Characterization of Antioxidant, Melanogenic activity of Fuctions by Sanmal and D-Mannuronic Acid (산말의 항산화 및 멜라닌 생성 억제활성)

  • This study investigated the functional study in anti-oxidant and anti-melanin activity by Desmarestia dudresnayi subsp. Tabacoides, Desmarestia viridis, Desmarestia ligulate leaf extracts. The DPPH radical scavenging activity of D. viridis extract showed a high antioxidant activity of 68.0 ± 1.9% at the concentration of 2.5 mg / mL, the ABTS and scavenging activity showed a high antioxidant activity of 84.6 ± 1.7% at the concentration of 2.5 mg / mL, and the NO radical scavenging activity was 2.5. It required a high antioxidant capacity of 91.6 ± 1.1% at mg/mL concentration. In addition, in B16F10 cells, at a concentration of 100 ㎍/mL, D. viridis extract is 35.0 ± 1.7% melanin among, which has the lowest amount of melanin among the three species. With these contents, it seems that there is a possibility that D. viridis could be developed as a cosmetic material. These results D. viridis considered that it can be used as a natural cosmetic material such as anti-oxidant and anti-melanin activity.

Effect of Yeonsan Ogye bioactive peptides on anti-oxidant indexes in rats' liver

  • Purpose: This study investigated the effect of bioactive Yeonsan Ogye peptides (YOPs) intake on changes in the hepatic anti-oxidant indexes in male rats. Methods: Sprague-Dawley male rats were divided into 3 groups and given a casein-based AIN-93G diet and distilled water ad libitum without any added YOPs (control), distilled water with 250 mg of YOPs (Y250), or 500 mg of YOPs (Y500) per kg of body weight for 4 weeks. YOP dose was decided as referred to in the referenced study where toxicity did not occur. The hepatic anti-oxidant indexes were determined using a commercial kit. Statistical analysis was performed using SPSS version 23.0 and are expressed as $mean{\pm}standard$ error of mean. Differences among the groups were evaluated by one-way analysis of variance followed by post hoc Duncan's multiple comparisons test. Results: There were no differences in the body weights, weight gain, food intake, food efficiency ratio, or organ weight, including liver, kidney, spleen, thymus, and epididymal fat, among all of the groups. The hepatic nitric oxide (NO) level in the Y500 group was lower than that in the control and Y250 groups, and the hepatic malondialdehyde (MDA) level was lower in the Y500 group than in the Y250 group. The differences in hepatic superoxide dismutase (SOD) and catalase (CAT) activities were not statistically significant between the groups. From these results we speculated that YOPs may have anti-oxidative abilities to regulate NO and MDA production without affecting SOD and CAT activities. Conclusion: YOPs are presumed to act as anti-oxidants in the animal or human body.

Bioconversion of Gentiana scabra Bunge increases the anti-inflammatory effect in RAW 264.7 cells via MAP kinases and NF-κB pathway

  • Mitogen-activated protein (MAP) kinases play an important role in cell growth and differentiation, as well as the modulation of proinflammatory cytokines. The objective of this study was to examine the increase in the anti-inflammatory effect of Gentiana scabra Bunge (GSB), due to bioconversion with the Aspergillus kawachii crude enzyme, via inhibition of the $NF-{\kappa}B$ signaling and MAP kinase pathways in RAW 264.7 cells. The expression of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 in RAW 264.7 cells treated with the GSB ethyl acetate fraction bioconverted with A. kawachii crude enzyme (GE-BA), was dramatically suppressed as compared to GSB ethyl acetate fraction non-bioconverted with the A. kawachii crude enzyme (GE-UA). The phosphorylation of p38, extracellular signal-regulated kinases, and inhibitory ${\kappa}B$ in RAW 264.7 cells treated with GE-BA was further suppressed, as compared to exposure to GE-UA. Moreover, the mRNA expression of interleukin 6, interleukin 1-beta, and tumor necrosis $factor-{\alpha}$ was further suppressed by GE-BA, compared to GE-UA. Similarly, anti-oxidant activities, such as 2,2-diphenyl-1-picrylhydrazyl hydrate and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) radical scavenging activity, of GE-BA were further increased compared to GE-UA. These observations demonstrate that the anti-oxidant and anti-inflammatory activities of GSB ethyl acetate fraction increases as a result from bioconversion with the A. kawachii crude enzyme.

