발암물질의 조기검색법 개발 및 Chemoprevention에 관한 연구

  • 발암물질의 조기검색법 개발 및 chemoprevention연구의 일환으로 발암물질과 DNA 및 단백질의 공유결합체인 발암물질-DNA 및 -단백질 adduct를 연구하였다. 발암물질(예, 밴조피렌)-단백질 adduct에 관한 연구에서는 시료(단백질)에 soluble protease를 이용하는 간편하고 손쉬운 ELISA(Enzyme Linked Immunosorbent Assay)분석법을 확립했다. 발암물질(예,벤조피렌,아플라톡신 B1) -DNA 및 -단백질 adduct를 이용한 발암성 조기검색법의 개발을 Ames test 및 염색체이상시험과 비교 연구한 결과 본 연구에서 새로이 개발한 DNA 및 Protein-adduct형성 시험법은 저농도에서 고농도에 이르기까지 뚜렷한 용량-반응 관계를 나타냈으며 Ames test 및 Chromosomal test에서 일어날 수 있는 false positive나 false negative의 결과를 나타낼 우려가 없었다. 벤조피렌-DNA adduct를 이용한 chemoprevention 연구에서는 항산화제로 알려진 비타민 E,C 및 $\beta$-carotene을 시험한 결과 용량의존적으로 벤조피렌-DNA adduct 형성을 억제하였다.

  • Antigenotoxicity test (SOS chromotest) and antimutagenecity test (Ames test) were carried out using water-soluble and methanolic extracts from Dianthi Herba. Antigenotoxic activity of methanolic extract against mutagens both MNNG and NQO was much more effective than that of water-soluble one. When the extract was added to the certain concentration $(100\;{\mu}l/tube)$, antigenotoxic activities against both mutagens were enhanced. Against the mutagen MNNG with Ames test, antimutagenic activity of the methanolic extract was better than that of water-soluble one. The 74.6% of inhibition ratio for revertant forming CFU/plate was shown at $300\;{\mu}l/plate$ of the methanolic extract.

  • To assess the genotoxicity of AS6, several classical toxicological tests were performed. In Ames test, AS6 did not show any transformation of revertant with or without S-9 metabolic activating system, indicating the lack of mutagenic effect of the compound. To assess clastogenic effect, in vivo micronucleus and in vitro chromosomal aberration assays were performed using male ICR mice and Chinese hamster lung (CHL) fibroblast cells, respectively. Chromosomal aberration was not induced regardless of the presence of S-9 metabolic activating system. In addition, AS6 did not cause any increase in the incidence of micronucleated polychromatic erythrocytes at any of the dose levels, suggesting little clastogenicity in vitro or in vivo. Taken together, these results demonstrate that AS-6 has no mutagenic effect in our test system.

  • To confirm the effects of binlang(Areca catechu L.) and tansen(Salvia miltriorrhiza bung) on the mutagenicity induced by hydrogen peroxide, SOS Chromotest with Escherichia coli PQ37 and Ames test with Salmonella typhimurium TA104 were perlormed. Methanol-soluble parts of their water extracts showed high inhibitory effect against the mutagenicity of hydrogen peroxide in two bacterial mutation assays. Step-wise fractionation of methanol-soluble part from tansen was done using ethyl acetate, butanol and water. Among these fraction was further partitioned by Sephadex LH-20 column chromtography, and 6 subfractions were obtained. The fraction III showed the strongest inhibitory effects against the mutagenic activities induced by hydrogen peroxide. The inhibition rates of fraction III at concentration of 500$\mu\textrm{g}$/assay were 28%, 30% and 15% against 4-NQO, MNNG and B(a)P, respectively. But the mutagenic potency of AFB1 was increased.

  • 2- [(4- Cyanophenyl)amino] -3-chloro-1, 4- naphthalenedione (NQ-Y15) was asssayed for its genotoxic potential by using Salmonella typhimurium reversion assay and in vitro chromosome aberration test on Chinese hamster lung cells. In the Ames test, NQ-Y15 induced his + revertants of Salmonella typhimurium TA 98 and TA1537, reaching levels twice the negative control values. But, NQ-Y15 induced only his+ revertants of Salmonella typhimurium TA1537 more than twice the control values under the condition with metabolic activation system. In the cytogenetic test on chinese hamster lung cells. NQ -Y15 showed significant chromosomal aberrations, but the incidence was significantly reduced in the presence of metabolic activation.

