Synthesis and Properties of Glycolic Acid Ester Type Cationic Surfactant (글리콜산 에스테르계 양이온 계면활성제의 합성 및 물성)

  • A cationic ester type surfactant, namely N-2-hydroxy-3-(2-hydroxyacetoxy) proply-N,N-dimethyldodecylaminium chloride(HPDA) was synthesized and confirmed by FT-IR and $^1H$-NMR spectroscopy. Surface tensions on the diluted aqueous solutions of the synthetic compounds were measured and evaluated critical micelle concentration. Surface tension was 33~34 dyne/cm in the range of $10^{-3}{\sim}10^{-2}mol/L$ and critical micelle concentration(c.m.c) value was $8.5{\times}10^{-3}mol/L$ by surface tension method. Emulsifying properties of the synthetic surfactant(HPDA) and Sodium lauryl sulfate (SLS), tetradecyl trimethyl ammonium bromide (TTAB) was tested. As a results, synthetic glycolic acid ester type compounds(HPDA) has been confirmed as a good emulsifier. The foaming power and stability of synthesized ester type surfactant was measured by Ross-Miles method.

  • Ordered mesoporous silica (MCM-41) materials with different textural properties were prepared using alkyl (dodecyl, cetyl, eicosane) trimethyl ammonium bromide (DTAB, CTAB, ETAB, respectively) as structure directing surfactants, functionalized with amine groups and used as adsorbent for the toxic metal ions, Cr (VI), As (V), Pb (II) and Hg (II). Amino functionalization of mesoporous MCM-41 was achieved by cocondensation of N-[3-(trimethoxysilyl)-propyl] aniline with tetraethyl orthosilicate. Adsorption isotherm and adsorption capacity of the amine functionalized materials for Cr (VI), As (V), Pb (II) and Hg (II) ions were followed by inductively coupled plasma mass spectrometry (ICP-MS). Results demonstrate that amine functionalized MCM-41 prepared with ETAB showed higher adsorption capacity for Cr (VI), As (V), Pb (II) and Hg (II) ions in comparison to MCM-41 prepared with CTAB and DTAB. The higher adsorption capacity for MCM-41(ETAB) was correlated with amine content in the material (determined by CHN analysis) and relative decrease in pore volume and pore diameter. X-ray diffraction (XRD) analysis, nitrogen adsorptiondesorption measurements and Fourier Transform infrared spectrometry (FTIR) were used to follow the changes in the textural parameters and surface properties of the mesoporous materials as a result of amine functionalization to correlate with the adsorption characteristics. The adsorption process was found to depend on the pH of the medium.

Dephosphorylation of Diphenyl-4-Nitrophenyl Phosphinate(DPNPIN) onto 2-Alkylbenzimidazolide Anion in TTABr Micellar Solution (TTABr 미셀 용액속에서 2-알킬벤즈이미다졸 음이온에 의해 추진되는 디페닐-4-니트로페닐 포스페네이트(DPNPIN)의 탈인산화반응)

  • This study is mainly focused on micellar effect of tetradecyltrimethyl ammonium bromide(TTABr) solution including alkylbenzimidazole(R-BI) on dephosphorylation of diphenyl-4-nitrophenylphosphinate(DPNPIN) in carbonate buffer(pH 10.7). Dephosphorylation of DPNPIN is accelerated by $BI^{\Theta}$ ion in $10^2$ M Carbonate buffer(pH 10.7) of $4{\times}10^{-4}$ M TTABr solution up to 80 times as compared with the reaction in Carbonate buffer by no benzimidazole(BI) solution of TTABr. The value of pseudo first order rate constant($k_{\psi}$) of the reaction in TTABr solution reached a maximum rate constant increasing micelle concentration. The reaction mediated by $R-BI^{\Theta}$ in micellar solutions are obviously slower than those by $BI^{\Theta}$, and the reaction rate were decreased with increase of lengths of alkyl groups. It seems due to steric effect of alkyl groups of $R-BI^{\Theta}$ in Stern layer of micellar solution. The surfactant reagent, TTABr, strongly catalyzes the reaction of DPNPIN with R-BI and its anion($R-BI^{\Theta}$) in Carbonate buffer(pH 10.7). For example, $4{\times}10^{-4}$ M TTABr in $1{\times}10^{-4}$ M BI solution increase the rate constant($k_{\psi}=99.7{\times}10^{-4}1/sec$) of the dephosphorylation by a factor ca. 28, when compared with reaction($k_{\psi}=3.5{\times}10^{-4}1/sec$) in BI solution(without TTABr). And no TTABr solution, in BI solution increase the rate constant($k_{\psi}=3.5{\times}10^{-4}1/sec$) of the dephosphorylation by a factor ca. 39, when compared with reaction ($k_{\psi}=1.0{\times}10^{-5}1/sec$) in water solution(without BI).

