새로운 기능성 식품소재로 그 용도가 확대되고 있는 당유도체 감미료 Fructo-oligo 당, Maltitol, Sorbitol 및 High maltose syrup의 수분 활성도 저하능력, 유산균 발효능력, 난우충성 및 상대감미도에 대하여 조사하였다. 수분활성도 저하능력은 Sorbitol이 다른 당보다 우수하였으며 Fructo-oligo 당, Maltitol은 설탕과 유사하였다. 여러 가지 유산균에 대한 발효능력은 glucose(Hydrol), Fructo-oligo 당, HMS 모두 우수하였으나 Maltitol은 Lactobacillus plantarum을 제외한 다른 유산균들의 생육을 억제하였다. Streptococcus mutans를 이용한 우충성 시험에서 Maltitol은 다른당에 비하여 뚜렷한 난우충성을 나타내었다. 당유도체들의 상대감미도는 당도수준이 높을수록 증가하는 경향을 나타내었으며 1% 설탕용액의 당도수준에서 mole 농도기준 상대감미도는 Neosugar 0.60, Sorbitol 0.21, Maltitol 0.50, HMS 0.27로 평가되었다.

Actinomycetes isolated from marine habitats represent a promising source of bioactive substances. Here, we report on the isolation, identification, productivity enhancement and application of the bioactive compounds of Streptomyces qinglanensis H4. Eighteen marine actinomycetes were isolated and tested for resistance to seven bacterial diseases. Using 16S rRNA sequencing analysis (GenBank accession number MW563772), the most powerful isolate was identified as S. qinglanensis. Although the strain produced active compound(s) against a number of Gram-negative and Gram-positive bacteria, it failed to inhibit pathogenic fungi. The obtained inhibition zones were 22.0 ± 1.5, 20.0 ± 1, 16.0 ± 1, 12.0 ± 1, 22.0 ± 1 and 24.0 ± 1 mm against Bacillus subtilis ATCC 6633, Escherichia coli ATCC 19404, Enterococcus faecalis ATCC 29212, Pseudomonas aeruginosa ATCC 9027, Candida albicans ATCC 10231 and Staphylococcus aureus ATCC6538, respectively. To maximize bioactive compound synthesis, the Plackett-Burman design was used. The productivity increased up to 0.93-fold, when S. qinglanensis was grown in optimized medium composed of: (g/l) starch 30; KNO3 0.5; K2HPO4 0.25; MgSO4 0.25; FeSO4·7H2O, 0.01; sea water concentration (%) 100; pH 8.0, and an incubation period of 9 days. Moreover, the anticancer activity of S. qinglanensis was tested against two different cell lines: HepG2 and CACO. The inhibition activities were 42.96 and 57.14%, respectively. Our findings suggest that the marine S. qinglanensis strain, which grows well on tailored medium, might be a source of bioactive substances for healthcare companies.
Objectives: Bacterial biofilm is regarded as a significant threat to the production of safe food and the arise of antibiotic-resistant bacteria. The objective of this investigation is to evaluate the quorum sensing inhibitory effect of Nepeta curviflora methanolic extract. Methods: The effectiveness of the leaves at sub-inhibitory concentrations of 2.5, 1.25, and 0.6 mg/mL on the virulence factors and biofilm formation of P. aeruginosa was evaluated. The effect of N. curviflora methanolic extract on the virulence factors of P. aeruginosa, including pyocyanin, rhamnolipid, protease, and chitinase, was evaluated. Other tests including the crystal violet assay, scanning electron microscopy (SEM), swarming motility, aggregation ability, hydrophobicity and exopolysaccharide production were conducted to assess the effect of the extract on the formation of biofilm. Insight into the mode of antiquorum sensing action was evaluated by examining the effect of the extract on the activity of N-Acyl homoserine lactone (AHL) and the expression of pslA and pelA genes. Results: The results showed a significant attenuation in the production of pyocyanin and rhamnolipid and in the activities of protease and chitinase enzymes at 2.5 and 1.25 mg/mL. In addition, N. curviflora methanolic extract significantly inhibited the formation of P. aeruginosa biofilm by decreasing aggregation, hydrophobicity, and swarming motility as well as the production of exopolysaccharide (EPS). A significant reduction in AHL secretion and pslA gene expression was observed, indicating that the extract inhibited quorum sensing by disrupting the quorum-sensing systems. The quorum-sensing inhibitory effect of N. curviflora extract appears to be attributed to the presence of kaempferol, quercetin, salicylic acid, rutin, and rosmarinic acid, as indicated by LCMS analysis. Conclusion: The results of the present study provide insight into the potential of developing anti-quorum sensing agents using the extract and the identified compounds to treat infections resulting from quorum sensing-mediated bacterial pathogenesis.
