Inhibitory Effect of 3-(4-Hydroxyphenyl)-1-(thiophen-2-yl) prop-2-en-1-one, a Chalcone Derivative on MCP-1 Expression in Macrophages via Inhibition of ROS and Akt Signaling

  • Chalcones (1,3-diaryl-2-propen-1-ones), a subfamily of flavonoid, are widely known to possess potent anti-inflammatory and anti-oxidant properties. In this study, we investigated the effect of 3-(4-Hydroxyphenyl)-1-(thio3-(4-Hydroxyphenyl phen-2-yl)prop-2-en-1-one (TI-I-175), a synthetic chalcone derivative, on endotoxin-induced expression of monocyte chemoattractant protein-1 (MCP-1), one of the key chemokines that regulates migration and infiltration of immune cells, and its potential mechanisms. TI-I-175 potently inhibited MCP-1 mRNA expression stimulated by lipopolysaccharide (LPS) in RAW 264.7 macrophages without significant effect on cell viability. Treatment of cells with TI-I-175 markedly prevented LPS-induced transcriptional activation of activator protein-1 (AP-1) as measured by luciferase reporter assay, while nuclear factor-${\kappa}B$ (NF-${\kappa}B$) activity was not inhibited by TI-I-175, implying that TI-I-175 suppressed MCP-1 expression probably via regulation of AP-1. In addition, TI-I-175 treatment significantly inhibited LPS-induced Akt phosphorylation and led to a significant decrease in reactive oxygen species (ROS) production by LPS, which act as up-stream signaling events required for AP-1 activation in RAW 264.7 macrophages. Taken together, these results indicate that TI-I-175 suppresses MCP-1 gene expression in LPS-stimulated RAW 264.7 macrophages via suppression of ROS production and Akt activation.

PEGylated Erythropoietin Protects against Brain Injury in the MCAO-Induced Stroke Model by Blocking NF-κB Activation

  • Cerebral ischemia exhibits a multiplicity of pathophysiological mechanisms. During ischemic stroke, the reactive oxygen species (ROS) concentration rises to a peak during reperfusion, possibly underlying neuronal death. Recombinant human erythropoietin (EPO) supplementation is one method of treating neurodegenerative disease by reducing the generation of ROS. We investigated the therapeutic effect of PEGylated EPO (P-EPO) on ischemic stroke. Mice were administered P-EPO (5,000 U/kg) via intravenous injection, and middle cerebral artery occlusion (MCAO) followed by reperfusion was performed to induce in vivo ischemic stroke. P-EPO ameliorated MCAO-induced neurological deficit and reduced behavioral disorder and the infarct area. Moreover, lipid peroxidation, expression of inflammatory proteins (cyclooxygenase-2 and inducible nitric oxide synthase), and cytokine levels in blood were reduced by the P-EPO treatment. In addition, higher activation of nuclear factor kappa B (NF-κB) was found in the brain after MCAO, but NF-κB activation was reduced in the P-EPO-injected group. Treatment with the NF-κB inhibitor PS-1145 (5 mg/kg) abolished the P-EPO-induced reduction of infarct volume, neuronal death, neuroinflammation, and oxidative stress. Moreover, P-EPO was more effective than EPO (5,000 U/kg) and similar to a tissue plasminogen activator (10 mg/kg). An in vitro study revealed that P-EPO (25, 50, and 100 U/mL) treatment protected against rotenone (100 nM)-induced neuronal loss, neuroinflammation, oxidative stress, and NF-κB activity. These results indicate that the administration of P-EPO exerted neuroprotective effects on cerebral ischemia damage through anti-oxidant and anti-inflammatory properties by inhibiting NF-κB activation.

Nrf-2 Mediated Antioxidative Effect of Korean and Chinese Safflower Seeds (한국산 중국산 홍화자의 Nrf-2 매개 항산화 효과)

  • Safflower (Carthamus tinctorius L.) seeds have been used in Korea and China for promoting bone formation and protection. This study was designed to examine the Nrf-2 mediated anti-oxidative effects of Korean and Chinese safflower seeds. Water and ethanol extracts of safflower seeds were treated to RAW 264.7 cells. Nrf-2 transcriptional activity was measured by reporter gene assay and western blot analysis. Semi-quantitive RT-PCR analysis was adopted to measure Nrf-2 dependent gene expressions. Water extracts of safflower seeds have strongly induced the activation of Nrf-2 transcription than ethanol extracts. Especially, water extracts of Korean safflower seeds has more strongly increased the expression of nuclear Nrf-2. Water extracts of Korea and China safflower seeds have also increased the expression of Nrf-2-dependent genes such as GCLC, NQO-1 and HO-1 in RAW 264.7 cells. However, all kinds of safflower seeds extracts did not increase intracellular ROS production. These results demonstrate that the antioxidant effects of safflower seeds are not related with ROS production, rather it is mediated by the direct activation of Nrf-2.

