Comparison of Filtration Efficiency of Membranes for Harvesting Microalgae using a Gravity-Filtration Device (중력 여과장치 이용 미세조류 수확을 위한 여과막의 효율성 비교)

  • Cost-effective microalgae harvesting methods are necessary for economical production of algal biodiesel. In this study, membranes with various pore sizes and materials were examined for their potentials in application to gravity-filtration of Tetraselmis sp. KCTC12432BP. For this test, 10 L of Tetraselmis sp. culture (2 g/L) was loaded on each membrane and filtration rates were measured. Among the tested materials, a woven cotton fabric showed the fastest water drain rate (0.73 L/hr) without serious cell leakage. Cell density of the concentrates after filtration was 6.8 g/L, indicating 3.4-fold concentration compared with the initial algal culture. The result suggests that the woven cotton fabric could serve as filtration membrane for harvesting Tetraselmis sp. among the tested ones.

Dietary Contributions of Phytoplankton and Zooplankton to Young Silver Carps (어린 백련어의 성장에 대한 동, 식물플랑크톤의 먹이기여도)

  • Two-monthold silver carps were cultured with seven algal species and zooplankton (Moina macrocopa) in the laboratory. The carps were reared in 10 L translucent cylindrical aquaria with algae and zooplankton for 8 days. The Moina effectively fed almost cultured algae: perfectly removed Cryptomonas (NIES-282) within 60 min. Both algal diets Cryptomonas and Fragilaria (NIES-391) significantly increased the Moina population ($r^2$>0.93, p<0.005), while Microcystis (MIES-90) and Oscillatoria (NIES-204) reduced the zooplankton ($r^2$=0.97, p<001). Fish removed about 50% of all algae for 52 hrs, even 60% of Microcystis still remained: all algae reduced ca. 5${\sim}$12% of initial weight. Furthermore, a continuous supply of algae with same density resulted in the death of fish, e.g. 11 days in cyanobacteria Microcystis. Therefore, the growth limitation of silver carp by algae indicates that zooplankton is of direct dietary contributor in planktivores feeding behavior.

  • 유해 남조 대발생(Harmful Algal blooms, HABs)이 담수호에 발생하면 마이크로시스틴과 같은 독성물질과 맛·냄새 물질을 생성하여 상수원이용과 친수활동을 방해한다. 그래서 유해 남조 대발생 전 유해남조 세포수를 예측하여 선제적 대응하는 것은 중요하다. 따라서 본 연구는 머신러닝기반 Random Forest(RF)를 활용하여 팔당댐 앞의 유해남조 세포수를 예측하는 모델을 개발하고 성능을 평가하고자 한다. 모델 구축을 위해 2012년 4월부터 2021년 12월까지의 팔당호(삼봉리, 경안천) 및 남북한강(의암댐~이포보)권역의 조류, 수질, 수리/수문, 기상 자료를 수집하여 입력 및 출력 자료로 이용하였다. 수집된 데이터에는 다양한 입력변수들이 있어 남조 세포수 예측 성능 비교를 위한 전체 26개 변수 적용과 통계학적으로 상관관계가 높은 12개 변수 적용을 통해 모델을 구축하였다. 입력, 출력 자료로 이용한 유해남조 세포수는 로그변환된 값으로 사용하였으며 일반적인 조류 시료 채취기간이 7일이므로 7일 후를 예측하기 위한 모델을 구축하였다. 구축한 모델의 성능은 실측데이터와 예측데이터의 R2로 산출하여 평가하였다. 전체 26개 입력변수로 모델 구축 후 학습 및 검증 수행 결과 R2의 학습 0.803, 검증 0.729로 나타났고, 유해남조 세포수와 유의미한 상관관계를 보이는 12개 입력변수로 모델 구축 후 학습 및 검증 수행 R2은 학습 0.784, 검증 0.731로 나타났다. 두 모델의 성능을 살펴본 결과 입력변수 개수의 변화에 따른 성능차이는 크지 않은 것으로 나타났으며, 남조세포수 예측을 위한 모델로서 활용가능함을 알 수 있었다. 향후 연구에서는 Random Forest 외 다른 기계학습 모델들과 딥러닝 모델을 통해 남조세포수 예측 성능이 높은 모델을 구축해볼 필요성이 있다.

