1. Objectives The purpose of this study is to prove the anti-aging effects of Sipyimigwanjung-tang. 2. Methods The SD rats used in this experiment were 6, 18 and 36 weeks old. A part of the 36weeks was grown to 52 and 68 weeks at labarotary. Each age group was again divided into three groups. These 15 groups consisted of 6 rats each. One group was given no treatment, another group was dosed 200 ${\mu}l$ of normal saline daily, and the last group was dosed 200 ${\mu}l$ of 1% Sipyimigwanjung-tang(SYG) and saline mixture. At the conclusion of the experiment, the age groups were relabelled accordingly(10w, 22w, 40w, 52w and 68w). After 4 weeks, the tissue of liver, heart, spleen, lung, kidney and brain was biopsied in order to measure the SOD, GSH, MDA. 3. Results and Conclusions In liver, the activity of the SOD of 52w-SYG was significantly increased than that of 10w, 22w and 40w-SYG, the level of the GSH of 40w-SYG was significantly increased than that of the normal group, the activity of the catalase of 68w-SYG was significantly increased than that of the normal and control group, the level of the MDA of 52w-SYG and 68w-SYG was significantly decreased than that of the normal and control groups. In heart, the level of the GSH of 22w-SYG was significantly increased than that of the normal and control group. In spleen, the level of the GSH of 52w-SYG was significantly increased than that of the normal and control group. In lung, the level of the GSH of 52w-SYG was significantly increased than that of the normal group and the level of 68w-SYG was significantly increased than that of the normal and control group. In kidney, the level of the GSH of 10w-SYG was significantly increased than that of the normal group. In brain, the level of the GSH of 68w-SYG was significantly increased than that of the normal and control group. The SYG inhibited the histology degeneration of brain tissue. These results suggest that oral administration of SYG (Sipyimigwanjung-tang) decoction has anti-aging effects in aged rats.
근육 양과 근력의 감소에 의한 근감소증은 다양한 병적 상태의 악화, 삶의 질 저하, 사망률의 증가를 동반하며, 기대수명이 증가함에 따라 앞으로도 유병률은 계속 증가할 것이다. 노화 과정에서 만성 산화 스트레스와 염증 반응의 증가는 골격근 소실의 주요 원인으로 작용하며, 에너지 대사에 필수적인 미토콘드리아의 기능 장애와 관련된 자가포식 및 세포사멸 신호의 교란은 근육 단백질의 손실을 가속화한다. 오래전부터 각종 질병의 예방 및 치료에 널리 사용되어온 동충하초의 주요 생리활성물질인 cordycepin의 약리학적 효과는 항산화 및 항염증 작용과 직접적인 관련이 있는 것으로 알려져 있다. 본 총설에서는 근감소증의 예방과 치료에의 적용을 위한 cordycepin의 세포사멸, 자가포식, 단백질의 이화작용 및 근육 재생에 중요한 위성세포의 활성에 대한 연관성을 제시하였다. 비록 현재까지 근감소증에 대한 cordycepin의 연구는 미진한 수준이지만, 그동안의 연구 결과에서 cordycepin은 노화로 인한 미토콘드리아 기능 약화를 억제하고 근육 단백질의 분해를 차단하는 데 기여할 수 있음을 알 수 있다. 또한 근세포 손상에 대한 cordycepin의 보호 효과는 항산화 및 항염증 활성과 밀접한 관련이 있음을 제안한다. 따라서 근세포의 노화방지에 관여하는 cordycepin의 분자생물학적 기전을 중심으로 보다 지속적인 기초 연구가 필요할 것으로 사료된다.
Sesame seed are one of the oldest oil-bearing crops known to man. Not only are they nutritious, they are physicologically beneficial. Because of these properties, the promotion of sesame as a crop is set to increase from now on. The lignan component of sesame, which has an antioxidative function, varies considerably. In addition, the seeds remain viable for longer than other crops. At low temperature, it has been reported that the seed remain viable for up to thirty years. This is due to the lignan content of the seeds. In the last twenty years, the mysteries of the natural chemistry of sesame seeds have been unravelled one by one. Up to now, the lignan component has been found to have an $\alpha$-tocopherol synergist effect, an anti-aging effect, a cancer suppressing effect, a hypertension reduction effect, an effect on the promotion of liver function, an effect on the control of the ration of unsaturated fat, and an effect on the synthesis of unsaturated fatty acids. The lignan component of sesame does not simply have an anti-oxidant function, but has diverse recently discovered physiological uses which make the study of lignan highly rewarding.
