The Indian Magical Herb 'Sanjeevni' (Selaginella bryopteris L.) - A Promising Anti-inflammatory Phytomedicine for the Treatment of Patients with Inflammatory Skin Diseases

  • Objectives: Selaginella bryopteris L. (family: Selaginaceae), is often used in traditional Indian systems of medicine for the prevention and cure of several disorders and for the treatment of patient with spermatorrhoea, venereal disease, constipation, colitis, urinary tract infections, fever, epilepsy, leucorrhoea, beri-beri and cancer. It is also used as a strength tonic. This study aimed to evaluate the mechanisms underlying the anti-inflammatory effects of topically administered aqueous, polar and non-polar methanolic fractions ($10mg/20{\mu}L$) of Selaginella bryopteris. Methods: An acute oral toxicity study of Selaginella bryopteris at doses from 250 to 2,000 mg/kg body weight (bw) was performed. Aqueous, polar and non-polar methanolic extracts ($10mg/20{\mu}L$) applied topically for 5 days were evaluated for their anti-inflammatory effects against 12-tetra-O-decanoyl phorbol acetate (TPA)- induced inflammation by using the redness in the ear, the ear's weight (edema), oxidative stress parameters, such as lipid-peroxide (LPO) and nitric oxide (NO), and the pro-inflammatory cytokines involved in inflammation, such as tumour necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$ and IL-6. Indomethacine ($0.5mg/20{\mu}L$) was used for the positive control. Results: Selaginella bryopteris produced no mortalities when administered orally at doses from 250 to 2,000 mg/kg bw. Topical treatment with the non-polar methanolic fraction ($10mg/20{\mu}L$) significantly suppressed redness ($2.4{\pm}0.5$) and edema ($30.4{\pm}1$) and effectively reduced the LPO level ($32.3{\pm}3.3$). The NO level was ($8.07{\pm}0.55$), and the $TNF-{\alpha}$, $IL-1{\beta}$, and IL-6 levels were decreased to $69.6{\pm}15.5$, $7.7{\pm}4.8$ and $82.6{\pm}5.9$, respectively. Conclusion: This study demonstrated for the first time the mechanisms underlying the anti-inflammatory effect of medicinal plants like Selaginella bryopteris and quantified the pharmacological interactions between them. The present study showed this herbal product to be a promising anti-inflammatory phytomedicine for the treatment of patients with inflammatory skin diseases.

Effect of Chungsangboha-tang on LPS induced Anti-inflammatory in THP-1 cells (LPS로 유발된 대식세포의 염증반응에 대한 청상보하탕(淸上補下湯)의 효과)

  • Background and Objective : Chungsangboha-tang (CSBHT) has analgesic, sedative, anti-convulsive and anti-histamine effects, so it alleviates the symptoms of asthma. For the comparison of anti-inflammatory effect(s) on CSBHT, PD098059 was used as a negative control. Materials and Methods : This study emphasized THP-1 cells, which had been well characterized as a human monocytic leukemic cell line. The cells resemble monocytes with respect to several criteria and can be differentiated into macrophage-like cells by treatment with PMA. By using the MTS assay, it was possible to prove the safety of CSBHT. Results : Results shows that the CSBHT did not affected cell survival within $10^{1}$ ng/ml to $10^{5}$ ng/ml. Especially, $10^{5}$ ng/ml CSBHT treated cells show 70% deduction of $TNF-{\alpha}$ gene expression against that of LPS treated group. Furthermore, $IL-1{\beta}$, IL-6, IL-8, IL-10 and $TNF-{\alpha}$ levels are down-regulated when treated with CSBHT with concentrations up to 100 ug/ml on monocyte-derived macrophages. Interestingly, CSBHT-treated samples showed that overall transcriptional activities were down-regulated to 20% of that of PD098059 ($TNF-{\alpha}$ inhibitor). At protein level, the expression of $TNF-{\alpha}$ showed similar results as that of transcriptional activity. Results show that the protein level decreased more in the CSBHT-treated group (487 ${\pm}$ 87 pg/ml) than in the LPS-treated group (703 ${\pm}$ 103 pg/ml). In addition, the protein level of IL-8 in the CSBHT treated-group (9.84 ${\pm}$ 3.28 ng/ml) decreased similar as the expression of the control and PD098059-treated groups. Conclusion : CSBHT affects immune response, especially allergic responses and suppression of inflammatory reaction. The results provide us an alternative way to care for clinical inflammatory diseases, not only asthma but also the other possible general inflammatory and allergic diseases.