Objective : Coptis rhizoma is traditional herb in Korean medicine, and Pelargonium sidoides extract has been used for relief of acute bronchitis in Western medicine. The present study examined the antibacterial effect of fusion formulation of Coptis rhizoma extract and Pelargonium sidoides extract against bronchial diseases bacteria. Methods : Test sample, fusion formulation of Korean and Western medicine, was prepared by mixing Coptis rhizoma extract and Pelargonium sidoides extract at a ratio of 1:2 (w/w). Antimicrobial properties of test sample were determined by agar diffusion assay and minimum inhibitory concentration (MIC) against bronchus diseases bacteria. Results : In agar diffusion assay, the highest amount of test sample (4 mg/disk) exhibited antibacterial activity against all microorganisms tested. Test sample showed the high activity for S. aureus (19.5 mm), C. diptheriae (16.5 mm), A. fumigatus (19.3 mm), F. nucleatum (22.7 mm) and Mycobactrium sp. (17.3 mm), whereas it showed a low activity for K. pneumonia (9.7 mm). The MIC value was determined as $250{\mu}g/m{\ell}$ against C. diptheriae. Test sample showed better growth inhibitory effects against S. aureus and A. fumigatus with the MIC valus of $125{\mu}g/m{\ell}$. Conclusion : These results suggest the possibility of application to chronic respiratory diseases of fusion formulation of Korean and Western medicine, which was prepared with Coptis rhizoma extract and Pelargonium sidoides extract.

A cDNA encoding a defensin-like peptide (Protaetiamycine) from the larvae of a beetle, Protaetia brevitarsis was cloned. The DNAs encoded the deduced propeptide of 79 amino acid residues with the predicted molecular weight of 8.4 kDa and PI of 8.24. Overall amino acid sequence of this protein has 39% similarity to that of Rhodnius prolixus defensin, 43% similarity to that of Acalolepta luxuriosa defensin, and 72% similarity to that of Oryctes rhinoceros defensin, suggesting that this gene is an insect defensin. In an attempt to apply the anti-bacterial peptide to the development of therapeutic agents, a 12-mer peptide amidated at its C-terminus, ACAAHCLAIGRG-$NH_2$ (Ala55-Lys66-$NH_2$, 12Pbn) was synthesized. This peptide showed some antifungal activity against Candida albicans. To increase antifungal activity, six 9-mer peptides were synthesized by modifying amino acid sequences of 12Pbn fragment. Among these peptides, 9Pbm3-9Pbm6 exhibited strong activity compared with Cecropin B and mellitin.
The effects of four extracts of medicinal herbs, Rubus coreanum, Acorus calamus, Morus alba and Rehmannia glutinosa on hair growth activity and acnes control were investigated. In the course of screening natural extracts for hair growth, we found that the extract of dried root of Rubus coreanum has the hair growth promoting effect. After topical application of these extracts to the back of C57BL/6 mice, the earlier conversion of telogen-to-anagen phase was induced. The growth of dermal papilla cells and mouse vibrissae hair follicle cultured in vitro, however, was not affected by treatment of these extracts. Furthermore these extracts do not possesspotent inhibitory effect on $5{\alpha}-reductase$ I and II activity and anti-bacterial effect on Escherichia coli , Propionibacterium acnes, Pityrosporum ovale, Staphylococcus aureus, Staphylococcus epidemidis, and Candida albicans. RT-PCR analysis showed that these extracts did notinduce mRNA levels of growth factors such as insulin-like growth factor-I, keratinocyte growth factor, hepatocyte growth factor and vascular endothelial growth factor in dermal papilla cells. These results suggest that Rubus coreanum has hair growth promoting effect. However, the effects of these materials on the hair growth promotion are not mediated through inhibition of $5{\alpha}-reductase$ I and II activity, stimulation of hair follicle cells and expression of growth factors in the dermal papilla cells.

The cotton leafworm, Spodoptera littoralis, is a major pest in Egypt and many countries worldwide, and causes heavy economic losses. As a result, management measures to control the spread of the worm are required. S. littoralis nucleopolyhedrovirus (SpliNPV) is one of the most promising bioagents for the efficient control of insect pests. In this study, a chitinase gene (chitA) of a 1.8 kb DNA fragment was cloned and fully characterized from SpliNPV-EG1, an Egyptian isolate. A sequence of 601 amino acids was deduced when the gene was completely sequenced with a predicted molecular mass of 67 kDa for the preprotein. Transcriptional analyses using reverse transcription polymerase chain reaction (RT-PCR) revealed that chitA transcripts were detected first at 12 h post infection (hpi) and remained detectable until 168 hpi, suggesting their transcriptional regulation from a putative late promoter motif. In addition, quantitative analysis using quantitative RT-PCR showed a steady increase of 7.86-fold at 12 hpi in chitA transcription levels, which increased up to 71.4-fold at 120 hpi. An approximately 50 kDa protein fragment with chitinolytic activity was purified from ChitA-induced bacterial culture and detected by western blotting with an anti-recombinant SpliNPV chitinase antibody. Moreover, purification of the expressed ChitA recombinant protein showed in vitro growth inhibition of two different fungi species, Fusarium solani and F. oxysporum, confirming that the enzyme assembly and activity was correct. The results supported the potential role and application of the SpliNPV-ChitA protein as a synergistic agent in agricultural fungal and pest control programs.
