Objective: This study aimed to develop and evaluate the effectiveness of a water-soluble microencapsulation method for probiotic strains using gum Arabic (GA) and skim milk (SKM) over a three-month storage period following processing. Methods: Four strains of Pediococcus acidilactici (BYF26, BYF20, BF9, and BF14) that were typical lactic acid bacteria (LAB) isolated from the chicken gut were mixed with different ratios of GA and SKM as coating agents before spray drying at an inlet temperature 140℃. After processing, the survivability and probiotic qualities of the strains were assessed from two weeks to three months of storage at varied temperatures, and de-encapsulation was performed to confirm the soluble properties. Finally, the antibacterial activity of the probiotics was assessed under simulated gastrointestinal conditions. Results: As shown by scanning electron microscopy, spray-drying produced a spherical, white-yellow powder. The encapsulation efficacy (percent) was greatest for a coating containing a combination of 30% gum Arabic: 30% skim milk (w/v) (GA:SKM30) compared to lower concentrations of the two ingredients (p<0.05). Coating with GA:SKM30 (w/v) significantly enhanced (p<0.05) BYF26 survival under simulated gastrointestinal conditions (pH 2.5 to 3) and maintained higher survival rates compared to non-encapsulated cells under an artificial intestinal juices condition of pH 6. De-encapsulation tests indicated that the encapsulated powder dissolved in water while keeping viable cell counts within the effective range of 106 for 6 hours. In addition, following three months storage at 4℃, microencapsulation of BYF26 in GA:SKM30 maintained both the number of viable cells (p<0.05) and the preparation's antibacterial efficacy against pathogenic bacteria, specifically strains of Salmonella. Conclusion: Our prototype water-soluble probiotic microencapsulation GA:SKM30 effectively maintains LAB characteristics and survival rates, demonstrating its potential for use in preserving probiotic strains that can be used in chickens and potentially in other livestock.
Environmental impact assessment survey reflecting farmers` opinion on the residence and production space in rural settlement area by ORD showed that more than 86% of respondents thought their reservoirs and waterways (small rivers) were getting seriously contaminated primarily by garbage and livestock manure. A typical rural settlement unit was taken to assess the impact of improper management of livestock manure in the farms on the water quality of small river flowing down along the villages where swine and dairy farms were situated in Daejook 2, 3-ri, Seolseong-myun, Icheon-gun. Nitrogen compounds such as NO$_3$-N, NO$_2$-N, NH$_3$-N, and phosphorus compound H$_x$PO$_4$, DO, BOD$_5$, COD, and microbial density were analyzed to evaluate water quality at five test sites designated along the water stream. Tests showed. for example, BOD$_5$ at site 4 was average 9.2mg/l which was about 3~8 times higher than that of observation site 2 and 3, at which most livestock houses were situated. This is a clear evidence that the nutrients of livestock manure illegally discharged to small river can lead to an eutrophication of the river at downstream. A soil absorption system with aeration could be one of alternatives to treat the contaminated wastewater by livestock manure. The place at downstream, inbetween observation site 1 and 2, could be the best construction site for the treatment facility from the standpoint of the overall treatment efficiency, An enclosed composting system can also be regarded as a good alternative for treatment of the sludge which is the by-product of the soil absorption system operation.
Objective: This study investigates the technological and sensory profile of boneless dry-cured ham with different contents of lactulose added as a prebiotic ingredient. Methods: In addition to the control samples (without the addition of lactulose), three treatments were formulated to contain 2%, 4%, or 6% lactulose. Technological (lactulose content, instrumental color and texture profile analysis) and sensory (acceptance and check-all-thatapplies tests) analyses were performed on the final product. Results: The lactulose content in the finished product (1.86%±0.23%, 3.16%±0.18%, and 2.51%±1.35%) was lower than the lactulose originally added (2%, 4%, and 6%, respectively). The addition of 4% and 6% lactulose made (p<0.05) the products darker (lower L⋆) and redder (lower h) with higher hardness and chewiness values, when compared to control samples. The additions of 2% and 4% lactulose reduce the appearance acceptability of the products, but overall the treatments were well accepted. Conclusion: The use of up to 4% lactulose as a prebiotic in the production of boneless dry-cured hams provides an alternative to improving its nutritional value with little alteration in the technological characteristics and still meeting the sensory characteristics desired by consumers.