본 연구에서는 낙타유를 유효성분으로 하여 리포좀을 제조하였고, 이를 이용하여 항노화 효능을 갖는 화장품 원료를 개발하고자 다양한 실험을 실시하였다. 제조된 낙타유 리포좀은 피부 섬유아세포에서 collagen과 hyaluronan synthase-3 (HAS-3)의 발현을 증가시키고 matrix metalloproteinase (MMP)-1의 발현을 감소시킬 뿐 아니라 elastase의 활성을 억제하여 주름 개선 기능을 갖는 것을 확인하였다. 또한 자외선으로부터 손상된 세포를 재생시키는 효과를 확인하였다. 이에 따라 낙타유를 함유한 리포좀은 항노화 소재로 활용할 수 있을 것으로 사료된다.
A novel retinol derivative, polyethoxylated retinamide (Medimin A) was synthesized, as an anti-aging agent. Collagen synthesis, skin permeation, stability, and toxicity of Medimin A were evaluated and compared with those of retinol and retinyl palmitate. In vitro collagen synthesis was evaluated by quantitative assay of [$^3H$]-proline incorporation into collagenase sensitive protein in fibroblast cultures. For in vitro skin permeation experiments, Franz diffusion cells (effective diffusion area: $1, 766{\;}\textrm{cm}^2$) and the excised skin of female hairless mouse aged 8 weeks were used The stabilities of retlnoids were evaluated at two different temperature ($25{\;}^{\circ}C$ and $40{\;}^{\circ}C$) and under UV in solubilized state and in OW emulsion. To estimate the safety, acute oral toxicity, acute dermal toxicity, primary skin irritation, acute eye irritation and human patch test were performed The effect of Medimin A on collagen synthesis was similar to that of retinol. The skin permeability of Medimin A was higher than those of retinol and retinyl palmitate. The Medimin A was more stable than retinol and retinyl palmitate. Medimin A was nontoxic in various toxicological tests. These results suggest that Medimin A would be a good anti-aging agent for enhancing bioavailability and stability.

BACKGROUND/OBJECTIVES: Doenjang, Korean traditional fermented soybean paste has been reported to have an anti-obesity effect. Because adipose tissue is considered a major source of inflammatory signals, we investigated the protective effects of Doenjang and steamed soybean on oxidative stress and inflammation in adipose tissue of diet-induced obese mice. MATERIALS/METHODS: Male C57BL/6J mice were fed a low fat diet (LF), a high-fat diet (HF), or a high-fat containing Doenjang diet (DJ) or a high-fat containing steamed soybean diet (SS) for 11 weeks. RESULTS: Mice fed a DJ diet showed significantly lower body and adipose tissue weights than those in the HF group. Although no significant differences in adipocyte size and number were observed among the HF diet-fed groups, consumption of Doenjang alleviated the incidence of crown-like structures in adipose tissue. Consistently, we observed significantly reduced mRNA levels of oxidative stress markers (heme oxygenase-1 and $p40^{phox}$), pro-inflammatory adipokines (tumor necrosis factor alpha and macrophage chemoattractant protein-1), macrophage markers (CD68 and CD11c), and a fibrosis marker (transforming growth factor beta 1) by Doenjang consumption. Gene expression of anti-inflammatory adipokine, adiponectin was significantly induced in the DJ group and the SS group compared to the HF group. The anti-oxidative stress and anti-inflammatory effects observed in mice fed an SS diet were not as effective as those in mice fed a DJ diet, suggesting that the bioactive compounds produced during fermentation and aging may be involved in the observed health-beneficial effects of Doenjang. CONCLUSIONS: Doenjang alleviated oxidative stress and restored the dysregulated expression of adipokine genes caused by excess adiposity. Therefore, Doenjang may ameliorate systemic inflammation and oxidative stress in obesity via inhibition of inflammatory signals of adipose tissue.
To evaluate the protective effect of collagen peptide-coated coffee extract on skin aging, cell viability was measured with a MTT assay using cultured CCD-986sk fibroblasts, and its effect on wrinkles in the skin of hairless mice induced by UVB-irradiation was examined. In addition, its effect on procollagen synthesis and anti-oxidative, and its inhibitory activity against collagenase, elastase, tyrosinase and MMP-1 were analysed. After the 30-minute topical treatment, the animals were exposed to UVB irradiation (60-100 mJ/cm2) for 4 weeks and its intensity increased during the period. Under the experimental conditions set in this study, the skin thickness of hairless mice significantly decreased (11.8-21.3%) compared to the control group. Based on these results, the prolonged oral intake of a collagen peptide mixture with coffee is expected to significantly increase the synthesis of procollagen in dermal fibroblasts, thereby contributing to the alleviation of wrinkling and lowered elasticity due to structural damage to the dermal layer caused by UV. The oral intake of collagen-coated coffee contributes to increasing collagen biosynthesis in a dose-dependent manner and alleviates the symptoms of thickened keratin caused by UV irradiation. However, it did not inhibit the enzymes involved in skin aging, whitening, wrinkle improvement, and antioxidation. Based on the these results, it can be concluded that the intake of collagen peptide-coated coffee extract can be utilized as an alternative material for the prevention or treatment of diseases associated with photoaging.
