A Study on the Source of Reductants for Nitrate Reduction in Rice (Oryza sativa cv. Tongil) Roots (벼(Oryza sativa cv. Tongil) 뿌리에 있어서 Nitrate 환원에 필요한 환원력의 공급원에 관한 연구)

  • There was a decrease in nitrate reductase activity (NRA) measured in vivo in rice roots (Oryza sativa cv. Tongil) grown in anaerobic culture solution. But it was reversed by addition of malonate to the in vivo nitrate reduction assay medium. Malonate increased the in vivo NRA during 2-5 hours incubation and decreased it in longer incubation hours. In vivo NRA was stimulated by addition of NaHCO3 to the assay medium, but not by Na2CO3. The stimulation of NRA by NaHCO3 was not observed in shoot removed rice roots. It is suggested that CO2 from NaHCO3 is carboxylated by phosphoenol pyruvate carboxylase, results in increasing the malate contents in the roots, and stimulates the in vivo NRA. NADH needed in nitrate reduction is supported by malate oxidation. In rice roots, it seems probable that malate oxidation in the mitochondria is more important to nitrate reduction than malae oxidation in cytoplasm.

Assessment of CH4 oxidation in macroinvertebrate burrows of tidal flats (갯벌의 무척추 동물 서식굴 내 메탄산화 평가)

  • In tidal flats that lack plants, methane ($CH_4$) fluxes are both positive (gas emission) and negative (gas "sinking") in nature. The levels of methanotroph populations significantly affect the extent of $CH_4$ sinking. This preliminary study examined $CH_4$ flux in tidal flats using a circular closed-chamber method to understand the effects of macroinvertebrate burrowing activity. The chamber was deployed over decapods (mud shrimp, Laomedia astacina and crab, Macrophthalmus japonicus) burrows for ~ 2 h, and the $CH_4$ and $CO_2$ concentrations were continuously monitored using a closed, diffuse $CH_4/CO_2$ flux meter. We found that Laomedia astacina burrow (which is relatively long) site afforded higher-level $CH_4$ production, likely due to diffusive emission of $CH_4$ in deep-layer sediments. In addition, the large methanotrophic bacteria population found in the burrow wall sediments has $CH_4$ oxidation (consumption) potential. Especially, nitrite-driven anaerobic oxidation of methane (AOM) may occur within burrows. The proposed $CH_4$-oxidation process was supported by the decrease in the ${\delta}^{13}C$ of headspace $CO_2$ during the chamber experiment. Therefore, macroinvertebrate burrows appear to be an important ecosystem environment for controlling atmospheric $CH_4$ over tidal flats.

A Proposal of Sequencing the Combined Processes for Resources Recovery and Nitrogen Removal from Piggery Waste (슬러리형 돈사폐수에서 자원회수와 질소제거를 위한 순차적 결합공정 제안)

  • The combined ADEPT(Anaerobic Digestion Elutriated Phased Treatment)-SHARON(Single reactor system for High Ammonium Removal Over nitrite)-ANAMMOX(Anaerobic ammonium oxidation) processes were operated for resources recovery and nitrogen removal from slurry-type piggery waste. The ADEPT process operated at an acidogenic loading rates of 3.95 gSCOD/L-day, the SCOD elutriation rate and acid production rate were 5.3 gSCOD/L-day and 3.3 gVFAs(as COD)/L-day, respectively. VS reduction and SCOD reduction by the hydrolysis were 13% and 0.19 $gSCOD_{prod.}/gVS_{feeding}$, respcetively. Also, the acid production rate was 0.80 $gVFAs/gSCOD_{prod}$. In methanogenic reactor, the gas production rate and methane content were 2.8 L/day($0.3m^3CH_4/kgCOD_{removal}@STP$) and 77%, respectively. With these operating condition, the removals of nitrogen and phosphorus were 94.1% as $NH_4-N$(86.5% as TKN) and 87.3% as T-P respectively.

