  • A potent immuno-stimulating activity was detected from the watersoluble and ethanol-precipitated crude extract (AG-1) of the root of Angelica gigas Nakai. The crude extract was fractionated into two fractions, an acidic AG-2 and a neutral AG-3 fraction by DEAE-cellulose adsorption. The two fractions contained polysaccharides of which M.W. were 10 Kdal and >2,000 Kdal respectively, proteins, and various inorganic components. The immunostimulating activities of two fractions were not reduced by proteinase K, acid or alkali treatment. The polysaccharides obtained from the root of A. gigas were mainly composed of arabinose, galactose, and galacturonic acid. These results indicated that immuno- stimulating components of the root of A. gigas was a kind of pectic polysaccharides or arabinogalactans.

  • 약초 당귀의 원산지를 판별하기 위하여 SAW 센서가 내장된 GC로 구축된 시스템을 이용하여 휘발성 성분을 분석하였다. 사용된 시료는 별도의 전처리 과정 없이 분석되어, 30초 이내에 얻어진 hertz 값(Frequency)과 이를 미분하여 표현한 크로마토그램을 $VaporPrint^{TM}$ 이미지 소프트웨어를 사용하여 패턴화하였으며 패턴인식을 통하여 비교한 결과, 수입산 당귀와 국내산 당귀의 패턴이 뚜렷하게 차이를 나타냄으로 원산지 판별을 할 수 있었다.

  • The proximate composition of 3 kinds of Umbelliferaeceae Wild Plants (Angelica gigas NAKAI., Ostericum sieboldii NAKAI. and Coriandrum sativum LINNAEUS) were analyzed and used for making pork sausages as a functional ingredient. In the proximate composition, crude protein content was the highest in Ostericum sieboldii, 37.87%, and crude lipid and crude ash contents were the highest in Coriandrum sativum. Vitamin C was contained in Angelica gigas at 0.30mg%. but was not detected in Ostericum sieboldii and Coriandrum sativum. $\beta$-carotene content in Angelica gigas was the highest at 19.96%, but selenium was not detected in any of them. Total phenol content in Coriandrum sativum was the highest as 1107.43mg%. TBA and VBN values of all sausages with wild plants increased gradually during four-week-storage and were lower than those of control. Coriandrum sativum-added sausage had the lowest TBA as well as VBN valuew. During the four-week-storage at 4$\^{C}$, TBA value and VBN value were in the very safe range at all sample. Total phenol contents in sausages appeared to have more antioxidative activity than vitamin C, $\beta$-carotene and selenium, and consequently decreased the TBA values of sausages.

감마선 조사 당귀(Angelica gigas Nakai)의 유효성분 안정성 및 유전독성학적 안전성 연구 (Evaluation on the Safety of ${\gamma}$-Irradiated Angelica gigas Nakai: Stability of Active Components and Safety in Genotoxicity Test)

  • In the present studies, we assessed the stability of active components and toxicological safety of irradiated Angelica gigas Nakai(Danggui). In order to confirm the stability of active components in the ${\gamma}$-irradiated roots of Danggui, the quantitative analysis of decursin and decursinol angelate of ${\gamma}$-irradiated sample was carried out by high performance liquid chromatographic (HPLC) methods using reverse phase columns and normal phase columns. From the root of Danggui, decursin and decursinol angelate were isolated by a silica gel column chromatography(toluene : ether (1 : 1), Hexane : EtOAc(15 : 1)). And then the structures were confirmed in the 1H and 13C-NMR analysis. The HPLC chromatograms of decursin and decursinol angelate in ${\gamma}$-irradiated Danggui were similar with those of non-irradiated sample. In the examination of in vitro genotoxicity of the water extract from ${\gamma}$-irradiated Danggui using Salmonella reversion assay(Ames test) and micronucleus test in Chinese hamster ovary (CHO) cells, mutagenicity was not exhibited in the two assays with or without metabolic activation. These resutls suggest that active components in the ${\gamma}$-irradiated Danggui should be stable and that the safety of ${\gamma}$-irradiated Danggui could be revealed in further test in vivo.

