Comparison of nutrition, anti-nutritional factors of rice straw and microbial composition in soil according to GM and non-GM rice field

  • The study was conducted to evaluate differences of growth characteristics of rice cultivated in two different regions (Cheonan and Jeonju). It focused on nutritional composition and anti-nutritional factors of rice straw produced from 21 rice varieties including GM rice (Iksan 483). The range of general nutrition ingredient is that crude was 0.97 ~ 3.2 %, carbohydrate was 67.45 ~ 80.01 %, crude protein was 1.46 ~ 4.81 %, crude ash was 6.52 ~ 18.96 %, crude fiber was 25.77 ~ 40.02 %, NDF was 51.84 ~ 67.77 %, ADF was 27.11 ~ 40.44 %, calcium was 0.49 ~ 5.18 mg/g and phosphorous was 0.26 ~ 2.77 mg/g. The general nutritional contents of GM rice were included above range. The range of phytic acid of rice straws cultivated in Cheonan and Jeonju was 0 ~ 0.056 mg/ml and 0 ~ 0.059 mg/ml, respectively. The phytic acid content of GM was 0.033 mg/ml, which was in the range of the content of rice straw in Cheonan and Jeonju. The range of trypsin inhibitor of rice straws cultivated in Cheonan and Jeonju was 0.061 ~ 0.461 TIU/mg and 0 ~ 1.278 TIU/mg, respectively. The trypsin acid content of GM was 0.461 TIU/mg, which was in the range of the content of rice straw in Cheonan and Jeonju. In addition, we investigated microbial community from each soil sample by using metagenomics sequencing based on rRNA microbial diversity in order to inspect indirect changes of soil environment with cultivation of GM rice. Metagenomics analysis was carried out using soil samples cultivated with GM and non-GM rice for before transplanting, young panicle differentiation stage, heading stage, and ripening stage. Beta diversity of microbial community in both soil environments were calculated by using Bray-Curtis distance method and showed low value with an average of 0.24 (dissimilarity = 1). As a result, it was confirmed that the cultivation of GM does not give a significant effect on the change of microbial composition in soil. Therefore, Our study demonstrates that there is no difference in the composition of soil microorganism due to GM and non-GM rice.

  • It is inevitable to use chemical germicidal agents like paraben, imidazolidinyl urea and phenoxyethanol to preserve the emulsions which is usually used in cosmetics. Although these chemical preservatives are good enough to reduce the microbiological contamination, they are irritative, allergenic to the skin. Several kinds of polyols are used in cosmetics as moisturizer and solvent. In this study, we evaluate the effects of polyols on anti-microbial activities, safety and resistant index. MIC(minimal inhibitory concentration) of polyols determined against 6 germs including Staphylococcus aureus. The order of MIC was PG $\cong$ DPG $\cong$ 1,3BG > HG > 1,2-PD > 1,2-HD $\cong$ 1,2-OD. The $2{\sim}3\;wt%$ of 1,2-HD(hexanediol) shows good anti-microbial effects in emulsions without allergenic response. Resistant index of 1,2-HD was less than 2 and this value was smaller than that of chemical preservatives. The mechanism of antimicrobilogical effect might be disturb the membrane of germs by investigating using electron microscope. Added to that, using this paradigm, low preservative contents, paraben-free system, and even preservative-free systems can be expected from these results.

