Mastoparan B (MPB), an antimicrobial cationic peptide isolated from the venom of the hornet Vespa basalis, is a basic amphipathic α-helical peptide composed of fourteen amino acid residues. In this study, we have investigated the algicidal effect of MPB against harmful algae blooms (HABs) casative Alexandrium tamarense, Chattonella marina, Cochlodinium polykrikoides and Gymnodinium catenatum. The algicidal effect of MPB showed in the concentration of 31.3 $\mu{g}$/mL to 500 $\mu{g}$/mL against 4 HAB species and observed cell lysis or cell ecdysis by microscopy. MPB reacted more sensitive to C. marina and C. polykrikoides than A. tamarense and G. catenatum. The algicidal study of MPB against HABs will provides much insight into development of new algicidal substances.
The monitoring activities at the National Fisheries Research and Development Institute (NFRDI) in Korea have been extended to include all the coastal waters of Korea after the outbreak of Cochlodinium polykrikoides blooms in 1995. We used several alternative methods including climatological analysis, spectral and optical methods which may offer potential detection of the major species of red tide in Korean waters. In the climatological analysis, NOAA, SeaWiFS, OCM satellite data was chosen using the known C. polykrikoides red tide bloom data and the area was mapped by helicopter reconnaissance and ground observation. The relationship between the distribution of sea surface temperature to C. polykrikoides bloom areas was studied. The anomalies of SeaWiFS chlorophyll a imageries against the imageries of non-occurring red tide for August, 2001 showed where the C. polykrikoides occurred. The anomalies of chlorophyll a concentrations from the satellite data during red tide outbreaks showed a similar distribution of C. polykrikoides in the red tide in August, 2001. The distribution between differences in sea surface temperatures during the day and at night also showed a possibility for red tide detection. We used a corrected vegetation index (CVI) to detect floating vegetation and submerged vegetation containing algal blooms. The results of from the optical absorption of C. polykrikoides in the ultraviolet band (340 nm) showed that if we use the optical characteristics from each red tide, we will be able to establish the feasibility of red tide detection.
Cochlodinium polykrikoides is a red-tide forming dinoflagellate that causes significant worldwide impacts on aquaculture industries and the marine ecosystem. There have been extensive studies on managing and preventing C. polykrikoides blooms, but it has been difficult to identify an effective method to control the bloom development. There is also limited genome information on the molecular mechanisms involved in its various ecophysiology and metabolism processes. Thus, comprehensive genome information is required to better understand harmful algal blooms caused by C. polykrikoides. We estimated the C. polykrikoides genome size using flow cytometry, with detection of the fluorescence of DNA stained with propidium iodide (PI). The nuclear genome size of C. polykrikoides was 100.97 Gb, as calculated by comparing its mean fluorescence intensity (MFI) to the MFI of Mus musculus, which is 2.8 Gb. The exceptionally large genome size of C. polykrikoides might indicate its complex physiological and metabolic characteristics. Our optimized protocol for estimating the nuclear genome size of a dinoflagellate using flow cytometry with PI can be applied in studies of other marine organisms.

Apidaecin Ⅰb, an antimicrobial peptide isolated from lymph fluid of the honeybee (Apis mellifera), is a basic non-helical peptide composed of eighteen amino acid residues. In this study, we have investigated the algicidal effect of Apidaecin Ⅰb against harmful algae blooms (HABs) causative Alexandrium tamarense, Chattonella marina, Cochlodinium polykrikoides and Gymnodinium catenatum. The algicidal effect of Apidaecin Ⅰb showed in the concentration of 12.5 $\mu{g}/mL$ to 50$\mu{g}/mL$ against 4 HAB species and observed cell lysis or cell ecdysis by microscopy. Apidaecin Ⅰb reacted more sensitive to C. marina than A. tamarense, C. polykrikoides and G. catenatum. The algicidal study of Apidaecin Ⅰb against HABs will provides much insight into development of new algicidal substances.
Isolation and identification of alga-lytic bacteria were carried out. Fifteen isolates of alga-lytic bacteria were screened by the double layer method using A. cylindrica NIES-19 as a sole nutrient and four isolates among them were compared with their alga-lytic activity. The isolate AK-05 exhibiting the highest alga-lytic activity was identified as Acidovorax temperans base on its 16S rDNA sequence. The culture supernatant of the isolate AK-05 was reliable for the alga-lytic. Alga-lytic activity assays of culture supernatant revealed that the major substances for alga-lytic activity were non-proteins and heat stable. The highest alga-Iytic activity was practical under alkaline conditions and at 25${\sim}$$30^{\circ}C$. It is indicating an advantage for the application of water blooms by cyanobacteria in eutrophic lakes where the pH is generally in alkaline region.
Cyanobacteria (blue-green algae) are not only the first oxygenic organisms on earth but also the foremost primary producers in aquatic environment. Massive growth of cyanobacteria, in eutrophic waters, usually changes the water colour to green and is called as algal (cyanobacterial) bloom or green tide. Cyanobacterial blooms are a result of high levels of primary production by certain species such as Microcystis sp., Anabaena sp., Oscillatoria sp., Aphanizomenon sp. and Phormidium sp. These cyanobacterial species can produce hepatotoxins or neurotoxins as well as malodorous compounds like geosmin and 2-methylisoborneol (MIB). In order to solve the nationwide problem of hazardous cyanobacterial blooms in Korea, the following technically and strategically sound approaches need to be developed. 1) As a long-term strategy, reduction of the nutrients such as phosphorus and nitrogen in our water bodies to below permitted levels. 2) As a short term strategy, field application of combination of already established bloom remediation techniques. 3) Development of emerging convergence technologies based on information and communication technology (ICT), environmental technology (ET) and biotechnology (BT). 4) Finally, strengthening education and creating awareness among students, public and industry for effective reduction of pollution discharge. Considering their ecological roles, a complete elimination of cyanobacteria is not desirable. Hence a holistic approach mentioned above in combination to addressing the issue from a social perspective with cooperation from public, government, industry, academic and research institutions is more pragmatic and desirable management strategy.
Korea has experienced 10 a Cochlodinium polykrikoides red tide outbreaks during the last 10 years (1993-2002). In climatological analysis, NOAA, SeaWiFS, OCM satellite data in 20th and 26th August 2001 were chosen using the known C. polykrikoides red tide bloom area mapped by helicopter reconnaissance and ground observation. The 26th August, 2001 SeaWiFS chlorophyll a anomaly imageries against the imageries of non-occurring red tide for August 20, 2001 showed the areas C. polykrikoides occurred. The anomalies of chlorophyll a concentration from satellite data between before and after red tide outbreaks showed the similar distribution of C. polykrikoides red tide in 26th August, 2001. The distribution of the difference in SST between daytime and nighttime also showed the possibility of red tide detection. We used corrected vegetation index (CVI) to detect floating vegetation and submerged vegetation containing algal blooms. The simple result of optical absorption from C. polykrikoides showed that if we use the optical characteristics of each red tide we will be able to get the feasibility of the red tide detection.

