N-nitrosoethylurea가 쥐 간세포의 항산화효소의 활성에 미치는 영향 (Effects of N-nitrosoethylurea on the Activities of Antioxidant Enzymes from Rat Liver Cell)

  • N-nitrosoethylurea(NEU)에 의한 지질 과산화물의 함량 변화와 알데히드 대사효소 및 항산화효소의 활성변화를 쥐 간세포에서 측정하였다. 알데히드 대사효소로는 alcohol dehydrogenase와 aldehyde dehydrogenase가 사용되었고 항산화효소로는 glutathione transferase, superoxide dismutase, glutathione reductase와 catalase가 사용되었다. 쥐 간세포에 다양한 농도의 NEU를 처리한 후 지질 과산화물의 함량변화를 측정하였다. 그 결과 6.25mM NEU에 의하여 지질 과산화물의 함량이 최대 2.5배 증가하였다. Alcohol dehydrogenase의 활성은 NEU처리에 의하여 대조군보다 최대 2.3배 증가하였고 aldehyde dehydrogenase의 활성은 약 2배 증가하였다. 전암성 병변의 지표로 이용되는 glutathione transferase와 catalase의 경우 NEU처리에 의한 활성증가가 미미하였다. 그러나 superoxide dismutase의 활성은 최대 1.5배 증가하였고, glutathione reductase의 활성은 약 3배 증가하였다. 그러므로 superoxide dismutase와 g1utathione reductase의 활성증가가 NEU의 독성에 대한 세포내 항산화 방어과정에서 중요한 역할을 할 것으로 추측된다

  • Chloral hydrate(CH), an intermediate metabolite of trichloroethylene(TRI) is reduced to trichloroethanol(TCE-OH), and is oxidized to trichloroacetic acid(TCA) by the nicotinamide adenine dinucleotide(NAD)-dependent enzymes such as alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) in liver. This study was performed to find out the change of activity of ADH and ALDH with increasing amount of TRI. Intraperitoneal injection of TRI were done to the male Sprague Dawely rats(mean body weight, $170{\pm}10g$) in com oil at the dosage of 150, 300, 600 mg/kg for 2 days. The results of experiments are following : 1. The contents of xenobiotic metabolic enzymes in liver are tended to be decreased with increasing amount of, but not significantlly (p>0.05). 2. Activity of ADH in microsome is decreased(p<0.05), and activity of ALDH is increased with amount of TRI(P<0.05). 3. Total trichloro-compounds(TTC) concentration in urine are increased with amount of TRI, but the ratio of between the TCE-OH and the TCA were not shown any critical change. These results suggests that the ALDH in microsome may be related to metabolism of TRI, but ADH was nothing less than the effected to metabolism of TRI.

인삼 사포닌이 효모 Alcohol Dehydrogenase에 미치는 영향 (The Effect of Ginseng Saponin on Yeast Alcohol Dehydrogenase)

  • Yeast alcohol dehydrogenates and ginseng saponin interaction has been investigated to understand the non-specific enzyme stimulating effect of the saponin of Panax ginseng C.A. Meyer. It was confirmed that several amphiphiles such as sodium dodecyl sulfate(SDS), Triton X-100, sodium taurodeoxycholate (Na-TDC) as well as ginseng saponin mixture and purified ginseng glycosides lowered Km values of yeast alcohol dehydrogenase (ADH) for ethanol and NAD in the presence of the above amphiphiles suggesting that the surface activity of the amphiphiles might play a significant role in the ADH catalyzed reactions. Conformational change of yeast alcohol dehydrogenase in the presence of the above amphiphiles at their optimal concentration for the maximum activity was studied. Circular dichroism (C.D) spectrum of yeast ADH showed that the conformational change of the enzyme occurred in the presence of above amphiphiles. Fluorescence data also showed that the hydrophobic area increased in the presence of above amphiphiles. Examination of the interaction between ADH and ginseng saponin using radioactive saponin showed that there might be a very weak interaction between them. From the above results, it was concluded that the non-specific enzyme stimulating effect of the saponin might be due to the change of polarity of the enzyme solution in the presence of the saponin.

만성 알콜 섭취로 인한 간내 알데히드 탈수소 효소 활성의 변동 (Alteration of the Aldehyde Dehydrogenase Activity by the Chronic Ethanol Administration)

  • The system most likely responsible for the accelerated metabolism of alcohol with chronic ingestion or at high blood ethanol levels, is the microsomal ethanol-oxidizing system(M EOS). While the increase in the MEOS with chronic ethanol ingestion is thought to be adaptive, it may also have serious adverse effects on the liver. The rates of the NADPH-dependent oxygen consumption by the liver microsomes from the prolonged ethanol fed rats were 2 times higher than the rates from the non-treated rats. With the alcohol ingestion, the total SH and nonprotein SH contents showed the significant decrease and at the same time, MDA in liver and GOT and GPT levels in blood showed the significant increase, which suggests the occurrence of liver damage due to the oxidative stress caused by chronic alcohol consumption. The mitochondrial aldehyde dehydrogenase(ALDH) activity was decreased by chronic ethanol ingestion, whereas the alcohol dehydrogenase activity and the cytosolic ALDH activity were not altered. These results suggest that the induction of cytochrome P450 by the chronic alcohol ingestion increases the oxidative stress which seems to result in the altered the physiological states of the liver including the ALDH activity, which may in turn to lead to the liver disease.