본 연구는 예부터 약용으로 사용되었거나, 현재 식품공전에 식품원료로 사용이 가능한 것으로 등록된 국내 산림지역에 자생하는 식물을 식품산업에 활용하고자 선행연구에서 우수한 항산화 활성을 보인 갈참나무 잎을 본 연구에서 사용하였다. 70% 에탄올을 이용하여 추출한 갈참나무 잎을 이용하여, hydrogen peroxide로 산화적 스트레스를 유도한 피부 섬유아세포에서의 세포 보호효과, 세포내 항산화 효과 및 항노화 효과를 측정하였다. 세포 독성을 평가한 결과 25, 50 및 $100{\mu}g/mL$의 갈참나무 잎 추출물을 처리하였을 때 모두 독성을 나타내지 않았으며, hydrogen peroxide로 산화적 스트레스를 유도한 상태에서는 세포를 보호하여 농도 유의적으로 세포생존율이 증가하였다. 특히, $100{\mu}g/mL$의 농도에서는 양성대조군으로 사용한 $50{\mu}M$ ascorbic acid 수준까지 세포 생존율이 증가하였다. 세포내 항산화 효과를 확인 하기위해 사용한 $H_2-DCFDA$ assay에서는 형광현미경과 형광흡광도 측정에서 모두 농도 유의적으로 세포내 ROS 저감 활성을 확인하였고 갈참나무 잎 추출물을 $100{\mu}g/mL$ 농도로 처리했을 때는 $50{\mu}M$ ascorbic acid와 비슷한 세포내 항산화 효과를 나타내었다. 또한 SA-${\beta}$-galactosidase assay를 이용한 갈참나무 잎 추출물의 피부 섬유아세포에 대한 항노화활성은 ROS 생성 억제 효과와 유사한 경향으로 갈참나무 잎 추출물의 농도 유의적으로 세포 노화 억제효과를 확인하였다. 이상의 결과를 종합하여 볼 때 갈참나무 잎 추출물이 hydrogen peroxide로 인한 산화적 스트레스 상태에서 세포 보호효과, 항산화 효과 및 항노화 효과가 관찰되어 기능성 식품원료로서의 활용도가 매우 넓을 것으로 판단된다.

Recent studies have highlighted the link between diseases and inflammation across our lifespan. Our sedentary lifestyle, high-calorie diet, chronic stress, chronic infections, and exposure to pollutants and xenobiotics, collectively intensify the course and recurrence of infections and inflammation in our bodies, promoting the prevalence of chronic diseases and aging. Given such phenomena and considering additional factors such as the frequency of prescription, and easy access to over-the-counter drugs, the need for anti-inflammatory therapeutics is ever-increasing. However, the readily available anti-inflammatory treatment option comes with a greater risk of side effects or high cost (biologics). Therefore in this growing competition of discovering and developing new potent anti-inflammatory drugs, we focused on utilizing the established knowledge of traditional medicine to find lead compounds. Since lead optimization is an indispensable step toward drug development, we applied this concept for the production of potent anti-inflammatory compounds achieved by structural modification of flavonoids. The derivative obtained through acetylation of myricetin, 3,3',4',5,5',7-hexaacetate myricetin, showed a greater inhibitory effect in the production of pro-inflammatory mediators such as nitric oxide, Prostaglandin E2, and pro-inflammatory cytokines like interleukin-6, interleukin1β, in lipopolysaccharide-stimulated RAW264.7 mouse macrophage cells compared to myricetin. The increased potency of inhibition was in conjunction with an increased inhibitory effect on inducible nitric oxide synthase and cyclooxygenase-2 proteins. Through such measures, this study supports lead optimization for well-established lead compounds from traditional medicine using a simpler and greener chemistry approach for the purpose of designing and developing potent anti-inflammatory therapeutics with possibly fewer side effects and increased bioavailability.
Taurine is an abundant amino acid in many animals, including humans. Relatively large amounts of taurine are found in leukocytes, heart, muscles, retinas, kidneys, bones, and liver. Taurine has antioxidant effects; it reacts with hydrogen peroxide to prevent oxidation of the cell membrane. Taurine enhances the effects of anticancer drugs, while also reducing side effects, and taurolidine, a taurine derivative, has been shown to exhibit anti-cancer effects without notable side effects in several types of cancer. Taurine aids in cholesterol metabolism by increasing the rate of synthesis of bile acids, and, thus, reduces triglyceride levels. In addition, taurine is involved in the growth and differentiation of nerve cells and is associated with some neurological disorders. Taurine aids in bone formation and prevents bone dissolution. Moreover, taurine prevents liver damage from a variety of drugs and, thus, protects the liver. Taurine is involved in the development and function of the retina and lens. It also has anti-atherosclerotic and anti-thrombotic effects that protect against cardiovascular disease. Taurine may have additional physiological functions, and warrants further investigation.