The effects of HYGB on various immunological factors related to pathogenesis of allergic dermatitis in NC/Nga mice (형개연교탕가미방의 아토피 피부염 병태모델에서의 면역조절능에 관한 연구)

  • Atopic dermatitis induced NC/Nga mice were used to investigate the efficacy of Hyunggaeyunkyotanggamibang(HYGB) on the recovery of dermatitic symptoms through its influence on the immune related factors. The results are as below: 1. HYGB treated group showed improvement of atopic dermatitis with naked eye observation, and significant decrease of dermatitis index was observed after 14 weeks. 2. HYGB treated group showed significant decrease of the ratio of CCR3+, B220+/IgE+, and CD11b+/Gr-1+ immune cells in dorsal skin by 41.7%, 21.5%, and 23.8%, respectively. 3. HYGB treated group showed an increase of CD19+ immune cells by 10.3% in PBMC, whereas CD3+, CD3+/CD69+, NKT+ immune cells were decreased by 4.3%, 42.9%, and 21.7%, respectively. 4. HYGB treated group showed an increase in the expression of IFN-$\gamma$ in the serum by 514.3%. However, the expressions of IL-4, IL-5, IL-6, IL-13, TNF-$\alpha$, MCP-1 and RANTES were decreased by 21.2%, 69.8%, 90.5%, 28.7%, 72.2%, 26.1%, and 19.9%, respectively. Also, the expression of IgE was decreased by 44.3%. 5. HYGB treated group showed a decrease of the expression of IL-4 and IL-5 by 43% and 44.3%. The results above indicated that HYGB clinically used for atopic dermatitis treatment has objective validity, and therefore can be provided as the basic data for EBM(Evidence based medicine) construction for anti-allergic and anti-inflammatory studies.

In vitro and in vivo anti-inflammatory and anti-allergic responses caused by water extract of Mori Cortex (상백피(桑白皮)가 항알러지 및 항염증반응에 미치는 영향)

  • Objective: This experimental study was performed to examine the in vitro and in viva anti-inflammatory and anti-allergic effects of Mori Cortex. Methods: Water extract of Mori Cortex was studied to its ability to stimulate or inhibit macrophage 264.7 cells to produce inflammatory and allergic mediators. Cytokines such as $IL-1{\beta}$, IL-6, IL-10 and $TNF-{\alpha}$ were measured by immunochemical assay. In vitro, the macrophages 264.7 were classified into four groups. One group was a normal group. The other group was a (-) control group stimulated with LPS. And the third group was a (+) control group pretreated for 1 hour with hydrocortisone. And the fourth group was a sample group pretreated for 1 hour with Mori Cortex. After pretreatment, macrophage were incubated with lipopolysaccharide(LPS) $100\;ng/m{\ell}$ for 12 hour and media collected and $IL-1{\beta}$, IL-6, IL-10 and $TNF-{\alpha}$ concentrations in supernatants were measured each by Enzyme linked immuno-soubent assay. Mori Cortex were used $50\;{\mu}g/m{\ell},\;100\;{\mu}g/m{\ell},\;250\;{\mu}g/m{\ell},\;500\;{\mu}g/m{\ell},\;and\;1,000\;{\mu}g/m{\ell}$. Hydrocortisones were used $10^{-8}M,\;10^{-7}M,\;10^{-6}M,\;10^{-5}M\;and\;10^{-4}M$. In vivo, the SD rats were classified into three groups. One group was a normal group injected with normal saline into the abdominal cavity. The other was a control group prescribed to compound 48/80 after normal saline injection. And the third was a sample group prescribed to compound 40/80 after Mori Cortex injection. Then, the release of histamine, IL-6 and $TNF-{\alpha}$ were measured. Results : In vitro, Man Cortex significantly increased the release of $IL-1{\beta}\;and\;TNF-{\alpha}$ by LPS-stimulated macrophage 264.7 cells. And it significantly decreased the release of IL-10. In IL-6, Mori Cortex of low concentration significantly decreased the release of IL-6, but that of high concentration acted in reverse. In vivo, Man Cortex didn't show significant inhibitory effects on the release of histamine and IL-6 in comparison with that of the control group. But it significantly increased the release of $TNF-{\alpha}$ in comparison with that of the control group.

