모델에 정맥내 주사하였을 때, 종양성장억제 효과를 보이지 않는 항체양에서도 I-131을 표지하여 치료를 시행했을 경우에는 효율적인 종양성장억제 효과를 보였다.
목적 : 인삼은 다양한 생물학적 작용이 있다. 그 중 항염증작용과 관련하여 염증성 사이토카인 분비 및 저산소 유도인자-1${\alpha}$ 활성화 조절 효과를 살펴보고자 한다. 방법 : phorbol myristate acetate (PMA)+A23187에 유도된 세포에서 염증성 cytokines 분비의 변화와 인간의 mast cell인 HMC-1 cells에서 hypoxia-inducible factor-1 (HIF-1)${\alpha}$의 작용을 관찰하였다. 결과 : PMA+A23187은 대조군과 비교해서 interleukin $(IL)-1{\beta}$, IL-6와 tumor necrosis factor $(TNF)-{\alpha}$의 분비를 증가시킨다. 또한 증가된 cytokines IL-1, IL-6, $TNF-{\alpha}$가 인삼의 처리에 의해 두드러지게 억제하는 것을 확인하였다. 인삼(5 ${\mu}g/ml$)은 $IL-1{\beta}$, IL-6, $TNF-{\alpha}$의 분비를 약 105.1${\pm}$9.7%, 95${\pm}$9.4%, 29.7${\pm}$4.5%,(P<0.05)로 최대로 억제하였고, PMA+A23187에 유도된 vascular endothelial growth factor (VEGF)와 granulocyte macrophage-colony stimulating factor (GM-CSF)의 분비를 41.3%와 75.7%로 각각 억제하였다. 그리고 저자는 인삼이 PMA+A23187로 유도된 HIF-1${\alpha}$ 발현과 HIF-1에 대해 DNA binding activity를 억제하고 있음을 관찰하였다. 결론 : 인삼이 HIF-1에서 염증반응을 억제함을 나타내고, 이는 인삼이 염증성 질환을 치료하는데 유익한 효과가 있음을 의미한다.
본 연구에서는 인간 유방암 세포 라인인 MDA-MB-231 세포에서 GD에 의해 EVs분비 억제에 의한 항암효과를 처음으로 확인하고자 하였다. MDA-MB-231 세포에 GD를 처리하였을 때 농도 의존적으로 세포의 증식률을 억제하는 것을 MTT assay를 통해 확인할 수 있었으며, ROS 염색과 apoptosis marker 단백질인 p-JNK단백질의 증가를 통해 GD에 의한 세포의 증식 억제 효과가 세포사멸에 의한 것임을 유추할 수 있었다. 또한 wound-healing assay, 세포 침윤 및 VEGF 농도를 측정한 결과 GD가 암세포의 이동, 전이 능력을 억제하는 것을 확인하였다. Nanosight를 통해서 MDA-MB-231 세포에서 분비되는 대조군 EVs 및 GD에 의해 변화된 EVs의 사이즈를 확인하였다. 마지막으로 GD를 처리한 MDA-MB-231 세포에서 분비된 EVs보다 GD를 처리하지 않은 대조군에서 분비된 EVs의 단백질 및 particles수가 유의적으로 감소하는 것을 확인을 하였다. 그리고 GD가 MDA-MB-231 세포에서 EVs분비를 감소시키는 것을 대표적인 exosome marker인 TSG101, CD63의 발현 감소로 확인할 수 있었다. 이러한 결과로 인해 GD가 암세포의 EVs 분비를 감소시켜 암세포의 성장 및 전이를 억제하였음을 확인하였다. 본 연구는 GD가 인간 유방암 세포인 MDA-MB-231 세포의 EVs 분비를 억제하는 효과가 있음을 제시하고 있다. 따라서 GD가 유방암의 화학요법 약물로 작용할 수 있음을 시사한다.
