People with obesity maintain low levels of inflammation; therefore, their exposure to foreign antigens can trigger an excessive immune response. In people with obesity or allergic contact dermatitis (ACD), symptoms are exacerbated by a reduction in the number of regulatory T cells (Tregs) and IL-10/TGF-β-modified macrophages (M2 macrophages) at the inflammatory site. Benefits of intermittent fasting (IF) have been demonstrated for many diseases; however, the immune responses regulated by macrophages and CD4+T cells in obese ACD animal models are poorly understood. Therefore, we investigated whether IF suppresses inflammatory responses and upregulates the generation of Tregs and M2 macrophages in experimental ACD animal models of obese mice. The IF regimen relieved various ACD symptoms in inflamed and adipose tissues. We showed that the IF regimen upregulates Treg generation in a TGF-β-dependent manner and induces CD4+T cell hypo-responsiveness. IF-M2 macrophages, which strongly express TGF-β and inhibit CD4+T cell proliferation, directly regulated Treg differentiation from CD4+T cells. These results indicate that the IF regimen enhances the TGF-β-producing ability of M2 macrophages and that the development of Tregs keeps mice healthy against ACD exacerbated by obesity. Therefore, the IF regimen may ameliorate inflammatory immune disorders caused by obesity.
In recent years, the safety of sunscreens has been challenged based on the reports of their adverse effect on users; dermatitis, allergic contact dermatitis and photo allergic contact dermatitis. The unscientific idea that higher SPF sunsreen is good for health mealeads many users to tend to prefer higher SPF sunscreen. In the toxicological aspect, the need to investigate the safety of sunscreens is steadily increasing. However, there were few studies on the correlation between sun protection factor (SPF) and the safety of sunscreens. The objective of this study was to assess whether there was a correlation between SPF and the safety of sunscreens. We measured in vitro SPF index using homosalate as a standard and examined the toxicity tests such as primary skin irritation tests, ocular irritation test and skin sensitization test. Homosalate (HS), octyl methoxycinnamate (OMC), octyl salicylate (OS), octocrylene (OC) as UVB organic filter and benzophenone-3 (BP3), butyl methoxy dibenzoil methane (BMDM) as UVA organic filter, and titanium dioxide (TD), zinc oxide (ZO) as inorganic filters were used. The skin irritation indexes in rabbits treated with HS, OMC, OS, BP3, and BMDM were significantly increased as SPF indices were increased. Neither ocular irritation in rabbits nor skin sensitization in guinea pigs were increased. It suggests that there might be a good correlation between SPF and the skin irritation indices of organic UV filters and skin irritation might be one of most sensitive index to assess the safety of sunscreens.
Allergic contact dermatitis (ACD) is an inflammatory skin disease and regarded as a prototype of T-cell mediated delayed-type hypersensitivity reactions. Aroma Oil Complex (AOC) is composed of lavender true oil, chamomile roman oil and tea tree oil. This study was performed to assess the effects of AOC in a canine model of ACD. ACD was induced on the back of dogs induced by sensitization and repeated application by 2,4-dinitro-1-chlorobenzene (DNCB). Topical treatment of AOC was applied once a day for 8 days and skin biophysical parameters including transepidermal water loss (TEWL), skin hydration, skin thickness and erythema index, were measured every two days during experimental periods. Histopathology and immunohistochemistry were performed to evaluate the anti-inflammatory effect. In skin biophysical parameters, TEWL, skin hydration, skin thickness and erythema index were significantly increased, with a maximum increase appeared on day 2 (p<0.05). After the completion of AOC treatment, skin biophysical parameters were significantly reached those of baseline in a time-dependent manner (p<0.05). In histopathology, marked increases of epidermal thicknesses were induced after DNCB challenge with numerous inflammatory cell infiltrations and edematous changes, decreases of connective tissue occupied regions in dermis. In addition, marked increases of cytokine - tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) and interferon-${\gamma}$ (IFN-${\gamma}$)-immunoreactivities in the dermis and of apoptotic markers - caspase-3 and PARP-immunoreactivities in the epidermis were observed in DNCB control as compared with intact control, respectively (p<0.01). The decrease of infiltrated inflammatory cells and related decreases of pro-inflammatory cytokine immunoreactivities were observed in AOC treated skin (p<0.01). Based on these findings, AOC may have anti-inflammatory and alleviatory effects in the allergic contact dermatitis.

In this study, we evaluated whether Lunasin Enriched Soybean (LES) suppressed 1-chloro-2,4-dinitrochlorobenzene (DNCB) induced allergic contact dermatitis in vivo. In the group treated with DNCB alone, erythema and hemorrhage in the skin were observed, while the application of LES after DNCB treatment modulated erythema and hemorrhage similar to the untreated group. In histopathological analysis of the skin, DNCB-treated groups showed increased skin thickness compared to the untreated groups, while the treatment of LES dose-dependently modulated skin thickness. Also, hypertrophy and hyperkeratosis of the epidermis, intracellular edema and infiltration of inflammatory cells were observed in the group treated with DNCB alone, while treatment of LES inhibited these inflammatory changes. DNCB also induced a significant up-regulation of iNOS and COX-2 expression, as compared to the untreated groups. However, the treatment of LES significantly decreased the down-regulation of iNOS and COX-2 expression, as compared to DNCB-treated groups. Also, LES reduced the IgE level in DNCB-stimulated groups. And lunasin from LES translocated the nucleus of the cells in the skin of Sprague-Dawley rats. Taken together, it is thought that LES has the potential to be used as an alternative treatment for allergic contact dermatitis.

NC/Nga mice with contact dermatitis induced using DNCB were applied with GJHB(祛風燥濕解毒方), and its influence on the expressions of IgE, immune related cytokines, and immunoglobulins were tested. The results were obtained as follows ; GJHB significantly redued the production of IgE compared to the control. GJHB significantly reduced the amount of IL-4 within the suspension of spleen cells, but significantly induced the production of $IFN-{\gamma}$, compared to the control. GJHB significantly reduced the production of IL-6 and $TNF-{\alpha}$ in the serum compared to the control. GJHB significantly suppressed the concentration of IgM, IgG2a, IgG2b in the serum. However, no significant decrease of IgG1 was observed. From the results above, GJHB suppressed the intermediate factors during the hypersensitive immune response as well as suppressing the generation of inflammation related cytokines. Therefore, anti-allergic effect of GJHB on the contact dermatitis through immune modulation was confirmed.
