후박 및 은행물 추출물을 함유한 치약의 임상 및 미생물학적 효과에 관한 연구 (Clinical And Microbiological Study On The Effect Of Magnoliae cortex And Ginkgo Biloba Extracts Containing Dentifrice In Gingivitis)

  • Previous studies have shown that Magnoliae cortex and Ginkgo biloba extracts were showed on the antimicrobial and anti-inflammatory action, in vitro. The purpose of this study was to evaluate on the effect of antimicrobial and anti-inflammatory activity of Magnoliae cortex and Ginkgo biloba extracts containing dentifrice in gingivitis. 70 subjects with gingivitis were divided into an experimental group which performed normal oral hygiene procedure with Magnoliae cortex and Ginkgo biloba extracts containing dentifrice and a control group which also performed normal oral hygiene procedure with the same dentifrice without the natural extracts and completed a doubleblind, cross-over study. At baseline and 3 weeks, subjects were assayed for clinical study by plaque index, gingival index, pocket depth, GCF rate, and microbiological study by subgingival dental plaque bacterial morphotypes by phase contrast microscopy, total anaerobes, total aerobes, Black pigmented bacteroides, A.actionomycetemcomitans, A.viscosus, C.rectus, Ssenguis; P.gingivalis, P.intennedia by bacterial culture and immunofluorescence microscopy. After 3 weeks using their respective dentifrices, reductions in the clinical indices of subjects were similar between the experimental dentifrice group and a control dentifrice group except for statistically significant much reductions in PI, GI, and GCF rate in the experimental dentifrice group as compared to control dentifrice group. Also statistically significant reductions in the motile rods and Spirochetes were found in both experimental group to compare with control group, however statistically much reduction in total anaerobes, Black pigmented bacteroides, and P.gingivalis, P.intennedia were found in the experimental dentifrice group as compared to control dentifrice group. This results indicates that Magnoliae cortex and Ginkgo biloba extracts containing dentifrice might be useful for elimination of gingival inflammation.

  • In order to investigate the anti-stress effect of Hyangbujapalmultnag in immobilization stress rats, the change of body weight, and, the humoral and cellular immune response were measured.The following results were obtained.1. The decrease of the body weight was significantly inhibited in test group, comparing with the control group.2 In the plaque fomation test, the number of the plaque in the control group was decreased but, the decreased in test group was significantly inhibited, comparing with the control group.3. In the hemagglutination titer, the control group was decreased on the serum antibody titer, but, the decreased in test group was significantly inhibited, comparing with the control group.4. In the footpad swelling response, the differences between the control and test group were not shown.

Oleanolic acid(OA)의 Streptococcus mutans에 대한 저해효과 (Identification of Inhibitory Effect on Streptococcus mutans by Oleanolic Acid)

  • 구강에 상주하는 미생물 중, Streptococcus mutans는 충치 및 치주염의 원인인 치아플라그를 형성하는데 중요한 역할을 한다. Phytochemical은 식물에서 추출된 화학성분으로서, 사람의 건강에 유익한 영양물질로서 많은 연구들이 진행되어왔다. 본 연구는 이 phytochemical이 중요 구강미생물인 S. mutans에 대한 효과를 살펴보았다. 최근에 Thomson seedless raisin에서 여러 phytochemicals가 추출되었는데, 그 중 oleanolic acid (OA)와 5-(hydroxymethyl)-2-furfural (HF)의 임상적으로 중요한 여러 미생물에 대한 항균활성효과를 확인한 결과, OA가 그람음성균들에게는 항균활성효과가 나타나지 않았고, 그람양성균들에만 항균활성효과를 보였다. 그러나, HF의 경우에는 모든 균주에 대해 항균활성을 나타내지 않았다. 또한, OA는 S. mutans 균주의 표면부착과 생균막의 형성을 저해하기도 하였다. 따라서, 이 연구결과들은 OA가 치아에 존재하는 S. mutans의 생육 및 여러 생리적 특성들을 저해하므로 항플라그제나 항충치약으로서의 활용가능성을 확인할 수 있었다.

