Beneficial effect of metformin on tolerance to analgesic effects of sodium salicylate in male rats

  • Background: Tolerance to the analgesic effects of opioids and non-steroidal anti-inflammatory drugs (NSAIDs) is a major concern for relieving pain. Thus, it is highly valuable to find new pharmacological strategies for prolonged therapeutic procedures. Biguanide-type drugs such as metformin (MET) are effective for neuroprotection and can be beneficial for addressing opioid tolerance in the treatment of chronic pain. It has been proposed that analgesic tolerance to NSAIDs is mediated by the endogenous opioid system. According to the cross-tolerance between NSAIDs, especially sodium salicylate (SS), and opiates, especially morphine, the objective of this study was to investigate whether MET administration can reduce tolerance to the anti-nociceptive effects of SS. Methods: Fifty-six male Wistar rats were used in this research (weight 200-250 g). For induction of tolerance, SS (300 mg/kg) was injected intraperitoneally for 7 days. During the examination period, animals received MET at doses of 50, 75, or 100 mg/kg for 7 days to evaluate the development of tolerance to the analgesic effect of SS. The hot plate test was used to evaluate the drugs' anti-nociceptive properties. Results: Salicylate injection significantly increased hot plate latency as compared to the control group, but the total analgesic effect of co-treatment with SS + Met50 was stronger than the SS group. Furthermore, the effect of this combination undergoes less analgesic tolerance over time. Conclusions: It can be concluded that MET can reduce the analgesic tolerance that is induced by repeated intraperitoneal injections of SS in Wister rats.

  • Objectives : Modified Gamgil-tang is a prescription commonly used for respiratory diseases. This thesis was carried out to check the treatment effects and diversity of drug formulation by comparing extraction method of ethanol and water of modified Gamgil-tang. Methods : All experiments were carried out with water and 50% ethanol extraction for comparison. In vivo experiment, hyaluronidase inhibitory effects and trypsin inhibitory effects were tested to measure the anti-inflammatory effects activity. Scavenging effects of DPPH free radical, xanthine oxidase inhibitory effects and inhibition on TBA-RS formation were experimented to measure anti-oxidative effects. With the in vivo experiment, ICR group mice and SD group rats were used as experimental animals. An anti-inflammatory effects experiment were carried out to measure the action on carrageenin-induced hind paw edema: analgesic effects were measured using writhing syndrome induced by 0.7% acetic acid in mice: antipyretic effect was measured using endotoxin, and inhibitory effects of increase vascular permeability induced by 0.5% histamine were measured. Results : For extraction of glycyrrhizin contents, ethanol extract was extracted 2 times of that of water extract. Anti-inflammatory effects showed high in ethanol extract. Anti-oxidative effects measured high in ethanol extract. No significant result was found in inhibition on TBA-RS formation. Analgesic effects were found to be similar in water and ethanol extract. Antipyretic effects were found to be stronger in water extract. Inhibitory effects of increase vascular permeability induced by 0.5% histamine showed stronger in ethanol extract. Conclusion : By measuring anti-inflammatory effects, analgesic effects, antipyretic effects, anti-oxidative effects, and histamine permeation inhibition effects both in water extract and ethanol extract after adding agents such as Mentha Herba, Gardenias Fructus, and propolis to existing Gamgil-Tang, ethanol extract was found to be more effective in anti-inflammatory effects, analgesic effects, anti-oxidative effects, and histamine permeation inhibition effects. The converse was found for antipyretic effect.

Effects of Glycine on the Development of Analgesic Tolerance to and Physical Dependence on Morphine in Mice

  • This study was performed to investigate the effects of glycine on the development of tolerance to and physical dependence on morphine. Repeated administration of morphine (10 mg/kg) developed tolerance and physical dependence. Glycine (100, 200 and 400 mg/kg) was administered intraperitoneally (i.p.) to mice for 7 days prior to the morphine injection. Analgesic effects were estimated by the tail flick methods. The inhibitory degree of the development of morphine-induced analgsic tolerance by i.p. administration of glycine was evidenced by the increase in analgesic response to morphine. Glycine inhibited the development of tolerance to morphine. In addition, we separately measured jumping response as the naloxone-precipitated withdrawal sign in mice that had received the same morphine. Glycine reduced the number of jumping behaviors in morphine dependent mice. These results suggest that glycine might be useful the prevention or treatment of morphine tolerance and physical dependence.