천연물 황금으로부터 여러 유기용매 추출물을 얻어 치아우식증 원인균인 S. mutans 에 대한 항균활성, 배지 내 환경변화, 그리고 부착억제에 대해 알아보았다. 1. 구강 질병인 치아우식증의 대표적 원인균 S. mutans에 대한 황금 ethyl acetate 추출물 (IPK-3)의 최소억제농도는 125 mg/ml 이었다. 2. 황금 IPK-3 추출물 50 mg/ml의 농도로 S. mutans의 배지 내 투여하였을 때 균의 생장 량과 최대 생장시간은 대조군보다 지연되었다. 3. 황금 IPK-3 추출물 50 mg/ml의 농도로 S. mutans의 배지 내 투여하였을 때, pH 변화는 대조군은 18시간에서 pH 5.63으로 급격한 변화를 보였으며 황금 IPK-3 추출물이 50 mg/ml의 배지에서는 6.50이상을 유지하여 pH가 변화가 없었다. 4. 황금 IPK-3 추출물을 첨가한 배지 내 탄수화물, 단백질 및 균체 외 다당류의 변화는 대조군에 비해 매우 낮게 생산되었다. 5. 균체를 형광염료(DAPI)로 염색하여 S-HA 부착억제을 확인한 결과, 대조군은 시간이 경과함에 따라 S.mutans의 hydroxyapatite(HA)에 부착 정도가 증가되었으나, 황금 IPK-3 추출물이 첨가된 시험군에서는 S-HA에 균의 부착이 매우 적었다. 이같이 황금의 IPK-3 (ethyl acetate 추출물)이 치아우식증의 원인균인 S. mutans 에 대한 항균활성효과 뿐만아니라 hydroxyapatite에 부착 억제 효과도 있었다.
본 연구는 식물성플랑크톤의 대량증식 조절과 관련된 해양성 살조능이 있는 박테리아의 분리와 유해조류에 대한 분리 균주의 살조능 특성에 주로 초점을 맞추고 있다. 해수 표면에서 자연적으로 발생하는 유해조류번성(HAB)은 전세계적으로 많은 환경문제를 일으키고 있다. 본 연구에서는 유해조류 성장을 억제하는 능력을 가진 40개의 박테리아 균주를 마산만, 진해만, 돌섬, 거제도, 통영 앞바다에서 분리하였다. 분리된 균주들은 다양한 유해조류인 Cochlodinium polykrikoides, Chattonella marina, Skeletonema costatum, Heterosigma akashiwo, Heterocapsa triquetra, Prorocentrum minimum, Scrippsiella trochoidea에 대한 살조특성을 추가로 조사하였다. 살조균주의 선별은 이중층 아가배지와 현미경 계수법을 이용하여 진행하였고 Pseudomonas, Vibrio, Bacillus, Pseudoalteromonas, Ruegeria, Joostella, Marinomonas, Stakelama, Porphyrobacter, Albirhodobacter의 속들에 속하는 균주들이었다. 가장 중요한 유해조류인 Co. polykrikoides에 대한 가장 강력한 살조능은 10% 배양상등액으로 6시간 처리했을 때 94%를 보이는 균주였다. 이 연구를 통해 살조효과를 보이는 새로운 속으로 Marinomonas sp. M Jin 1-8, Stakelama sp. ZB Yeonmyeong 1-11 & 1-13, Porphyrobacter sp. M Yeonmyeong 2-22, Albirhodobacter sp. 6-R Jin 6-1를 새롭게 찾았다. 결론적으로 이들 해양박테리아를 이용하면 식물성플랑크톤 번성을 제어하는데 중요한 역할을 할 것으로 예상된다.
본 연구에서는 작약(Paeonia lactiflora Pall.) 종자 오일과 유박의 생리활성의 비교를 통해 작약 종자 유박 활용에 가능성을 평가하고자 수행되었다. 작약 종자 유박 추출물의 추출 수율은 22-45%로 추출용매에 따른 차이를 보였다. 총 폴리페놀 및 플라보노이드와 radical소거능은 오일보다 유박 추출물에서 유의하게 높았다. 작약 유박 추출물의 항균활성은 양성대조군에 비해 우수하였으며, 이는 48시간까지 지속되었다. 또한 LPS로 유도된 염증 반응은 유박 추출물 처리시 현저하게 감소하여 작약 종자 유박의 항염증 활성이 입증되었다. 본 연구를 통해 작약 종자 오일 뿐만 아니라 유박 추출물의 우수한 항산화, 항균 및 항염증 활성을 나타내어 바이오헬스 소재로서 작약 종자 유박의 활용 가능성이 확인되었다.