Protective effects of Acanthopanax divaricatus extract in mouse models of Alzheimer's disease

  • BACKGROUND: Acanthopanax divaricatus var. albeofructus (ADA) extract has been reported to have anti-oxidant, immunomodulatory, and anti-mutagenic activity. MATERIALS/METHODS: We investigated the effects of ADA extract on two mouse models of Alzheimer's disease (AD); intracerebroventricular injection of ${\beta}$-amyloid peptide ($A{\beta}$) and amyloid precursor protein/presenilin 1 (APP/PS1)-transgenic mice. RESULTS: Intra-gastric administration of ADA stem extract (0.25 g/kg, every 12 hrs started from one day prior to injection of $A{\beta}1$-42 until evaluation) effectively blocked $A{\beta}1$-42-induced impairment in passive avoidance performance, and $A{\beta}1$-42-induced increase in immunoreactivities of glial fibrillary acidic protein and interleukin (IL)-$1{\alpha}$ in the hippocampus. In addition, it alleviated the $A{\beta}1$-42-induced decrease in acetylcholine and increase in malondialdehyde levels in the cortex. In APP/PS1-transgenic mice, chronic oral administration of ADA stem extract (0.1 or 0.5 g/kg/day for six months from the age of six to 12 months) resulted in significantly enhanced performance of the novel-object recognition task, and reduced amyloid deposition and IL-$1{\beta}$ in the brain. CONCLUSIONS: The results of this study suggest that ADA stem extract may be useful for prevention and treatment of AD.

Protective Effect of Vitis amurensis Stems and Leaves Extract on Hydrogen Peroxide-induced Oxidative Neuronal Cell Damage in Cultured Neurons (과산화수소수로 유도된 배양 뇌신경세포손상에 대한 왕머루 잎과 줄기 추출물의 보호효과)

  • Vitis amurensis (VA; Vitaceae) has long been used in oriental herbal medicine. It has been reported that roots and seeds of VA have anti-inflammatory and antioxidant effects. In the present study, the protective effect of ethanol extract from stems and leaves of VA on hydrogen peroxide (${H_2}{O_2}$) (100 ${\mu}M$)-induced neuronal cell damage was examined in primary cultured rat cortical neurons. VA (10-100 ${\mu}g$/ml) concentration-dependently inhibited ${H_2}{O_2}$-induced apoptotic neuronal cell death measured by 3-[4,5-Dimethylthiazol-2-yl]-2,5-diphenyl-tetrazolium bromide (MTT) assay and Hoechst 33342 staining. VA inhibited ${H_2}{O_2}$-induced elevation of intracellular $Ca^{2+}$ concentration (${[Ca^{2+}]}_i$) and generation of reactive oxygen species (ROS), which were measured by fluorescent dyes. Pretreatment of VA also prevented glutamate release into medium induced by 100 ${\mu}M$ ${H_2}{O_2}$, which was measured by HPLC. These results suggest that VA showed a neuroprotective effect on ${H_2}{O_2}$-induced neuronal cell death by interfering with ${H_2}{O_2}$-induced elevation of ${[Ca^{2+}]}_i$, glutamate release, and ROS generation. This has a significant meaning of finding a new pharmacological activity of stems and leaves of VA in the CNS.

Investigation of the effect of Lithospermi Radix on tight-junction related genes in HaCaT cells (자초(紫草) 열수추출물이 각질형성세포 HaCaT의 세포 연접 관련 유전자의 발현에 미치는 영향 연구)

  • Objectives : The aim of this research was to determine the diverse effects of Lithospermi Radix Water Extract (LR) on human keratinocyte HaCaT cells, and to examine whether those effects could be applied to the human skin. Methods : We examined effect of LR on the cell viability of using the MTS assay in human keratinocyte HaCaT cells. The antioxidation effect of LR was analyzed relative to the well-known antioxidant resveratrol, using an ABTS assay. Quantitative RT-PCR analysis revealed that, in HaCaT cells, LR influenced the mRNA expression of tight-junction genes associated with skin moisturization. Furthermore, a wound-healing assay demonstrated altered cell migration in LR-treated HaCaT cells. Result : The cytotoxicity was confirmed to be higher in LR at a concentration of $800{\mu}g/m{\ell}$ using the MTS assay in HaCaT cells. In comparison to $100{\mu}M$ resveratrol, $1,600{\mu}g/m{\ell}$ LR showed either a similar or superior antioxidation effect. LR treatment in HaCaT cells reduced the mRNA expression levels of claudin 3, claudin 4, claudin 6, claudin 8, and ZO-2 to less than 0.80-fold, whereas JAM-A and Tricellulin mRNA expression level increased more than 1.33-fold. In addition, HaCaT cells migration was decreased to 83.9% by LR treatment. Conclusions : LR of antioxidation activity will have an anti-aging effect on the skin by reducing oxidative stress. Further studies are required to address the implications for human skin, given LR's effects of altering mRNA expression of tight junction-related gene and decreasing cell migration of HaCaT cells.

