Isolation and Quantitative Determination of Anemarsaponin B from the Rhizomes of Anemarrhena asphodeloides Bunge (지모(Anemarrhena asphodeloides Bunge)의 근경으로부터 Anemarsaponin B의 분리 및 함량분석)

  • A method for isolation and qantitative determination of anemarsaponin B from the rhizomes of Anemarrhena asphodeloides has been developed. Isolation of anemarsaponin B was achieved by silica gel and RP-18 column chromatography. The HPLC method for quantitative determination of anemarsaponin B provided a method for standardization of the crude drug. It suggested that the content of anemarsaponin B in Anemarrhena asphodeloides is about 0.12-1.48%.

Physical Mapping of rDNAs Using McFISH in Anemarrhena asphodeloides Bunge (지모에서 McFISH를 이용한 rDNAs의 물리지도 작성)

  • Anemarrhena asphodeloides, a medicinal plant, has chromosome number of 2n=2x=22. To characterize the somatic metaphase chromosomes, physical mapping of 45S and 5S rDNAs using McFISH (multi-color fluorescence in situ hybridization) was applied. Two pairs of 45S rDNA loci were detected on the terminal regions of the short arm of chromosomes 1 and 3. A pair of 5S rDNA signal was observed on the short arm of chromosome 3. 5S rDNA site seemed to be the same locus as one of the 45S rDNA site. McFISH was very useful tool for the localization and identification of rDNAs on the metaphase chromosomes in A. asphodeloides.

Effects of Peduncle Cutting on Yield and Crude Saponin Content in Anemarrhena asphodeloides Bunge (화경제거가 지모의 수량 및 조사포닌함량에 미치는 영향)

  • In this study, effects of wiling peduncle on production yield and crude saponin content in Anemarrhena asphodeloides Bunge were investigation. The peduncle cutting were conduct on May 30, June 15, June 30, non-cutting of seed propagation type and young plant propagation type in Anemarrhena asphodeloides Bunge. The highest rhizome yield was 276.9g/plant on May 30, cutting puduncle in seed propagation type. The rhizome yield at seed propagation type was increased about 40.8%, compared to that at non-cutting. Crude saponin content in rhizome was 7.50% on May 30, cutting puduncle in seed propagation type. The crude saponin content at seed propagation type was increased about 32.2%, compared to that at non-cutting.

  • Anemarrhena asphodeloides BUNGE is one of important medicinal crops, which has been collected or/and cultivated for its rhizomes. The main medicinal ingredient of the A. asphodeloides Bunge rhizomes is a saponin, which is known to have medical values for diaphoresis, sedatives and biuresis. However, studies on cultural methods and breeding works on this plant have not been made in detail. To increase productivity and to improve quality of crops, it is important to collect cultivated and wild lines, to classify them based on morphological and genetic characteristics, and to select superior pure lines at first. Therefore, total 20 A. asphodeloides lines collected were cultivated at the fieldof Chungnam Provincial Administration of Rural Development in 1995 to study the agronomic characteristics and to classify them based on morphological characteristics. Characteristics related with reproductive organ such as the number of spikes per plant and peduncle length showed greater variations than vegetative organrelated characteristics such as leaf length and rhizome length based on the coefficient of variation. Vigorous shoot growth resulted in better development of reproductive organs such as peduncle length and number of spikes per plant. However, the development of spikes was negatively correlated with chlorophyll content. Characteristics of underground parts were more significantly correlated with spike characteristics than aerial part characteristics. A. asphodeloides tested in this study were classified into 2 groups (group A and group B) based on centroid linkage cluster analysis. Group A showed more vigorous shoot growth with more leaves and spikes per plant, longer peduncle length, thicker peduncle diameter and higher shoot dry weight than group B. Group A could be further classified into 2 sub-groups based on leaf size, spike length and peduncle diameter, while group B also could be classified based on number of leaves, number of spikes and shoot dry weight.

  • Cytogenetic analysis of Anemarrhena ashodeloides BUNGE, which is one of medicinal plants belonging to Haemodoraceae was carried out using Feulgen staining. The somatic metaphase chromosome number was identified to 2n=22 (x=11) and the size of chromosomes ranges from $1.27-3.80\;{\mu}m$. Three pairs of chromosomes were relatively long in total length and the others were short. The karyotype was bimodal in chromosome length and arm ratios. The chromosome complement comprise eight pairs of metacentric (chromosome 2, 3, 6, 7, 8, 9, 10, and 11), two pairs of submetacentric (chromosome 4 and 5), and one pair of subtelocentric (chromosome 1).

Effects of Different Mulching Materials on Rhizome Yield and Crude Saponin Contents in Anemarrhena asphodeloides Bunge (피복비닐 종류에 따른 지모의 근경수량 및 조사포닌 함량)

  • In this study effect of different mulching materials on yield and crude saponin contents in Anemarrhena asphodeloides Bunge. The results of this study are summarized as follows: The rhizome yield of seed propagation type in transparent PE mulching cultivation was of which increased 16.7% more than those of non-mulching cultivation. The rhizome yield of young-plant propagation type in green PE mulching and transparent PE mulching cultivations were of which increased 29.4% and 26.5%, respectively more than those of non-mulching cultivation. The contents of methanal extract of seed propagation type in transparent PE mulching and black PE mulching cultivations were of increased each 31.0%, 15.4% more than those of non-mulching cultivation. Crude saponin contents of seed propagation type in transparent PE mulching cultivation was increased up to 30.3% compared to those of non-mulching cultivation. However, the crude saponin contents of seed propagation type in green PE mulching and black & white PE mulching cultivations were not affected by the non-mulching cultivation. Crude saponin contents of young-plant propagation type in transparent PE mulching cultivation was increased 24.0% and 15.4% more than those of green PE mulching and black PE mulching cultivations.