목 적 : 급성 허혈성 신손상 후 손상된 세뇨관 상피세포의 재생은 순환하거나 국소적으로 생성된 성장인자나 cytokine, 생리적 인자 및 주위 환경들의 조절에 의해 매개되는 것으로 알려져 있다. 따라서 급성 허혈성 신손상 후 신기능 회복에 따른 혈중 IGF-I의 변화와 전체 신장조직과 각 부위에 따른 IGF-I, -II, VEGF, $TGF-{\beta}1$, CTGF의 mRNA 발현의 변화를 알아보는데 연구의 목적이 있다. 방 법 : 체중이 250-300 g의 백서를 대상으로 급성 허혈성 신손상을 시켜 유발된 급성 신부전 모델을 이용하여 급성 신부전의 진행과정에 따라 혈청 IGF-I치와 여러 성장인자 발현의 변화를 측정하였다. 급성 신부전 진행은 혈청 크레아티닌을 측정하여 평가하였고 혈청 IGF-I 농도는 iodinated IGF-I과 polyclonal anti-IGF-I 항체를 이용한 방사면역 측정법을 이용하여 측정하였다. 신장에서 성장인자의 발현은 RT-PCR을 이용하였고 신장조직에서의 RNA 추출은 통상적인 방법에 준하였다. 신장에서의 IGF-I과 CTGF의 분포는 anti-IGF-I 항체와 anti-CTGF 항체를 이용한 면역조직화학법을 이용하였다. 결 과 : 1) 양측 신동맥을 결찰한 후 1일째부터 체중 감소와 혈청 크레아티닌치의 증가가 관찰되었는데 이러한 변화는 3일째에 가장 현저하였다. 혈청 IGF-I은 신손상 후 1일 째 현저하게 감소하였고 신손상 3일째에 더 증가되었다가 5일째부터는 신손상 전으로 회복되었다. 2) 신기능 회복에 따른 성장인자의 변화는 거의 유사하였다. IGF-I, IGF-II, $TGF-{\beta}1$ 및 VEGF의 mRNA 발현은 허혈성 신손상 후 1일째에 현저하게 감소하였고 신손상 3일째에는 신손상 전에 비해 증가하였으며 7일째에는 거의 신손상 전의 수준으로 회복되었다. 3) IGF-I과 IGF-II는 정상 신장에서는 주로 수질부 특히 바 깥쪽 수질부에서 발현되었고 허혈성 신손상 1일째에 현저하게 감소하였다가 3일째에는 정상 수준으로 회복되었다. $TGF-{\beta}1$은 정상에서 신수질부에서 주로 발현되었고 1일째는 현저하게 감소하였다가 회복되는 소견을 보였다. CTGF와 VEGF는 정상 신장의 수질부와 피질부 모두에서 발현되었으며 신손상 1일째에 현저하게 감소되었다가 3일째부터 정상 수준으로 회복되는 소견을 보였다. 4) 정상 백서의 신장에서 IGF-I은 주로 피질부위의 신세뇨관에 분포하였고 사구체에서는 발견되지 않았는데 급성 허혈성 신손상 후 1일째 신장의 피질부위에서는 IGF-I가 현저히 감소되었다. CTGF는 정상 백서의 신장에서는 주로 혈관에 분포하는 소견을 보였는데 신손상 3일째에는 세뇨관에서 현저한 증가가 관찰되었다. 결 론: 이번의 연구로 저자들은 IGF-I, IGF-II, $TGF-{\beta}1$, CTGF, VEGF 등과 같은 성장인자가 급성 허혈성 신손상 후 신기능과 세뇨관세포의 회복과정에 발현의 변화가 초래된다는 사실을 알 수 있었다. 따라서 신기능 회복을 촉진시키기 위해 여러 성장인자를 급성신부전 치료제로 사용할 수 있을 것으로 생각되고 이에 대한 연구가 더 필요할 것으로 생각된다.
This study was performed to investigate the anti-photoaging effects of Persimmon leaf tea(PLT) in hairless mice(SKH-1) exposed to UVB irradiation. The animals were divided into non-treated group (normal, N) and UV-radiated groups. UV-radiated groups were divided into only UV-radiated group(control, C) and UV-radiated and PLT treated experimental groups[first extraction treated group(PLT-I), second extraction treated group(PLT-II), and third extraction treated group(PLT-III)]. Three PLT treated experimental groups of mice were treated with both oral administration(300 mg/Kg B.W./day) and topical application (100 ul of 2% conc./mouse/day) for 4 weeks. Anti-photoaging effects of Persimmon leaf were evaluated by anti oxidative reaction, stereomicroscopic and microscopic observations. The expression of photoaging skin related factors including mast cell tryptase, proliferating cell nuclear antigen (PCNA) and vascular endothelial growth factor (VEGF) was examined by immunohistochemical staining. Treatment of PLT-I, -II, -III prevented the wrinkle formation as well as epidermal hyperplasia, inflammatory cells, disruption of collagen in photoaged skin induced by UVB radiation. It also reduced the PCNA and VEGF expression in the UVB irradiated dorsal skin. Furthermore, it significantly decreased the number of mast cells in the UVB irradiated dermis(p<0.05 and p<0.01). On the effects of oxidative stress and antioxidant function on the treatment with water extract from Persimmon leaf tea(PLT), the activity of superoxide dismutase(SOD) was significantly increased in PLT-III group(p<0.05), and catalase(CAT) was significantly increased in PLT-I and PLT-III groups(p<0.05), and PLT-II group(p<0.001). These extracts showed relatively antioxidant activity and protective effect on UVB-induced oxidative stress in hairless mice(SKH-1). Our results suggest that Persimmon leaf tea may serve as an useful radical scavenging antioxidant and anti-photoaging skin agents in the UVB irradiated skin.