Whitening and anti-wrinkle effect of Pinus koraiensis leaves extracts according to the drying technique (건조방법에 따른 잣나무(Pinus koraiensis) 잎 추출물의 주름개선 및 미백 효과)

  • This study provide activity for beauty food of water and 80 % ethanol extracts from Pinus koraiensis leaves. Total phenolic content of extracts from Pinus koraiensis leaves were each 12.22 mg/g (Drying under hot air) and 17.93 mg/g (Drying under shade), 14.36 mg/g (Lyophilization) in water extracts (WE) and 11.9 mg/g and 20.63 mg/g, 17.96 mg/g in 80 % ethanol extracts (EE). The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of extracts from Pinus koraiensis leaves was 96.20 % in EE from drying under shade at extracts concentration. The 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical decolorization activity of extracts from drying under shade was 99.85 % in WE and 99.80 % in EE at extracts concentration. The antioxidant protection factor (PF) extracts from drying under shade type was 9.63 PF in WE and 10.48 PF in EE at extracts concentration. The thiobarbituric acid reactive substance from Pinus koraiensis leaf was 89.39 % in EE from drying under shade at extracts concentration. The elastase inhibition activity of EE for anti-wrinkle effect showed an excellent wrinkle improvement effect, showing 71.46 % in EE from lyophilization. Collagenase inhibition activity of EE from drying under shade was 97.48 % in extracts. Tyrosinase inhibition activity which was related to anti-melanogensis was observed. The tyrosinase inhibitory effect of extracts from lyophilization was confirmed to be 60.4 % in EE more than another drying methods at extracts concentration. Through out all results, it can be expected Pinus koraiensis leaves extracts to use as a functional material for anti-oxidant and functional beauty food.

Evaluation of Anti-oxidant, Anti-microbial and Anti-thrombosis Activities of Fruit, Seed and Pomace of Schizandra chinensis Baillon (오미자 열매, 씨, 착즙 후 박의 항산화, 항균 및 항혈전 활성 평가)

  • In this study, for the efficient use of the byproduct of the omija (Schizandra chinensis Baillon: SC) processing industry, the ethanol extracts of the fruit (F), seed (S), and pomace (P) of SC were prepared, and their useful bioactivities were evaluated. For F-SC, S-SC, and P-SC, the extraction yields were 28.3%, 22.1%, and 7.2%, respectively, and the polyphenol contents were 8.81, 37.22, and 9.20 mg/g, respectively. The total flavonoid content in P-SC (4.31 mg/g) was 3.5-fold higher than that in F-SC (0.76 mg/g). In an antioxidation activity assay, P-SC showed stronger radical scavenging activities against DPPH anion, ABTS cation, and nitrite and stronger reducing power activities than the other extracts. The calculated concentration required for 50% radical scavenging activity, $RC_{50}s$, of P-SC for DPPH anion, ABTS cation, and nitrite was 226.2, 192.5, and $92.5{\mu}g/ml$, respectively. In an antimicrobial activity assay, F-SC, S-SC, and P-SC showed similarly strong growth inhibitions against Bacillus subtilis and P. vulgaris at a concentration of 0.5 mg/disc. F-SC and P-SC showed 15-fold extended time in thrombin, prothrombin, and activated partial thromboplastin time assays at a concentration of 5 mg/ml. The anticoagulation activity of P-SC (2.5 mg/ml) was comparable to that of aspirin (1.5 mg/ml). Furthermore, F-SC and S-SC showed very good platelet aggregation inhibitory activities. F-SC, S-SC, and P-SC did not show significant hemolysis against human red blood cell up to a concentration of 0.5 mg/ml. These results suggest that S-SC and P-SC, both of which are byproducts of the omija processing industry, show strong potential as novel antioxidant, antimicrobial, and antithrombosis agents.