Effects of Sesami Semen on Anti-Allergic Inflammation Mechanism related with Atopic Dermatitis (흑지마(黑芝麻)가 알러지성 염증 반응에 미치는 영향)

  • This study aimed to investigate the effects of Sesami Semen on the itching and anti-allergic inflammation mechanism related with cytokine, chemokine, histamine, $\beta$-hexosaminidase, NF-$\kappa$B, and free radical, and it was concluded as follows : 1. Sesami Semen did not show any cytotoxicity at the range of con-centration (1-250 ${\mu}g/m\ell$) on the human fibroblast cell (hFCs). 2. Sesami Semen reduced the gene expressions of IL-8, TNF-$\alpha$, IL-6 mRNA comparing with control. 3. Sesami Semen reduced the levels of IL-6, IL-8, MCP-1 within THP-1 cell depending on the concentration, and especially significantly reduced the the levels of IL-6, MCP-1 at all the concentration. 4. Sesami Semen significantly decreased the histamine secretion on HMC-1 at all the concentration. 5. Sesami Semen decreased the $\beta$-Hexosaminidase secretion 6.2% at 10 ${\mu}g$/ml conc., 58.3% at 100 ${\mu}g$/ml conc. and 63.2% at 200 ${\mu}g$/ml conc., respectively. And IC50 (${\mu}g$/ml) was 158.25 ${\mu}g$/ml. 6. Sesami Semen significantly suppressed the activity of NF-$\kappa$B promoter depending on the concentration. 7. Sesami Semen decreased the production of reactive oxygen species (ROS) and DPPH generation depending on the concentration. As judged with above results, the effects of Sesami Semen on the anti-allergic inflamation would be recognized, and it could be applied on the medicinal sources for prevention or treatment of several allergy disease. And more studies are needed furthermore with the seperation of effective materials.

  • To assess the relationship between anti-oxidant and immunomodulatory activities of Diospyros kaki Thunb. fruit, we harvested persimmons on the first Friday of every month from July to October, and conducted a DPPH assay, a T-bet promoter assay, and an ELISA for IL-4 determination. Anti-oxidant activity increased as fruit weight rose. When the fruit was extracted with five different solvents, and the extracts examined for radical scavenging activity, such activity exhibited a pattern similar to that seen when anti-oxidant activity was assessed. T-bet promoter activity decreased on ripening, whereas IL-4 expression increased, as tested in a DNFB-induced animal model. Collectively, the results suggest that inflammation-inhibitory activity, valuable in treatment of some immune diseases, rises as persimmons ripen.

The Effect of Bangpungtongsung-San on Model of Allergic Rhinitis (알러지성 비염 병태 모델에 대한 방풍통성산(防風通聖散)의 항알러지 효과)

  • Background and Objective : Allergic rhinitis is a IgE-mediated hypersensitive reaction at nasal mucosa. In oriental medicine, Bangpungtongsung-San is clinically widely used for the treatment of the allergic rhinitis. The objective of this study is to investigate the effects of Bangpungtongsung-San on allergic rhinitis experimentally. Material and Methods : BALB/c mouse were divided into there groups: intact, control, experimental groups. Control and experimental group were induced allergic rhinitis by Ovalbumin as· the method of Levin and Vaz. Experimental group was orally administered the Bangpungtongsung-San for 28days. Total IgE, Interleukin-4, Interleukin-5 and $Interferon-{\gamma}$ were measured at three groups. The statistical significance was examined by Independent Samples T test. Results : 1. The experimental group shows increase of $IFN-{\gamma}$ by 17% compared with control group but there was no statistical significance. 2. The experimental group shows increase of IL-4 and IL-5 compared with control group but there was no statistical significance. 3. The experimental group shows increase of Total IgE and diminution of OVA-specific IgE by 32% compared with control group but there was no statistical significance. Conclusion : The effect of Bangpungtongsung-San on allergic rhinitis is not immunological but seems to be anti-imflammatory and anti-stress effect.