Harmful cyanobacterial blooms (HCBs) are caused by the rapid proliferation of cyanobacteria and are believed to be exacerbated by climate change. However, the extent to which HCBs will be stimulated in the future due to increased temperature remains uncertain. This study aims to predict the future occurrence of cyanobacteria in the Nakdong River, which has the highest incidence of HCBs in South Korea, based on temperature rise scenarios. Representative Concentration Pathways (RCPs) were used as the basis for these scenarios. Data-driven model simulations were conducted, and out of the four machine learning techniques tested (multiple linear regression, support vector regressor, decision tree, and random forest), the random forest model was selected for its relatively high prediction accuracy. The random forest model was used to predict the occurrence of cyanobacteria. The results of boxplot and time-series analyses showed that under the worst-case scenario (RCP8.5 (2100)), where temperature increases significantly, cyanobacterial abundance across all study areas was greatly stimulated. The study also found that the frequencies of HCB occurrences exceeding certain thresholds (100,000 and 1,000,000 cells/mL) increased under both the best-case scenario (RCP2.6 (2050)) and worst-case scenario (RCP8.5 (2100)). These findings suggest that the frequency of HCB occurrences surpassing a certain threshold level can serve as a useful diagnostic indicator of vulnerability to temperature increases caused by climate change. Additionally, this study highlights that water bodies currently susceptible to HCBs are likely to become even more vulnerable with climate change compared to those that are currently less susceptible.

A number of algal bloom models (red-tide models) have been developed and applied to simulate the redtide growth and decline patterns as the interest on the phytoplankton blooms has been continuously increased. The quantitative error analysis of the model is of great importance because the accurate prediction of the red-tide occurrence and transport pattern can be used to setup the effective mitigations and counter-measures on the coastal ecosystem, aquaculture and fisheries damages. The word "red-tide model" is widely used without any clear definitions and references. It makes the comparative evaluation of the ecological models difficult and confusable. It is highly required to do the performance test of the red-tide models based on the suitable classification and appropriate error analysis because model structures are different even though the same/similar words (e.g., red-tide, algal bloom, phytoplankton growth, ecological or ecosystem models) are used. Thus, the references on the model classification are suggested and the advantage and disadvantage of the models are also suggested. The processes and methods on the performance test (quantitative error analysis) are recommend to the practical use of the red-tide model in the coastal seas. It is suggested in each stage of the modeling procedures, such as verification, calibration, validation, and application steps. These suggested references and methods can be attributed to the effective/efficient marine policy decision and the coastal ecosystem management plan setup considering the red-tide and/or ecological models uncertainty.

As the occurrence of harmful algal blooms (HABs) have become severe in precious water resources, the development of efficient harmful algae treatment methods is considering as an important environmental issue for sustainable conservation of water resources. To treat HABs in water resources, various conventional physical and chemical methods have been utilized and showed treatment efficiency, However, these methods can lead to discharging of cyanotoxins into the water bodies by chemical or physical algal cell lysis or destruction. Thus, to overcome this limitation, the development of safe HABs treatment methods is required. In the present study, adsorption technology was investigated for the removal of harmful algal species, Microcystis aeruginosa from aqueous phases. Industrial waste biomass, Corynebacterium glutamicum biomass was valorized as biosorbent (PEI-modified alginate/biomass composite fiber; PEI-AlgBF) for M. aeruginosa through immobilization with alginate matrix and cationic polymer (polyethylenimine; PEI) coating. The functional groups characteristic of PEI-Alg was determined using FT-IR analysis. By adsorption process used PEI-AlgBF, 52 and 67% of M. aeruginosa could be removed under the initial density of M. aeruginosa 200×104 cells mL-1 and 50×104 cells mL-1, respectively. As the increasing surface area of PEI-AlgBF, the removal efficiency was increased. In addition, we could find that adsorptive removal of M. aeruginosa has occurred without any M. aeruginosa cell lysis and destruction.
