Screening for Aldose Reductase Inhibitory Activity of Extracts of the Marine Plants from Korea (한국산 해산식물의 추출물로부터 Aldose Reductase 억제활성의 검색)

  • We examined MeOH extracts of the marine plants from Korea for their inhibitory activities on rat lens aldose reductase in vitro. Among the MeOH extracts tested, the extracts of Enteromorpha prolifera, Ecklonia cava, Pelvetia siliquosa and Salicornia herbacea exhibited a rat lens aldose reductase inhibition (IC$_{50}$: 3.04, 8.84, 11.42 and 4.99 $\mu$g-ml$^{-1}$, respectively) in vitro.

  • Sparganium stoloniferum Buch.-Ham. (Sparganiaceae) is a perennial herb which has been used as treatments for menstrual disorder, galactagogue and endometriosis in folk remedies. The Sparganii Rhizoma processed, heated with vinegar, used to different purpose compared with origin in medicine. In order to estimate the different effects between processed (PSR) and unprocessed (USR) Sparganii Rhizoma, extraction and systemic fractionation of the rhizomes were archived. Inhibitory effects of the extracts as well as the fractions of PSR and USR on rat lens aldose reductase and on rat platelet aggregation were investigated for the prevention or the treatment of chronic diabetic complications. Their antioxidant effects, measured using lipid peroxidation of liver tissue and radical scavenging activities on 1,1-diphenyl-picrylhydrazyl in vitro, were evaluated. The most of biological activities tested except rat platelet aggregation, fractions from PSR were shown to exhibit stronger activities than those from USR. It seems that caused by changing of the chemical components by heating process in conditioned with acetic acid. Compounds isolated were shown to significant inhibitory activity on rat lens aldose reductase. Inhibitory activity of compounds isolated on rat lens aldose reductase have been tested in vitro. $IC_{50}$/ (6.31 $\mu$M) of cerebroside I (V) was nearly equal to that (6.32 $\mu$M) of a reference compound, tetrametbylene glutaric acid (TMG).

  • Aldose reductase was purified to electrophoretic homogeneity from porcine liver. The purified enzyme was a monomer of 36 kDa. The enzyme was strongly inhibited by $Cu^{2+}\;and\;Mg^{2+}$ ions. Incubation of the enzyme with pyridoxal 5'-phosphate led to complete inhibition of enzymatic activity, suggesting that lysine residue is involved at or near the active site of the enzyme. The enzyme exhibited a broad substrate specificity. Furthermore, the enzyme was capable of decolorizing Alizarin, an anthraquinone dye.

  • The present study was undertaken to elucidate the hypoglycemic effect and inhibitory effect of Cudrania tricuspidata root bark on aldose reductase activity. C. tricuspidata MeOH ext. 1,000 mg/kg showed a significant blood glucose lowering effect on alloxan-induced hyperglycemic rats and increasing body weight. C. tricuspidata MeOH ext. showed a potent inhibitory effect on bovine lens aldose reductase.

  • Carvomethone, (+) pulegone, (-)isopulegol and (-) menthone, which are natural and widely distributed in the plantkingdom, were examined for their Lens Aldose Reductase inhibitory effects. All monoterpenes tested showed that the mild inhibitory effects. Inhibition percents of four monoterpenes were in the range of 23-42% at $10^{-3}M$ and 5-21% at $10^{-4}M$.

  • The inhibition of aldose reductase (AR) by the extracts from sixteen noxious and exotic plants was examined. Among them, Aster pilosus showed the highest inhibition of AR ($IC_{50}$ value $0.11{\mu}g/mL$). This study showed that A. pilosus with promising AR-inhibitory activities can be utilized for the development of natural therapies for treating and managing diabetic complications.

  • Inhibitory effects of the whole extract and isolated compounds from the leaves of Vaccinium koreanum (Ericaceae) on rat lens aldose reductase (RLAR) and on platelet aggregation were investigated for the prevention or the treatment of chronic diabetic complications. Arbutin, isolated from n-butanol fraction of methanol extract, exhibited a potent inhibitory effects on rat platelet aggregation induced by ADP $(IC_{50}=0.12\;{\mu}M)$ and collagen $(IC_{50}=0.039\;{\mu}M)$ and showed same inhibitory activities with positive control, tetramethylene glutaric acid, on RLAR.

  • We examined methanol extracts prepared from the Korean plants for their inhibitory effects on rat lens aldose reductase (RLAR) activity in vitro. Among 41 plant extracts, the extracts of Lagerstroemia indica, Punica granatum, Eurya japonica, Liquidambar styraciflua, and Vitis coignetiae exhibited highest inhibitory potency, even more potent than tetramethylene glutaric acid (TMG), known as one of typical aldose reductase inhibitors $(IC_{50}\;value,\;0.51\;{\mu}g/ml)$. Especially, the extract of Lagerstroemia indica (Lythraceae), exhibited the most inhibitory potency $(IC_{50}\;value,\;0.069\;{\mu}/ml)$ on RLAR.

  • Quantitative structure activity relationship has been probed for spirosuccinimide-fused tetrahydropyrrolo[1,2-a]pyrazine-1,3-dione derivatives acting as aldose reductase inhibitors. While the spirosuccinimide compounds contain a chiral center, the aldose reductase inhibition assay was performed with racemic mixtures in the published work. As the physicochemical descriptors of the QSAR analysis must be evaluated for a definite molecular structure, we devise a new 'racemic' descriptor as the arithmetic mean of the (R)-enantiomer descriptor and the (S)-enantiomer descriptor. The resultant QSAR model derived from the racemic descriptors outperforms the original QSAR models, closely reproducing the observed activity of optically pure enantiomers as well as racemic mixtures.