Anti-inflammatory Effects of Various Mushrooms in LPS-stimulated RAW264.7 Cells

  • Mushrooms have been widely cultivated and consumed as foods and herbal medicines owing to their various biological properties. However, few studies have evaluated the anti-inflammatory effects of mushrooms. Here, we investigated the effects of mushroom extracts (MEs) on lipopolysaccharide (LPS)-induced inflammation in macrophages (RAW264.7 cells). First, we extracted MEs with either water or ethanol. Using LPS-treated RAW264.7 cells, we measured cell proliferation and NO production. Gene expression of tumor necrosis $factor-{\alpha}$ ($TNF-{\alpha}$), interleukin (IL)-6 (IL-6), and $IL-1{\beta}$ was assessed by RT-PCR, and protein abundance of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) and phosphorylation of p65 were determined by immunoblotting. MEs prepared using both water and ethanol inhibited LPS-induced inflammation in RAW264.7 cells. Nitric oxide (NO) levels induced by LPS were reduced by treatment with MEs. Isaria japonica Yasuda water extracts and Umbilicaria esculenta (Miyoshi) Minks ethanol extracts significantly decreased the mRNA expression of inflammation-related cytokine genes including $TNF-{\alpha}$, IL-6, and $IL-1{\beta}$. Similarly, the protein abundance of iNOS and COX-2 was also decreased. The phosphorylation of p65, a subunit of nuclear $factor-{\kappa}B$ was at least partly suppressed by MEs. This study suggests that mushrooms could be included in the diet to prevent and treat macrophage-related chronic immune diseases.

The Effects of Vitex rotundifolia Linne fil. Extract on the Inflammatory and Allergic Reactions (만형자 추출물이 염증 및 알레르기 반응에 미치는 영향)

  • Objectives : This study was carried out to investigate the anti inflammatory and anti allergy effects of Vitex rotundifolia Linne fil. extract(VRE). Results : 1. In vitro test, VRE was used to determine the modulation of cytokine secretion, the activation of inflammatory and allergic factor and the inhibition of gene expression. The cell survival rate of Raw 264.7 and Jurkat T cells didn't decrease and accordingly cytotoxicity wasn't observed. In anti-allergic assay, the secretion of IL-2, TNF-${\alpha}$, IL-4, IL-5 and IFN-${\gamma}$ were suppressed on Jurkat T cells induced by dust mites. And the gene expression of COX-2 was suppressed in HMC-1 stimulated by calcium ionophore A23187. In anti-inflammatory assay, the gene expression of TNF-${\alpha}$, COX-2 were suppressed on LPS-activated Raw 264.7 cells. And the secretion of IL-6 and IL-8 were suppressed on EoL-1 cells induced by dust mites. P38 and ERK activation of MAPK decreased generally. VRE showed potent inhibitory activity of NO production. 2. In vivo test, we used NC/Nga mouse induced by atopic dermatitis to observe the effects of VRE on the weight, water and feed, blood test, weight of organs, total IgE and histological change of main organs. Quantity of water and feed were not changed, therefore it didn't affect the weight directly, and no change was observed in related main organs, thus maybe there is no organ toxicity by test substances. And the symptoms were decreased significantly, and the thickness of epithelial cell layer and the number of mast cells were inhibited significantly by the difference of dosage. The number of total complete blood cells and IgE in serum were not changed significantly. Conclusion : These results suggest that VRE has anti-inflammatory and anti-allergic effects. Therefore VRE could be used effectively on improvement or treatment of atopic dermatitis. However, further study is needed to prove which component of VRE indicates effective pharmacological action.

Synergic Effect of Quercetin and Astragalin from Mulberry Leaves on Ani-inflammation (상엽 유래 퀘세틴과 아스트라갈린의 항염증에 대한 상승효과)

  • The leaf of mulberry (Morus alba L) has long been used in Oriental medicine for the prevention or treatment of several diseases. However, little is known about the inhibitory effects of a single compound isolated from the mulberry leaves on inflammatory response. We are isolate a single compound of quercetin (3,3',4',5,7-pentahydroxyflavone) and astragalin (kaempferol-3-O-glucopyranoside) from the mulberry leaves, and then investigate the anti-inflammatory effects of quercetin, astragalin or quercetin plus astragalin in lipopolysaccharide (LPS)-stimulated murine peritoneal macrophages. Each compound suppressed the production of inflammatory mediators (NO, $PGE_2$ and IL-6) in LPS-stimulated murine peritoneal macrophages in a dose-dependent manner. Especially, the cotreatment of quercetin (2.5 ${\mu}M$) and astragalin (2.5 ${\mu}M$) markedly suppressed the production and the expression of inflammatory mediators. These suppressive effects were synergistically increased by their combination. These results suggest that the combination of quercetin and astragalin from the mulberry leaves may be useful for therapeutic drugs against inflammatory immune diseases, probably by suppressing the production of inflammatory mediators.
