참당귀(Angelica gigas Nakai)잎 용매추출에 따른 생리활성 (Biological Activities of Extracts from Leaf of Angelica gigas Nakai)

  • Antioxidant properties and antioxidant activities were analyzed for water extracts and 50% and 70% ethanol extracts of the leaf of Angelica gigas Nakai. The polyphenol and flavonoid contents in water, 50%, and 70% ethanol extract of the leaf of Angelica gigas Nakai, it was found that the polyphenol contents were 18.75 mg GAE/g, 28.95 mg GAE/g, and 34.73 mg GAE/g, respectively, and flavonoid contents were respectively. The DPPH IC50 scavenging activity was 45.84 mg/mL, 36.44 mg/mL, 19.11 mg/mL, respectively, and theABTS+ radical scavenging ability (1 mg/mL) was 28.73%, 22.79%, and 12.70%, respectively. Tyrosinase inhibitory activity, 70% ethanol extract, 50% ethanol extract, and water extract 33.14%, and 4.53%, respectively. Nitrite scavenging activity, 70% ethanol extract, 50% ethanol extract, and water extract were in the order of 1 mg/mL scavenging activity, 36.43%, 34.80%, and 18.85%, respectively.

Antioxidant and ACE Inhibitory Activity of Cultivated and Wild Angelica gigas Nakai Extracts Prepared Using Different Extraction Conditions

  • The purpose of this study was to investigate the biological activities of cultivated Angelica gigas Nakai (CAG) and wild Angelica gigas Nakai (WAG) extracts prepared by extraction with water, 30% ethanol, 60% ethanol, or 90% ethanol. The electron donating ability of the WAG extracts was higher than that of the CAG extracts and 0.1% and 1.0% solutions of the comparative substance, L-ascorbic acid. The superoxide dismutase-like activity of the CAG extracts was higher than that of WAG extracts. Superoxide dismutase-like activity was highest (33.95%) in the CAG water extract. The total polyphenol content was highest in the 60% ethanol extracts of WAG. The nitrite scavenging ability of the CAG and WAG extracts was highest at a pH of 1.2. The tyrosinase inhibitory effect was highest (43.72%) in the water extract of WAG. The angiotensin converting enzyme inhibitory activity was highest (83.84%) in the 60% ethanol extract of WAG. The results of the present study will be useful for understanding the antioxidant and angiotensin-converting enzyme inhibitory activities of Angelica gigas Nakai extracts.

국내산 참당귀를 이용한 다식 제조에 관한 연구 (Development of Traditional Korean Snack, Dasik Using Angelica gigas Nakai)

  • 참당귀는 민방에서 월경통과 산후진정 등의 부인병 치료에 이용되어온 것으로 알려져 있다. 본 연구는 이러한 참당귀를 이용하여 제조한 우리 전통 한과류인 다식에 대하여 관능검사를 실시하였고 수분, 회분, 조단백질과 조지방 등의 일반성분을 분석하였으며, 참당귀의 phenolic compounds와 flavonoid 및 항산화활성 등을 측정하였다. 참당귀 다식의 관능검사 결과 참당귀 분말을 20% 함유한 흑임자다식이 동양의 참당귀 분말을 함유한 콩가루다식보다 높은 선호도를 나타내었다. 특히, 흑임자 분말에 참당귀 분말 20% 첨가군은 30대 및 40대에서 가장 선호도가 높았다. 참당귀 분말을 20% 첨가한 흑임자다식은 수분함량이 11.85%와 회분함량이 3.4%였고, 조단백질 14.1%, 조지방 23.44%의 함량을 나타내었다. 또한 참당귀의 phenolic compounds 함량은 2.23 mg/g, flavonoid 함량은 9.9 mg/g이었으며 항산화활성은 methanol reflux sample을 0.1 g/$m\ell$의 농도로 측정했을시 53.14%로 나타났다.

당귀 추출물 정맥 주사가 Middle Cerebral Artery Occlusion 모델 흰쥐에서 Gliosis 억제에 미치는 영향 (The Effect of Intravenous Injection of the Water Extract of Angelica gigas Nakai on Gliosis in the Middle Cerebral Artery Occlusion Rats)

  • Objectives : Gliosis becomes physical and mechanical barrier to axonal regeneration. Reactive gliosis induced by middle cerebral artery occlusion is involved with up-regulation of CD81 and GFAP (Glial fibrillary acidic protein). The current study is to examine the effect of the Angelica gigas Nakai(intravenous injection. 100 mg/kg twice in a day) on CD81 and GFAP of the rat in the brain after middle cerebral artery occlusion. Methods : Cerebral infarction was induced by middle cerebral artery occlusion. And after intravenous injection of water extract of Angelica gigas Nakai, the size of cerebral infarction was measured. Examination of optical microscope were also used to detect the expression of CD81 and GFAP in the brain of the rat. Results : The following results were obtained : We found that size of cerebral infarcion induced by MCAO (Middle Cerebral Artery Occlusion) in rats were decreased after intravenous injection of Angelica gigas Nakai. We injected the extract of Angelica gigas Nakai to the MCAO in rats, and the optical microscope study showed that Angelica gigas Nakai had effect on protecting the cells of hippocampus. We found that GFAP, CD81 and ERK of the brain in rats with cerebral infarction after MCAO were meaningfully decreased after intravenous injecting Angelica gigas Nakai. We found that c-Fos expression of the brain in rats with cerebral infarction after MCAO were significantly increased after intravenous injecting Angelica gigas Nakai. Conclusions : These results indicate that Angelica gigas Nakai could suppress the reactive gliosis, which disturbs the astrocyte regeneration in the brain of the rat with cerebral infarction after MCAO by controlling the expression of CD81 and GFAP. And the effect may be modulated by the up-regulation of c-Fos and ERK.

