쇠뜨기(Equisetum arvense) 추출물의 항산화 활성 분석 및 ${\alpha}-glucosidase$ 저해활성 (Studies for Component Analysis, Antioxidative Activity and ${\alpha}-glucosidase$ Inhibitory Activity from Equisetum arvense)

  • 쇠뜨기 추출물을 이용하여 당뇨병에 대한 기능성 의약품 개발을 목적으로 쇠뜨기 가루의 화학성분, 항산화 활성을 조사하였다. 일반성분은 탄수화물이 53.20%로 탄수화물의 함량이 가장 높았다. 그 밖에 조회분, 조단백질, 수분, 조지방 순으로 나타났으며, 각각 20.42%, 15.32%, 8.85%, 2.21%로 나타났다. 쇠뜨기 가루에 가장 많은 무기성분으로는 K, Ca등 2종류가 기타원소에 비해 다량 함유되어 있었다. 이중 가장 많이 검출된 것은 K로 3768.12 mg/100 g으로 가장 많았으며, Ca은 1,130.44 mg/100 g로 나타났다. P, Mg, Na는 $180{\sim}110\;mg/100g$이며, 가장 적은 양을 보인 것은 Zn이 2.19 mg/100 g로 나타났다. 구성당은 총 2종이 분리 동정되었으며, 그 중 glucose가 13,351.88 mg/100 g, mannose는 1,616.31 mg/100 g으로 나타났다. 지방산 중 가장 많이 포함하고 있는 것은 linolenic acid$(C_{18:3})$가 42.83%으로 나타났으며 linoleic acid$(C_{18:2})$ 23.60%, palmitic acid$(C_{16:0})$ 19.00% 순으로 나타났다. 또한 총 지방산 중 포화지방산이 25.57%로 불포화 지방산이 포화지방산에 비해 3 배정도 많이 함유하고 있는 것으로 드러났다. DPPH 라디칼 소거법에 의한 쇠뜨기 추출물들의 항산화 활성은 70% ethanol 추출물의 항산화 활성$(IC_{50})$$168.1\;{\mu}g/ml$로 가장 높게 나타났고, 다음으로 70% methanol 추출물이 $187.8\;{\mu}g/ml$으로 나타났으며, chloroform 추출물이 $194.2\;{\mu}g/ml$, methanol, 물, ethanol추출물 순으로 나타났다. ${\alpha}-Glucosidase$ 측정값은 70% methanol 추출물에서 82.05%의 높은 저해 활성을 보였다.

Protective effects of perilla oil and alpha linolenic acid on SH-SY5Y neuronal cell death induced by hydrogen peroxide

  • BACKGROUND/OBJECTIVE: Oxidative stress plays a key role in neuronal cell damage, which is associated with neurodegenerative disease. The aim of present study was to investigate the neuroprotective effects of perilla oil (PO) and its active component, alpha-linolenic acid (ALA), against hydrogen peroxide $(H_2O_2)$-induced oxidative stress in SH-SY5Y neuronal cells. MATERIALS/METHODS: The SH-SY5Y human neuroblastoma cells exposed to $250{\mu}M$ $H_2O_2$ for 24 h were treated with different concentrations of PO (25, 125, 250 and $500{\mu}g/mL$) and its major fatty acid, ALA (1, 2.5, 5 and $25{\mu}g/mL$). We examined the effects of PO and ALA on $H_2O_2$-induced cell viability, lactate dehydrogenase (LDH) release, and nuclear condensation. Moreover, we determined whether PO and ALA regulated the apoptosis-related protein expressions, such as cleaved-poly ADP ribose polymerase (PARP), cleaved caspase-9 and -3, BCL-2 and BAX. RESULTS: Treatment of $H_2O_2$ resulted in decreased cell viability, increased LDH release, and increase in the nuclei condensation as indicated by Hoechst 33342 staining. However, PO and ALA treatment significantly attenuated the neuronal cell death, indicating that PO and ALA potently blocked the $H_2O_2$-induced neuronal apoptosis. Furthermore, cleaved-PARP, cleaved caspase-9 and -3 activations were significantly decreased in the presence of PO and ALA, and the $H_2O_2$-induced up-regulated BAX/BCL-2 ratio was blocked after treatment with PO and ALA. CONCLUSIONS: PO and its main fatty acid, ALA, exerted the protective activity from neuronal oxidative stress induced by $H_2O_2$. They regulated apoptotic pathway in neuronal cell death by alleviation of BAX/BCL-2 ratio, and down-regulation of cleaved-PARP and cleaved caspase-9 and -3. Although further studies are required to verify the protective mechanisms of PO and ALA from neuronal damage, PO and ALA are the promising agent against oxidative stress-induced apoptotic neuronal cell death.

