Comparative effects of systemic administration of levofloxacin and cephalexin on fracture healing in rats

  • Objectives: This study aimed to compare the effects of systemic administration of levofloxacin or cephalexin on fracture healing in rats. Materials and Methods: In this animal study, tibial fractures not requiring fixation were artificially induced in 30 male Wistar albino rats using a 1.1 mm surgical bur. The rats were randomly divided into 6 groups (n=5). Groups 1 and 2 received daily subcutaneous saline injections. Groups 3 and 4 received subcutaneous injections of 25 mg/kg levofloxacin twice daily. Groups 5 and 6 received daily subcutaneous injections of 20 mg/kg cephalexin. The rats in Groups 1, 3, and 5 were sacrificed after 1 week, while the rats in Groups 2, 4, and 6 were sacrificed after 4 weeks. The score of fracture healing was determined through histological assessment of sections from the fracture site according to Perry and colleagues. Data were analyzed by Kruskal-Wallis and Mann-Whitney tests. Results: The mean score of fracture healing at 4 weeks was significantly higher than that at 1 week in the saline, levofloxacin, and cephalexin groups (P<0.001). At 1 week, no significant difference was noted among the three groups of saline, levofloxacin, and cephalexin in the mean score of fracture healing (P=0.360). However, this difference was significant at 4 weeks (P=0.018), and the mean score in the saline group was significantly higher compared to that in the levofloxacin group (P=0.015). Conclusion: It is recommended not to prescribe levofloxacin for more than 1 week after surgical management of bone fractures due to its possible adverse effects on fracture healing.

홍화씨 Youghurt 급여가 난소절제 Rat의 골다공증에 미치는 영향 (Effects of Safflower Seed Yoghurt on the Osteoporosis in Ovariectomized Rat)

  • 홍화씨 yoghurt가 골다공증에 미치는 영향을 조사하기 위하여 sham-operation되어 6주간 정상식이한 군을 대조구로 하였고, 난소 절제하여 6주간 저칼슘식이한 군을 정상식이군(G I), yoghurt를 첨가하여 사육한 군(G II) 및 1%${\sim}$4%홍화씨yoghurt를 첨가하여 사육한 군(GIII${\sim}$GVI)으로 구분하여 실험식이를 급여하였다. 사료섭취량, 체중 증가량 및 사료효율을 알아보았고, 왼쪽 대퇴골의 무게, 밀도, 길이, 강도, 회분 및 무기질 함량을 조사하였다. 또한 오른쪽 대퇴골의 조직을 SEM으로 관찰하였다. 사료섭취량은 대구조와 실험구간 유의적인 차이는 없었다. 사료효율 및 체중 증가량은 홍화씨 yoghurt 식이구가 유의적으로(P < 0.05) 높았으며, 특히 GV와 GVI구가 높게 나타났다. 왼쪽 대퇴골의 밀도, 강도, 회분 및 미네랄 함량은 대조구와 G I간에 유사한 경향을 나타내었고, 홍화씨 첨가수준이 높을수록 유의적으로 높게 나타났다. 반면 무게와 길이는 전 실험구에 있어서 거의 일정하게 나타났다. 전자현미경으로 측정한 결과 4% 홍화씨 yoghurt 식이구(GVI)가 가장 골다공증 치유효과가 높은 것으로 나타났다. 이상의 결과 홍화씨 yoghurt 식이가 난소 절제 및 저칼슘식이를 한 rat의 골다공증에 미치는 효과에 있어서 정상식이 및 yoghurt 식이구보다 홍화씨 yoghurt 식이구가 높은 치유효과를 보였으며 가장 우수한 것으로 나타났다.