  • Dissolved air flotation (DAF) is an effective solid/liquid separation process for low density particles, such as algal flocs, humus materials and clay particles produced from low turbidity water. The fraction of humic substances for natural organic matters (NOMs) are considered problematic in water because it can readily react with chlorine to form harmful by-products (trihalomethanes) and can be exposed to undesirable color, tastes and odors in drinking water. A broad class of NOMs includes fulvic acid, humic acid and humin. This paper will discuss the results from a study that performed with a DAF pump process using synthetic wastewater contained humic substance. Batch jar tests were performed to evaluate coagulant dose and recycle ratio on flotation efficiency.

  • Algal growth studies of Chlorella strains were conducted in a batch mode with bench type experiments. Carbon dioxide fixation rates of the following green microalgae were determined: Chlorella sp. H84, Chlorella sp. A2, Chlorella sorokiniana UTEX 1230, Chlorella vulgaris, and Chlorella pyrenoidosa. C. vulgaris, among other strains of microalgae, showed the highest growth rate (1.17 optical density/5 days). Cultivating conditions for C. vulgaris that produced the highest growth rate were at concentrations of 243 ${\mu}g\;CO_2$/mL, 10 mM ammonia, and 1 mM phosphate, with an initial pH range of 7-8.

Ecological Characteristics of Some Algal Populations along Environmental Gradients of Zinc (아연환경구배에 의한 녹조류개체군생장의 생태학적 특성)

  • The maximum relative growth rate of algae treated with Zinc was shown as follows: 15, 8, 6, 3 and -5% per day for the rather sensitive Chlorella sp. populations, or 14, 7, 5 and 4% per day for the Pleurococcus sp. populations, and 22, 20, 13, 9 and 7% per day for the more resistant Scenedesmus spinosus populations, respectively for the culture medium with 0, 1, 5, 10 and 20 ppm of Zinc treatment. With mixed cultures of Chlorella sp. and Scenedesmus spinosus populations, the growth of the Chlorella sp. population overcame that of the S. spinomsus population from the cultures treated with relatively low concentration of Zinc. On the contrary, the population growth of the latter resistant species overcame that of the former sensitive species when the concentration of Zinc was above 5 ppm Zn of the medium. This paper describes the results of further investigations of the effects evaluated by direct cell counts method, optical density comparisons, oxygen production and consumption determinations and the measurements of the fate of Zinc treated in the solutions.

Chemical Properties of Sea Tangle (Saccharina. japonica) Cultured in the Different Depths of Seawater (양식 다시마의 수심과 부위에 따른 화학적 성분 특성)

  • To develop the technology for cultivation of high-density seaweed, sea tangle was cultured from varying depths(0.5 m, 3 m) of seawater at Gijang and Wando area. Proximate composition, component sugar, total amino acid, fatty acid composition, and element composition of different parts of sea tangle (Saccharina japonica) have been examined. Significant differences were found in the amount of crude protein and ash content in lower, middle, and upper parts of algal blades. The upper parts of the sea tangle was rich in crude protein, while lower parts was rich in crude ash. Crude lipid content was higher in the middle parts than those of the other parts. The component sugars were not significantly different from all parts of algal blades. The highest content of most of the amino acids were found in the upper parts of the blades. The amount of saturated fatty acids concentrated mostly in lower parts of blades, while the content of polyunsaturated fatty acids concentrated in the upper parts. The highest N element contents were found in upper part of algal blades. However, the contents of those chemical component were not affected by the depths of seawater.