Anti-oxidant, Anti-aging, and Whitening Effects of Viscum album var. coloratum In Vitro (한국산 겨우살이의 항산화, 항노화 및 미백 효과)

  • In this study, we investigated the anti-oxidant, anti-aging, and skin whitening effects of Korean mistletoe (Viscum album var. coloratum). The mistletoe fraction was composed of four types: hexane (HX), ethyl acetate (EA), butanol (BU), and methylene chloride (MC). In total phenol content assay, HX showed the highest phenol content among four fractions. In addition, EA significantly increased 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging and catalase-like activities, and MC significantly increased superoxide dismutase (SOD)-like activity. When we compared IC50 value in the hyaluronidase and elastase inhibition assay, MC had the lowest IC50. In addition, we also performed tyrosinase inhibition assay to demonstrate the possibility of Korean mistletoe as a cosmetic component. HX showed the highest tyrosinase inhibition rate among the fractions.

Anti-oxidant and Anti-inflammatory Potentials of Sasa quelpaertensis Leaf Residue Extracts (제주조릿대 잎 잔사 추출물의 항산화 및 항염 활성)

  • Sasa quelpaertensis Nakai is a native Korean plant that grows only on Mt. Halla of Jeju Island. Its leaf is used for a popular bamboo tea due to its various health-promoting properties, and it has been increasingly used as food and cosmetic ingredients. To utilize the S. quelpaertensis leaf efficiently, the preparation method for phytochemical-rich extract (PRE) using the leaf's residue was previously reported, which was produced after hot water extraction. This study was undertaken to evaluate the anti-oxidant and anti-inflammatory potential of PRE and its solvent fractions. The ethyl acetate fraction of PRE (EPRE) showed higher DPPH, ABTS, and superoxide radical scavenging activities, and it effectively inhibited intracellular reactive oxygen species (ROS) and nitric oxide (NO) production in lipopolysaccharides (LPS)-stimulated RAW 264.7 cells. EPRE also induced the expression of heme oxygenase-1 (HO-1) by increasing the level of nuclear factor E2-related factor 2 (Nrf2) in a nuclear fraction. The inhibiting effect of EPRE on LPS-induced NO production was partially reversed by the HO-1 inhibitor (zinc protoporphyrin, ZPP), suggesting that HO-1 is involved in suppressing NO production. Taken together, the results suggest that EPRE has potential as a promising anti- oxidant and anti-inflammatory agent.

In vitro correlation and analysis of anti-oxidant and anti-inflammatory activities by fruit ripening of peach cultivars (복숭아 품종별 성숙정도에 따른 항산화 및 항염증 효과간의 상관관계)

  • This study was performed to investigate the correlation between anti-oxidant and anti-inflammatory activities in ripe and unripe fruits of three peach cultivars: Miwhang (MH), Kanoiwa hakuto (KH) and Cheonhong (CH). The unripe fruits had higher levels of total phenols and flavonoids contents than those in the ripe fruits of all the three cultivars. The unripe fruits of CH showed the highest levels of total phenols, flavonoids and antioxidant activities among the fruit samples analyzed. Nitric oxide inhibition values in RAW 264.7 cells for the unripe fruits of MH and KH were 30 and 29%, respectively. However, the inhibition was not observed in unripe CH and the ripe fruits of either cultivar. Total phenols and flavonoids contents showed high linear correlations with the anti-oxidant activities whereas the anti-inflammatory activity had low linear correlations with them.

  • Tremendous natural product extracts were used as a herb medicine remedy or therapy for centuries. Because these extracts have various biological activities, we examined the effects of Gryllotalpa orientalis extract for anti-oxidant and anti-inflammation activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing ability of plasma (FRAP), hydroxy radical scarvenging and cyclooxygenase-2 promoter assays. Gryllotalpa orientalis extracts were prepared from solvents such as distilled water (DW), dimethyl sulfoxide (DMSO), ethanol and methanol. The results showed that Gryllotalpa orientalis extracts have potent DPPH (methanol extract), FRAP (DW extract) and hydroxy radical scavenging (DW and methanol extracts) activity than any other extracts used. A significant inhibition of cyclooxygenase-2 (Cox-2) promoter activity was detected in the presence of DMSO extract or ethanol extract. Collectively, the present results suggested that Gryllotalpa orientalis extract could be used for anti-oxidant and/or anti-inflammation agent for human or agricultural purposes.