  • These studies were conducted an attempt to investigate effects of 'Paeryungtang' and 'Kamipaeryungtang' water extracts on diuretic, antipyretic, antiinflammatory and analgesic actions. The results of these studies were summarized as follows; Increase in urinary volume, urinary $Na^+$ excretions were significantly recognized in normal rat. Increase in urinary volume, urinary $Na^+\;and\;Cl^-$ excretions were significantly shown in rat with 2mg/kg $HgCl_2$-induced acute renal failure. Antipyretic, anti-inflammatory and analgesic effects of 'Paeryungtang' and 'Kamipaeryungtang' were recognized in mice, rats and rabbits.

  • Lonicera japonica is widely distributed in Southeast Asia and has been traditionally used as an anti - inflammatory, diuretic, abscessic, antipyretic, and antidotic agent. Lonicera japonica was investigated for its anti-inflammatory and analgesic effects using several in vivo models. SKLJI was purified for i.v. injection from Lonicera japonica as a potent anti-inflammatory and analgesic fraction, after activity-guided fractionation study. (omitted)

  • Experimental studies were done to investigate analgesic and antiphlogistic actions of Violae Herba. Its water extract showed a significant dose dependent analgesic effect against writhing syndrome induced by acetic acid as well as in hot plate test in mice. The extract also showed a significant inhibitory effect against paw-edema induced by egg-white in rats.

신경병증성 통증 증후군의 관리를 위한 부가적 진통제로서의 Paroxetine (Paroxetine, as an Adjuvant Analgesic for the Management of Neuropathic Pain Syndrome)

  • Background: Tricyclic antidepressants (TCA) have been used for various pain syndromes for their analgesic effects. They, however, often have anticholinergic side effects and therefore search for more selective drugs with fewer side effects is justified. Paroxetine, a selective serotonin reuptake inhibitor devoid of autonomic side effects, was evaluated for its role as an analgesic adjuvant in the management of neuropathic pain. Method: According to individual diagnostic group as diabetic neuropathy, postherpetic neuralgia, central pain syndrome and cancer related plexopathy, 10 patients per each group were equally accumulated. Patients have been stabilized in their analgesic regimen at least four weeks prior to enrollment into study. TCA, if taken, was discontinued for two weeks for wash out period. Baseline four point verbal pain intensity score was obtained and oral administration of paroxetine 20 mg was initiated. At two weeks follow-up visit, pain intensity scores, pain improvement scores judged by family, drug efficacy, tolerability and overall evaluation were assessed. The incidence of side effects were also obtained. Result: After two weeks of treatment, pain intensity scores decreased in 77.5% of patients and no patients experienced aggravation. These findings were objectively reflected in pain improvement scores judged by family members. But, the number of nonresponders was different among groups. In drug efficacy, tolerability and overall evaluation, the proportions of patients who scored as excellent or good were 75%, 80% and 80% respectively. Incidence of side effects was 27.5%, but the side effects spontaneously disappeared after discontinuation of medication. Conclusion: Paroxetine, a selective serotonin reuptake inhibitor, appears to be effective as adjuvant analgesic for the management of various neuropathic pain syndromes.

咽喉疾患에 應用되는 必用方甘桔湯 및 加味必用方甘桔湯의 效能에 關한 實驗的 硏究 (Experimental Studies on the Anti-inflammatory, Analgesic Effects of Pilyongbangkamgil-Tang and Kamipilyongbangkamgil-Tang on Rats and Mice.)