Function of Blending Essential Oil in the Development of Anti-Dandruff Products

  • Purpose: In this paper, we show our blending ratio of 10 types of Essential Oils that survives beneficial bacteria and kills harmful bacteria in the scalp, and we investigate the possibility of application of our blending ratio to the development of anti-dandruff products and the possibility of being used as a raw material for clinical beauty and customized cosmetics. Methods: The scalp microorganisms used in our study were M. furfur, S. epidermidis, E. coli, and P. nitroreducens. There are a total of 10 Essential Oils such as True Lavender, Lime, Roman chamomile, Rosemary camphor, Cedarwood, Geranium, Clove, Tea tree, Palmalosa, and Peppermint. The antibacterial test of the blended Essential Oil was carried out according to the test method of the standardized evaluation methodology of "Food and Food Additives Code". Since M. furfur is related to the growth of sebum in the scalp, in this study we used the fnLNB and the fnLNA with 20 ㎖ of whole fat cow milk added. Results: The blending ratio of EO, which inhibits dandruff-causing bacteria such as M. furfur, S. epidermidis, E. coli, and does not inhibit P. nitroreducens showing dominant growth in a healthy scalp, was B8(Clove 0.2%, Roman chamomile 0.5%, Tea tree 0.3%), B9(Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.3%), B10(Clove 0.1%, Geranium 0.1%, Palmarosa 0.1%, Roman chamomile 0.5%, Tea tree 0.2%). Conclusion: It is thought that the blending ratio of BEO obtained as a result of this study can provide a basis for use as an alternative to antibiotics in developing anti-dandruff drugs and emerge as a new alternative to solve scalp microbial imbalance. In order for EO to be used as a useful raw material for anti-dandruff preparation, researches on 1) Standardization (the effects of products differ according to the types, regions, climate, extraction methods, etc.), 2) Antimicrobial effects, 3) Safety, etc., must be established.

  • 피부 염증은 흉터, 노화 뿐만 아니라 아토피와 같은 질환으로 발전할 수 있어 이를 조절하는 것이 매우 중요하다. 본 연구에서는 최근 인체에서 염증을 조절하는 것으로 알려진 specialized pro-resolving mediators (SPMs)의 in vitro 합성과 화장품 적용 가능성을 확인하였다. 대두의 lipoxygenase를 이용하여 mono 또는 di-hydroxy docosahexaenoic acid가 혼합된 시료 S-SPMs를 제작하였고 효능 평가에 이용하였다. 먼저, UVB로 염증을 유도한 세포에서 TNF-α와 IL-6의 발현이 S-SPMs에 의해 감소하고, 미세먼지에 의해 유도된 nitric oxide (NO)의 생성 역시 감소하는 것을 확인하여 S-SPMs의 항염 효능을 확인하였다. 또한, S-SPMs을 처리한 조건에서 malondialdehyde (MDA) 생성이 감소하여 지질 과산화 억제능이 있음을 확인하였고 S-SPMs에 의한 matrix metalloproteinase-1 (MMP-1)의 발현 감소, procollagen type I의 함량 증가를 통해 collagen 분해를 억제하고 반대로 합성은 촉진함을 확인하였다. 끝으로 filaggrin과 loricrin의 발현이 S-SPMs에 의해 증가한 것을 확인하여 피부 장벽 강화 효능을 확인하였다. 위 결과를 토대로 S-SPMs은 피부의 염증 억제와 함께 손상회복, 주름개선 및 장벽 강화를 위한 소재로 활용 가능함을 확인하였다.

Anti-bacterial Effect of Oenothera lamarckiana Aerial Part Extract

  • Ingestion of food contaminated with microorganism, if not always, could lead to severe health problem. Preservatives has been added to food to prevent food from being contaminated with microorganism. But, these have potential to threaten the health. Therefore, much effort has been taken to find the safe materials showing the anti-microbial activity. In this study, we investigated the anti-bacterial activity of Oenothera lamarckiana aerial part extract against eight bacteria strain. In paper disc assay, extract inhibited the growth of Staphylococcus aureus, Methicillin-resistant S. aureus, Bacillus cereus and Shigella dysenteriae at 200 μg/disc, but not against Escherichia coli, E. coli O157:H7, Salmonella Typhi and S. enteritidis. Minimum inhibitory concentration (MIC) against Staphylococcus aureus, Methicillin-resistant S. aureus, Bacillus cereus and Shigella dysenteriae is 250, 250, 500 and 500 μg/mL, respectively. Compared with reported MIC of other plant resources, O. lamarckiana aerial part extract showed the relatively high anti-bacterial activity. O. lamarckiana aerial part could be suitable for the preservative development. But, it still remains to be studied to evaluate safety and so on.

