Quality characteristics of Doenjang prepared with sweet potato (고구마를 이용한 된장의 품질 특성)

  • The effect of sweet potato on the quality of Doenjang was investigated during fermentation. Viable cells of yeast decreased gradually after 4 weeks of fermentation, but those of aerobic bacteria increased in the late stage. Amylase activity of Doenjang was higher in the late stage of fermentation, while neutral protease maintained high activity during fermentation. Hunter L and b values of Doenjang decreased gradually during fermentation, while a value was increased. The pH of Doenjang decreased gradually until 10 weeks of fermentation, and the titratable acidity was low in the sweet potato added groups. The acid value was low in the Shinyulmi sweet potato added Doenjang. Water activity and oxidation-reduction potential of Doenjang decreased during fermentation. Reducing sugar of Doenjang decreased in the middle stage of fermentation, and it was low in sweet potato added groups. The alcohol content of Doenjang decreased after 2 weeks of fermentation. Amino and ammonia-type nitrogen of Doenjang increased during fermentation and reached the maximum after 10 and 12 weeks of fermentation, respectively. After 12 weeks fermentation, 8% of Shinyulmi sweet potato added Doenjang was more favorable taste, flavor and overall acceptability (p<0.05) than the control or the Yeonwhangmi sweet potato added groups.

  • This study reports a numerical analysis of the internal flow characteristics of the integrated urea-SCR muffler system with the various geometries of the multi-perforated tube which is set up between the muffler inlet and in front of SCR catalysts. The multi-perforated tube is generally used to disperse uniformly the urea-water solution spray and to make better use of the SCR catalyst, resulting in the increased $NO_x$ reduction and decreased ammonia slip. The effects of the multi-perforated tube orifice area ratios on the velocity distributions in front of the SCR catalyst, which is ultimately quantified as the uniformity index, were investigated for the optimal muffler system design. The steady flow model was applied by using a general-purpose commercial software package. The air at the room temperature was used as a working fluid, instead of the exhaust gas and urea-water solution spray mixture. From the analysis results, it was clarified that the multi-perforated tube geometry sensitively affected to the formation of the bulk swirling motion inside the plenum chamber set in front of the SCR catalyst and to the uniformity index of the velocity distribution produced at the inlet of the catalyst.

The Effect of $MnO_2$ Addition on the $V_2O_5/TiO_2$ Catalytic Filters for NO Reduction (NO 환원반응을 위한 $V_2O_5/TiO_2$계 촉매필터의 $MnO_2$ 조촉매 효과)

  • Nitrogen oxides (NO, $NO_2$ and $N_2O$) have been controlled effectively by the SCR catalysts coated on monolith or honeycomb in commercial sites with ammonia as reductant at high temperature range $300{\sim}400^{\circ}C$. However, the catalytic filter has much merit on the point of controlling the particles and nitrogen oxides simultaneously. It will be more advanced-system if the catalytic working temperature is reduced to the normal filtration temperature of under $200^{\circ}C$. This study has focus on the development of the catalytic filter working at the low temperature. So the additive effect of the components such as Pt and Mn (which are known the catalytic component of $V_2O_5/TiO_2$ was investigated. The $V_2O_5-WO_3$ catalytic filter exhibited high activity and selectivity at $250{\sim}320^{\circ}C$ showing more than 95% NO conversion for the treatment of 600 ppm NO at face velocity 2 cm/s. The Pt-$V_2O_5-WO_3$ catalytic filter shifted the optimum working temperature towards the lower temperature ($170{\sim}200^{\circ}C$). And NO conversion was 100% and higher than that of $V_2O_5-WO_3$ catalyst at $250{\sim}320^{\circ}C$. The $MnO_X-V_2O_5-WO_3$ catalytic filter showed the wide temperature range of $220{\sim}330^{\circ}C$ for more than 95% NO conversion. This is a remarkable advantage when considered the $MnO_X$ catalytic filter presents the maximum activity at $150{\sim}250^{\circ}C$ and $V_2O_5-WO_3$ catalytic filter shows the maximum activity at $250{\sim}320^{\circ}C$.

Effects of Biologically Active Materials Prepared for Several Minerals and Plants on the Growth of Rumen Microbes (무기물성 및 식물성 생리활성 물질이 반추위 미생물의 성장에 미치는 영향)

  • In order to know the effects of scoria, germanium, charcoal, ginger, stevia, and CLA(Conjugated Linoleic Acid) as biologically active materials on pathogenic microbes and rumen anaerobic microbes, the growth rate of pathogens (including Escherichia coli O157, Salmonella paratyphi, Listeria monocytogenes and Staphylococcus aureus) and in vitro lumen microbial growth, gas production, ammonia concentration, carboxymethyl-cellulase (CMCase) activity, and microbial populations were investigated. The growth of pathogenic microbes was inhibited by the supplement of 0.10% ginger. Ginger had powerful antimicrobial properties on all the pathogens used in this experiments. Additionally in the antibacterial assay by paper disc method, we could observe the clear zone of similar area with the positive control(antibiotics) for E. coli as applied with the 10% stevia or the 10% CLA only. The supplements of ginger, stevia and CLA in vitro rumen fermentation inhibited populations of rumen bacteria and protozoa. Particularly supplement of ginger resulted in remarkable reduction of the protozoa population, which means it might serve as a source inhibiting material of methane creation in the rumen.