Background: Antibiotic resistance is a significant public health concern around the globe. Antimicrobial peptides exhibit broad-spectrum and efficient antibacterial activity with an added advantage of low drug resistance. The higher water content and 3D network structure of the hydrogels are beneficial for maintaining antimicrobial peptide activity and help to prevent degradation. The antimicrobial peptide released from hydrogels also hasten the local wound healing by promoting epithelial tissue regeneration and granulation tissue formation. Objective: This study aimed at developing sodium alginate based hydrogel loaded with a novel antimicrobial peptide Chol-37(F34-R) and to investigate the characteristics in vitro and in vivo as an alternative antibacterial wound dressing to treat infectious wounds. Methods: Hydrogels were developed and optimized by varying the concentrations of crosslinkers and subjected to various characterization tests like cross-sectional morphology, swelling index, percent water contents, water retention ratio, drug release and antibacterial activity in vitro, and Pseudomonas aeruginosa infected wound mice model in vivo. Results: The results indicated that the hydrogel C proved superior in terms of cross-sectional morphology having uniformly sized interconnected pores, a good swelling index, with the capacity to retain a higher quantity of water. Furthermore, the optimized hydrogel has been found to exert a significant antimicrobial activity against bacteria and was also found to prevent bacterial infiltration into the wound site due to forming an impermeable barrier between the wound bed and external environment. The optimized hydrogel was found to significantly hasten skin regeneration in animal models when compared to other treatments in addition to strong inhibitory effect on the release of pro-inflammatory cytokines (interleukin-1β and tumor necrosis factor-α). Conclusions: Our results suggest that sodium alginate -based hydrogels loaded with Chol-37(F34-R) hold the potential to be used as an alternative to conventional antibiotics in treating infectious skin wounds.
In order to find an alternative source of inoculum to caecal content for studying the fermentation activity of rabbit hindgut, caecal content and faeces of 25 hybrid Hyla rabbits were used as inocula for an in vitro gas production trial. About 1 g of three substrates (dehydrated alfalfa meal, dehydrated beet pulp, barley) was weighed, in quadruplicate per inoculum, in 120 ml bottles; 75 ml of anaerobic medium and 4 ml of reducing solution were added and bottles were placed at $39^{\circ}C$. Caecal content and faeces were diluted respectively 1:2 (CI) and 1:8 (FI) with anaerobic medium and were introduced in the respective bottles (10 ml). Gas production was recorded 20 times at 2-24 h intervals throughout fermentation (96 h). The fermentation characteristics (i.e. degraded organic matter, OMd; potential gas production, A; fermentation rate, Rmax; time at which it is reached, Tmax; pH, volatile fatty acid, VFA) were studied by inoculum and feedstuffs. The feedstuffs, according to their chemical composition, showed very different fermentation characteristics. In particular, OMd, A and Rmax allowed feedstuff classification as follows: barley>beet pulp>alfalfa. The inocula differ (p<0.05) in Tmax, were higher for CI (15.53 vs. 11.96 h) and in VFA production. In particular, CI produced higher levels of acetate (38.9 vs. 33.4 mM/g OM incubated, p<0.01) and isobutyrate (0.72 vs. 0.42, p<0.01) but less propionate (7.1 vs. 10.3, p<0.01) and butyrate (11.3 vs. 14.0, p<0.01). However, the trend of gas production, similar for the inocula according to the fermented substrate, and the good regression equation to estimate some caecal fermentation parameters from faeces suggest that, after standardisation, the faeces could be used as an alternative inoculum for gas tests in rabbit.
Duroc 품종은 돼지 사육에 있어 산육성과 육질 향상을 위해 이용되고 있다. 본 연구는 육종에 많이 이용되는 Duroc 품종의 모계 특이적인 서열의 검색과 계통유전학적 유연관계의 정립을 위하여 미토콘드리아 유전체의 전체 염기서열을 결정하고 집단 내 다형성을 조사하였다. mtDNA 전체 서열의 길이는 16,584-bp 이고, D-loop과 tRNA, rRNA 유전자 영역에서는 삽입/결실이 확인되었다. 4개의 coding gene (COⅡ, COⅢ, ND3, ND4)에서 불완전한 종결코돈을, ND4L과 ND2 유전자는 선택적 개시코돈 양상을 보였다. Duroc 집단에 대한 분석 결과 조절영역에서의 특이적인 11-bp 중복 단위가 일부 개체(15.2%)에서 발견되었고, ND2의 개시코돈과 CYTB 유전자에서도 다형현상을 보였다. 각각의 유전자 영역에서의 다형성은 서로 연관되어 있었고, 그 결과 Duroc 집단은 크게 두 가지 haplotype으로 구분되었다. 계통수에서 Duroc mtDNA 서열은 유럽계열 cluster에 위치하였으나, haplotype 분석과 기존에 연구결과들을 종합해 보면 Duroc 품종은 여러 모계선조 집단에서 기원한 것으로 보이며, 유럽과 아시아 계열 모두가 품종 형성에 이용된 것으로 사료된다된 것으로 사료된다.