재배지 환경에 따른 참당귀 생육 및 지표성분 함량 비교 (Comparison of Growth and Contents of Active Ingredients of Angelica gigas Nakai under Different Cultivation Areas)

  • 참당귀(Angelica gigas Nakai)는 미나리과(Apiaceae) 당귀속(Angelica L.)에 속하는 다년생 식물로, 뿌리에 효능이 많아 약용 목적으로 재배된다. 본 연구에서는 주로 생산되는 참당귀 재배지의 특성에 대한 연구와 이에 따른 참당귀의 생장 및 지표성분 함량과의 상관관계를 구명하여 참당귀의 안정적인 한약재 수급을 위한 생산량 및 유용성분 함량 증가에 적합한 재배 환경에 대한 정보를 제공하는데 있다. 참당귀의 근직경은 평창 두일리가, 지근 개수는 평창 탑동리가 가장 많았다. 생중량은 평창 탑동리가 가장 높았으며, 건중량도 이와 같은 경향을 보였다. 재배지의 토성은 사질식양토(Sandy Clay Loam)였으며, 유기물 함량은 탑동리에서 3.63%로 가장 높은 함량을 보였다. 전 질소 함량은 봉화 남회룡리에서 0.19%로 가장 높았으며, 유효인산 함량은 고선리에서 높았다. 총 decursin 함량은 두일리에서 5.31%로 가장 높았다. 결과적으로 평창 두일리에서 참당귀의 생육이 가장 우수하였으며, decursin 함량 또한 높았다. 당귀의 수량이나 성분은 재배지역의 환경조건에 크게 영향을 받는데, 해발이 높은 지역에서 재배하는 것이 지표성분 함량과 생산량 증가에 유리하다고 사료되며, 환경조건과 생육 및 지표성분 함량과의 상관관계에 대한 기초자료가 될 것이라고 판단된다. 또한, 최적의 재배환경의 확립과 식물의 생리작용에 대한 연구의 기초가 될 수 있을 것이라고 판단된다.

당귀 열수 추출물을 이용한 막걸리의 품질 특성 (Quality Characteristics of Makgeolli using Angelica gigas Nakai Water Extracts)

  • The aim of this study was to investigate the quality characteristics of makgeolli that was produced by using different percentages (control, 3%, 5%, 7%, 9% by rice weight) of Angelica gigas Nakai water extracts during fermentation at $25^{\circ}C$ for 7 days. Yeast cell count in makgeolli steadily increased and showed the highest value at day 4 of fermentation, since then, the count decreased consistently. The final alcohol contents were higher than 15% in all samples. Soluble contents and antioxidation activity were significantly increased in percentage as Angelica water extracts increased (p<0.05). As the amount of Angelica water extracts increased, total polyphenol contents increased as well. Soluble contents and total polyphenol contents were increased according to the fermentation period. Total acidity increased up to day 3, but slowly decreased afterwards. However, the pH rapidly decreased by day 3, but then increased steadily. Reducing sugar amount was increased sharply by day 2 of fermentation and then, diminished gradually. Hunter's color value was not significantly different among samples however, they were significantly different between day 2 samples of fermentation and day 3 or more samples (p<0.05). In sensory evaluations, the sample produced using 5% Angelica water extracts received the highest score in herbal flavor, herbal taste, sourness, sweetness, and overall acceptance. Makgeolli using Angelica gigas Nakai water extracts showed excellent characteristics in antioxidative activity and higher total polyphenol content. Makgeolli produced using 5% Angelica water extracts received the highest score in sensory evaluation. Therefore, makgeolli produced using 5% Angelica water extracts was considered to be the most suitable for manufacturing makgeolli.