The Study of Reaction Characteristics of V/W/TiO2 Catalyst Using Se-TiO2 Support On NH3-SCR Reaction (Se-TiO2 지지체를 이용한 V/W/TiO2 NH3-SCR 촉매의 반응 특성 연구)

  • In this study, an experiment and a reaction characteristic study were conducted to enhance the reaction activity of V2O5/WO3/TiO2 at 300 ℃ or less by adding selenium to the support, in a selective catalytic reduction method using ammonia as a reducing agent to remove nitrogen oxides. Se-TiO2 and TiO2 were synthesized using the sol-gel method, and used as a support when preparing V2O5/WO3/TiO2 and V2O5/WO3/Se-TiO2 catalysts. The reaction activity of our catalyst was compared with that of a commercial catalyst. The denitration efficiency of the catalyst using TiO2 prepared by the sol-gel method was lower than that of the catalyst prepared using commercial TiO2, but was improved by the addition of selenium. Thus, the effect of selenium addition on the catalyst structure was analyzed using BET, XRD, Raman, H2-TPR, and FT-IR measurements and the effect of the increase in specific surface area by selenium addition and the formation of monomer and complex vanadium species on reaction characteristics were confirmed.

Complete genome sequence of Marinobacter salarius HL2708#2 isolated from a lava sea water environment on Jeju Island (제주용암 해수 환경에서 분리한 Marinobacter salarius HL2708#2의 유전체 해독)

  • During screening of microbes for compounds having cosmetic benefits, we isolated Marinobacter salarius HL2708#2 from lava seawater on Jeju Island, Republic of Korea. The complete genome sequence was determined. Strain HL27080#2 features a circular chromosome of 4,304,603 bp with 57.21% G+C content and a 244,163 bp plasmid with 53.14% G+C. There were 4,180 protein coding sequences identified, along with 49 transfer RNA and 18 ribosomal RNA noncoding genes. The genome harbored genes for the utilization of alcohol, maltose/starch, and monosaccharide as sole carbon sources. Genes responsible for halophilic characteristics and heavy metal resistance could be annotated, as well as aromatic and alkane hydrocarbons. Contrary to the prior report that M. salarius is negative for nitrate and nitrite reduction, nitrate/nitrite reductase along with nitrate/nitrate transporters and nitronate monooxygenase were evident, suggesting that strain HL2708#2 may be able to denitrify extracellular nitroalkenes to ammonia.

Ruminal pH pattern, fermentation characteristics and related bacteria in response to dietary live yeast (Saccharomyces cerevisiae) supplementation in beef cattle

  • Objective: In this study we aimed to evaluate the effect of dietary live yeast supplementation on ruminal pH pattern, fermentation characteristics and associated bacteria in beef cattle. Methods: This work comprised of in vitro and in vivo experiments. In vitro fermentation was conducted by incubating 0%, 0.05%, 0.075%, 0.1%, 0.125%, and 0.15% active dried yeast (Saccharomyces cerevisiae, ADY) with total mixed ration substrate to determine its dose effect. According to in vitro results, 0.1% ADY inclusion level was assigned in in vivo study for continuously monitoring ruminal fermentation characteristics and microbes. Six ruminally cannulated steers were randomly assigned to 2 treatments (Control and ADY supplementation) as two-period crossover design (30-day). Blood samples were harvested before-feeding and rumen fluid was sampled at 0, 3, 6, 9, and 12 h post-feeding on 30 d. Results: After 24 h in vitro fermentation, pH and gas production were increased at 0.1% ADY where ammonia nitrogen and microbial crude protein also displayed lowest and peak values, respectively. Acetate, butyrate and total volatile fatty acids concentrations heightened with increasing ADY doses and plateaued at high levels, while acetate to propionate ratio was decreased accordingly. In in vivo study, ruminal pH was increased with ADY supplementation that also elevated acetate and propionate. Conversely, ADY reduced lactate level by dampening Streptococcus bovis and inducing greater Selenomonas ruminantium and Megasphaera elsdenii populations involved in lactate utilization. The serum urea nitrogen decreased, whereas glucose, albumin and total protein concentrations were increased with ADY supplementation. Conclusion: The results demonstrated dietary ADY improved ruminal fermentation dose-dependently. The ruminal lactate reduction through modification of lactate metabolic bacteria could be an important reason for rumen pH stabilization induced by ADY. ADY supplementation offered a complementary probiotics strategy in improving gluconeogenesis and nitrogen metabolism of beef cattle, potentially resulted from optimized rumen pH and fermentation.

