Effects of long-term endurance exercise and Salvia miltiorrhiza vinegar on body composition and insulin resistance in high fat diet-induced obese rats (장기간의 지구성 운동과 단삼식초 섭취가 고지방식 유도 비만 흰 쥐의 신체구성과 인슐린 저항성에 미치는 영향)

  • The purpose of this study was to examine the effects of long-term endurance exercise and salvia miltiorrhiza vinegar on body composition and insulin resistance of high-fat diet (30% carbohydrate, 50% fat and 20% protein) induced obese rats. After 8 weeks of high fat diet (50% of total calories), rats were divided into 4 groups (sedentary group, n=10; exercise group, n=10; Salvia miltiorrhiza vinegar group, n=10; exercise+Salvia miltiorrhiza vinegar group, n=10) for 8 weeks. Body weight, body composition, diet intake volume, oral glucose tolerance test, plasma total cholesterol were measured. The results showed that Salvia miltiorrhiza vinegar plus endurance exercise training for 8 weeks significantly improved body weight control, visceral fat weight, and insulin resistance. However, only Salvia miltiorrhiza vinegar treatment did not significantly improve body composition and insulin resistance. In addition, there was no additive by the combination of Salvia miltiorrhiza vinegar and endurance exercise in insulin, body fat, and total cholesterol. The reduction of body fat, glucose, insulin and cholesterol by combination was resulted from the exercise. These results suggest that Salvia miltiorrhiza vinegar has slight effect on anti-hyperglycemia and anti-obesity.

Protective Effects of Korean Red Ginseng against Alcohol-induced Hepatosteatosis (알코올에 의해 유발된 지방변성증에서 홍삼의 보호효과)

  • Alcohol-induced fatty liver (steatosis) results from excessive generation of reducing equivalents by ethanol metabolism. Generally, chronic ethanol treatment causes hepatosteatosis by regulating sterol regulatory element-binding protein 1c (SREBP-1c), which increases the synthesis of hepatic lipids. The effect of ethanol on SREBP-1c is mediated through mammalian sirtuin-1 (SIRT-1), a NAD+-dependent protein deacetylase that regulates hepatic lipid metabolism. Ginseng is a widely used herbal medicine that is used in Asia for its anti-diabetes and anti-obesity effects. The pharmacological and therapeutic effects of ginseng are primarily produced by bioactive constituents known as ginsenosides. Here, we examined the regulatory effects of Korean red ginseng (KRG) extracts on SREBP-1c and SIRT-1 on lipid homeostasis in AML-12 mouse hepatocytes. AML-12 cells were treated with ethanol and/or KRG extracts (0 - 1,000 μg/ml). Lipid droplets were assayed using Oil red O staining, and western blotting was used to measure SIRT-1 and SREBP-1 expression. Treatment with KRG extracts restored SIRT-1 expression and reduced SREBP-1c expression in ethanol-treated cells. We also showed that KRG extract and ginsenosides Rb2 and Rd significantly decreased SREBP-1 acetylation in ethanol-treated cells. These results show that treatment with KRG extract and its active ginsenoside constituents Rb2 and Rd protected against alcohol-related hepatosteatosis via regulation of SIRT-1 and downstream acetylation of SREBP-1c, which altered hepatic lipid metabolism.

Silibinin Inhibits Adipogenesis and Induces Apoptosis in 3T3-L1 Adipocytes (Silibnin의 지방세포분화 억제 및 세포사멸 유도 효과)

  • $C/EBP{\beta}$ and $C/EBP{\delta}$ are required for the initiation of adipogenesis and induce the expression of key adipogenic regulators, such as $PPAR{\gamma}$ and $C/EBP{\alpha}$. In the present study, we have examined the effects of silibinin and its possible molecular mechanisms in regulating adipocyte differentiation and expression of $C/EBP{\beta}$ and $C/EBP{\delta}$ in the early stage of adipogenesis. Silibinin statistically significantly inhibits intracellular lipid accumulation and the mRNA expression of various genes involved at different stages during adipogenesis. Silibinin also suppresses expression of lipoprotein lipase (LPL), fatty acid binding protein 4 (AP2), and adiponectin in 3T3-L1 adipocytes. Thus, the anti-adipogenic effect of silibinin seems to originate from the ability to inhibit the expression of $C/EBP{\beta}$ and $C/EBP{\delta}$. Furthermore, silibinin decreases cell viability for differentiation period and induces apoptotic cell death through capspase-3 activation.