  • 잔나비걸상 Elfvingia applananta 자실체의 수용성성분 EA의 varicella zoster virus(Oka strain; VZV/oka)에 대한 항바이러스효과를 plaque reduction assay에 따라 실험한 결과 EA는 용량의존적으로 plaque 형성을 억제하였으며 $EC_{50}$$464.14\;{\mu}g/ml$이었다. EA와 단백질성 항바이러스제인 interferon(IFN) alpha 및 IFN gamma와의 병용시험 결과, 단독처리시와 병용처리시에 m(slope) value가 서로 유사한 값을 보였으므로 이들은 상호 배타적인 약물(mutually exclusive drug)임을 알 수 있었고, IFN alpha와의 병용시 combination index(CI)는 f(a)가 0.50에서 0.90인 유효농도 범위내에서 $0.83{\sim}1.09$의 값을 나타내어 부분적 상승 또는 상가효과를 보였으며, IFN gamma와의 병용시에는 $1.20{\sim}1.24$를 나타내어 길항효과를 보였으므로, IFN alpha와의 병용이 IFN gamma와의 병용보다 더 우수한 병용효과를 나타내었다.

서양산 고추냉이 추출물이 함유된 세치제의 효능에 대한 임상적 실험연구 (A CONTROL OF PLAQUE, GINGIVITIS AND ANTI-CARIES EFFECT OF DENTIFRICE CONTAINING HORSERADISH EXTRACTS)

  • 식물성 자연추출물 중 하나인 고추냉이 추출물을 배합한 세치제의 구강환경 개선 효과를 알아보기위해 총72명을 대상으로 교차실험 및 이중맹검법을 실시하여 치면세균막지수, 치은염지수 및 우식활성도검사 등을 비교 검토한 결과 다음과 같은 결론을 얻었다. 1. 치면세균막지수는 초기값에 비해 대조군은 28%, 고추냉이 추출물 배합세치군은 58%의 억제효과를 나타내었다. 2. 치은염지수는 초기값에 비해 대조군은 26%, 고추냉이 추출물 배합세치군은 40%의 억제효과를 나타내었다. 3. Dentocult-SM을 이용한 우식활성도 평가에서는 초기값에 비해 대조군은 4%, 고추냉이 추출물 배합세치군은 69%의 억제효과를 나타내어 고추냉이 추출물 배합세치군이 대조군에 비해 유의할만한 Streptococcus mutans 억제효과를 나타내었다. 이상의 결과들로 보아 고추냉이 추출물을 배합한 세치제는 치면세균막 억제와 치은염의 감소 그리고 우식활성도 억제에 중요한 역할을 하여 구강환경 개선에 뛰어난 효과를 보이는 것으로 사료된다.

전갈(全蝎)이 노령(老齡)에 따른 mouse의 면역(免疫) 기능(機能)에 미치는 영향(影響) (Effects of Buthus martensi Karsch on immune response in mice of different ages)

  • To clarify the activating effects of Buthus martensi Karsch on immunological function, its effect on primary and secondary antibodies production in mice of various ages was investigated. Buthus martensi Karsch increased the number of both antibody producing cells(anti-IgM and anti-IgG producing plaque forming cells, PFC) and phagocytic activity of peritoneal macrophage. Futhermore, these phenomena were significantly increased with aging in mice. Buthus martensi Karsch also increased natural killer cell activity concerning to cancer immunology. These results suggest that Buthus martensi Karsch markedly increases the reduced activity in the elderly and activates the immune response in senescence mice.

  • To clarify the activating effects of Scolopendrae corpus on immunological function, its effect on primary and secondary antibodies production in mice of various ages was investigated. 동물성 소염진통제 (II) - 반묘 및 먹가래 다당체분획의 소염.진통 및 면역조절작용 - (Anti-inflammatory Agents from Animals(II) - Anti-inflammatory, Analgesic and Immunoregulatory Activities of Mylabris sidae and Epicauta gorhami Polysaccharide Fractions -)

  • Effect of Mylabris sidae(MS) and Epicauta gorhami(EG) polysaccharide fractions on the inflammation and immune responses were studied in vivo. MS and EG contained cantharidin about 0.61 and 0.65%, respectively. It was shown that MS and EG polysaccharide fractions at a oral dose of 100 mg/kg have the significant anti-inflammatory and analgesic activity; They inhibited significantly the carrageenin-induced inflammation and acetic acid-induced writhing syndrome. They accelerated significantly the carbon clearance and the phagocytosis of colloidal carbons by Kupffer cells in liver, but they at a oral dose of 100 mg/kg suppressed significantly the Arthus reaction in the sheep red blood cell(S-RBC)-sensitized mice in accordance with the inhibition of haemaglutinin titer, haemolysin titer and plaque-forming cells. On the other hand, they at a oral dose of 200 mg/kg accelerated slightly the oxazolone-induced dermatitis in rats and delayed hypersensitivity in the S-RBC-challenzed mice in consistent with the increase of rosette forming cells. As the above results, it exhibited that MS and EG polysaccharide fractions inhibited the humoral immune responses, but they accelerated the function of macrophages and cellular immune responses. EG polysaccharide fraction had more active than MS polysaccharide fraction.