치유농업 이용의도에 영향을 미치는 변인 고찰 (A Study on the Variables Affecting the Intention to Use Healing Agriculture)

  • 최근 급격히 증가하는 노인들과 정신적 휴식이 필요한 도시인들의 신체적 정신적 건강과 안녕감을 유지하기 위한 방법으로써 치유농업에 관한 다양한 연구가 이루어지고 있다. 본 연구는 치유농업 이용의도에 영향을 미치는 변인고찰을 위하여 노후준비도, 농촌치유 공급조건, 농촌치유 제공서비스를 독립변수로 정하여 연구하였다. 본 연구의 목적은 첫째, 치유농업 이용의도에 영향을 미치는 다양한 요인들을 실증적 분석을 통하여 파악하고자 한다. 둘째는 다양한 요인들 중에서 어느 변인이 이용의도에 더 많은 영향을 미쳤는지를 검증하고자 한다. 셋째, 이들 검증결과를 바탕으로 치유농업 활성화 정책을 제언하는 데 있다. 본 연구를 위해서 30세 이상 성인 남녀를 대상으로 설문조사를 실시하여 최종 356부를 분석대상으로 확정하였다. 연구가설의 검증은 선형회귀분석을 통하여 검증하였다. 분석결과는 다음과 같다. 첫째, 노후준비도의 신체적 준비와 정서적 준비 그리고 경제적 준비는 이용의도에 유의한 영향을 미치는 것으로 조사되었다. 둘째, 농촌치유 공급조건의 자연경관, 접근성, 안전성은 이용의도에 유의한 영향을 미치는 것으로 조사되었다. 그러나 경제성과 전문성은 기각되었다. 셋째, 농촌치유 제공서비스의 농작물 재배, 동물매개, 치유시설은 이용의도에 유의한 영향을 미치는 것으로 검증되었다. 이러한 연구를 통해, 치유농업의 이용의도를 높이기 위해서는 소비자의 노후준비가 잘되어 있어야 하며, 농촌치유 공급조건에서는 접근성과 안전성이, 농촌치유제공 서비스에서는 치유시설이 잘 갖춰져야 치유농업을 이용하려는 소비자의 이용의도를 높일 수 있다는 결론을 도출하였다.

Effect of microthreads on removal torque and bone-to-implant contact: an experimental study in miniature pigs

  • Purpose: The objective of this study was to evaluate the effect of microthreads on removal torque and bone-to-implant contact (BIC). Methods: Twelve miniature pigs for each experiment, a total of 24 animals, were used. In the removal torque analysis, each animal received 2 types of implants in each tibia, which were treated with sandblasting and acid etching but with or without microthreads at the marginal portion. The animals were sacrificed after 4, 8, or 12 weeks of healing. Each subgroup consisted of 4 animals, and the tibias were extracted and removal torque was measured. In the BIC analysis, each animal received 3 types of implants. Two types of implants were used for the removal torque test and another type of implant served as the control. The BIC experiment was conducted in the mandible of the animals. The $P_1-M_1$ teeth were extracted, and after a 4-month healing period, 3 each of the 2 types of implants were placed, with one type on each side of the mandible, for a total of 6 implants per animal. The animals were sacrificed after a 2-, 4-, or 8-week healing period. Each subgroup consisted of 4 animals. The mandibles were extracted, specimens were processed, and BIC was analyzed. Results: No significant difference in removal torque value or BIC was found between implants with and without microthreads. The removal torque value increased between 4 and 8 weeks of healing for both types of implants, but there was no significant difference between 8 and 12 weeks. The percentage of BIC increased between 2 and 4 weeks for all types of implants, but there was no significant difference between 4 and 8 weeks. Conclusions: The existence of microthreads was not a significant factor in mechanical and histological stability.