생리활성지방산;그 대사와 기능 (Physiologically Active Fatty Acids their Metabolism and Function)

  • Essentiality was proposed in the field of lipid by Burr and Burr in 1929. When rats were raised on the fat-free diet, their growth retarded and their skin and tails showed the characteristic deficient symptoms, which were relieved by the addition of ${\omega}6(n-6)$ polyunsaturated fatty acids as linoleic(LA) and arachidonic(AA) acids to the basal diet. LA is dehydrogenated to ${\gamma}-linolenic$ acid(GLNA) by ${\Delta}6$ desaturase, then GLNA is 2 carbon chain elongated by elongase to $dihomo-{\gamma}-linolenic$ acid(DGLNA), which is desaturated by ${\Delta}5$ desaturase to AA. These acids are called LA family or ${\omega}6(n-6)$ polyunsaturated fatty acids(PUFA). ${\alpha}-Linolenic$ acid(ALNA) is converted through the series of desaturation and elongation steps to docosahexaenic acid(DHA) via eicosapentaenoic acid(EPA). These acids belong to ALNA family or ${\omega}3(n-3)$PUFA. Human who consume large amounts of EPA and DHA, which are present in fatty fish and fish oils, have increased levels of these two fatty acids in their plasma and tissue lipids at the expense of LA and AA. Alternately, vegetarians, whose intake of LA in high, have more elevated levels of LA and AA and lower levels of EPA and DHA in plasma lipids and in cell membranes than omnivores. AA and EPA are metabolized to substances called eicosanoids. Those derived form AA are known as prostanocids(prostaglandins and prostacyclins) of the 2-types and leukotrienes of the 4-series, whereas those derived from EPA are known as prostanoids of the 3-types and leukotrienes of the 5-series. DGLNA is a precursor of the 1-types of prostaglandins. The metabolites of AA and EPA have competitive functions. Ingestion of EPA from fish or fish oil replaces AA from membrane phospholipids in practically all cells. So this leads to a more physiological state characterized by the production of proatanoids and leukotrienes that have antithrombic, antichemotactic, antivasoconstrictive and antiinflammatory properties. It is evident that ${\omega}3$ fatty acids can affect a number of chronic diseases through eicosanoids alone.

Nutritional characteristics of horsemeat in comparison with those of beef and pork

  • This study was conducted to determine the nutritional characteristics of horsemeat and bone meal in comparison with those of beef and pork presented by Dietary Reference Intakes For Koreans. Longissimus muscle and large metacarpal bone samples were collected from 20 fattened Jeju horses. Muscle samples were subjected to proximate analysis, assays for fatty acid profile and minerals, and bone samples to mineral assays. Horsemeal had similar levels of protein (21.1 vs 21.0 or 21.1%) and lower levels of fat (6.0 vs 14.1 or 16.1%) compared with beef or pork, respectively. Horsemeat had much higher levels of palmitoleic (8.2 vs 4.4 or 3.3%) and $\alpha-linolenic$ (1.4 vs 0.1 or 0.6%) acids than beef or pork, respectively. Linoleic acid was much higher in horsemeat (11.1%) and pork (10.1%) than in beef (1.6%). PUFA:SFA and n-6:n-3 ratios in horsemeat were 0.29 and 10.2, respectively. There were no big differences in mineral contents between horsemeat, beef and pork. For daily recommended mineral intakes of male adults (Dietary Reference Intakes For Koreans), phosphorus, sodium, potassium, iron, zinc and copper can be provided up to 24, 2.5, 6.7, 21, 26 and 40%, respectively, by 100 g raw horsemeat, but calcium and manganese levels are negligible. Horse cannon bone had much higher mineral contents especially in calcium (10,193 mg/100 g), phosphorus (5,874 mg/100 g) and copper (0.79 mg/100 g). Thiamin, riboflavin, niacin and retinol contents were 0.20, 0.21, 1.65 mg/100 g and $30{\mu}g/100g$, respectively. But ascorbic acid and beta-carotene were not detected. Our data demonstrated that higher levels of palmitoleic and $\alpha-linolenic$ acid in horsemeat than in beef and pork may be beneficial for human health. Horsemeat and bone meal are a good source of some minerals and vitamins.