  • According to oriental clinic effect findings hitherto, Pilyongbangkamgil-Tang has been curative effects on chronic pharnygitis, acute tensillitis and angina. Auther tried to make clear the anti-inflammatory effect on rat paw which has become edema formation by $5\%$ of acetic acid/saline solution, and the analgesic effect on mouse thorough method of acetic acid because the medicine decoction of Kamipilyongbangkamgil-Tang, Pilyongbangkamgil-Tang, and the extracted powder of Pilyongbangkamgil-Tang are administered to rat and mouse. In present report, anti-inflammatory, analgesic effect of Pilyongbangkamkil-Tang, the extracted powder of the above prescription and Kamipilyongbangkamgil-Tang were estimated by the above test. The results are summarized as follows. 1. Kamipilyongbangkamgil-Tang (liquid), Pilyongbangkamgil-Tang and the extracted powder of Pilyongbangkamgil-Tang were tested for analgesic effects; Kamipilyongbangkamgil-Tang has the most effective analgesic function, then Pilyongbangkamgil-Tang, then the extracted powder, and I found that the t-test of those above prescriptions, in this order, should come to the result of voluntariness P〈0.001, P〈0.02, and P〈0.05 respectively. 2. Analgesic effects of pilyongbangkamgil-Tang and the extracted powder against the mouse pain induced by acetic acid have been strengthened by increase of double dosage and 4 fold dosage. 3. With Pilyongbangkamgil-Tang and Kamipilyongbangkamgil-Tang treated for rat, the increasing and the inhibitory rate of rat paw edema formation showed significantly statistical values, and the anti-inflammatory effect of the extracted powder of Pilyongbangkamgil-Tang is not supported so long than Pilyongbangkamgil-Tang. 4. The anti-inflammatory effect of Kamipilyongkamgil-Tang on the edema formed at the rat paw appeared significantly statistical value than the other sample.

Analgesic Effects of Toad Cake and Toad-cake-containing Herbal Drugs -Analgesic effects of toad cake-

  • Objectives: This study was conducted to clarify the analgesic effect of toad cake and toad-cake-containing herbal drugs. Methods: We counted the writhing response of mice after the intraperitoneal administration of acetic acid as a nociceptive pain model and the withdrawal response after the plantar surface stimulation of the hind paw induced by partial sciatic nerve ligation of the mice as a neuropathic pain model to investigate the analgesic effect of toad cake and toad-cake-containing herbal drugs. A co-treatment study with serotonin biosynthesis inhibitory drug 4-chloro-DL-phenylalanine methyl ester hydrochloride (PCPA), the catecholamine biosynthesis inhibitory drug ${\alpha}$-methyl-DL-tyrosine methyl ester hydrochloride (AMPT) or the opioid receptor antagonist naloxone hydrochloride was also conducted. Results: Analgesic effects in a mouse model of nociceptive pain and neuropathic pain were shown by oral administration of toad cake and toad-cake-containing herbal drugs. The effects of toad cake and toad-cake-containing herbal drugs disappeared upon co-treatment with PCPA, but not with AMPT or naloxone in the nociceptive pain model; the analgesic effect of toad-cake-containing herbal drugs also disappeared upon co-treatment with PCPA in the neuropathic pain model. Conclusion: Toad cake and toad-cake-containing herbal drugs have potential for the treatments of nociceptive pain and of neuropathic pain, such as post-herpetic neuralgia, trigeminal neuralgia, diabetic neuralgia, and postoperative or posttraumatic pain, by activation of the central serotonin nervous system.

  • Comparative studies were made on the analgesic effect of salicylamide, used individually and combined with parasympatholytics (propantheline and atropine) and antihistaminics (tripelennamine, diphenhydramine) as regards the analgesic effect (in thermal contact method) were examined by its oral administration with each combined drug to mouse (three assumption cross-over test), and the following effects were found. 1. The increasing order of the parasympatholytics to the analgesic effect of salicylamide is as follows: propantheline>atropine. 2. The increasing order of the antihistaminics to the analgesic effect of salicylamide is as follows: chlorpheniramine>diphenhdramine>tri pelennamine. In the ratio '1 : 1' salicylamide to parasympatholytics and antihistaminics, the analgesic effect of salicylamide was more increase than the other ratio in this study.
