Production of Biobutanol by Clostridium beijerinckii from Water Hyacinth (부레옥잠을 이용한 Clostridium beijerinckii의 Biobutanol 생산)

  • Biofuel has been considered as promising renewable energy to solve various problems that result from increasing usage of fossil fuels since the early 20th century. In terms of chemical and physical properties as fuel, biobutanol has more merits than bioethanol. It could replace gasoline for transportation and industrial demand is increasing significantly. Production of butanol can be achieved by chemical synthesis or by microbial fermentation. The water hyacinth, an aquatic macrophyte, originated from tropical South America but is currently distributed all over the world. Water hyacinth has excellent water purification capacity and it can be utilized as animal feed, organic fertilizer, and biomass feedstock. However, it can cause problems in the rivers and lakes due to its rapid growth and dense mats formation. In this study, the potential of water hyacinth was evaluated as a lignocellulosic biomass feedstock in biobutanol fermentation by using Clostridium beijerinckii. Water hyacinth was converted to water hyacinth hydrolysate medium through pretreatment and saccharification. It was found that productivity of water hyacinth hydrolysate medium on biobutanol production was comparable to general medium.

  • In this study, alginate-based hydrogel membranes composed of hydrogel beads and highly tough hydrogel matrix including moisturizing oil and natural emulsifier were prepared and their elution characteristics were evaluated. As a result, it was confirmed that the elution rate of the moisturizing oil component can be controlled within a desired range by controlling the composition of the hydrogel bead and the tough hydrogel matrix. In particular, it has been confirmed that by combining tough hydrogel having a structure of interpenetrating polymer network (IPN) and hydrogel beads, the physical stability of the membranes can be improved and the elution rate of the moisturizing oil can also be controlled more finely.

Studies on the Various Utilization of Microbial Formulation for the Production of Vegetable Crops (원예작물(園藝作物) 생산성(生産性)에 미치는 미생물(微生物) 제제(製劑)의 복합적(複合的) 이용연구(利用硏究))

  • The carrier materials used for the development of bacterial inoculants to be effective in field were made with various carrier materials of two major forms, alginate bead and powder inoculants. Inoculants were prepared after mixing those carrier materials with Pseudomonas fluorescens SSL3 and Bacillus subtilis B5, and the treatment effects of each inoculants was investigated on cucumber, tomato, pepper and potato. Survival density of SSL3 and B5 in various carrier materials for duration of storage and the bead inoculants were better than the powder. In the powders, survival rate increased in carrier materials treated 5% skimilk. The growth condition of microorganisms in carrier materials is good at powder. When they were preserved in the long period, contamination is problem. Scanning(200 to 600nm) of the P. fluorescens SSL3 supernatant in centrifuged MKB broth incubated for 48h had two main peaks, pyochelin(300nm) and pyoverdin(400nm). The potato yield in field experiments of spring, treated with bead formulas showed increase of 22~29% in whole potato breeds as compared with control, because the bead formulas degraded, and released the antibiotic microorganisms in slow and constant rate. In the pot experiment, there were significant difference in soil, wheatbran, and bead formed wheatbran.

Stabilization of Lactobacillus with Double Matrix Capsulation (더블매트릭스 캡슐화에 의한 유산균의 안정화)

  • This study using yogurt of food in order to increase the conservativeness and stability of lactobacillus, to be deliciously flavored and give it visual differential effect, were described regarding making the preparing method of double matrix capsulation in food industries. Our study group was especially made to soft and moisture cream using 5wt% of sucrose ester emulsifier as first capsulation. Double matrix capsulation was formed with the best stabilized bead type capsules when it blended 1:3 ratio of chitosan and alginate. The bead diameter size was about 2.5~4mm (mean diameter: 3mm). Activity of lactobacillus containing cream for depending on various pH variations showed that alkalinity ($pH=10.3{\pm}0.3$) condition was higher than acidity ($pH=4.3{\pm}0.3$) and neutrality ($pH=7.12{\pm}0.2$) conditions. After a month, it also was certified to the activity of lactobacillus in incubated at $37^{\circ}C$ in culture medium. As application of food industry, we developed the containing lactobacillus capsule and 5 colored kinds of double matrix capsulation in yogurt cream. As for above mentioned those results, one of tool to stabilize the living lactobacillus, doubled matrix capsulation greatly be expected to contribute to food industry. Furthermore, it can be expected to apply the drug delivery system (DDS) to active ingredients of stabilizing technologies at drug and cosmetic industries.