Isolation of Antioxidative Products and Evaluation of the Pancreatic Lipase Inhibitory Activity of (+)-Catechin via Thermal Treatment ((+)-Catechin 가열처리에 의한 항산화 활성물질 분리 및 pancreatic lipase 저해활성 평가)

  • Thermal processing of (+)-catechin was carried out at $121^{\circ}C$ for different reaction times (1, 2, 3, 6, and 12 h). The reacted products, compounds (1) and (2), were isolated and quantified via HPLC analysis. The antioxidant properties of processed (+)-catechin and its isolated compounds for different reaction time was measured via radical scavenging assays using DPPH and $ABTS^+$ radicals. Additionally, the anti-obesity efficacy of the thermal treated (+)-catechin was evaluated via porcine pancreatic lipase assay. The reacted (+)-catechin for 3 h had a slightly higher antioxidant capacity than that the parent (+)-catechin. Products 1 and 2, which were isolated from the reacted mixture during 3 h, showed an antioxidant capacity, and these two compounds may be responsible for the antioxidant capacity of processed (+)-catechin. Simple thermal treatment of (+)-catechin can be used to produce (+)-epicatechin (1) and protocatechuic acid (2) with enhanced antioxidant and anti-adipogenic effects.

We have performed this test in order to develop a skin care device for home in order to compensate for this point.In this paper, we develop a low-level laser to create a personal skin care products and sought to incorporate them into the skin cosmetic.Expand the pores by using the low-level laser to the skin by to the dermal layer of the skin was penetrated aim experiment the ampoule, and by a comparison of the medical low-level laser reliability and determine the effectiveness or absence of the performance and efforts to commercialize.

  • Kaempferia parviflora (Krachaidam, Zingiberaceae) is used as a traditional Thai medicine for treating various ailments, including allergy, fatigue, sexual dysfunction, and ulcer. Moreover, it is also used to promote longevity and as a nerve tonic. The aim of this study was to develop functional yoghurt containing Kaempferia parviflora and to examine the physicochemical characteristics of yoghurt supplemented with different concentration of K. parviflora. To this end, we examined the pH and sensory evaluation of yoghurt containing K. parviflora. The pH of this yoghurt decreased whereas the TA increased with incubation time (up to 5 h) without altering the amounts of K. parviflora added. In the sensory evaluation, the taste, flavor, color, and overall acceptability decreased in proportion to the amount of K. parviflora added. Among the experimental group, high scores were achieved by yoghurt containing 1% K. parviflora compared with that by the control group. We conclude there is an urgent need for additional research on the production of this multi-functional yogurt (with properties including anti-obesity and anti-inflammatory), using an optimum concentration of K. Parviflora.

Evaluation of biological activities of plasma-treated phloridzin (플라즈마 처리 phloridzin 반응물의 생리활성 평가)

  • Phloridzin is a predominant member of the chemical class of dihydrochalcones and mainly found in apple. The biological activity of phloridzin treated with dielectric barrier discharge (DBD) plasma was evaluated to investigate whether exposure to plasma can be used as a tools to enhance the biological activity of natural resources. DBD plasma treatment of phloridzin was carried out for three different exposure times. The antioxidant effects of degraded phloridzin for different reaction time were evaluated via radical scavenging assay using DPPH radical. In addition, the anti-diabetic and anti-obesity properties of the degraded phloridzin were measured based on ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities. Phloridzin treated for 60 min showed significantly higher radical scavenging, ${\alpha}$-glucosidase and pancreatic lipase inhibitory activities compared to the tested positive controls. Degradation of phloridizin induced by DBD plasma might be responsible for enhancing the biological activity of phloridizin.