발암원을 투여한 쥐에서 식이지방이 대장의 종양발생과 세포증식에 미치는 영향 (Effect of Different Dietary Fats on Colon Tumor Incidence and in vivo Cell Proliferation in Colonic Mucosa of MNU-Treated Rats)

  • The study was designed to observe the effect of different dietary fats on the incidence of colorectal tumor and in vivo cell proliferation in colon carcinogenesis. Male Sprague Dawley rats were intrarectally infused with chemical carcinogen(methylnitrosourea, MNU) and fed 16%(w/w) fat diet containing one of dietary fats(beef tallow, corn oil, perilla oil) for 30 weeks. To measure in vivo cell proliferation, the incorporation of 5-bromo-2-deoxyuridine(BrdU) into DNA was localized using the monoclonal anti-BrdU antibody. Large number of tumors were found in the distal colon and tumor incidence was increased in the order of perilla oil(57.7%)$\alpha$-linolenic acid rich in perilla oil could have a protective effect against colon cancer compared to saturated fatty acid or n-6 linoleic acid.

돼지 정자의 동결보존 시 α-Linolenic Acid의 효과 (Effects of α-Linolenic Acid in Frozen-thawed Boar Spermatozoa)

  • The aim of this study was to evaluate effect of ${\alpha}$-linolenic acid (ALA) on viability, acrosome reaction and mitochondrial intact in frozen-thawed boar sperm. The boar semen was collected by gloved-hand method and cryopreserved in 20% egg yolk freezing extender containing ALA (0, 3, 5, and 10 ng/mL) with 0.05% ethanol. The frozen-boar spermatozoa were thawed at $37.5^{\circ}C$ for 45 sec in water-bath. The spermatozoa samples were evaluated the plasma membrane integrity, acrosome reaction, and mitochondrial integrity using flow cytometry. In results, population of live sperm with intact plasma membrane was significantly higher in control and 3 ng/mL ALA treatment group than ethanol group (p<0.05). In contract, dying sperms were higher in ethanol group than 3 ng/mL ALA treatment (p<0.05). Acrosomal membrane damage in all sperm population was reduced in 3 ng/mL ALA groups compared with ethanol treatment (p<0.05). However, acrosome damage in live sperm population was no significant difference among the all treatment groups. Mitochondrial integrity was not influenced by ALA treatments in both of live and all sperm population. In conclusion, this results show that supplement of ALA during the cryopreservation process could reduce the membrane damages including plasma and acrosomal membrane, whereas ALA did not influence to mitochondria in boar spermatozoa. Therefore, these results suggest that ALA can protect against the membrane damage derived cryo-stress, and cryopreservation efficiency of boar semen would be improved by use of ALA.

Dimethylhydrazine을 투여한 쥐에서 식이 지방이 혈장 지질 조성과 조직의 과산화물형성 및 항산화효소 수준에 미치는 영향 (Effects of Dietary Fats on Plasma Lipids and the Level of Lipid Peroxidation and Antioxidant Enzymes in Rats Treated with Dimethylhydrazing)

  • This study was designed to compare the effect of different dietary fats on plasma lipids, the degree of lipid peroxidation and the activity of antioxidant enzymes in RBC and liver rats treated with or wighout 1, 2-dimethylhydrazing (DMH). Male Sprague Dawley rats, at 7 weeks-old, were divided into control and DMH-treated grous, and each group was again subdivided into four were perilla oil (PO), blend fat (BF) containing ten different kinds of dietary oil, beef tallow (BT), corn oil (CO). At the same time, each rat was injected intramusculary with saline(for control) or DMH twice a week for 6 weeks to give total dose of 180 mg/kg body weight. Compared with BT feeding, BF reduced plasma total choesterol level and PO and Co reduced plasma TG levels (p<0.05). DMH injection decreased plasma cholesterol in all dietary groups. However, PO decreased tocopherol levels and increased TBARS levels in RBC compared to BT. The degree of hemolysis in PO group was higher than that of BT group (p<0.05 only in control group. Fatty acid composition of hepatic microsome was reflected by dietary fatty acid profile. The peroxidizability index and TBARS level in hepatic micorsome were significantly increased but tocopherol level was lowered in PO group compared to BT group. Activites of superoxide dismutase and glutathione peroxidase in RBC and hepatic cytosol were not influenced y dietary fats and DMH treatment(p<0.05). Overall, perilla oil rich in $\omega$3 $\alpha$-linolenic acid could be a very important dietary source in reducing plasma lipids and blend fat was also good dietary oil mixture in reducing plasma cholesterol. However, the degree of lipid peroxidation was greater in tissue by perilla oil feeding and it is very difficult to use only perilla oil as oil source for meal preparation, so that it could be suggested to use more perilla oil and fish to give an equal effect of blend fat in order to reduce the risk factors against cardiovascular disease.

