The anti-atopic effect of natural carbonated hot spring water on DNCB-induced NC/Nga mice under different temperature (DNCB로 아토피가 유도된 NC/Nga생쥐에서 천연 탄산 온천수의 온도별 항아토피 효과)

  • Objectives : The aim of this study is to investigate the effect of natural carbonated hot spring water (NCHW) on DNCB-induced NC/Nga mice, an atopic dermatitis (AD) model. Methods :At first, NC/Nga mice were prepared and induced to have atopic lesion on their back skin by dinitrochlorobenzene (DNCB). They swam for 20 minutes everyday in tanks where the one is filled with $28^{\circ}C$ NCHW, the other is with $25^{\circ}C$ carbonated hot spring water and another is with artificial carbonated hot spring water (ACHW). After 3 weeks, We assessed the skin clinical score and macroscopic appearance, total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and histological changes. Results : There are meaningful results of improving atopic lesion-state by relieving the count of total IgE, IgG1 levels, WBC differential counting, IFN-${\gamma}$, IL-4, IL-13, TNF-${\alpha}$ production levels and recovering skin clinical score in the group with $28^{\circ}C$ NCHW in comparison with the other groups. Conclusions : The NCHW may have potential as an effective treatment for atopic dermatitis.

Purification of Anti-allergic Compound from Mori Cortex Radicis Extract (상백피 추출물 중의 항알러지 활성 성분의 분리)

  • Mori Cortex Radicis, the root bark of mulberry tree, has been used in the treatment of bronchial asthma and other lung diseases in traditional medicine. There was a recent repor t that the water soluble part with molecular weight of above 10,000 has anti-allergic activity. Therefore, we intended to isolate and purify the anti-allergic compound from hot water extract of the Mori Cortex Radicis. Crude extract of Mori Cortex Radicis was prepared by hot-water extraction, and anti-allergic compound was further purified by alcohol precipitation, successive ultrafiltration, anion exchange chromatography and gel filtration chromatography. This compound had homogeneity which was shown by the sharp single peak in HPLC chromatogram (TSK-GEL G400OPW column, RI detector). The molecular weight of the compound was estimated as 23Kda on the basis of calibration curve plotted against protein standards. This compound was identified as complex of sugar, protein and lignin (19.2: 5.9: 72.7), and proteolysis could not decrease the anti-allergic activity but mild delignification decreased the activity remarkably. Therefore, we concluded that the anti-allergic compound of Mori Cortex Radicis was a lignin-carbohydrate complex.

  • Experimental Studies were done to research the clinical effects of Hyunggaeyeugyotang on the Anti-allergic effect in mice and rats. The results obtained as follows; 1. In the effect of Hyunggaeyeungyotang on vascular permeability responses to intradermal histamine in rats, Hyunggaeyeugyotang revealed Significant effect. 2. In the effect of Hyunggaeyeungyotang on vascular permeability responses to intradermal serotonin in rats, Hyunggaeyeungyotang revealed significant effect. 3. In the 48hr homologous passive cutaneous anaphylaxis in rats provoked by the IgE-like antibody against egg albumin, Hyunggaeyeungyotang revealed significant effect. 4. In the effect of Hyunggaeyeungyotang on Delayed-type hypersensitivity responses to picryl chloride in mice, Hyunggaeyeungyotang revealed none of significant. 5. In the effect of Hyunggaeyeungyotang on Delayed-type hypersensitivity responses to SRBC in mice, Hyunggaeyeungyotang revealed none of significant. According to above-stated results, Hyunggaeyeungyotang is concluded to be effective as anti-allergic regimen and recommended to be used for treatment of allergic disease.

  • We investigated the effects of butanol fraction of Asterias amurensis (BFA) on the anti-allergic activity. BFA inhibited both cyclooxygenase-2 dependent prostaglandin $D_2$ and 5-lipoxygenase dependent leukotriene $C_4$ generation in a concentration-dependent manner with $IC_{50}$ values of 174.6 and 22.2 ${\mu}g$/ml, respectively. In addition, BFA also inhibited the degranulation in a dose dependent manner. Furthermore, oral administration of BFA inhibited IgE-mediated passive cutaneous anaphylaxis in mice. These results suggested that BFA may be useful in regulating mast cell-mediated allergic diseases.