Antioxidant Activity and Melanin Inhibitory Effects of Yambean (Pachyrhizus erosus) Extract (얌빈 추출물의 항산화 효능과 멜라닌 생성 억제효과)

  • Objectives : Yam bean (Pachyrhizus erosus) possess various nutrients, it has been widely used as traditional cosmetic material in Indonesia. The aim of this study was to investigate the anti-oxidant activity and the anti-melanogenic effect of Yambean (Pachyrhizus erosus) extract and its fractions. Methods : The anti-oxidant activity of yam bean extract assessed based on total polyphenol, flavonoid contents, DPPH and ABTS radical scavenging assay. To evaluate anti-melanogenic effects and cytotoxicity of Yambean extract and its fractions, B16F10 melanoma cell was used. Results : In results, total polyphenol content of yam bean water extract (YW) and Yambean 70% ethanol extract (YE) were $1.18{\pm}0.03mg/g$ (mg of gallic acid/g of sample), $1.16{\pm}0.01mg/g$. Total flavonoid contents of YW, YE were $3.55{\pm}0.06mg/g$ (mg of naringin/g of sample), $1.78{\pm}0.03mg/g$. Moreover, YE scavenged DPPH and ABTS effectively in $4mg/m{\ell}$ compared to YW. Cytotoxicity of YE and its fractions in B16F10 melanoma cell was measured using MTT assays. It had no cytotoxicity up to $500{\mu}g/m{\ell}$. Melanin accumulation in B16F10 melanoma cell was induced using alpha-melanocyte stimulating hormone (${\alpha}-MSH$) and 3-isobutyl-1-methylxanthine (IBMX). B16F10 melanoma cell treated with $10-500{\mu}g/m{\ell}$ YE and hexane, ethyl acetate, butanol, $H_2O$ fractions for 24h. Non treated B16F10 melanoma cell (Control) markedly increased melanin contents. In contrast, YE ethylacetate fraction effectively suppressed melanin accumulation in a dose-dependent manner. Conclusion : In conclusion, these results suggest that Yambean extract has the potential as a cosmetic material which possess anti-oxidant and anti-melanogenic activities.

Antioxidative, Anti-inflammatory Effects of Jibaekjihwang-tang (zhibaidihuang-tang) on Osteoarthritic Rat Model (지백지황탕(知栢地黃湯)의 골관절염 Rat 모델에 대한 항산화, 항염증 효과)

  • Objectives This study intended to evaluate antioxidative, anti-inflammatory effects of Jibaekjihwang-tang on monosodium iodoacetate (MIA)-induced osteoarthritic rat model and investigate the potential mechanism. Methods Jibaekjihwang-tang (100 or 200 mg/kg body weight) was orally administered once daily for 2 weeks days from day 7 after intra-articular MIA injection. And blood analysis, the histologic examinations were performed. Moreover, protein expressions related to anti-oxidant and cartilage degradation and anti-inflammatory cytokines were measured by western blot analysis in cartilaginous tissue. Results Jibaekjihwang-tang reduced serum inflammatory cytokines such as tumor necosis factors-α and interleukin-6. Furthermore, the increase of anti-oxidant enzymes reversed the oxidative stress caused by MIA. Meanwhile, Jibaekjihwang-tang suppressed MIA-induced inflammation and cartilage degradation in cartilaginous tissue. Conclusions Jibaekjihwang-tang alleviated MIA-induced inflammation. Jibaekjihwang-tang was associated with a protective effect on cartilage and by reducing inflammation and cartilage degradation. These findings provide new approaches for understanding osteoarthritis therapy.

  • This paper is concerned with the studies on the effective components of anti-oxidant activity, with a view to demonstrate the anti-aging activity of Korean ginseng. Feeding the extract of Korean ginseng or its effective component to mice inhibited strongly the induction of lipid peroxidation produced by ethanol intoxication. From the extract of Korean red ginseng, one effective component Compound A, mp.143, $C_6H_6O_3$ was isolated by chromatographic purification and its chemical structure was determined as 2-meth-y1-3-hydroxy-${\gamma}-pyrone(maltol).$