Application of tubular membrane to thickening process as a pre-treatment of anaerobic digester (관형막을 이용한 혐기소화조의 전처리 농축공정으로써 적용가능성 연구)

  • The purpose of this study is to estimate the applicability of a stable anaerobic digester using a separator membrane to the preprocessing thickening process. The results of the experiments showed about a 47.16% weight loss rate for the sludge under anaerobic condition, and about 41.17% under intermittent aeration condition. The concentrations of rejection water were $SCOD_{Cr}$ 25 mg/L, T-N 16.6 mg/L, and T-P 1.4 mg/L on the average under the intermittent aeration condition, which were lower than the concentrations of rejection water under an anaerobic condition. As for the factors affecting the reduction of the flux, correlation analyses of TTF, MLSS, $SCOD_{Cr}$, and $EPS_{Protein}$, $EPS_{Polysacchride}$ resulted in -0.97, -0.95, -0.84 and -0.86, -0.95, respectively, which showed that TTF had the highest correlation to the reduction of the flux. In addition, it was concluded that MLSS, $SCOD_{Cr}$ and $EPS_{Protein}$, $EPS_{Polysacchride}$ also have close correlations. The results are considered to show that, in the case of the process using a tubular membrane in the preprocessing process of an anaerobic digester, an intermittent aeration condition of the thickener considering the contamination of the membrane and load of rejection water is appropriate for the stable preprocessing process.

Removal of Simultaneously Biological Organic, Nitrogen, and Phosphorus Removal in Sequencing Batch Reactors using Night-soil (연속회분식 반응기(Sequencing Batch Reactor)를 이용한 분뇨중 유기물과 질소 및 인의 동시제거)

  • Sequencing Batch Reactor(SBR) experiments for organics and nutrients removal have been conducted to find an optimum anaerobic/anoxic/aerobic cycling time and evaluate the applicability of oxidation-reduction potential(ORP) as a process control parameter. In this study, a 61 bench-scale plant was used and fed with night-soil wastewater in K city which contained TCODcr : 10, 680 mg/l, TBm : 6, 893 mg/l, $NH_4^+-N$ : 1, 609 mg/l, $PO_4^{3-}-P$ : 602 mg/l on average. The cycling time In SBRs was adjusted at 12 hours and 24 hours, and then certainly included anaerobic, aerobic and inoxic conditions. Also, for each cycling time, we performed 3 series of experiment simultaneously which was set up 10 days, 20 days and 30 days as SRT From the experimental results, the optimum cycling time for biological nutrient removal with nlght-soil wastewater was respctively 3hrs, 5hrs, 3hrs(anaerobic-aerobic-anoxic), Nitrogen removal efficiency was 77.9%, 77.9%, 81.7% for each SRT, respectively. When external carbon source was fed in the anoxic phase, ORP-bending point indicating nitrate break point appeared clearly and nitrogen removal efficiency increased as 96.5%, 97.1%, 98.9%. Phosphate removal efficiency was 59.8%, 64.571, 68.6% for each SRT. Also, we finded the applicability of ORP as a process control parameter in SBRs.

Metabolic Flux Shift of Weissella kimchii sk10 Grown Under Aerobic Conditions

  • The sk10 isolated from kimchi was identified as W. kimchii on the basis of l6s-rDNA sequencing. Studies were made to analyze the metabolic flux shift of the sk10 on glucose under aerobic growth conditions. The sk10 produced 38.2 mM acetate, 16.3 mM ethanol, and 33.2 mM lactate under aerobic conditions, but 2.4 mM acetate, 48.0 mM ethanol, and 44.1 mM lactate under anaerobic conditions. The NADH peroxidase (NADH-dependent hydrogen peroxidase) activity of sk10 grown under aerobic conditions was 11 times higher than that under anaerobic conditions. Under the low ratio of $NADH/NAD^+$, the metabolic flux toward lactate and ethanol was shifted to the flux through acetate kinase without NADH oxidation. The kinds of enzymes and metabolites of sk10 were close to those in the pathway of Leuconostoc sp., but the metabolites produced under aerobic growth conditions were different from those of Leuconostoc sp. The stoichiometric balance calculated using the concentrations of metabolites and substrate was about 97%, coincident with the theoretical values under both aerobic and anaerobic conditions. From these results, it was concluded that the metabolic flux of W. kimchii sk10 was partially shifted from lactate and ethanol to acetate under aerobic conditions only.

Nitrite Accumulation of Anaerobic Treatment Effluent of Slurry-type Piggery Waste (슬러리상 돈사폐수의 혐기성 처리수의 아질산성 질소 축적)

  • The effluent from anaerobic digestion process of slurry-type piggery waste has a characteristic of very low C/N ratio. Because of high nitrogen content, it is necessary to evaluate nitrogen removal alternative rather than conventional nitrification-denitrification scheme. In this study, two parallel treatment schemes of SBR-like partial nitritation reactor coupled with anaerobic ammonium oxidation (ANAMMOX) reactor, and a nitritation reactor followed by nitrite denitrification process were evaluated with a slurry-type piggery waste. The feed to reactors adjusted with various $NH_4-N$ and organics concentration. The nitrite accumulation was successfully accomplished at the loading rate of about $1.0kgNH_4-N/m^3-day$. The $NO_2-N/NH_4-N$ ratio 1~2.6 in nitritated effluent that operated at HRT of 1 day indicated that SBR-like partial nitritation was applicable to ANAMMOX operation. Meanwhile, the nitrite accumulation of 87% was achieved at SBR operated with HRT of 3 days and $0.4mgO_2/L$ for denitritation. Experimental results further suggested that HRT (SRT) and free ammonia(FA) rather than DO are an effective control parameter for nitrite accumulation in piggery waste.