  • The tyrosinase inhibition activity and mutagenicity as assessed by the Ames test on phenolic antioxidants (5-Caffeoyl quinic acid, 3,4-Dihydroxy cinnamic acid, Quercetin 3-O-${\beta}$-D-glucopyranose, Kaempferol 3-O-${\beta}$- D-glucopyranose) and the ethyl acetate fraction isolated from mulberry leaves were examined. The ethyl acetate fraction and chlorogenic acid exhibited weaker tyrosinase inhibitory activities than kojic acid. In addition, the ethyl acetate fraction from mulberry leaves, containing phenolic antioxidants, showed no mutagenicity by the Ames test.

Mutagenicity of the Essential Oils in Ames Test (Ames Test에 의한 정유의 돌연변이원성)

  • Mutagenic acivity of essential oils was tested using Salmonella typhimurium TA100 in the presence or absence of 59 fraction prepared from the mouse liver. Growth inhibitory effect of the oils on the bacteria was measured to warrant the mutagenic effect. Most oils were (round to be very strongly toxic against the bacteria at a high dose $(2,000{\mu}g/plate)$. At lower doses than this concentration, the Curcuma longa oil was found to be the most mutagenic with S9 fraction whereas it was not mutagenic without the fraction suggesting that this oil could undergo activation for the mutagenicity by cytochrome P45O. However, the mutagenicity of the Eugenia caryohpylata oil was disappeared under S9 fraction. Other oils obtained from Cinnamomum cassia, Chrysanthemum sibiricum, Paeonia moutan the flower of Artemisia princeps var. Orientalis, Allium sativum, were not mutagenic. This result suggested that antimutagenicity assay on the essential oil is necessary for the biological available substances.

Antimutagenic Effect of Extracts from Wild Mushrooms (야생버섯 추출물의 항돌연변이 효과)

  • The inhibitory effects of methanol extracts from wild mushrooms on mutagenicity induced by benzo(a)pyrene (B(a)P) and N-methyl-N-nitro- nitrosoguanidine(MNNG) were investigated using Salmonella typhimurium reversion assay. In Ames test, The methanol extracts of 11 mushrooms did not show any mutagenicity but The methanol extracts of Lactarius piperatus, Naematoloma fasciculare and Innotus xeranticus showed 40∼80% of inhibitory effect on the mutagenicity induced by indirect mutagen of B(a)P and also showed 60∼80% of antimutagenic activity toward MNNG irrespective of their concentrations.

  • Kyoaesamultang(KAT) has been used as an important prescription for various diseases including threatened abortion, associated with pregnancy in traditional medicine. In oder to identify the safety of KAT, this study was designed to determine mutagenicity and hepato-toxicity. In Rec-assay, Bacillus subtills H-17($Rec{^+}$) and M-45($Rec{^-}$) strains were used to clarify the DNA damage property. In Ames test, Salmonella typhimurium TA98 and TA100 were used for mutagenicity testing. In SOS umu test, Salmonella typhimurium TA1535 containing plasmid pSK1002 was used as a tester strain, and the levels of umu operon expression were monitored by measuring the $\beta$-galactosidase activity. From tested results, KAT did not show DNA damage and mutagenicity. On the other hand, hepato-toxicity of KAT to female ICR mice was monitored by the measurements of s-GOT, s-GPT and LDH activities after oral feeding for 15days. KAT showed 34% increase of s-GOT and s-GPT activities, also exhibited 35% increase of LDH activity in mice sera.

Study on the Mutagenicity of Drinking Water (음용수의 변이원성에 관한 조사연구)

  • This is a study on the risk assessment of drinking water using mutagenicity testing. The tests have been carried with the raw water, treated water, and drinking water (tap water) in Kwangju and Mokpo areas. The Ames preincubation test was carried concentrating samples using by Sep-Pak PLUS cartriges in Salmonella typhimurium TA100 and TA98. The samples were tested with several chemical water quality analysis. The THMs have not been measured in raw water, but measured treated water and tap water at a value of 7.135-12.473 $\mu$g/l. It was observed that the number of revertants colonies increased in treated water and tap water on TA100 without S9 and showed weak mutagenicity on TA98 without S9. Indirect mutation was not seen in TA100 and TA98 with S9. The results indicated that formed substances of treatment process's of water that increased mutagenicity.
