Effects of Ampelopsis Radix Pharmacopuncture Solution on Nitric Oxide and Prostaglandin $E_2$ Production in Macrophages (백렴약침액이 대식세포에서 NO와 Prostaglandin 생성에 미치는 영향)

  • Objectives : Recently, Pharmacopuncture therapy has been used for the treatment of inflammatory diseases such as rheumatoid arthritis. Especially, we have been interested in chemical mediators concerned with inflammation such as prostaglandin, nitric oxide. The purpose of this study is investigated that the effect of Ampelopsis Radix Pharmacopuncture solution in RAW 264.7 macrophages, performed several experimental items : those are Prostaglandin $E_2$, Nitric Oxide and Cyclooxygenase-2. Methods : The cytotoxicity of Ampelopsis Radix Pharmacopuncture solution in RAW 264.7 macrophages were measured by MTT assay. In order to observe cyclooxygenase-2 mRNA expression in lipopolysaccharide and interferon-gamma stimulated RAW 264.7 macrophages, RT-PCR was used. Prostaglandin $E_2$ production and Nitric Oxide production was measured by nitric oxide detection kit and Prostaglandin $E_2$ assay kit. Results : 1. The MTT assay demonstrated that cytotoxic effect of Ampelopsis Radix Pharmacopuncture solution in RAW 264.7 macrophages was not appeared. 2. Ampelopsis Radix Pharmacopuncture solution inhibited nitric oxide production in lipopolysaccharide and interferon-gamma stimulated RAW 264.7 macrophages. 3. Ampelopsis Radix Pharmacopuncture solution inhibited cyclooxygenase-2 mRNA expression in lipopolysaccharide and interferon-gamma stimulated RAW 264.7 macrophages. 4. Ampelopsis Radix Pharmacopuncture solution inhibited Prostaglandin $E_2$ production in lipopolysaccharide and interferon-gamma stimulated RAW 264.7 macrophages. Conclusions : On the basis of these results, It was shown that Ampelopsis Radix Pharmacopuncture solution was able to inhibit the production of $PGE_2$ and NO, as well as COX-2 mRNA expression. Our results may provide new mechanism by which Ampelopsis Radix Pharmacopuncture solution accounts for its beneficial effect on accelerating wound healing and anti-inflammation.

  • Adriamycin is a major cancer chemotherapeutic agent against a me range of human neoplasms. However, its clinical application is limited since It has a variety of side effects, bone marrow suppression and cardiotoxity, and this toxicity appears by free radical. This study investigated the effect of Ampelopsis radix on the toxicity of adriamycin. The methanol fraction reduced slightly adriamycin induced lipid peroxidation and superoxide production at the dose of 50mg /kg. 1.p., respectively. During the adriamycin administration. Protein bound-SH, nonprotein bound-SH, and glutathione-5-transferase did not change, but methanol fraction treated group were markedly increase. These results indicated that Ampelopsis radix has a major influence on the thiol group and related enzyme activity on the antioxidative effects.

The Effects of Ampelopsis Radix on Allergic Inflammation in PMA-stimulated Human Mast Cells (백렴의 알레르기 염증반응에 대한 억제효과)