Comparative analysis of Bombyx batryticatus and Bombyx mori on α-glucosidase inhibition and their bioactive compositions

  • This study aimed to evaluate and compare the inhibitory effects of extracts from Bombyx batryticatus (BBE) and Bombyx mori (BME) on α-glucosidase, DPP-4, and LDL oxidation activities, focusing on their potential applications in managing postprandial hyperglycemia and metabolic syndrome. The results demonstrated that both BBE and BME effectively inhibited α-glucosidase and LDL oxidation, with BBE exhibiting higher inhibitory activity than BME. HPLC analysis identified linolenic acid, linoleic acid, linolenic acid ethyl ester, pheophorbide a, and pyropheophorbide a as key compounds contributing to these effects. Notably, the identified unsaturated fatty acids and pheophorbides showed strong α-glucosidase inhibitory activity, surpassing that of acarbose, a standard diabetic drug. These results suggest that, in addition to the previously reported 1-DNJ and fibroin proteins, unsaturated fatty acids and chlorophyll-derived pheophorbides may play significant roles in glycemic control. Compounds, particularly those from BBE, present promising opportunities for the development of natural therapeutic agents for diabetes management. The study concludes that BBE and BME have strong potential as functional ingredients in future diabetes treatment strategies, possibly offering enhanced efficacy over conventional inhibitors.

SMP종자 전처리에 의한 담배 종자 발아과정의 지방산 변화 (Changes of Fatty Acid during Germination by Seed Pretreatment, SMP, in Tobacco)

  • 본 연구에서는 발아력향상을 위해 담배품종 Nicotiana tabacum cv. KF109와 cv. KB108 종자에 종자전처리방법 중 하나인 solid matrix priming (SMP)처리를 하여 전처리효과를 알아보고 전처리과정 중 담배종자 발아시 에너지원으로 사용되는 지방산의 함량 변화를 조사한 결과 아래와 같다. 1. 발아시험결과 전처리에 의한 처리간 발아율 차이는 보이지 않았으나, 발아세, T50은 KF109의 경우 9일처리, KB108의 경우 7일처리에서 가장 높았다. 2. 종자내 지방산을 분석한 결과 처리기간동안 함량의 변화가 큰 지방산의 종류는 palmitic acid, stearic acid, oleic acid+elaidic acid, linoleic acid+linolelaidic acid, $\alpha-linolenic$ acid였다. 3. 지방산 종류에 따라 다소 차이가 있으나 각 지방산의 함량은 KF109의 경우 8일 또는 9일에서, KB108의 경우 6일 또는 7일까지 증가하였가 이후 감소하였다. 4. 특히 지방산 중 oleic acid+elaidic acid와 linoleic acid+linolelaidic acid 함량의 변화가 가장 컸으며 다른 지방산에 비하여 함량도 높았는데 KF109와 KB108품종의 경우 8일과 6일처리까지 각각 증가하여 최고에 이른 후 급격히 감소하는 결과를 보여 종자전처리 진행과정을 확인하는 데에 이용 가능할 것으로 판단된다.

특이지방산의 탐색과 붓꽃류에서 중쇄지방산(myristic acid)의 확인 (The Composition of Useful Medium Chain Fatty Acids in Eight Plant Species)

  • 산업적으로 활용성이 높은 특이 지방산을 탐색하기 위하여 8종의 국내 자원식물을 재료로 하여 지방산 조성을 조사한 결과 붓꽃(Iris tectorum) 등 3가지 식물에서 myristic acid(14 : 0)와 lauric acid(12 : 0)등의 중쇄지방산이 함유되어 있음을 확인하였다. 특히 붓꽃에는 myristic acid가 전체 지방산의 75.9%를 차지하였다. 독일붓꽃(Iris germanica)에는 myristic acid와 lauric acid가 전체 지방산의 57%와 15.5% 씩 함유되어 있었다. 꽃창포(Iris ensata) 뿌리에는 lauric acid와 capric acid(10 : 0)가 각각 10.7%와 9.5% 함유되어있었으나 회향(Foeniculum vulgare)과 사상자(Torilis Japonica)의 종실에는 전체 지방산(mg/g dry wt)이 각각 193.3 mg/g과 128.2 mg/g의 높은 함량을 나타내었다. 붓꽃의 부위별 지방산 조성에서는 종실에 linoleic acid(18 : 2)가 64.5%, 잎에 ${\alpha}-linolenic$ acid(18 : 3)가 48.9% 그리고 곁뿌리(lateral root)에는 caprylic acid(8 : 0)와 capric acid가 각각 9.5%와 8.6%씩 함유되어 있었으며 붓꽃과 독일붓꽃의 미숙종자에는 myristic acid가 각각 10.8%와 15.6% 함유되었다.