Quality characteristics and bioactivity analysis of habanero red pepper (Capsicum chinense Jacq.) through acetic acid bacteria fermentation (하바네로 고추 초산발효물의 품질 특성 및 생리활성 분석)

  • Habanero red pepper (HRP)-fermented liquid with Acetobacter pasteurianus SRCM101474 was prepared, the quality characteristics measured and bioactivity analysis were measured. As the fermentation period increased, the number of viable cells increased, pH decreased, and acidity increased. After fermentation, the organic acid content of HRP was confirmed to be acetic acid with the highest content of 33413.54 ppm. Analysis of capsaicin and dihydrocapsaicin showed that the increase was significantly raised as acetic acid fermentation progressed. The total phenol and flavonoid contents as well as the antioxidant activity increased as the fermentation period passed. In addition, α-glucosidase inhibitory and pancreatic lipase inhibitory activities were significantly increased after fermentation. It was confirmed that it can be used as a functional material by measuring the physiological activities of antioxidant, anti-obesity, and anti-diabetes through fermentation, and the possibility of development as a spicy material and application product using it as capsaicin increases after fermentation.

Evaluation of the Relevance of Nutritional Status and Dietary Inflammation Index to Blood Glucose Levels in Middle-aged Women: in terms of 2013-2018's Korean National Health and Nutrition Survey Data (중년 여성의 혈당수준에 따른 영양상태 및 식이염증지수의 융합적 관련성 평가: 2013-2018 국민건강영양조사 자료 이용)

  • This study targeted 4,572 middle-aged women to examine the relationship between nutritional status and dietary inflammatory index according to blood glucose level using data from the Korean National Health and Nutrition Examination Survey (KNHANES). Data were analyzed using complex survey chi-square, General Linear Model and logisitc regression in SPSS Win 25.0 program. Women with high blood glucose (normal blood sugar→diabetes) had high rates of obesity and blood TG/HDL-cholesterol ratio. On the other hand, the Mean Adequacy Ratio (10 nutrients) and the intake of anti-inflammatory foods: beans, seeds, mushrooms, and fruits, were lower in the diabetic category. When we analysed the association between blood glucose and the Dietary Inflammatory Index, the risk of pre-diabetes and diabetes was significantly higher in the most pro-inflammatory diet category (Q5) compared to the most anti-inflammatory diet category (Q1). These findings suggest that nutritional education emphasizing the intake of various foods should be effectively conducted effectively in order to improve blood glucose among middle-aged women.

Evaluation of the Biological Activity Affected by Extracting Solvents of Rosemary (Rosmarinus officinalis L.) (로즈마리(Rosmarinus officinalis L.) 극성별 용매 추출물의 생리활성 검증)

  • Rosemary (Rosmarinus officinalis L.) is widely used as a food material. Although various physiological activities of rosemary have been reported, there have been no studies on the physiological activity of solvent extracts with different polarities. Rosemary extracts were obtained by extraction of dried powder using 0%, 25%, 50%, 70%, and 95% ethanol (EtOH) in distilled water, methanol, ethyl acetate, and hexane. As these ratios of EtOH are generally chosen by default and scarcely optimized, we investigated the impact of the composition of EtOH in distilled water on extract-related characteristics, such as DPPH free radical scavenging and ${\alpha}$-glucosidase inhibition, on the differentiation of 3T3-L1 adipocytes and inhibition of tyrosinase. Adipogenesis inhibition was highest at 70% EtOH. DPPH scavenging activity and inhibition of tyrosinase activity were reduced with 50% EtOH in water. However, inhibition of ${\alpha}$-glucosidase activity was higher in 50% EtOH in water. The best solvents in terms of DPPH scavenging activity, inhibition of tyrosinase and ${\alpha}$-glucosidase, and differentiation of adipocytes obtained with different concentrations of EtOH, although a lower similar activities were found with 50% ethanol. Considering the extraction solvents, a ratio of EtOH in water gives different content and constituents of compounds. These differences will give activities inhibition of adipogenesis, tyrosinase, ${\alpha}$-glucosidase activity, and DPPH scavenging activity.