Inhibition of Cytopathic Effect of Human Immunodeficiency Virus-1 by Water-soluble Extract of Ganoderma lucidum

  • To examine components of Ganoderma lucidum for anti-human immunodeficiency virus (HIV) activity, the aqueous extracts of its basidiocarps were separated into high-molecular-weight (HMF) and low-molecular-weight (LMF) fractions. These fractions were used in XTT [2,3-bis (2-methoxy-4-nitro-5-sulfophenyl)-5-[(phenylamino) carbonyl]-2H-tetrazolium hydroxide] antiviral assay which can quantitatively measure cytopathic effects of HIV-1 on CEM, human T lymphoblastoid cell line. The CEM cell line added with serial diluted HMF or LMF was cultured in the absence or presence of HIV-1. The results showed that the LMF of the aqueous extract strongly inhibited cytopathic effect of the target cell induced by HIV-1. When two-fold serially diluted LMF ranging from $40.97{\mu}g/ml$4 to 125.00 .mu.g/ml was added to the virus-free culture system, no toxicity on the target cells was detected in all the concentrations tested. However, when it was added to the HIV-infected culture system, the viabilities of the target cell reached a plateau recovering its viabilities to 71.7% and 82.5% in experiment-1 and -2 at 15.60 .mu.g/ml, respectively. The cell viabilities were then gradually decreased but maintained at more than 50% above 31.20 .mu.g/ml concentration. On the contrary, HMF did not prevent any HIV-induced cytopathic effect at any concentrations tested on this cell line. From these results, negligible toxicities were observed by both HMF and LMF of G. luciolum, and recovery of cell viability in HIV infected target cell was induced only by LMF of the carpophores.

도라지 추출물의 구강미생물에 대한 항균효과 (Anti-microbial Activity of Platycodon Grandiflorum Extracts Against Oral Microbes)

  • Objectives: 도라지 추출액을 이용하여 구강미생물에 대한 Platycodon grandiflorum 추출물의 항균 효과를 실험하고자 한다. Methods: 항균 활성 및 최소 저해 농도는 한천 희석법을 사용하여 측정하였다. 추출물 (Platycodon grandiflorum 추출물과 껍질이 없는 Platycodon grandiflorum 추출물)을 준비하고 0.0625 mg/ml, 0.125 mg/ml, 0.25 mg/ml, 0.5 mg/ml, 1 mg/ml의 농도로 한천 플레이트를 만들었다. Results: 치아우식균보다 치은염을 유발시키는 A. naeslundii, A. odontolyticus, A. viscosus 세 균주에서 더 우수한 항균효과를 나타내었으며, 껍질이 없는 도라지 추출물 보다 사포닌 함량이 더 많은 껍질이 있는 도라지 추출물에서 우수한 항균효과를 나타낸 것을 확인하였다. Conclusions: 도라지 추출물 결과에서 보듯이 천연 추출물이 구강질환을 유발시키는 치아우식균 및 잇몸질환균에 대해 항균효과가 있을 수 있음을 시사한다.

울릉도 자생식물인 섬괴불나무(Lonicera morrowii A.Gray) 잎 추출물의 생리활성 (Physiological activities of leaf extract of Lonicera morrowii A.Gray, a plant native to Ulleungdo)