Effect of scaling and root planing on the expression of anti-inflammatory cytokines (IL-4, IL-9, IL-10, and IL-13) in the gingival crevicular fluid of electronic cigarette users and non-smokers with moderate chronic periodontitis

  • Purpose: The aim of this cross-sectional study was to investigate the effect of scaling and root planing (SRP) on the expression of anti-inflammatory cytokines (interleukin [IL]-4, IL-9, IL-10, and IL-13) in the gingival crevicular fluid (GCF) of electronic cigarette users and non-smokers with moderate chronic periodontitis (CP). Methods: Electronic cigarette users and non-smokers with CP were included in the study. Full-mouth plaque and gingival indices, probing depth (PD), clinical attachment loss (CAL), and marginal bone loss (MBL) were assessed. The GCF was collected, and its volume and levels of IL-4, IL-9, IL-10, and IL-13 were assessed. These parameters were evaluated at baseline and 3 months after SRP. The sample size was estimated, and comparisons between groups were performed. P<0.05 was considered to indicate statistical significance. Results: Thirty-six electronic cigarette users (47.7±5.8 years old) and 35 non-smokers (46.5±3.4 years old) with CP were included. At baseline, there were no differences in plaque index (PI), PD, CAL, MBL, and GCF IL-4, IL-9, IL-10, and IL-13 between electronic cigarette users and nonsmokers. At the 3-month follow-up, there were no significant differences in PI, gingival index (GI), PD, CAL, and MBL in electronic cigarette users compared to baseline, while there were significant reductions in PI, GI, and PD among non-smokers. At the 3-month follow-up, GCF IL-4, IL-9, IL-10, and IL-13 levels were significantly elevated in both groups (P<0.05) compared to baseline. The increases in GCF IL-4, IL-9, IL-10, and IL-13 levels were significantly higher in non-smokers (P<0.05) than in electronic cigarette users at the 3-month follow-up. Conclusions: Levels of GCF IL-4, IL-9, IL-10, and IL-13 increased after SRP in electronic cigarette users and non-smokers with CP; however, the anti-inflammatory effect of SRP was more profound in non-smokers than in electronic cigarette users.

Antimicrobial and Anti-Biofilm Activities of the Methanol Extracts of Medicinal Plants against Dental Pathogens Streptococcus mutans and Candida albicans

  • Several medicinal plants are ethnomedically used in Korea as agents for treating infection, anti-inflammation, and pain relief. However, beyond typical inhibitory effects on cell growth, little is known about the potential anti-biofilm activity of these herbs, which may help to prevent cavities and maintain good oral health. This study aimed to investigate the antimicrobial and anti-biofilm activities of the methanol extracts of 37 Korean medicinal plants against dental pathogens Streptococcus mutans and Candida albicans, which synergize their virulence so as to induce the formation of plaque biofilms in the oral cavity. The antimicrobial activities were investigated by broth dilution and disk diffusion assay. The anti-biofilm and antioxidant activities were evaluated based on the inhibitory effect against glucosyltransferase (GTase) and the DPPH assay, respectively. Among 37 herbs, eight plant extracts presented growth and biofilm inhibitory activities against both etiologic bacteria. Among them, the methanol extracts (1.0 mg/ml) from Camellia japonica and Thuja orientalis significantly inhibited the growth of both bacteria by over 76% and over 83% in liquid media, respectively. Minimum inhibitory concentration (MIC) values of these methanol extracts were determined to be 0.5 mg/ml using a disk diffusion assay on solid agar media. Biofilm formation was inhibited by more than 92.4% and 98.0%, respectively, using the same concentration of each extract. The present results demonstrate that the medicinal plants C. japonica and T. orientalis are potentially useful as antimicrobial and anti-biofilm agents in preventing dental diseases.