Optimization of HPLC Method and Clean-up Process for Simultaneous and Systematic Analysis of Synthetic Color Additives in Foods (식품 중 타르색소의 동시분석 및 계통분석을 위한 HPLC 분석조건 및 정제과정 확립)

  • To develop a method for separation process using Sep-pak $C_18$, simultaneous and systematic analysis of 8 permitted and 11 non-permitted synthetic food colors in Korea, optimization of analysis conditions for reverse phase ion-pair high performance liquid chromatography was carried out. For the best result of Sep-pak $C_18$ separation the pH of color standard mixture solution was $5{\sim}6$ and 0.1% HCl-methanol solution were set as eluent. The colors eluated from Sep-pak $C_18$ cartridge were determined and confirmed by high performance liquid chromatography with a photodiode array detector at 420 nm for yellow colors type, at 520 nm for red colors type, at 600 nm for blue and green colors type and at 254 nm for mixed colors. Conditions for HPLC analysis were as follows: column, Symmetry $C_18$ (5 m, 3.9 mm $i.d.{\times}150\;mm$); mobile phase, 0.025 M ammonium acetate (containing 0.01 M tetrabutylammonium bromide) : acetonitrile : methanol (65 : 25 : 10) and 0.025 M ammonium acetate(containing 0.01 M tetrabutylammonium bromide) : acetonitrile : methanol (40 : 50 : 10); flow rate, 1 mL/min. It takes 35 minutes for simultaneaus analysis and 18 minutes for systematic analysis. The detection limits range of each colors were $0.01{\sim}0.05\;{\mu}g/g$.

  • This study is mainly focused on micellar effect of cetyltrimethyl ammonium bromide(CTABr) solution including alkylbenzimidazole(R-BI) on dephosphorylation of isopropyl-4-nitrophenylphosphinate(IPNPIN) in carbonate buffer(pH 10.7). The reactions of IPNPIN with R-$BI^{\ominus}$ are strongly catalyzed by the micelles of CTABr. Dephosphorylation of IPNPIN is accelerated by $BI^{\ominus}$ ion in $10^{-2}$ M carbonate buffer(pH 10.7) of $4{\times}10^{-3}$ M CTABr solution up to 89 times as compared with the reaction in carbonate buffer by no benzimidazole(BI) solution of $4{\times}10^{-3}$ M CTABr. The value of pseudo first order rate constant($k_{\Psi}$) of the reaction in CTABr solution reached a maximum rate constant increasing micelle concentration. Such rate maxima are typical of micellar catalyzed bimolecular reactions. The reaction mediated by R-$BI^{\ominus}$ in micellar solutions are obviously slower than those by $BI^{\ominus}$, and the reaction rate were decreased with increase of lengths of alkyl groups. It seems due to steric effect of alkyl groups of R-$BI^{\ominus}$ in Stern layer of micellar solution. The surfactant reagent, CTABr, strongly catalyzes the reaction of IPNPIN with R-BI and its anion(R-$BI^{\ominus}$) in carbonate buffer(pH 10.7). And no CTABr solution, in $1{\times}10^{-4}$ M BI solution increase the rate constant($k_{\Psi}=3.9{\times}10^{-4}\;sec^{-1}$) of the dephosphorylation by a factor ca.39, when compared with reaction ($k_{\Psi}=1.0{\times}10^{-5}\;sec^{-1}$) in water solution(without BI). This predicts that the reactivities of R-$BI^{\ominus}$ in the micellar pseudophase are much smaller than that of $BI^{\ominus}$. Due to the hydrophobicity and steric effect of alkyl group substituents, these groups would penetrate into the core of the micelle for stabilization by van der Waals interaction with long alkyl groups of CTABr.

