  • Decursin is a major component of the root of Angelica gigas(Umbelliferae), which has been traditionally used in Korea as a tonic and to treat anemia, hemiplegia, and women's diseases. The objective of this study is to identify the anti-cancer mechanism induced by decursin on apoptosis of human leukemia and lymphoma cells. Cytotoxicity of decursin on U937, HL-60, MOLT-4, THP-1 cells showed the significant effects. First of all, $IC_{50}$ of decursin on four cell lines was 27.1, 32.4, 17.4, $15.1{\mu}M$, respectively. So $IC_{50}$ in THP-1 cells was the smallest among 4 cell lines treated with decursin($15.1{\mu}M$). In order to understand the apoptosis-mechanism by decursin, we examined the gene expression of bcl-2(anti-apoptotic), bax(pro-apoptotic) and p53(tumor suppressor)after treating the THP-1 cells with decursin(10, 50 and $100{\mu}M$). It was found bcl-2 gene was decreased dose dependently, the expression level of bax gene of THP-1 cells treated with $100{\mu}M$ of decursin was about 3 times higher than those of control, and p53 gene was increased In the same concentration($100{\mu}M$), p53 gene was increased dose dependent manner. In protein express, bcl-2 and p53 protein showed a tendency to decrease. bax was increased about 4 fold. Therefore decursin is a useful chemotherapeutic agent against leukemia.

  • This experiment was conducted to obtain the basic information for the increase ofgermination percent by the treatment of stratification, soaking and $Gibberellin(GA_3)$ in Angelica gigas Seeds caltivated in 1989. The germination percent was increased by 30% with stratification, and the effect of low temperature did not differ significantly. The optimum soaking temperature and periods were found to be 4days at $20^{\circ}C\;and\;6dys\;l0^{\circ}C$ of water temperature. When exchanged water every day during soaking period, inhibitors were more extracted than non-exchange one. The exogenous application of $GA_3$ also increased germination percent about 20% campared with control, and the reasonable level was 2ppm in soaked seed and 5ppm in non-soaked seed.

  • This study was carried out to control the decrease of yield and the deterioration of quality of Angelica gigas roots by bolting. This phenomenon resulted from lignificartion and dwarfishness of roots because of bolting formed for inflorescence and fruition. Seedlings were grown for 60,90 and 120 days in nursery of the green house, and transplanted in main field to compare with traditional seedling that grew for 1 year. Growth of the petiole length compare with length and leaf width of aerial parts in nursery showed a remarkably progress, and root characters as root length, root diameter and root weight grew more vigorously in the order of nursery periods. Bolting was not entirely formed in plots transplanted the seedlings which grown for short-time; 60, 90 and 120days, but showed the high bolting rate, 64.3%,in traditional seedling plots. Yield was higher in plots that transplanted short-time growing seedlings than that transplanted traditional seedlings, and among 3 kinds of nursery periods yields were high in the order of nursery periods. For the increase of yield and decrease of bolting rate, it was needed to transplant small seedlings of 3-4g/plant which grew in nursery for 90-120 days.

  • Angelica gigas has been a major medicinal crop in Korea for a long time. This study was to determine optimum seeding time and bolting response by shifting seeding time between November 10, 1993, and August 10, 1994 using promising line, Suwon 2. Days to emergence were similar among seeding times from April 10 to August 10 in Suwon. Seedling duration was shortened 92 days when seeded on July 10 compared with the conventional seeding time of April 10. Productivity of seedling ranging from 0.31 to 0.70cm in diameter was highest of 730 plants per $m^2$ in July 10 seeding. When seeded in July 10, bolting rate was 2.4 percent as compared with 2.6 percent in conventional seeding (April 10). The optimum seeding time was July 10. Seedling quality, survival rate after planting in field, and root yield were greatly influenced by seeding time.

  • This studies were conducted to basic information on growth characteristics and root yield in progeny the derived from different bolting years and to establish optimum seed production time in Angelica gigas. Plant height of early and mid-growing stages had decreased with each year increase in bolting year, but showed no significance in late growing stage at different bolting years. Root characteristics including root length, root diameter and dry root weight was slightly good and it had high root/ shoot ratio in third year seed production. respectively. Bolting rate showed that first year seed 38.1%, second year seed 10.4% and third year seed 1.2%, respectively. Therefore, optimum seed production time revealed third year seed for reduce bolting rate. Root yield at different bolting years showed average 1,690kg per hectare as fresh root weight on first year seed, 2,860kg on second year and 2,940kg on third year seed, respectively. A significant positive correlation was appeared between bolting rate and plant height in mid-growing stage. On the other hard, there was highly significant negative correlation between bolting rate and root yield.

  • The purpose of this study was to investigate the biological activities of an aqueous extract of Angelica gigas (Ag) fermented by Saccharomyces cerevisiae (Sc). First, the soluble solids of the F/3 group, in which the Ag was fermented by Sc for 3 days, decreased from $1^{\circ}Bx$ to $0.9^{\circ}Bx$. On the other hand, the pH increased with the number of days of fermentation. The result of a TLC experiment confirmed that it gradually decomposed into a low-molecular weight sugar form upon fermentation. The total phenolic compounds and flavonoid contents were higher in the fermented group than in the non-fermented group. K and Ca contents were increased by fermentation in the following order: F/3, NF, and F/0 groups. Decursin and decursinol angelate contents were highest in the F/3 group. The DPPH (${\alpha}$, ${\alpha}{\prime}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging activity of the NF, F/0, and F/3 groups were 41.89%, 39.51%, and 60.26%, respectively. The inhibition activities of tyrosinase and lipoxygenase were stronger in the F/3 group than in the NF group. This experiment showed that the fermentation of Ag Nakai can lead to an increase in its antioxidant ability, physiological activity, whitening and anti-inflammatory effects. Thus, this oriental herbal medicine can be developed into a functional material that can be utilized in the development of cosmetic products in future.