Objectives : The purpose of this study was to investigate effects of Houttuyniae Herba water extract (HC) on calcium release and production of various inflammatory mediators such as nitric oxide (NO), interferon-inducible protein (IP)-10, platelet derived growth factor (PDGF)-BB, keratinocyte-derived chemokine (KC), vascular endothelial growth factor (VEGF), interleukin (IL)-4, and IL-5 in lipopolysaccharide (LPS)-induced RAW 264.7 mouse macrophages. Methods : NO production was measured by Griess reagent assay. Intracellular calcium level was measured with Fluo-4 assay. Levels of cytokines were measured by High-throughput multiplex bead array cytokine assay based on xMAP (multi-analyte profiling beads) technology. Results : HC significantly decreased NO production for 24 hrs incubation at the concentrations of 10, 25, 50, 100, and $200{\mu}/mL$ in LPS-induced RAW 264.7 (P < 0.05). HC significantly decreased production of IP-10, KC, VEGF, and PDGF-BB for 24 hrs incubation at the concentrations of 50, 100, and $200{\mu}/mL$ in LPS-induced RAW 264.7 (P < 0.05). HC also significantly decreased intracellular calcium release for 24 hrs incubation at the concentrations of 25, 50, 100, and $200{\mu}/mL$ in LPS-induced RAW 264.7 (P < 0.05). But HC did not show any significant effect on production of IL-4 and IL-5 in LPS-induced RAW 264.7. Conclusions : The results suggested that HC has anti-inflammatory property related with its inhibition on the production of NO, IP-10, KC, VEGF, and PDGF-BB in LPS-induced macrophages via calcium pathway.
본 연구에서는 피부재생 화장품 소재로 활용하고자 저분자화 시킨 Polydeoxynucleotide (PDRN)의 창상 치유 효과를 조사하였다. 이를 위하여 연어 정소 유래 PDRN 단백질 제거공정, 내독소 제거공정을 거쳐 순수분리 정제하였고 분자량 저감공정을 거쳐 기존 PDRN 보다 피부 침투율을 높인 고순도 PDRN을 제조하였다. 상처 치료 과정 중 PDRN 처리에 의한 효능을 평가하기 위해 sprague-dawley rats (SD)의 배부에 bioxy punch를 이용한 4개의 창상을 유발하고, 시료를 포함한 총 5종의 실험시료를 마리당 $500{\mu}L$씩 도포한 후 7일 간격으로 4주간 피부조직 변화를 관찰하였다. 상처에 PDRN을 도포한 후, 절개된 상처의 표피화와 수축이 더 빨라졌고, 창상면적에 있어서 PDRN의 도포는 양성대조군인 $Fucidin^{(R)}$ 도포군과 비교하여 유의하게 줄어들었다. 염색한 조직의 현미경 관찰 결과에서는 양성대조군이 가장 빠르게 재상피화가 이루어졌으며, 그 다음으로는 PH군, PD군, HA군으로 교원질 재합성 및 형성 수준을 보였다. 또한, 병변의 형질전환성장인자($TGF-{\beta}$) 및 혈관 내피성장인자(VEGF) 등의 성장인자에서도 염색 조직의 결과와 유사하게 나타났다. 이러한 결과를 종합하여 볼때, 저분자화된 PDRN은 창상에 치료효과가 있다고 판단되며, 화장품 및 의료산업 분야의 기능성 소재로 활용 가능할 것으로 판단되어 진다.

RNA interference provides an effective tool for developing antitumor therapies. Cell-penetrating peptides (CPPs) are delivery vectors widely used to efficiently transport small-interfering RNA (siRNA) to intracellular targets. In this study, we investigated the efficacy of the cancer-specific CPP carrier BR2 to specifically transport siRNA to cancer-target cells. Our results showed that BR2 formed a complex with anti-vascular endothelial growth factor siRNA (siVEGF) that exhibited the appropriate size and surface charge for in vivo treatment. Additionally, the BR2-VEGF siRNA complex exhibited significant serum stability and high levels of gene-silencing effects in vitro. Moreover, the transfection efficiency of the complex into a cancer cell line was higher than that observed in non-cancer cell lines, resulting in downregulated intracellular VEGF levels in HeLa cells and comprehensively improved antitumor efficacy in the absence of significant toxicity. These results indicated that BR2 has significant potential for the safe, efficient, and specific delivery of siRNA for diverse applications.