  • Objectives: In this study, we investigated the inhibitory effects of Ampelopsis Radix methanol(AR-M) extract on allergic inflammation in activated human mast cells and its potential therapeutic or toxic effects. Methods: Ampelopsis Radix(AR) was extracted with 80% methanol. HMC-1 cells, a human mast cell line, were treated with different concentrations of AR-M extract, and then stimulated with PMA plus A23187. The cell toxicity of AR-M extract was determined by MTT assay. The concentrations of $PGE_2$ and cytokines were measured by ELISA. The gene expression of COX-2 and its protein levels were determined by RT-PCR and Western blot. The phosphorylation of ERK MAPK and the NF-${\kappa}B$ activation were determined by Western blot. Results: AR-M extract was significantly inhibited the production of PGE2 and inflammatory cytokines(TNF-${\alpha}$, IL-6 and IL-8) in PMA/A23187-stimulated HMC-1 cells. AR-M extract also attenuated the mRNA expression of COX-2 and its protein induction. Furthermore, AR-M extract attenuated PMA/A23187-induced phophorylation of ERK1/2 MAPK and the NF-${\kappa}B$ p65 subunit translocation into nuclear of HMC-1 cells. AR-M extract significantly decreased PMN A23187-induced release of histamine in a dose-dependent manner. Conclusions: These results indicate that Ampelopsis Radix shows the property of anti-allergic inflammation In vitro through suppressing the production of inflammatory mediators released from mast cells, suggesting have a potential for the treatment of allergic diseases.

  • In the result of investigating traditional chinese medical literatures to understand definite immunopharmacologic effects of drugs for clearing away heat and detoxicating such as Ampelopsis Radix, Rhapontici Radix, Cremastrae Appendiculatae Tuber, Rhaseoli Radiati Semen, Potentillae Discolohs Herba, Potentillae Chinensis Herba, Chrysanthemi Indici Flos, Lomcerae Caulis, we could reach conclusions as follows: 1. Rhapontici Radix, Chrysanthemi Indici Flos can increase voracity of leukocytes, macrophages and increase to produce IL-2 by splenocytes. 2. Potentillae Chinensis Herba, Chrysanthemi Indici Flos, Lonicerae Caulis can inhibit activities of B lymphocytes and have anti-inflammatory effects. 3. Drugs for clearing away heat and detoxicating almost have antibiotic, anti-inflammatory effects, and so can be applied to many inflammatory immune diseases. 4. Drugs for clearing away heat and detoxicating also have antifebrile, diuretic, detoxicating effects. Above results indicates that Drugs for clearing away heat and detoxicating have immunosuppressive effect, so that can be applied to many inflammatory immune diseases.

  • Objectives: This experimental study was carried out to evaluate the immune modulating and anti-tumor activity of Ampelopsis Radix extracts (ARE). Materials and Methods: To elucidate the effects of ARE on the macrophage and NK cell activity, we analyzed NO production, NK cytotoxicity and gene expressions of cytokine related with macrophage and NK cell activity. Results: ARE activated and promoted macrophages to product NO in part. And, ARE has significant properties to activate macrophages and NK cells by promoting related cytokines like IL-1, IL-12, IFN-$\gamma$, iNOS and TNF-$\alpha$ gene expressions. We also observed that ARE promoted protein expression of IFN-$\gamma$, and TNF-$\alpha$ in mice splenocytes. Conclusions: ARE is an effective herbal drug for immune modulating and anti-cancer by promoting activity of macrophages and NK cells.

Study on Pathology of Taeeumin Medicinal Herb in Sasang Constitutional Medicine (태음인 약재의 사상의학적 병리 연구)

  • Objective This study was designed to define the efficacy of medicinal herbs of Taeeumin in Sasang constitutional medicine. Method We searched and analyzed original text such as Donguisusebowon Sinchuk edition written in 1901, Donguisusebowon Gabo edition written in 1894, Donguisusebowon Sansang Chobongwon, and posthumous manuscripts left by Je-ma Lee published by the Ministry of Health of North Korea. Results Seven herbs such as Ephedrae Herba, Coicis Semen, Castaneae Semen, Fel Ursi, Mori Cortex Radicis, Ginkgonis Semen, and Farfarae Flos regulate the mechanism of Esophagus-Cold (Wiwanhan). Three herbs such as Melonis Pedicellus, Ailanthi Radicis Cortex, and Quisqualis Fructus regulate the mechanism of Cold Lung-Dry (HanpaeJo). Sixteen herbs such as Rhei Rhizoma, Angelicae Tenuissimae Radix, Puerariae Radix, Cimicifugae Rhizoma, Angelicae Dahuricae Radix, Moschus, Gleditsiae Spina, Mume Fructus, Aurum, Glycine Semen Germinatum, Ampelopsis Radix, Cornu rhinocerotis, Antelopis Cornu, Bomeolum, Bezoar Bovis, and Typhae Pollen regulate the mechanism of Liver-Heat (Ganyeol). Three herbs such as Chrysanthemi Indici Flos, Nelumbinis Semen, and Spirodelae Herba regulate the mechanism of Heat Lung-Dry (YeolpaeJo). Conclusion Forty four herbs of Taeeumin regulate the mechanisms of Esophagus-Cold (Wiwanhan), Cold Lung-Dry (HanpaeJo), Liver-Heat (Ganyeol), Heat Lung-Dry (YeolpaeJo) and correct the energy-fluid pathology of Taeeumin.