Inhibitory Effect of Scutellaria baicalensis Root Extract on Chemical Mediator Release and Immune Response

  • Inhibitory effect of Scutellaria baicalensis ethanol extracts (SR) on chemical mediator release and immunoglobulin (Ig) production from Sprague-Dawley rats originated cells as type I allergic reaction was examined. SR showed concentration-dependent inhibition on basal and concanavalin A (ConA)-stimulated Ig production. In the mesenteric lymph node lymphocytes, the inhibitory effect of SR on the IgE production in the presence of Con A was stronger than these on IgA and IgG production. Moreover, tumor necrosis factor-alpha $(TNF-{\alpha})$ production-inhibiting effect of SR in the presence ConA was observed. However, SR did not affect the production of $interferon-{\gamma}$. SR also inhibited histamine release from the peritoneal exudate cells stimulated with a calcium ionophore A23187. In the case of leukotriene B4, SR markedly inhibited it at the concentration of 100 mg/ml. From these results, ethanol extracts obtained from Scutellaria baicalensis may have an anti-allergic effect on the intestinal system of rats.

Helicobacter pylori Infection and Gastroduodenal Pathology in Children with Upper Gastrointestinal Symptoms (상부 위장관 증세가 있는 소아의 위십이지장병변 및 Helicobacter pylori 감염)

  • Purpose: This study was undertaken to evaluate the gastroduodenal pathology and Helicobacter pylori infection in children with upper gastrointestinal symptoms. Methods: One hundred and seven pediatric patients with upper gastrointestinal symptoms were undergone endoscopy at the Gyeongsang National University Hospital from June 1990 to April 1991. Histopathologic examination was done by H & E staining of gastric antral biopsy specimen and gastritis was defined according to the Sydney System. Tissue H. pylori status was evaluated with the urease test using Christensen's urea broth and H & E or Warthin-Starry silver staining of gastric antral biopsy specimen. IgG Immunoblotting were also performed to detect specific anti-H. pylori antibody in these patients. Results: The reasons for endoscopy were recurrent abdominal pain, acute abdominal pain, sallow face, hunger pain, and frequent nausea. Variable degrees of gastric mucosal hyperemia were found in most of the patients. Gastric hemorrhagic spots, gastric ulcer, duodenal ulcer, duodenal erosion, and hemorrhagic duodenitis were rare endoscopic findings. Histologic chronic gastritis was found in 88% of 107 patients. Histologic chronic duodenitis was observed in all 99 patients whose tissue were available. Gastric tissue H. pylori was positive in 57% of 107 patients by one of the ureasetest, H & E staining and Warthin-Starry silver staining. However, gastric tissue H. pylori detection rate was lower in the younger age groups. Anti-H. pylori IgG antibodies were detectable in 96% of 107 patients. Conclusion: Chronic gastroduodenitis and anti-H. pylori IgG antibody were ubiquitous in children with upper gastrointestinal symptoms.

Detection of Multi-class Pesticide Residues Using Surface Plasmon Resonance Based on Polyclonal Antibody

  • The detection of carbamate (carbofuran, carbaryl, benfracarb, thiodicarb, and methomil) and organophosphate (diazinon, cadusafos, ethoprofos, parathion-methyl, and chlorpyrifos) pesticide residues with very low detection limits was carried out using surface plasmon resonance (SPR) based equipment. The capacity to develop a portable SPR biosensor for food safety was also investigated. The applied ligand for the immunoassays was polyclonal goat anti-rabbit immunoglobulin (IgG) peroxidase conjugate. Concentration tests using direct binding assays showed the possibility of quantitative analysis. For ligand fishing to find a proper antibody to respond to each pesticide, acetylcholinesterase (AChE), and glutathione-S-transferase (GST) were tested. The reproducibility and precision of SPR measurements were evaluated. With this approach, the limit of detection for pesticide residues was 1 ng/mL and analysis took less than 11 min. Thus, it was demonstrated that detecting multi-class pesticide residues using SPR and IgG antibodies provides enough sensitivity and speed for use in portable SPR biosensors.

$HgCl_2$ Dysregulates the Immune Response of Balb/c Mice (수은에 의한 마우스의 면역반응 조절장애)