Objectives : This study was carried out to know the effects of Gwanjul8-bang(here in after reffered to GJ8) on the inhibition of arthritis induced by collagen on DBA/1J mice. Methods : For this purpose, GJ8 was orally administered to mouse with arthritis induced by collagen II. Cytotoxicity, hepatotoxicity, arthritis index, value of immunocyte in draining lymph node(DLN) and paw joint, cytokine in serum were measured in vivo. Results : 1. The arthritis index was decreased significantly. 2. In total cell counts of DLN and paw joint, the cells in DLN increased significantly while there was a significant decrease in paw joint. 3. In lymph nodes, $CD19^+$, $CD3^+$, $CD4^+/CD25^+$ cells increased significantly, and $B220^+/CD23^+$ cells decreased significantly. 4. In joints, $CD3^+$, $CD4^+$, $CD11b^+/Gr-1^+$ cells decreased significantly. 5. The levels of $TNF-{\alpha}$, IL-6, IL-17, MCP-1 and vascular endothelial growth factor(VEGF) in serum was significantly decreased compared with control. 6. Anti-collagen II in serum was significantly decreased compared with control. 7. The degree of arthritis induced damage of joint of GJ8 group is slight compared with control group in histopathologic observation(hematoxylin & eosin(H&E), Masson-Trichrome(MT) staining). Conclusions : Comparison of the results for this study showed that GJ8 had immunomodulatory effects. So we expect that GJ8 should be used as a effective drugs for not only rheumatoid arthritis but also another auto-immune disease. Therefore there seems to be many clinical research needed in the future.

① 이용자는 회원가입 신청 또는 회원정보 변경 시 실명으로 모든 사항을 사실에 근거하여 작성하여야 하며,
허위 또는 타인의 정보를 등록할 경우 일체의 권리를 주장할 수 없습니다.
② 당 사이트가 관계법령 및 개인정보 보호정책에 의거하여 그 책임을 지는 경우를 제외하고 회원에게 부여된
ID의 비밀번호 관리소홀, 부정사용에 의하여 발생하는 모든 결과에 대한 책임은 회원에게 있습니다.

제 4 장 서비스의 이용
제 12 조 (서비스 이용 시간)

원칙으로 합니다. 단, 당 사이트는 시스템 정기점검, 증설 및 교체를 위해 당 사이트가 정한 날이나 시간에
서비스를 일시 중단할 수 있으며, 예정되어 있는 작업으로 인한 서비스 일시중단은 당 사이트 홈페이지를
통해 사전에 공지합니다.
② 당 사이트는 서비스를 특정범위로 분할하여 각 범위별로 이용가능시간을 별도로 지정할 수 있습니다. 다만
이 경우 그 내용을 공지합니다.
제 13 조 (홈페이지 저작권)
① NDSL에서 제공하는 모든 저작물의 저작권은 원저작자에게 있으며, KISTI는 복제/배포/전송권을 확보하고
있습니다.
② NDSL에서 제공하는 콘텐츠를 상업적 및 기타 영리목적으로 복제/배포/전송할 경우 사전에 KISTI의 허락을
받아야 합니다.
③ NDSL에서 제공하는 콘텐츠를 보도, 비평, 교육, 연구 등을 위하여 정당한 범위 안에서 공정한 관행에
합치되게 인용할 수 있습니다.
④ NDSL에서 제공하는 콘텐츠를 무단 복제, 전송, 배포 기타 저작권법에 위반되는 방법으로 이용할 경우
저작권법 제136조에 따라 5년 이하의 징역 또는 5천만 원 이하의 벌금에 처해질 수 있습니다.
제 14 조 (유료서비스)
① 당 사이트 및 협력기관이 정한 유료서비스(원문복사 등)는 별도로 정해진 바에 따르며, 변경사항은 시행 전에
당 사이트 홈페이지를 통하여 회원에게 공지합니다.
② 유료서비스를 이용하려는 회원은 정해진 요금체계에 따라 요금을 납부해야 합니다.
제 5 장 계약 해지 및 이용 제한
제 15 조 (계약 해지)
회원이 이용계약을 해지하고자 하는 때에는 [가입해지] 메뉴를 이용해 직접 해지해야 합니다.
제 16 조 (서비스 이용제한)
① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는
경우 서비스 이용을 제한할 수 있습니다.
- 2년 이상 서비스를 이용한 적이 없는 경우
- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.
[부 칙]
1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