Anti-proliferative and Apoptosis Inducing Effect of Momordin I on Oral Carcinoma (KB) Cells

  • Treatment of oral cancers with chemotherapeutic agents become evaluated as an effective method to reduce cancer cell proliferation. Anti-proliferative and anti-oral cancer activities of momordin I on oral cancer cells were evaluated in this study. Momordin I was originally purified from a natural product, Ampelopsis radix and showed the antiproliferative activity against oral carcinoma, KB cells. Obtained $IC_{50}$ value was approximately $10.4{\mu}g/ml$. Time-and dose-dependent chromosomal DNA fragmentations were observed in momordin I-treated KB cells. Flow cytometry analysis showed time-dependent apoptotic cell appearance after treatment of momordin I. Approximately 18.6% apoptotic cells were observed at 72 hours after $20{\mu}g/ml$ of momordin I treatment. These observation were consistent with the results obtained in DNA fragmentation analysis. These data suggest that momordin I has anti-proliferative effect and induces cell death in KB cells through apoptosis.

  • Objective : The aim of this study is to evaluate anti-cancer effects of $Ampelopsisradix$ Extract (AE) on human lung cancer A549 cells. Method : The apoptotic activities and cell growth arrest activities of AE were measured using 3-[4,5-dimethyl-thiazol-2-yl]-2,5-diphenyltetrazolium bromide (MTT) assay. The molecules involved in apoptotic process were assessed by western blotting. Result : Treatment of AE potently reduced cell viability in a dose-dependent manner in A549 cells. AE (100-500 ${\mu}g/m{\ell}$) resulted in apoptosis via activation of caspase 9 following PARP cleavage in a time-and dose-dependent manner. The levels of Bax and Bad levels were increased by AE with a concomitant decrease of Bcl-xL. In addition, AE at the low dose (30 ${\mu}g/m{\ell}$) significantly inhibited cell growth in the presence of serum. Conclusion : AE has the potential as a therapeutic agent against lung cancer.

Inhibitory Effects of Momordin I Derivatives on the Formation of Fos-Jun-AP-1 DNA Complex

  • In our previous studies, we have observed that curcumin and momordin I isolated from Ampelopsis radix inhibit the formation of Fos-Jun-activation protein-1 (AP-1) DNA complex. We have screened more effective compounds which have a 5-membered ring framework like momordin I and have modified disaccharide or carboxylic acid portions in momordin I. We synthesized momordin I derivatives according to the published method with slight modification. Synthetic momordin I derivatives showed remarkable inhibitory activities on Fos-Jun-AP-1 DNA complex formation results in in vitro assays. The $IC_{50}$ values of momordin I derivatives were about 4.0 $\mu$M in an electrophoretic mobility shift assay (EMSA). This value is about 125 times higher than that of curcumin and about 12 times higher than that for curcumin derivative C1, and moreover about 30 times higher than that for momordin I. We found momordin I derivatives (a) and (b) are the strongest inhibitory compound for Fos-Jun-AP-1 DNA complex formation.