V2O5WO3/TiO2 Catalyst Prepared on Nanodispersed TiO2 for NH3-SCR: Relationship between D ispersed Particle Size of TiO2 and Maximum Decomposition Temperature of NOx (NH3-SCR용 나노분산 TiO2 담체상에 제조된 V2O5WO3/TiO2 촉매: TiO2 분산입도와 NOx 최대 분해온도와의 상관성)

  • For the selective catalytic reduction of NOx with ammonia (NH3-SCR), a V2O5WO3/TiO2 (VW/nTi) catalyst was prepared using V2O5 and WO3 on a nanodispersed TiO2 (nTi) support by simple impregnation process. The nTi support was dispersed for 0~3 hrs under controlled bead-milling in ethanol. The average particle size (D50) of nTi was reduced from 582 nm to 93 nm depending on the milling time. The NOx activity of these catalysts with maximum temperature shift was influenced by the dispersion of the TiO2. For the V0.5W2/nTi-0h catalyst, prepared with 582 nm nTi-0h before milling, the decomposition temperature with over 94 % NOx conversion had a narrow temperature window, within the range of 365-391 ℃. Similarly, the V0.5W2/nTi-2h catalyst, prepared with 107 nm nTi-2h bead-milled for 2hrs, showed a broad temperature window in the range of 358~450 ℃. However, the V0.5W2/Ti catalyst (D50 = 2.4 ㎛, aqueous, without milling) was observed at 325-385 ℃. Our results could pave the way for the production of effective NOx decomposition catalysts with a higher temperature range. This approach is also better at facilitating the dispersion on the support material. NH3-TPD, H2-TPR, FT-IR, and XPS were used to investigate the role of nTi in the DeNOx catalyst.

Effect of Four Medicinal Plants on In Vitro Ruminal Fermentation and Methane Emission (약용식물 4종의 in vitro 반추위 발효 성상 및 메탄 저감에 대한 영향)

  • The objective of this study was to the effect of four medicinal plants (Rheum palmatum, Pharbitidis semen, Reynoutria japonica, Tribulus semen) supplementation on methane reduction and ruminal fermentation in in vitro batch culture method. Each medicinal plant was supplemented 5% on a substrate basis in the bottle, then filled with buffered rumen fluid. Incubation was conducted for 24 hours in a shaking incubator (39℃, 120 rpm). The ruminal pH values were not significantly different between the control and treatment groups. However, the digestibility of the feed was significantly higher in the group supplemented with medicinal plants than control group. Methane production (mL/g of digested dry matter) and total gas production (mL) was significantly lower in the treatment group compared to the control group in Tribulus semen group. Total volatile fatty acids concentration were significantly higher in all treatment groups than control group, and acetate concentration was significantly higher in all treatment groups than control group except for Rheum palmatum group. Propionate concentration was significantly higher in all treatment groups than control group, while butyrate concentration was significantly higher in Rheum palmatum group than control group. Ammonia nitrogen concentration was significantly higher in all treatment groups than control group. In conclusion, the addition of medicinal plants did not negatively impact rumen fermentation, and the results indicate that Tribulus semen has potential as a feed additive for reducing methane emissions.

Effect of Gaseous Chlorine Dioxide on Sterilization in Industrial Food-holding Cabinets (이산화염소가스를 이용한 식품산업용 소독장에서의 살균효과)

  • The aim of this study was to investigate the effect of different concentrations of chlorine dioxide ($ClO_2$) on sterilization and deodorization of food-holding cabinets under different exposure times. For the measuring sterilization and deodorization, a 6.5 L chamber and a 625 L cabinet with circulation systems were used. Two bacteria (Staphylococcus aureus KCTC1916 and Escherichia coli KCTC 1682) that were artificially inoculated in the plate respectively were put into the 6.5 L chamber and the 625 L cabinet. The $ClO_2$ gas was produced by ampules. In the 6.5 L chamber, neither of the two bacteria was detected after 24 hours treatment by $ClO_2$ gas. Moreover, the deodorization rate against ammonia and phenol was 94% and 70%, respectively, but deodorization against formaldehyde was not effective. When the concentration reached maximum (6 ampule, 4.6 ppm) levels in the cabinet, it lasted for approximately 2 h and then decreased slowly. When a circulator was used, the gas concentration was very low (6 ampule, 0.8 ppm) and the antibacterial activity against S. aureus and E. coli was low. The level of reduction against S. aureus and E. coli was 2.98 log CFU/plate and 6.06 log CFU/plate, respectively, in the cabinet after 24 h without a circulator. The reduction against S. aureus KCTC1916 and E. coli KCTC1682 was 2.69 log CFU/plate and 4.41 log CFU/plate for 24 h, respectively.