흰쥐에 Toluene과 Alcohol의 병행투여가 Toluene 대사 효소활성에 미치는 영향 (Effect of the Combination of Ethanol with Toluene Treatment for a Short Time Period on the Toluene Metabolizing Enzyme Activity)

  • 흰쥐를 대조군과 체중 100g 당 ethanol 0.3ml를 1일 1회 4일간 복강내로 투여한 실험군, 체중 100g당 toluene 0.3ml를 1일 1회 4일간 복강내로 투여한 실험군 및 ethanol 투여 2시간 후 toluene을 투여한 군으로 나누어 실험을 행하여 다음과 같은 결과를 얻었다. Alcohol 투여군, toluene투여군 모두 간 microsomal aniline hydroxylase(AH) 및 aminopyrine demethylase(AD) 활성도가 대조군 보다 유의한 증가를 보였다. Toluene과 ethanol의 병 행군은 toluene 투여군 보다 AH 및 AD 활성도가 다소 낮게 나타나는 경향을 보였다. 간조직 중 benzylalcohol dehydrogenase 활성도 역시 alcohol 및 toluene 투여군 모두 대조군에 비하여 높게 나타났으며 toluene과 alcoho의 병행투여군은 toluene 투여군 보다 본 효소 활성도가 다소 낮게 나타나는 경향을 보였다. 한편 간조직 중 benzaldehyde dehydrogenase 활성도는 alcohol 및 toluene 투여군 모두 대조군에 비하여 증가되는 경향을 보였으며 alcohol과 toluene의 병행투여군은 toluene 투여군 보다 유의한 감소를 보였다. 또한 간조직 중 xanthine oxidase 활성도는 alcohol과 toluene 병행 투여군이 toluene 투여군 보다 유의하게증가되었다. 이상 실험성적을 종합해 볼 때 alcohol의 전처치는 toluene 대사 효소 활성도를 억제시켜 toluene 대사에 영향을 미칠 것으로 생각된다.

꾸지뽕나무 뿌리 추출물의 알코올 대사 효소 활성 및 혈액 산성화 기전 조절을 통한 숙취해소 효과 (Effects of Cudrania Tricuspidata Root Extract (CTE) on Ethanol-Induced Hangover via Modulating Alcohol Metabolizing Enzyme Activities and Blood Gas Levels in Rats)

  • 본 연구에서는 알코올을 섭취한 흰쥐에서 알코올 대사와 혈액가스 불균형에 대한 꾸지뽕 나무 뿌리 추출물 (이하 CTE)의 효과를 확인하고자 하였다. 알코올을 10 ml/kg의 농도로 흰쥐에게 경구 투여하였고, 섭취 30분 후 CTE를 500 mg/kg 및 750 mg/kg의 농도로 투여하였다. 혈액은 알코올 섭취 1시간, 3시간, 5시간 후 꼬리 정맥을 통하여 채취하였다. 양성 대조군은 시판되고 있는 숙취해소 음료 (컨디션)를 사용하였다. 약물 처리는 모두 단회 경구 투여로 진행되었다. CTE의 섭취는 흰쥐의 혈액과 간 조직 내 알코올 대사 효소인 알코올 분해효소 (alcohol dehydrogenase)와 아세트알데히드 분해효소(acetaldehyde dehydrogenase)를 증가 또는 유지시킴으로써 혈중 알코올 농도를 감소시켰다. 또한, CTE는 알코올섭취에 의한 혈액가스 (pH, $CO_2$, $HCO_3{^-}$, lactic acid)의 불균형을 회복시켰다. 결론적으로, CTE는 현재 시판되고 있는 숙취해소음료 보다 우수한 숙취 해소 효과를 나타내었다. 따라서 본 연구자는 CTE를 숙취 해소 및 예방을 위한 천연물 유래의 안전하고 새로운 물질로써 제시하는 바이다.