A Study on the Stability Test for the Cream Containing Suaeda Asparagoides Extract (나문재 추출물 함유 크림의 안정성 평가에 관한 연구)

  • In the previous study, the anti-oxidant activity of extract/fraction of Sueada asparagoides (SA) was investigated and the results showed that the ethylacetate (EtOAc) fraction and its aglycone fraction had the best performance on the free radical scavenging activity, reactive oxygen species scavenging (ROS) activity and cell protective activity (J. Soc. Cosme. Scientists Korea, 33(3), 145 (2007)). In this study, the stability of cream containing 0.3% SA EtOAc extract (called extract below) was evaluated. pH, viscosity and absorbance (363 nm) were measured under the 4 different temperatures ($0^{\circ}C,\;25{\circ}C,\;37{\circ}C\;and\;45{\circ}C$) and under the sun light at the 4 week intervals during the 12 weeks in total. The control cream without containing the extract did not show pH change under the different temperatures mentioned above. However, the pH of the cream the extract was decreased 0.08 at the temperature ranges of $0^{\circ}C\;to\;37^{\circ}C$. Under the $45^{\circ}C$ and sun light condition, the pH was decreased 0.51 and 0.66, respectively. The cream containing the extract did not show absorbance change at the temperature ranges of 0 to $37^{\circ}C$ for 12 weeks. Instead, the absorbance of the cream treated under $45^{\circ}C$ and sun light condition was decreased 7.6 % and 7.4 %, respectively. This decrease in absorbance is relatively small compared to the 48.3 % decrease of the extract sampled from the cream using ethanol solution. This indicates that the extract is stabilized in the cream. After treating the cream for 12 weeks under the different temperatures, the viscosity was measured for the cream containing the extract and control cream. The values were increased by 1,748 cPs in average compared to the initial value for the former and by 951 cPs in average for the latter. On the other hand, the viscosity of control cream treated under the sun light for 12 weeks was significantly decreased (4,022 cPs) relative to the cream containing the extract, which showed 2,484 cPs increase in viscosity. This indicates that the SA extract contributes to the stability of the emulsion product by protective effect to maintain the viscosity of the cream against sun light. In addition, any change in color or smell was not observed through 12 weeks of the experimental time period. Thus, it is concluded that it is still not clear in the stability of the cream containing the extract when it is stored for the long time. Accordingly, it is suggested that further study is needed to provide more information to the manufactures, who are seeking for the application of the extract to improve the anti-oxidant activity and stability of cosmetic products.

Effect of Temperature and Gas Permeability of Functional Packing Films on Storability of Fresh-cut Salicornia herbacea Classified by Size (저장온도와 기능성 필름의 기체 투과도가 다양한 크기의 퉁퉁마디 신선편이의 저장성에 미치는 영향)

  • These studies were identified the effect of four types of non-perforated breathable (NPB) packing film and three sizes on storage ability of fresh-cut for ready to eat packaging at $5^{\circ}C$ for 25 days and $10^{\circ}C$ for 15 days storage in Salicornia herbacea. The fresh weight loss was less than 2% in every films at $5^{\circ}C$ condition after 25 days storage, and the $10^{\circ}C$ also had same result on 15 days storage except 100,000 cc NPB film. Compare with storage after 15 days, storage condition at $5^{\circ}C$ had shown better result under the 1% fresh weight loss rate. The 5,000 cc and $5^{\circ}C$ condition had been shown the characteristics of MA packaging in carbon dioxide and oxygen concentrations. The ethylene concentration in vacuum film was higher 10 to 100 times than in NPB film treatments during storage. But ethylene concentration was not statistically significant differences among size treatments. Every conditions had been measured the anti-oxidant activity by DPPH method after storage at $5^{\circ}C$ for 25 days and $10^{\circ}C$ for 15 days. S. herbacea at $5^{\circ}C$ had been more than twice of activity compare with that at $10^{\circ}C$. 100,000 cc NPB film had been higher contents of anti-oxidant activity at $5^{\circ}C$ and $10^{\circ}C$. As the fresh-cut sizes, 3 cm and 5 cm sizes had changed depending on film types but 10 cm were not effected by the film types in the DPPH activity. When panel test had been tried to measure the visual quality and off-flavor after storage, $5^{\circ}C$ with a filme of 5,000 cc treatment had established higher value than other treated conditions. As these results, it's may be suggested that the $5^{\circ}C$ with 5,000 cc non-perforated breathable film for MA storage in Salicornia herbacea at fresh cut distribution system. Fresh cut size 10 cm with 100,000 cc NPB film also had the good quality for 15 dyas storage at $10^{\circ}C$, and this result can be applied for short term distribution system in Korea.