동종요법을 이용한 개의 분리불안과 안과질환 치료 4례 (The Effect of the Pulsatilla 30C as Homeopathy for Ophthalmic Diseases with Concomitant Separation Anxiety)

  • Homeopathic Pulsatilla 30C was evaluated in 4 clinical cases of ophthalmic complications with concomitant separation anxiety in dogs. Separation anxiety in dogs is excessive anxiety regarding to the separation from owners and is one of the most common behavioral problems in dog. In addition, ocular diseases is sometimes related with abnormal behaviors. Holistic approach is good alternative method to treat dogs with physical problem associated behavior problem. Homeopathy is one of the complementary and alternative medicines and bases on healing of patients themselves which includes mind healing, not the disease itself. Among the material medical remedy of homeopathy, the Pulsatilla is used as homeopathic remedy for ocular problems, earache, cough, cold, and anxiety from lost attention. These case studies described the successful treatment of ocular diseases and separation anxiety by application of Pulsatilla 30C as homeopathy in 4 dogs. During the homeopathy, these dogs did not receive any other treatments, and were performed the physical exam to check the ocular problems. These dogs were completely cured of physical and behavior problem within 10 days. On following up, any clinical signs were not observed at one month after the last therapy. In addition, as from the treatment the anxiety problem of patients has been much better improved. According to these case studies, we know that an application of homeopathic remedy may be useful therapeutic choice to treat both problems of physical and behavior.

Mitochondria-Targeted Vitamin E Protects Skin from UVB-Irradiation

  • Mitochondria-targeted vitamin E (MVE) is designed to accumulate within mitochondria and is applied to decrease mitochondrial oxidative damage. However, the protective effects of MVE in skin cells have not been identified. We investigated the protective effect of MVE against UVB in dermal fibroblasts and immortalized human keratinocyte cell line (HaCaT). In addition, we studied the wound-healing effect of MVE in animal models. We found that MVE increased the proliferation and survival of fibroblasts at low concentration (i.e., nM ranges). In addition, MVE increased collagen production and downregulated matrix metalloproteinase1. MVE also increased the proliferation and survival of HaCaT cells. UVB increased reactive oxygen species (ROS) production in fibroblasts and HaCaT cells, while MVE decreased ROS production at low concentration. In an animal experiment, MVE accelerated wound healing from laser-induced skin damage. These results collectively suggest that low dose MVE protects skin from UVB irradiation. Therefore, MVE can be developed as a cosmetic raw material.

화상 및 창상에 대한 자근.치자 복합제제의 경피 흡수 및 치료효과 (Transdermal Drug Delivery & Therapeutic Effect of the Preparations of Lithospermi Radix and Gardeniae Fructus Extracts on the Burn & Wound Healing)

  • Research was undertaken to compare the pharmacological activity of Lithospermi radix (LR) reported as an oriental medicine for classical uses. LR contains naphthoquinone pigments : shikonin, acetylshikonin, isobutylshikonin, etc. LR is used for the treatment of excision wound, burn, eczema, blister, scarlatina and septicemia as antifebrile, antidotic and antiphlogistic. Gardeniae fructus (GF) has been used for the treatment to jaundice, hepatic disease, anti-inflammatory and analgesic effects, and it contains crocin, geniposide and its derivatives. The therapeutic effects of burn and excision wound healing from LR & GF hydrogel with $Nano-ATP^{\circledR}$ (GLN) were investigated. To evaluate the therapeutic value of various hydrogels, thermal burn model and excision wound mouse model were used. The burn and wound reduction rate and therapeutic period were measured to calculate the healing extent after 5 experiments. The 2nd degree burn was prepared on hairless mouse back skin and dressing with collagen. The burn and wound reduction rate of GLN hydrogel treated group decreased more rapidly than that of other gel group in animal model. Furthermore therapeutic periods of GLN hydrogel treated group was shorter than that of other gel group. In anti-inflammatory test, GLN hydrogel treated group decreased edema rapidly than that of other gel group. These results suggest that the GLN hydrogel treatment has an therapeutic effect on burn and excision wound healing.