Semi-continuous cultivation of the mixotrophic dinoflagellate Gymnodinium smaydae, a new promising microalga for omega-3 production

  • Omega-3 fatty acids, including eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA), are polyunsaturated fatty acids beneficial to human health. A limited number of microalgae have been used for commercial omega-3 production, which necessitates the identification of new microalgae with high omega-3 contents. We explored the fatty acid composition and EPA and DHA contents of the mixotrophic dinoflagellate Gymnodinium smaydae fed with the optimal algal prey species Heterocapsa rotundata. Cells of G. smaydae were found to be rich in omega-3 fatty acids. In particular, the DHA content of G. smaydae was 21 mg g-1 dry weight, accounting for 43% of the total fatty acid content. The percentage of DHA in the total fatty acid content of G. smaydae was the highest among the reported microalgae except for Crypthecodinium cohnii. Moreover, to determine if the prey supply interval affected the growth rate of G. smaydae and its fatty acid content, three different prey supply intervals (daily, once every 2 d, and once for 4 d) were tested. Daily prey supply yielded the highest total fatty acid and DHA contents in G. smaydae. Furthermore, we successfully produced high-density G. smaydae cultures semi-continuously for 43 d with daily prey supply. During the semi-continuous cultivation period, the highest density of G. smaydae was 57,000 cells mL-1, with an average growth rate of 0.7 d-1. Taken together, the percentage of EPA and DHA in the total fatty acid content was maintained in the range of 54.2-56.9%. The results of this study support G. smaydae as a promising microalgal candidate for commercial DHA production and demonstrate that daily supply of prey can efficiently produce high-density G. smaydae cultures for more than a month.

Antialgal Interactions of Biological Control Agents on Cyanobacterium and Diatom Blooms in vitro (유해조류 제어를 위한 두 가지 이상의 생물제재 및 효과)

  • Antialgal bacteria and ciliates were tested alone and in combination for their abilities to decrease the densities of the warm-weather cyanobacterium, Microcystis aeruginosa, and the cold-weather centric diatom, Stephanodiscus hantzschii. Our results indicate that the density of M. aeruginosa was effectively suppressed by the bacterium, Streptomyces neyagawensis, and the heterotrich ciliate, Stentor roeselii. However, co-treatment with both bio-agents stimulated the algal density rather than decreasing it, suggesting that S. neyagawensis and S. roeselii may have an antagonistic relationship. Additional experiments revealed that the density of S. hantzschii was effectively suppressed by the bacterium, Pseudomonas putida, and by the above mentioned strain of S. roeselii. Co-treatment with both bio-agents had a higher antialgal activity than treatment with each alone, indicating that the bio-agents may act synergistically. These results suggest that the anti-alge efficacy of co-treatment with multiple biological control agents is likely to differ depending on the bio-agents and target organisms.

Green algae dominance quickly switches to cyanobacteria dominance after nutrient enrichment in greenhouse with high temperature

  • In order to understand the mechanisms of conversion between different algal dominance, an experiment was performed in a greenhouse from 22 June to 10 July 2011. The experiment included a treatment group subjected to three instances of nutrient enrichment and a control with no nutrient enrichment. The initial water was dominated by Ankistrodesmus of Chlorophyta. The average water temperature at 08:30 h and 14:00 h during the experiment was $31.6^{\circ}C$ and $34.6^{\circ}C$, respectively. The results showed that the total nitrogen (TN), total phosphorus (TP), dissolved total nitrogen (DTN), dissolved total phosphorus (DTP), and soluble reactive phosphorus (SRP) concentrations in the treatment were significantly higher than in the control (P < 0.05). However, the TN/TP and DTN/DTP in the control was higher than in the treatment (P < 0.05). The dominant algae in the control did not change during the experiment, while the dominant algae in the treatment switched to Planktothrix of Cyanophyta on day 9. The chlorophyll a (Chl-a), wet weight of all algae, wet weight of Cyanophyta, and percentage of Cyanophyta in the control were all significantly lower than in the treatment (P < 0.05). Amounts of zooplankton, especially rotifers, were present at the end of the experimental period. The density of rotifers between the control and treatment was not significantly different (P > 0.05), while the copepod density in the treatment was higher than in the control (P < 0.05). We conclude that green algae dominance quickly switches to cyanobacteria dominance after nutrient enrichment in a greenhouse with elevated temperature.