Growth Characteristics and Content of Sarsasapogenin in Different Plant Parts of Anemarrhena asphodeloides Bunge (知母의 生育特性과 部位別 Sarsasapogenin 含量)

  • This study was carried out to find the growth characteristics and content of sarsasapogenin in different plant parts of Anemarrhena asphodeloides Bunge. Five native-cultivars were collected and evaluated for several agronomic traits. The collected native-cultivars were classified into two seed-attached peduncle and vegetative propagation types. Seed-attached peduncle lines were predominance of growth traits than vegetative propagation. For the content of sarsasapogenin in each part was investigated, BuOH extarct of 'timo' was developed on silica gel 60G plate using elution solvent($CHC1_2$: Methyl ethyl Ketone : EtOH = 11 : 2 : 0.5). The developed plate were examined using Dual Wavelength Zig - Zag using. Content of sarsasapogenin in main root and lateral root were respectively 1.67mg/g and 1.31mg/g.

Antimicrobial, Antioxidant and Cellular Protective Effects against Oxidative Stress of Anemarrhena asphodeloides Bunge Extract and Fraction (지모 뿌리 추출물과 분획물의 항균활성과 항산화 활성 및 세포보호 연구)

  • Extracts and fractions of Anemarrhena asphodeloides Bunge were prepared and their physiological activities and components were analyzed. Antimicrobial activities of the ethyl acetate and aglycone fractions were $78{\mu}g/ml$ and $31{\mu}g/ml$, respectively, for Staphylococcus aureus and $156{\mu}g/ml$ and $125{\mu}g/ml$, respectively, for Pseudomonas aeruginosa. 1,1-Diphenyl-2-picrylhydrazyl free radical scavenging activities ($FSC_{50}$) of 50% ethanol extract, ethyl acetate fraction, and aglycone fraction of A. asphodeloides extracts were $146.2{\mu}g/ml$, $23.19{\mu}g/ml$, and $71.06{\mu}g/ml$, respectively. The total antioxidant capacity ($OSC_{50}$) in an $Fe^{3+}$-EDTA/hydrogen peroxide ($H_2O_2$) system were $17.5{\mu}g/ml$, $1.5{\mu}g/ml$, and $1.4{\mu}g/ml$, respectively. The cytoprotective effect (${\tau}_{50}$) in $^1O_2$-induced erythrocyte hemolysis was 181 min with $4{\mu}g/ml$ of the aglycone fraction. The ${\tau}_{50}$ of the aglycone fraction was approximately 4-times higher than that of (+)-${\alpha}$-tocopherol (${\tau}_{50}$, 41 min). Analysis of $H_2O_2$-induced damage of HaCaT cells revealed that the maximum cell viabilities for the 50% ethanol extract, ethyl acetate fraction, and aglycone fraction were 86.23%, 86.59%, and 89.70%, respectively. The aglycone fraction increased cell viability up to 11.53% at $1{\mu}g/ml$ compared to the positive control treated with $H_2O_2$. Analysis of ultraviolet B radiation-induced HaCaT cell damage revealed up to 41.77% decreased intracellular reactive oxygen species in the $2{\mu}g/ml$ aglycone fraction compared with the positive control treated with ultraviolet B radiation. The findings suggest that the extracts and fractions of A. asphodeloides Bunge have potential applications in the field of cosmetics as natural preservatives and antioxidants.

Screening of Antioxidative Activity of Hot-Water Extracts from Medicinal Plants (한약재 열수추출물의 항산화효과 검정)

  • Interrelation between the antioxidative activity of hot-water extracts of 130 medicinal plants and their cellular antimutagenic activity was investigated. Antioxidative activity was evaluated by assaying electron-donation to DPPH free radical and scavenging of hydroxyl radical $({\cdot}OH)$ generated through Fenton rection, respectively. All medicinal plants examined in this study exhibited markedly electron-donating ability and radical scavenging ability in each assay system. The results demonstrated the fact that Pilbal (Piper longum L.) is the strongest in electron-donating activity, on the other hand, that Seokgok (Dendrobium moniliforme L.) is the strongest in ${\cdot}OH$ scavenging activity. When evaluated their antioxidative activities, 24 medicinal plants including Jimo (Anemarrhena asphodeloides Bunge) were found to be the medicinal plants carrying strong antioxidative activity, which exhibited more than 50% activity compared to the control group in both electron-donating and free radical scavenging. The experiment was also performed to examine whether 11 medicinal plants having significant antimutagenicity damage DNA in the presence of $Cu^{2+}$, showing the fact that all samples tested, except Taeksa (Alisma canaliculatum All. Br.), Paekjain (Nitraria sibirica Pall) and Ohyak (Lindera strychifolia Sieb. et Zucc. Villar) are capable of inducing DNA strand break. We also found that Taeksa and Paekjain strongly block DNA strand break induced by chemical mutagen mitomycin C.