중합체 매개 용융압출에 의한 참당귀 나노복합체의 제조 (Fabrication of Nano-composites from the Radix of Angelica gigas Nakai by Hot Melt Extrusion Mediated Polymer Matrixs)

  • Background: The objective of this study was to make colloidal dispersions of the active compounds of radix of Angelica gigas Nakai that could be charaterized as nano-composites using hot melt extrusion (HME). Food grade hydrophilic polymer matrices were used to disperse these compound in aqueous media. Methods and Results: Extrudate solid formulations (ESFs) mediated by various HPMCs (hydroxypropyl methylcelluloses) and Na-Alg polymers made from ultrafine powder of the radix of Angelica gigas Nakai were developed through a physical crosslink method (HME) using an ionization agent (treatment with acetic acid) and different food grade polymers [HPMCs, such as HP55, CN40H, AN6 and sodium alignate (Na-Alg)]. X-ray powder diffraction (XRD) analysis confirmed the amorphization of crystal compounds in the HP55-mediated extrudate solid formulation (HP55-ESF). Differential scanning calorimetry (DSC) analysis indicated a lower enthalpy (${\Delta}H=10.62J/g$) of glass transition temperature (Tg) in the HP55-ESF than in the other formulations. Infrared fourier transform spectroscopy (FT-IR) revealed that new functional groups were produced in the HP55-ESF. The content of phenolic compounds, flavonoid (including decursin and decursinol angelate) content, and antioxidant activity increased by 5, 10, and 2 times in the HP55-ESF, respectively. The production of water soluble (61.5%) nano-sized (323 nm) particles was achieved in the HP55-ESF. Conclusions: Nano-composites were developed herein utilizing melt-extruded solid dispersion technology, including food grade polymer enhanced nano dispersion (< 500 nm) of active compounds from the radix of Angelica gigas Nakai with enhanced solubility and bioavailability. These nano-composites of the radix of Angelica gigas Nakai can be developed and marketed as products with high therapeutic performance.

참당귀잎 추출물을 첨가한 요구르트의 품질 특성 (Quality Characteristics of Yogurt Supplemented with Angelica gigas Nakai Leaf Extract)

  • 참당귀잎 추출물을 일정비율로 첨가한 발효유를 제조하고, 이화학적 특성과 항산화능 및 관능평가 등 품질특성을 분석하였다. pH는 4.40-4.45 범위를 보였고, 산도는 0.96%-0.98%로 참당귀잎 추출물 첨가에 따른 유의적인 차이를 보이지 않았다. 점도는 참당귀잎 추출물의 첨가에 따라 다소 감소되는 경향을 보였지만, 저장중 안정성에는 영향을 주지 않았다. 유산균수는 참당귀잎 추출물의 첨가량 증가에 따라 다소 억제되는 경향을 보였지만, 0.1%-0.3%의 범위에서는 1.9×109-3.2×109 CFU/mL의 높은 수준을 보였다. 요구르트 중의 decursin 함량 분석은 0.1%-0.3%의 첨가량에 따라 0.26 ㎍/g, 15.23 ㎍/g 및 23.57 ㎍/g으로 비례하여 정량분석 되었다. 유기산 생성은 lactic acid의 생성량이 가장 높게 나타났으며, 첨가비율에 따른 유의성은 보이지 않았다. 요구르트의 항산화능은 참당귀잎 추출물의 첨가로 높게 나타났으며, 또한 첨가량이 증가할수록 항산화효과도 높게 나타나는 효과를 보였다. 관능평가 결과, 참당귀잎 추출물 0.1% 첨가구가 무첨가구와 유사한 수준으로 높게 평가되었으며, 0.2% 첨가구의 경우도 6.31-6.50의 보통이상 수준으로 평가되었다. 이상의 결과와 같이 참당귀잎 추출물의 첨가농도는 0.2% 이내 범위가 적합하였다.