Soft and Hard Tissue Augmentation with/without Polydeoxyribonucleotide for Horizontal Ridge Deficiency: A Pilot Study in a Dog Model

  • Purpose: To investigate the effects of simultaneous soft and hard tissue augmentation and the addition of polydeoxyribonucleotide (PDRN) on regenerative outcomes. Materials and Methods: In five mongrel dogs, chronic ridge defects were established in both mandibles. Six implants were placed in the mandible, producing buccal dehiscence defects. The implants were randomly allocated to one of the following groups: 1) control: no treatment; 2) GBR: guided bone regeneration (GBR) only; 3) GBR/PDRN: GBR+PDRN application to bone substitute particles; 4) GBR/CTG: GBR+connective tissue grafting (CTG); 5) GBR/VCMX: GBR+soft tissue augmentation using volume stable collagen matrix (VCMX); and 6) group GBR/VCMX/PDRN: GBR+VCMX soaked with PDRN. The healing abutments were connected to the implants to provide additional room for tissue regeneration. Submerged healing was achieved. The animals were euthanized after four months. Histological and histomorphometric analyses were then performed. Results: Healing abutments were gradually exposed during the healing period. Histologically, minimal new bone formation was observed in the dehiscence defects. No specific differences were found between the groups regarding collagen fiber orientation and density in the augmented area. No traces of CTG or VCMX were detected. Histomorphometrically, the mean tissue thickness was greater in the control group than in the other groups above the implant shoulder (IS). Below the IS level, the CTG and PDRN groups exhibited more favorable tissue thickness than the other groups. Conclusion: Failure of submerged healing after tissue augmentation deteriorated the tissue contour. PDRN appears to have a positive effect on soft tissues.

Red Deer Antler Extract Accelerates Hair Growth by Stimulating Expression of Insulin-like Growth Factor I in Full-thickness Wound Healing Rat Model

  • In order to investigate and evaluate the effects of red deer antlers on hair growth in the full-thickness wound healing model, Sprague-Dawley rats were given incision wounds through the full thickness of their dorsal skin and deer antler was applied for 40 days. At specified intervals thereafter (4, 8, 16, 32 and 40 days), the animals were sacrificed and the wound site skins were excised, processed, and sectioned. At post-injury days 16, 32 and 40, longer and more active new hair appeared around the healing wound of antler-treated skin. Histological studies showed that the antler extract markedly increases the depth, size, and number of hair follicles. Expression of IGF-I (insulin-like growth factor) mRNA was detected by RT-PCR and real time RT-PCR. The result showed that the expression of IGF-I (days 16, 32, and 40) was obviously up-regulated in antler-treated skins compared to control skins. Similar results were seen in the ELISA analysis to quantify the IGF-I expression. These results support the notion that wound healing can cause hair growth by enhancing the expression of IGF-I. Deer antler extract appears to have the potential to promote hair growth and could be used in hair growth products.

비타민 D$_3$와 인산칼슘의 토끼 대퇴골 골절치유 효과에 대한 골수강내 정맥 조영술 (Intraosseous Phlebography for Determination of the Effect of Vitamin D$_3$ and Calcium Phosphate on Fractured Bone Healing in Rabbit)

  • Intraosseous phlebography was performed to determine the healing effect of the dosing treated with vitamin-D$_3$ and/or calcium phosphate on the fractured bone in male rabbits. Femoral shafts of 16 adult male rabbits were fractured and these animals were divided into 3 treated groups and 1 control group l. The 1st treated group of rabbits was dosed with vitamin-D$_3$(VD), the 2nd treated group of rabbits was given calcium phosphate(CP), and the 3.d treated group of rabbits received vitamin-D$_3$ and calcium phosphate. The control group of rabbits was dosed with saline alone. Radiographs were taken weekly in each rabbit over 6 weeks period and the results obtained were as follows. 1. Sinusoidal vein was visualized at 3rd week in eve animal received CP and VC, at 5 week in animals received VD and in control animals. 2. Main nutrient vein was observed more clearly in treated groups than in control group of animals. 3. These results indicate that venous reconstruction of the fractured bone in groups of rabbits received CP and VC was earlier and more complete than those of other groups of rabbits. 4. Therefore, intraosseous phlebography is suitable for determination of the bone healing effect of vitamin-D$_3$ and/or calcium phosphate in femoral fractured rabbits.