농약을 포함한 화학물질의 독성을 평가하기 위하여 많은 실험동물이 사용되고 있다. 최근 동물실험에 대한 규제 강화와 실험동물의 윤리적 보호를 위해 동물실험을 대체하려는 동물대체시험법 연구가 활발히 진행되고 있다. 우리나라에서 화학농약 등록 시 급성경구독성시험, 급성경피독성시험, 피부자극성시험 등 여러 시험에서 실험동물을 이용하여 인축독성평가를 하고 있다. 세계적인 추세에 맞춰 농약의 안전성 평가를 위한 동물대체시험법 연구제시와 규제가 시급한 실정이다. 본 연구는 국제적으로 승인된 대체시험법 중 인공피부모델을 이용한 피부자극성평가 적용 가능성을 검토하기 위하여 실시하였다. 시험물질은 기존의 토끼를 이용한 피부자극성시험에서 피부반응을 평점화하여 산출한 피부 1차 자극지수(P. I. I.) 0-7(구분:없음-강도) 값을 가진 농약제품 16종을 선발, 인공피부(KeraSkin$^{TM}$)를 이용하여 피부자극성평가를 수행하였다. 그 결과, 중도이상의 농약인 경우 세포생존율 50% 이하로 자극성 물질로 나타났으며, 경도 이하의 농약인 경우 세포생존율 50% 이상으로 비자극성 물질로 나타났다. 피부의 자극성이 있는 경도에서 강도의 농약의 경우 현재 농촌진흥청 고시 농약표시 기준에 따라 주의사항을 표시하게 되어 있고(RDA, 2012b), 농약의 피부자극성의 경우 민감도가 사람에 따라 많은 차이를 보이기 때문에 피부자극성 대체시험법 적용여부는 추후 추가적인 검토가 필요할 것으로 판단되며, 앞으로 농약안전성평가를 위한 동물실험을 대체할 수 있는 시험법 검증연구를 통해 농약등록 시험기준과 방법을 개선할 필요성이 있다고 사료된다.
Hypertension is the chronic disease that the 16% of total population are suffering, and it needs to be studied to find alternative treatment because of the tolerance and side effect of medications that may bother some patients. in this paper, we verified practicality of implantable electrical stimulator that can readily change stimulus magnitude and frequency. And this device is possible to stimulate baroreflex or parasympathetic nerve. Therefore we performed in vitro tests and animal experiment for device's operating conditions. This device consist of implantable electrical stimulator and extracorporeal control/monitoring system. Stimulator was designed to make 1Hz~100Hz pulses and it can change continuous or periodic pulse train type. And this device can control stimulator's function and monitor stimulator's status and patients' blood pressure at exterior of body using ZigBee module as wireless telecommunication. We verified that stimulator have error rate under 5% at 50mm depth of organs and, stimulator makes high-efficiency energy with closer position of two electrodes. Also we can confirm the performance of device that decreasing blood pressure and heart rate of a rat by electrical stimulation.
생체내 피부 자극은 일련의 복합적인 생리 화학적 변화를 수반한다. 이러한 생체내 현상을 보다 민감하게 반영하고, 동물 시험 경감 측면 및 정확성, 재현성을 보완하기 위하여 자극 물질 검색을 위한 대체 시험법이 필요하게 되었다. CAMVA는 이러한 필요성에 의해 고안된 방법의 하나로써 약 10일된 유정란의 복합적인 혈관이 융합된 장뇨막내에 자극 물질을 투여하고 일정 시간 후, 막 내 혈관의 충혈, 출혈, 응집현상 등의 변화를 통해 자극 정도를 평가하는 in vitro 시험계이다. 본 연구에서는 화장품 영역에서 중요한 위치를 차지하는 계면활성제의 자극 정도를 검색하여 in vitro 시험법인 CAMVA와 in vivo 시험법간의 상관성을 조사하였으며, 그 결과 매우 높은 상관성을 나타냄을 알 수 있었다. 그러므로 CAMVA는 안 자극 시험의 대체 시험법 뿐만 아니라 피부자극의 예측에도 적용 가능한 유용한 시험법이라 생각된다.
본 연구에서는 성체 한우의 도축과정 중에 채혈된 혈액으로부터 분리 정제한 혈청의 생물학적 안전성을 검증하기 위하여 세균, 마이코플라즈마 및 바이러스의 검출과 동정을 실시하였다. 검사를 실시한 대부분의 혈청에서 전염성 병원체가 검출되지 않았고 두 개의 혈청에서 토양 유래의 것으로 추정되는 Bacillus thuringiensis가 검출 되었다. 본 연구의 결과는 국내에서 도축되는 한우는 생물학적 안전성을 담보할 수 있는 최소한의 기본적인 안전장치를 통해 도축된다는 사실을 보여주며 한우의 도축과정에서 부산물로 얻을 수 있는 혈액을 안전한 생물학적 제제로 개발할 수 있다는 가능성을 보여주는 것으로 생각된다.