Effects of Nitrogen Fertilization on Leaf Yield and Pyranocurmarine Accumulation in Angelica gigas Nakai

  • Angelica gigas Nakai is one of the most widely used herbal medicines and is known to have many pharmaceutical effects including an anti-oxidant, anti-cancer etc. This study was carried out to investigate an effect of fertilization on leaf yield, production of dry-matter and accumulation of pyranocurmarine compounds such as decursin (DE) and decursinol angelate (DA) in Angelica gigas Nakai. Effect of fertilization was determined from response surface regression equation composing of 2 by 3 factorial arrangement of urea, sodium dihydrogen phosphate and potassium chloride. Yield of leaf in Angelica gigas Nakai significantly increased until 100 days after transplanting. Production of leaf also tended to increase with increasing nitrogen fertilization. Model of regression equation showed that leaf production depended upon nitrogen ($Pr>{\mid}t{\mid}$ : 0.087, 0.256 and 0.079). Also, statistical results between nitrogen application level and production of dry-matter showed significant relationship (p<0.05) and contents of dry-matter was highest in 10 kg 10a-1 treatment on 24 Sep. Active compound isolated and purified from leaf and root of Angelica gigas Nakai was identified as DE and DA by gas chromatograph-mass spectrophotometry (GC-MS). Concentration of DA as prevalent compound in leaf was highest on 20 Aug. but decreased on 24 Sep. Amount of DE and DA accumulated in Angelica gigas Nakai significantly increased with growth stages and nitrogen level. The result of our investigation imply that nitrogen fertilization is important factor for production of leaf and accumulation of pyranocurmarine in Angelica gigas Nakai as a medicinal/food materials.

당귀 부산물의 급여가 재래닭의 생산성과 육질에 미치는 영향 (Effects of Feeding Angelica gigas By-products of Performance and Meat Quality of Korean Native Chicks)

  • The objective of this experiment was to investigate the effect of feeding Angelica gigas Nakai by-products on performance and meat quality of Korean native chicks(KNC). Five hundred forty KNC were placed into 0, 0.2, and 0.4% Angelica gigas supplements with four replications between males and females. Weight gain, feed conversion ratio(FCR), breast meat lipid, fatty acid and protein contents were measured from 17 to 20 weeks old. Egg production, feed intake and FCR were examined from 22 to 29 weeks at four weeks at four weeks intervals. Basal diets based on corn and soybean meal contained 15% crude protein and 2,850㎉/kg ME for the growing period, 16.5% and 2,800㎉/kg for the laying period. Experiments were designed in a one way analysis. Weight gain of female chicks fed 0.2% Angelica gigas by-products increased significantly compared to that of other treatments for the growing period, but was not consistency in male groups. Moisture, protein, fat content were not statistically different among all treatments. Fatty acid composition(C16:0, C18:0) of chicks fed 0.2% Angelica gigas by-products was higher than other treatments. In laying period, hens fed 0.2 or 0.4% Angelica gigas Nakai by-products seemed to increase the egg production and significantly improved FCR compared to that of control(P〈0.05). Birds fed 0.4% treatment showed significantly the lowest feed intake and FCR of all treatments(P〈0.05). The results of these studies indicated that dietary supplemental Angelica gigas by-products may have a role to improve the performance of KNC.

Cytotoxic Coumarins from the Roots of Angelica gigas NAKAI

  • Some known coumarins, decursin, nodakenetin, umbelliferone, 7-demethylsuberosin, columbianetin, decursinol angelate and decursinol, showing significant cytotoxic activities against P388 cell lines,were isolated from the roots of Angelica gigas (Umbelliferae) . 7-Demethylsuberosin and columbianetinwere obtained from Angelica gigas for the first time. Chernotaxonornic difference about coumarins com-ponents between the roots of Angelica gigas and those of A. acutiloba is also discussed.

멜라노마 세포에서 당귀추출물의 MITF, TRP-1, TRP-2, tyrosinase mRNA 발현 억제 효과 (Inhibitory Efficacy of Angelica gigas Nakai on Microphthalmia-associated Transcription Factor (MITF), Tyrosinase Related Protein-1 (TRP-1), Tyrosinase Related Protein-2 (TRP-2), and Tyrosinase mRNA Expression in Melanoma Cells (B16F10))

  • 아시아에서 한방약초로 널리 알려진 당귀 추출물의 미백활성을 알아보기 위하여 tyrosinase 저해활성을 측정한 결과 1,000 ${\mu}g/ml$의 농도에서 70% 이상의 활성을 나타내었다. 또한 당귀 추출물에 대한 멜라노마 세포(B16F10)의 세포생존율을 확인한 결과 500 ${\mu}g/ml$의 농도에서 99% 이상의 세포생존율을 확인할 수 있었다. 미백 관련 인자인 MITF, TRP-1, TRP-2 및 tyrosinase의 mRNA 발현량을 측정한 결과 50 ${\mu}g/ml$의 농도에서 각각 85.7%, 123.9%, 68.8%, 208%로 당귀 추출물을 처리하지 않은 군보다 감소하였음을 확인할 수 있었다. 이러한 연구 결과에 따라 당귀 추출물이 melanin 합성과 관련이 있는 유전자 발현의 억제효과가 있음을 확인할 수 있었으며, 미백 화장품 소재로서의 가능성을 확인하였다.