Alcohol 섭취 쥐에서 Alchol 대사효소계에 미치는 복어추출물의 효과 (Effect of Puffer Fish Extract on the Hepatic Alcohol Metabolizing Enzyme System in Alcohol-Treated Rat)

  • The present study was undertaken to clarify the effect of Puffer fish skin extract (PF) on the hepatic alcohol metabolism in rats. It was observed that alcohol concentration in blood had been markedly decreased by the pretreatment of PF for two weeks. Activities of alcohol dehydrogenase (ADH) and microsomal ethanol-oxidizing system (MEOS) were significantly incrased (more than 20% of control) by pretreatment of PF for two weeks and acute alcohol intoxication (5 g/kg) on final day. When rats were fed with subacute toxic state by alcohol (25v/v % , once a day for six weeks), activities of ADH and MEOS were significantly increased by additional treatments of PF for final two weeks. But the catalase activity was not affected by any of both case. And also activities of ADH and MEOS in vitro were not changed . These results suggest that PF treatemnt prompted the recovery from alcohol intoxication.

Comparison of silkworm powder from 3 Bombyx mori varieties on alcohol metabolism in rats

  • Increased alcohol consumption is a burden on the world because it is associated with various health problems. However, the effects of silkworms on alcohol metabolism have not been studied yet. The hard-to-eat mature silkworms have become easier to ingest recently due to the development of technology, steam-lyophilising mature silkworm larvae. In this study, we investigated and compared the effects of SMSPs from three silkworm varieties, Baekokjam, Golden-silk and Yeonnokjam weaving white, golden, and light green cocoons on alcohol metabolism in vivo. Sprague-Dawley rats pretreated with three SMSPs (0.1 g/kg or 1 g/kg body weight) or normal diet (AIN-76A) for 2 weeks were subjected to intragastric administration of absolute ethanol (3 g/kg body weight, 3 h). Three SMSPs did not affect the final body weight and liver weight. All 3 SMSPs were effective to reduce the enzymes in alcohol metabolism, alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH), and liver damage and enzymes involved in liver damage, aspartate aminotransferase (AST) and alanine aminotransferase (ALT). Among SMSP from 3 varieties of silkworm, preadministration of 1 g/kg Baekokjam SMSP showed the most effective suppressive effect on the activities of ADH, ALDH, AST and ALT. The Baekokjam SMSP contained higher amounts of beneficial amino acids than Golden-silk or Yeonnokjam SMSP. These results suggest that Baekokjam SMSP might be used as a new and promising candidate for improving alcohol metabolism and liver injury through promoting rapid alcohol metabolism.

한약재 추출물 함유 음료가 알코올 투여 흰쥐의 알코올 대사 관련 효소에 미치는 영향 (Effects of Medicinal Herbal Drink on Alcohol Metabolic Enzyme in Drunken Rats)

  • Alcohol is the most widely psychoactive drug and has known in almost all civilization since ancient time. Recently increase consuming alcoholic beverages, alcohol is on of the major public health problems in the world. Alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) play important roles in the metabolism of alcohols and aldehydes. The drink consists of medicinal herbs, Puerariae Radix, Phyllostachyos Folium, Citri Pericarpium, Polygonati Rhizoma, Rehmanniae Rhizoma (Vinegar), which have been widely used in oriental medicine. This study was designed to investigate effects of medicinal herbal drink (MHD) on alcohol metabolism in drunken SD rats subjects. In experiment, rats were treated to ethanol (EtOH, 3 g/kg, PO) at 60 min. after saline (CON) or MHD (1 ml/kg, PO) administration. The blood alcohol concentration (BAC), blood acetaldehyde concentration (BALC) activities of ADH, ALDH, AST and ALT were significantly decreased in MHD group than in control group as a time-dependent manner. And drinking water volume in MHD group with duplicate treatment, were significantly decreased than in CON group. These results suggested that MHD intake could give an influence upon the reduction in BAC and BALC may alleviate acute ethanol-induced hepatotoxicity by altering alcohol metabolic enzyme activities.

The Effects of Calcium Nutrition on the Activities of Lactate Dehydrogenase, Alcohol Dehydrogenase and Other Enzymes in Melon (Cucumis melo L.) Seedlings Subjected to Flooding

  • With transient flooding followed by poor or slow drainage plant roots may become reduction conditions because the root zone was fully filled with water. This study was examined the effects of calcium treatment in the early growth stage on biochemical changes in leaves and roots of melon (Cucumis melo L.) seedlings kept under flooding condition for 72 h. The activities of lactate dehydrogenase more gradually enhanced in the roots than those of leaves of melon seedlings treated with calcium. The activities of alcohol dehydrogenase associated with alcohol fermentation under low oxygen conditions continuously increased in the leaves and roots of seedlings untreated with calcium under flooding at least 72 h but those was constant within at least 12 h in treated with calcium. These results showed that calcium supplying in the early growth stage mitigated alcohol fermentation of melon seedlings kept under flooding condition for 72 h. Activities of nitrate reductase and acid phosphatase in the leaves and roots of seedlings in treated with calcium somewhat higher than those of non-treated with calcium. The activities of sucrose phosphate synthase and fructose-1,6-bisphosphatase of leaves of seedlings in treated with calcium more higher than those of non-treated with calcium. These results indicated that calcium nutrition mitigate the reduction of activities of some enzymes of melon seedling kept under flooding condition for 72 h.