Evaluation of Nitritation of High Strength Ammonia with Variation of SRT and Temperature using Piggery Wastewater (SRT와 온도 변화를 통한 돈사폐수 내 고농도 암모니아의 아질산화 평가)

  • The purpose of this study is to investigate the high strength ammonia oxidation of piggery wastewater. Laboratory scale reactors was operated using influent of piggery wastewater and effluent of anaerobic digester from piggery wastewater at $35^{\circ}C$ and $20^{\circ}C$. Results of various operating conditions were compared and analyzed. After analyzing the results, effluent of anaerobic digester from piggery wastewater required shorter Solid retention time (SRT) than influent of piggery wastewater. In terms of the temperature, stable ammonia removal and denitrification was achieved on the both of the condition. At the temperature of $35^{\circ}C$, nitrite conversion rate was better than $20^{\circ}C$. It can be concluded that treating the piggery wastewater using anaerobic digester on the condition of the temperature at $35^{\circ}C$ is more efficient on the nitritation of the piggery wastewater.

Rates and Controls of Organic Matter Mineralization and Benthic Nutrient Release in the Coastal Sediment Near Lake Shihwa (시화호 인근 연안 퇴적물의 유기물 분해 특성, 저층 영양염 용출 및 조절요인)

  • We investigated geochemical constituents of pore-water and sediment, rates of organic carbon (Corg) oxidation and sulfate reduction (SR), and benthic nutrient flux (BNF) to elucidate characteristic of Corg oxidation and its control in the coastal area near Lake Shihwa. The study sites were selected in the vicinity of Soraepogu (E0), Songdo tidalflat (E1) and Oido dock (E3) and in front of floodgate Shihwa tidal plant (E5). The Corg contents in the sediments and concentrations of ammonium and phosphate in pore water exhibited the highest value at EO, and gradually decreased toward the outer sea (E1, E3, E5). Rates of anaerobic Corg oxidation (260.6 mmol C m-2 d-1) and SR (91.4 mmol S m-2 d-1) at E0 were 4-9 and 6-54 times higher than at the site of outer sea (E1, E3, E5). Rates of SR at E3 and E5 accounted for 11-23% of anaerobic Corg oxidation, whereas it comprised 47-70% of anaerobic Corg oxidation at E0 and E1. Rates of Corg oxidation and SR showed a highly positive correlation with the concentration of dissolved organic carbon (r2 = 0.795 and 0.777, respectively). The BNF at E0, E1, and E3 accounted for 120-510% and 26-178%, respectively, of the N and P required for primary production in the water column. Overall results suggest that the Corg oxidation in the sediment controlled by concentration of dissolved organic carbon in the pore water and the excessive Corg oxidation stimulates the benthic nutrient flux, which may cause a phytoplankton bloom in the water column.

Simultaneous Enrichment of Novel Filamentous-Like Bacterial Population in Lab-Scale Granular Anaerobic Ammonia Oxidation (Anammox) Sequencing Batch Reactor (실험실 규모 입상 혐기성 암모늄 산화 연속회분식 반응조 내의 신종 사상균 동시 농화 배양에 관한 연구)

  • Enriching anammox bacteria (AMX) in a lab-scale granular sequencing batch reactor using local digester centrate, we observed the significant enrichment of the filamentous-like bacterial population. These bacteria were revealed as novel bacterial species (termed CHL) belonging to Chlorobi/Bacteroidetes phyla via Denaturing Gradient Gel Electrophoresis (DGGE). Further, niche differentiation of AMX and CHL quantification was observed in granule and filament biomass, suggesting AMX was dominant in the granule and CHL was dominant in the filament. Therefore, it was confirmed the structural role of CHL was indeed to aid the granule formation of the AMX. In parallel, the physiological role of CHL was suspected to degrade biopolymers in the digester centrate using nitrate as an electron acceptor.