Optimized Condition of Genomic DNA Extraction and PCR Methods for GMO Detection in Potato (유전자재조합 감자의 검정을 위한 DNA분리 및 PCR검출의 최적조건 탐색)

  • To compare the quality of genomic DNA extracted from potato for PCR detection, four different methods, such as silica-based membrane method, silica-coated bead method, STE solution treatment, and CTAB-phenol/chloroform method, were evaluated. Also, to remove an excessive carbohydrate from the potato, ${\alpha}$- and ${\beta}$-amylase were used individually and in combination. When used both silica-based membrane method and silica-coated bead method combined with enzymes, the genomic DNAs were extracted from the raw potato with high purity for PCR. However, the silica-coated head method combined with enzyme treatment was the most efficient for extraction of the genomic DNA from the frozen fried potatoes. When applied with STE solution, the highly purified DNA was extracted from the raw potatoes without enzyme treatment in adequate yield for PCR. In cases of processed potatoes, such as frozen-fried potato and fabricated potato chips, CTAB-phenol/chloroform method is mostly feasible for DNA extraction and PCR efficacy at high sensitivity. As the results of PCR amplification, 216bp of PCR product was detected on 2% agarose gel electrophoresis, but any amplicons derived from New leaf and New leaf Y gene was not detected in any sample.

Preparation of Photocatalysts by Hydrothermal Precipitation Method and Their Photocatalytic Performance of Brilliant Blue FCF (수열합성법에 의한 광촉매 제조 및 Brilliant Blue FCF 분해 성능)

  • Experimental research on the preparation of photocatalyst for the decomposition of brilliant blue FCF ($C_{37}H_{31}O_9N_2S_3Na_2$) was performed. $TiO_2$ and ZnO powders were prepared from titanium (IV) sulfate and zinc acetate at low reaction temperature and atmospheric pressure by hydrothermal precipitation method without calcination. In addition, $TiO_2$ was prepared with cationic surfactant CTAB (Hexadecyltrimethyl ammonium bromide) at the same conditions. The physical properties of prepared $TiO_2$ and ZnO, such as crystallinity, average particle size and absorbance, were investigated by XRD, Zeta-potential meter and DRS. And, the photocatalytic degradation of brilliant blue FCF has been studied in the batch reactor under UV radiation. For the photocatalysts prepared without CTAB, $TiO_2$ has smaller particle size and larger absorbance and photocatalytic reaction rate than ZnO. And $TiO_2$, prepared with CTAB whose concentration is 1/10 of that of precursor, shows 15% higher than that prepared without CTAB in final photocatalytic degradation ratio of brilliant blue FCF.

Factors Affecting the Formation of Iodo-Trihalomethanes during Chlorination in Drinking Water Treatment (정수처리에서 염소 처리시 요오드계 트리할로메탄류 생성에 영향을 미치는 인자들)

  • Effects of bromide ($Br^-$) and iodide ($I^-$) concentrations, chlorine ($Cl_2$) doses, pH, temperature, ammonia nitrogen concentrations, reaction times and water characteristics on formation of iodinated trihalomethanes (I-THMs) during oxidation of iodide containing water with chlorine were investigated in this study. Results showed that the yields of I-THMs increased with the high bromide and iodide level during chlorination. The elevated pH significantly increased the yields of I-THMs during chlorination. The formation of I-THMs was higher at $20^{\circ}C$ than $4^{\circ}C$, $10^{\circ}C$ and $30^{\circ}C$. In chloramination study, addition of ammonium chloride ($NH_4Cl$) markedly increased the formation of I-THMs. Among the water samples collected from seven water sources including wastewater treatment plant (WWTP) effluent water (EfOM water), prepared humic containing water (HA water) and algal organic matter (AOM) containing water (AOM water), EfOM water generated the highest yields of I-THMs ($12.31{\mu}g/mg$ DOC), followed by HA water ($4.96{\mu}g/mg$ DOC), while AOM water produced the lowest yields of I-THMs ($0.99{\mu}g/mg$ DOC). $SUVA_{254}$ values of EfOM water, HA water and AOM water were $1.38L/mg{\cdot}m$, $4.96L/mg{\cdot}m$ and $0.97L/mg{\cdot}m$, respectively. The I-THMs yields had a low correlation with $SUVA_{254}$ values ($r^2$ = 0.002).