  • The studies reported here were undertaken to investigate the effects of mercury chloride on immune system of Balb/c mouse employing a flexible tier of in vitro and in vivo assays. Mercury chloride inhibited the proliferative responses of spleen cells to lipopolysaccharide, pokeweed mitogen, and phytohemagglutinin as a dose-dependent manner. This inhibitory effect was observed not only when $HgCl_2$ was added 2nd or 3rd day of 3 days culture period but also when spleen cells was pretreated with $HgCl_2$ for 2 hours. Mercury chloride, however, potentiated the production of IgM and IgG from spleen cells. During the $HgCl_2$ administration by drinking for 3 weeks, the weight gain of mice was significantly blunted than that o control group mice, while no overt signs related to mercury toxicity were noted in any mice of experimental group. There was no change in thymus and spleen weights, and in histological findings of kidney, bone marrow of femur, thymus, spleen, and popliteal lymph node after 3 weeks of mercury exposure. However, $HgCl_2$ induced a significant increase of total serum IgM, IgG including $IgG_1,\;IgG_{2a}\;and\;IgG_{2b}$, and IgE in Balb/c mice. Treatment in vivo with anti-IL-4 monoclonal antibody significantly abrogated the $HgCl_2$-induced increase in total serum IgG1 and IgE. Whereas $HgCl_2$ potentiated total serum IgM and IgG, there was no difference in total serum hemagglutinin to SRBC (Sheep Red Blood Cell) between experimental and control group mice when these mice were immunized with SRBC. All these findings observed in Balb/c mice suggest that mercury perturbates well-orchestrated regulation of immune responses before developing histopathological changes in lymphoid tissues.

  • Objective : This Experimental study was done to investigate the Effect of Taklisodok-um and Hwangryunhaedok-tang(TH) on Atopic Dermatitis. Methods : We assessed effects of TH on the IgE, IL-4, IL-5, IL-6, IgM, IgG1, IFN-${\gamma}$ in vivo, on the IL-4, IL-5, CCR3 in the skin tissues of ear and dorsum with NC/Nga mice. And we assessed effects of TH on the COX-2, IL-$1{\beta}$, TNF-${\alpha}$, IL-6 with RAW 264.7 cell. Results and Conclusion : 1. IgE, IL-4, IL-5, IL-6, IgM, IgGl levels in the serum of TH treated NC/Nga mouse group were decreased compared to the untreated control mice. IFN-r showed a increase in the experimental group compared to the untreated control group. The spleen weight of TH treated NC/Nga mice was decreased compared to the untreated control group. 2. mRNA expression levels of IL-4, IL-5 and CCR3 in the skin tissues of TH treated NC/Nga mice were decreased, and expression levels of IL-6 in the skin tissues of TH treated NC/Nga mice were decreased compared to the untreated control group. IFN-${\gamma}$ mRNA expression levels were increased compared to the untreated control group. 3. Judged from that IL-$1{\beta}$, TNF-${\alpha}$, IL-6 express of gene, effect of inflammatory Cytokines revelation were decreased compared to the untreated control group. 4. Depend on the strength of TH, inflammatory RAW 264.7 in the serum of TH were inhibited compared to the untreated control serum that leaded a COX-2 activity model. 5. Histological observation of the ear and skin tissues showed that the extents of inflammation and infiltrated immune cells in the epidermis and dermis of TH treated NC/Nga mice were highly reduced compared to the untreated control group.

Effect of Sopung-san Extracts on the Passive Cutaneous Anaphylaxis of Wistar Rats (Type I Allergic Dermatitis) (소풍산(消風散) 추출물이 수동 피부 아나팔락시스(1형 알러지 피부염)에 미치는 효과)

  • Purpose : The objective of present study is to detect the effect of Sopungsan aqueous extracts (SPS) on the passive cutaneous anaphylaxis (PCA; type I allergic dermatitis) Method : 500, 250 and 125mg/kg of SPS were orally administered 12 hr-interval before antigen challenge (total 4 times administered). PCA reactions were induced using rat anti-ovalbumin (OVA) serum contain IgE (titer 1:32) as sensitization and OVA as antigen challenge. 30 min after antigen challenge, the diameter of blue-dye spots (evans blue) and leaked amount of dye were observed with histology and histomorphometry at the PCA induced sites. In addition, serum total IgE and histamine levels were also observed by ELISA, respectively. The effects of SPS were compared with dexamethasone 1mg/kg treated rats in the present study. Results : As results of PCA reaction, vasodilation related increase of diameter of blue-dye spot and amount of leaked dye were observed with swelling and edematous changes in the dermis of PCA induced sites. However, these changes on PCA reactions were dramatically and dose-dependently decreased by treatment of SPS as compared with vehicle control. In addition, serum elevations of IgE and histamine were also dose-dependently inhibited by treatment of SPS as compared with vehicle control respectively. The effects of SPS 500mg/kg were similar to that of dexamethasone 1mg/kg in the present study. Conclusion : Base on the results of the present study, it is concluded that SPS has favorable effect on the PCA-induced allergic dermatitis, and SPS 500mg/kg showed similar favorable effects as compared with dexamethasone 1mg/kg. The present findings demonstrate that SPS can be effective for the prevention and treatment of allergic dermatitis.