  • 본 연구에서는 섬괴불나무 잎으로부터 phenol성 화합물의 생리활성 탐색을 통하여 기능성 소재로 활용 가능성을 살펴보았다. 인체에 무해한 용매인 열수와 ethanol로 추출 했을 때 열수는 3.53 mg/g, 40% ethanol은 2.82 mg/g의 TPC를 나타내었으며, 추출물을 이용하여 항산화 활성, xanthine oxidase (XOase), angiotensin converting enzyme (ACE), α-glucosidase 억제효과 및 항균 활성을 검정하였다. 그 결과 섬괴불나무 잎의 열수와 ethanol 추출물 50-200 ㎍/mL TPC에서 농도의존적으로 DPPH 유리라디칼 소거활성 및 PF가 증가하는 경향을 보였고, 50 ㎍/mL의 저농도에서도 모두 매우 높은 활성을 나타내었다. XOase 저해효과에서는 200 ㎍/mL TPC에서 열수와 ethanol 추출물이 각각 76.35, 99.83%의 높은 저해효과 나타냈으며, ACE 저해효과 또한 200 ㎍/mL TPC에서 열수와 ethanol 추출물 각각 79.06, 87.14%의 저해효과를 확인하였다. α-Glucosidase 저해효과를 측정한 결과 200 ㎍/mL TPC에서 열수와 ethanol 추출물 각각 80.45, 63.58%의 저해효과를 보였다. 생육저해환으로 항균활성을 측정 결과 열수 추출물 200 ㎍/100 μL TPC에서 H. pylori, P. acne균에 대해 각각 11.5, 18.5 mm의 clear zone을 형성 하였고, ethanol 추출물 200 ㎍/100 μL 농도에서는 P. acne균에 대해 10 mm의 clear zone을 형성하였다. 따라서 본 연구를 통하여 섬괴불나무 잎 추출물의 항산화 활성, xanthine oxidase (XOase), angiotensin converting enzyme (ACE), α-glucosidase 억제효과 및 항균 활성이 있다고 판단되었다.

  • With the improvement of economic status and the desire for beauty, the interest in health and skin care is increasing. For these demands, since ages medicinal plants are in vogue. A variety of plants, cosmetics and foods with novel bioactive ingredients for skin care and beauty are under constant research and development. Skin is influenced by various factors such as Ultra-violet rays, stress, hormones and aging which together lead skin to lose elasticity, changes in pigmentation and wrinkle formation. Many medicinal plants have proven effects in skin care and beauty treatment. From this list of medicinal plants, one which is famous for its beauty, flavor and fragrance is Rosa damascene. Rosa damascene has many therapeutic action and postulated pharmacological studies such as anti-arthritic, anti-microbial, cardio protective, anti-inflammatory, antioxidant, analgesic, immune-modulator, gastro-protective, and skin ameliorative effect. Research in the field of Cosmetology has proven the effect of Rosa damascene in rehydrating skin, reducing scars and stretches, acne management, lowering skin pigmentation, delaying wrinkling and is recommended as a skin vitalizing agent. In this review, the morphology, chemical constituents, and some pharmacological activity are discussed.

Bioconversion Products of Whey by Lactic Acid Bacteria Exert Anti-Adipogenic Effect

  • Microbial bioconversion using lactic acid bacteria (LAB) provides several human health benefits. Although whey and whey-derived bioactive compounds can contribute to an improvement in human health, the potential anti-obesity effect of whey bioconversion by LAB has not been well studied. This study aimed to investigate whether bioconversion of whey by Pediococcus pentosaceus KI31 and Lactobacillus sakei KI36 (KI31-W and KI36-W, respectively) inhibits 3T3-L1 preadipocyte differentiation. Both KI31-W and KI36-W reduced intracellular lipid accumulation significantly, without decreasing 3T3-L1 preadipocyte proliferation. In addition, obesity-related transcription factor (peroxisome proliferator-activated receptor γ) and genes (adipocyte fatty acid-binding protein and lipoprotein lipase) were down-regulated significantly in 3T3-L1 cells in the presence of KI31-W and KI36-W. Collectively, these results suggest that bioconversion of whey by LAB exhibits anti-adipogenic activity and may be applied as a therapeutic agent for obesity.