추출법에 따른 참당귀의 부위별 정유성분 비교 (Comparison of Volatile Compounds in Plant Parts of Angelica gigas Nakai by Extracting Methods)

  • Volatile flavor compounds from the shoot and root of Angelica gigas Nakai were extracted by HE (Hydrodistillation extraction), SDE (Simultaneous steam distillation & extraction), and SFE (Supercritical fluid extraction system), and analyzed by GC-MS. The amount and the number of chemical components in essential oils from shoot and root by SFE was the higher than those by other extraction methods. Respectively, thirty one constituents were identified from the essential oil of the shoot and root by HE, twenty seven and twenty three constituents were identified from the essential oil of shoot and root by SDE, thirty one and forty five constituents were identified from the essential oil of shoot and root by SFE. The result showed large differences in extraction methods and in plant parts of Angelica gigas Nakai. Also, the bioactive compounds in root part was identified as nodakenin and decursinol (11.95% and 8.42%, respectively) by SFE. These results suggested that SFE was the best extraction method for the increasing of extraction yield, the determination of volatile components and the increasing of bioactive compounds in the shoot and root of Angelica gigas Nakai.

참당귀(Angelica gigas Nakai) 중 Decursin 및 Decursinol Angelate의 분리 및 정제 (Isolation and Purification of Decursin and Decursinol Angelate in Angelica gigas Nakai)

  • 생약재로 사용 중인 Angelica gigas Nakai의 decursin 및 decursinol angelate를 60% ethanol로 최종적으로 추출 후 recycling HPLC 및 HPLC/MS에 의해 각각을 분리하였다. 60% ethanol로 $-20^{\circ}C$에서 추출하였을 경우 95% 순도의 decursin 및 decursinol angelate를 얻을 수 있었고 10번 이상의 recycle를 통해서 순수한 decursin 및 decursinol angelate를 분리할 수 있었다. 분리한 decursin 및 decursinol angelate를 HPLC/MS를 통해 분자량을 확인하였을 경우 m/z=329($[M+H]^+$) 및 m/z=351($[M+Na]^+$)과 같은 결과를 얻을 수 있었다. 이 분리 방법을 통해 decursin 및 decursinol angelate를 통한 의약품 및 식품 등에 관한 여러 연구에 있어 두 물질 각각에 대한 약리학적 효과를 도출하는데 기여할 것이며, 물질표준화에 기여할 것으로 사료된다.

신약 개발을 위한 참당귀(Angelica gigas Nakai) 추출 Decursin과 Decursinol Angelate의 약리 작용 (Pharmacological Effect of Decursin and Decursinol Angelate from Angelica gigas Nakai)

  • Traditionally, Cham dang-gui (Angelica gigas Nakai) is one of the most popular herbal medicines in Asian countries including Korea. A. gigas has been used as a functional food product for treatment anemia, women's health care, a sedative, an anodyne or a tonic agent. Decursin and decursinol angelate isolated from the roots of A. gigas are pyranocoumarin compounds. Recently, as the global herbal medication market is increasing, investigations about pharmacological effects of decursin and decursinol angelate are rapidly increasing. We summarized previous studies about pharmacological effects of decursin and decursinol angelate, and reviewed relation with pharmacological effects of decursin and decursinol angelate on human disorder, focused on the approach for new drug development. Pharmacological effects of decursin and decursinol angelate were classified as anti-tumor activity, anti-bacterial activity, improvements of the circulating system, inhibition of cytochrome P-450 activity, anti-inflammation activity, anti-oxidant activity and cognitive-enhancing activites. The activity of A. gigas with improvement of the circulating system may have wide therapeutic potential for circulatory diseases, including diabetes, hyperlipidemia and atherosclerosis. Also, anti-inflammation activity A. gigas may be beneficial for the treatment and prevention of asthma, atopic dermatitis and rheumatism arthritis. This relation could potentially lead to the development of herbal new drugs. In order to development a new drug containing decursin and decursinol angelate, it is also necessary to consider the safety profile, and the information in this review would contribute to development a new drug from herbal medicine.