The Effects of Negative- and Positive- Charged Surfactants on In vitro DM Digestibility and the Growth of Ruminal Mixed Microorganisms (양(+) 이온성 및 음(-) 이온성 계면활성제 첨가가 반추위 혼합 미생물에 의한 In vitro 건물소화율 및 미생물 성장에 미치는 영향)

  • In order to investigate the effects of supplemental ionic surfactants in in vitro ruminal fermentation, N-Lauroylsarcosine sodium salt(N-LSS) and sodium dodecyl sulfate(SDS) for negative(-) ionic surfactant, and hexadecylpyridinium chloride monohydrate(HPCM) and hexadecyltrimethyl ammonium bromide(HTAB) for positive (+) ionic surfactant were supplemented by 0.05% and 0.1% into the Dehority’s artificial medium containing rice straw(1mm) as a substrate. In vitro DM digestibility, the growth of rumen mixed microbes, pH, cumulative gas production and SEM(Scanning Electron Microscopy) observation of microbial attachment on rice straw particle were investigated through the experiment composing 9 treatments (two supplemental levels of two positive ionic(+) surfactant, two supplemental levels of two negative(-) ionic surfactant) including the control. The sample collection was at 6, 12, 24, 48 and 72 h post fermentation with 3 replications per treatments. DM digestibility in treatments supplemented (+) or (-) surfactants almost stopped afterward 12 h fermentation, in vitro DM digestibility at 72 h post fermentation in the ionic surfactants was at half level of that of the control(P<0.05). Accumulative gas production in in vitro was less(P<0.05) with addition of ionic surfactants compared to the control. The amount of rumen mixed microbes recovered from in vitro incubation fluid pleateaued at 12 h post fermentation for the positive (+) ionic surfactants, but steadily increased as fermentation time elapsed for the control. Rumen microbial growth rate was significantly(P<0.05) low in the negative(-) ionic surfactant compared to the control. pH of the incubation fluid was ranged from 6.02 to 7.20, and was the highest in the negative(-) ionic surfactants, and was the lowest in the control(P<0.05). In SEM observation, rumen microbial population attached on rice straw particle was less with addition of ionic surfactants than the control. In conclusion we could not found any positive effects of negative- and positive- charged surfactants on rumunal fermentation characteristics and rumen microbial growth rates.

  • The reactions of p-nitrophenyldiphenylphosphate (p-NPDPP) with anions of benzimidazole (BI) and its 2-alkyl derivatives (R-BI) are strongly catalyzed by the micelles of cetyltrimethyl ammonium bromide (CTABr). On the other hand, the first order rate constants $(k'_{R-BI^-})$ and the second order rate constants $(k_{m(R-BI^-)})$ of the reactions mediated by R-$BI^-$in the micellar pseudophase are much smaller than those mediated by $BI^-$. In order to explain the slower rates of the micellar reactions mediated by R-$BI^-$, we compared the concentration-ratios ([R-$BI^-$]/[$BI^-$]) with the first order rate constant-ratios $(k'_{R-BI^-}/k'_{BI^-})$ and the second order constant-ratios $(k_{m(R-BI^-)}/k_{m(BI^-)})$ for the reactions taking place in the micellar pseudophase. The rate constant-ratios were much smaller than the concentration-ratios. For example in a 5 ${\times}10^{-4}$M butyl-BI solution, the two ratios were 0.089 and 0.430 (for the first order) respectively, and in a $10^{-4}$M butyl-BI solution the former was 0.100 (for the second order). This predicts that the reactivities of R-$BI^-$ in the micellar pseudophase are much smaller than that of $BI^-$. Based on the values of several kinetic parameters measured for dephosphorylation of p-NPDPP mediated by R-$BI^-$, a schemetic model is proposed. Due to the hydrophobicity and the steric effect of the alkyl substituents, these groups would penetrate into the core of the micelle for stabilization by van der Waals interaction with long cetyl groups of CTABr. Consequently, the movements of R-$BI^-$ bound to the micelle should be restricted, leading to decreased collison frequencies between the nucleophiles and p-NPDPP. We refer this as an "anchor effect". This effect became more predominent when a larger alky group in R-BI was employed and when a greater concentration of R-BI was used.