The Removal of Heavy Metals from CCA Treated Wood by Aspergillus Niger-alginate Bead (CCA처리목재의 고정화 Aspergillus niger에 의한 중금속 제거)

  • One way of using wood safely and soundly for a long time is use of preservative-treated wood. Although, preservative-treated wood present to us durability and safety during using periods, it is also important to safe disposal after its service periods because of toxic components. Biological methods could be applicable to its disposal methods and better safe than chemical methods in the aspect of environmental problems. This study applied biological methods to remove the heavy metals from end-used CCA treated wood. The Aspergillus niger was used for this study which make black stain on the wood surface by it's spore. This study investigated the growing of A. niger on the CCA treated wood chips and the removal efficiency of heavy metals from CCA treated wood chips using immobilizing A. niger. A. niger could growing on the CCA treated wood chips and A. niger remove the effective elements of CCA treated wood effectively.

Removal of Inorganic Arsenic from Steamed Hijiki Sargassum fusiforme Concentrate Using Activated Carbon (활성탄을 이용한 톳(Sargassum fusiforme) 자숙농축액 내 무기비소 저감 기술)

  • The edible sargasso seaweed hijiki Sargassum fusiforme is known to have high concentration of arsenic, which is a threat to human health, particularly due to inorganic arsenic. In this study, various methods were used to remove inorganic arsenic from steamed hijiki concentrate. The highest concentration of arsenate [As(V)] in both raw and processed hijiki during steamed hijiki manufacturing process was within the range of 8.213-14.356 mg/kg, and it is a potential source of inorganic arsenic, which can result in re-contamination and cause environmental pollution. The removal efficiencies of the various removal methods were within the range of 57.3-83.4%, and 19.0% reduction was achieved using activated carbon and alginate bead. Further, activated carbon showed the best adsorption effect of inorganic arsenic. Therefore, we suggest that activated carbon is a suitable efficient method for removing inorganic arsenic and has low operational costs in field applicability.

Optimization of γ-Aminobutyric Acid (GABA) Production Using Immobilized Lactobacillus plantarum K154 in Submerged Culture of Ceriporia lacerata (Ceriporia lacerata 배양액과 고정화 Lactobacillus plantarum K154를 이용한 감마아미노뷰티르산 생산 최적화)

  • The production of GABA was optimized by co-cultivation of immobilized Lactobacillus plantarum K154 (ILK) with Ceriporia lacerata cultures. The mycelial culture of C. lacerata was performed in a defined medium containing 3% glucose, 3% soybean flour, and 0.15% $MgSO_4$ in a submerged condition for 7 days at $25^{\circ}C$, resulting in the production of 29.7 g/L mycelia, 3.1 g/L exopolysaccharides, 2% (w/w) ${\beta}$-glucan, 68.96 unit/mL protease, and 10.37 unit/mL ${\alpha}$-amylase. ILK in C. lacerata culture showed viable cell counts of $3.13{\time}10^9CFU/mL$ for immobilized cells and $1.48{\time}10^8CFU/mL$ for free cells after 1 day. GABA production in the free and immobilized cells was 9.96 mg/mL and 6.30 mg/mL, respectively, after 7 days. A recycling test of ILK in the co-fermentation was consequently performed five times at $30^{\circ}C$ for 15 days, resulting in the highest production of GABA. GABA could also be efficiently overproduced by co-cultivation with the produced polysaccharides, ${\beta}$-glucan, peptides, and probiotics.
