Protective Effect of Fucoidan Extract from Ecklonia cava on Hydrogen Peroxide-Induced Neurotoxicity

  • We evaluated the antioxidant activity and neuronal cell-protective effect of fucoidan extract from Ecklonia cava (FEC) on hydrogen peroxide ($H_2O_2$)-induced cytotoxicity in PC-12 and MC-IXC cells to assess its protective effect against oxidative stress. Antioxidant activities were examined using the ABTS radical scavenging activity and malondialdehyde-inhibitory effect, and the results showed that FEC had significant antioxidant activity. Intracellular ROS contents and neuronal cell viability were investigated using the DCF-DA assay and MTT reduction assay. FEC also showed remarkable neuronal cell-protective effect compared with vitamin C as a positive control for both $H_2O_2$-treated PC-12 and MC-IXC cells. Based on the neuronal cell-protective effects, mitochondrial function was analyzed in PC-12 cells, and FEC significantly restored mitochondrial damage by increasing the mitochondrial membrane potential (${\Delta}{\Psi}m$) and ATP levels and regulating mitochondrial-mediated proteins (p-AMPK and BAX). Finally, the inhibitory effects against acetylcholinesterase (AChE), which is a critical hydrolyzing enzyme of the neurotransmitter acetylcholine in the cholinergic system, were investigated ($IC_{50}$ value = 1.3 mg/ml) and showed a mixed (competitive and noncompetitive) pattern of inhibition. Our findings suggest that FEC may be used as a potential material for alleviating oxidative stress-induced neuronal damage by regulating mitochondrial function and AChE inhibition.

Comparison of 14C-radioactivity in rice-paddy soil exposed to atmospheric and elevated CO2 conditions after 14C-carbaryl treatment

  • This study was performed to investigate if elevated $CO_2$ affects the residue pattern of $^{14}C$ in the soil environment after $^{14}C$-carbaryl treatment $^{14}C$-carbaryl was applied on the rice plant-grown greenhouse soil exposed to atmospheric and elevated $CO_2$ conditions. $^{14}C$-radioactivity was measured in the rhizospheric soil and rice straw samples six months after $^{14}C$-carbaryl application. Significantly high radioactivity was observed in the soil exposed to atmospheric $CO_2$ as compared to that in the soil exposed to elevated C(h. Background level of radioactivity was observed in rice plant samples. These observations suggest the possibility that elevated $CO_2$ may affect residual radioactivity of $^{14}C$-carbaryl in the soil rather than that in the plant.

Effect of Heating on Polymerization of Pig Skin Collagen Using Microbial Transglutaminase

  • Polymerization of heated or unheated pig skin collagen using microbial transglutaminase (MTGase) was investigated. Pig skin collagen samples were heated or left unheated, then enzymatically polymerized with MTGase. SDS-PAGE was conducted to confirm the intermolecular polymer and the results showed similar bands between samples without MTGase and unheated samples with MTGase. The polymerized product of pig skin collagen was not formed in unheated samples, even when MTGase was added during incubation. Different results were obtained from samples heated at $80^{\circ}C$ and $100^{\circ}C$ for 2 min, whereas the SDS-PAGE pattern indicated that a polymer band was generated in both cases. The heat treatment successfully modified the native structure of collagen and also made collagen more reactable in the MTGase polymerization system. Scanning Electron Microscope (SEM) investigation of pig skin collagen showed a biopolymer structure through intermolecular collagen crosslinking, while there were no intermolecular crosslinks in samples not treated with MTGase. There were no significant differences in fibril diameter between treated samples and controls. These results suggest that heat treatment of native pig skin collagen enhanced the polymerization capability of MTGase.

Establishment of a diagnostic method for porcine proliferative enteropathy using polymerase chain reaction (중합효소연쇄반응을 이용한 돼지 증식성 장염 진단기법 확립)

  • Porcine Proliferative Enteropathy(PPE) is an infectious enteric disease and a major cause of economic loss in swine industry due to weight loss, poor growth and sudden death in growing and finishing pigs at 6 to 20 weeks of age. PPE has been diagnosed by clinical signs, syndrom and lesions in the intestine in Korea. However, the diagnostic method had several problems in the detection of infected or carrier pigs. Therefore, in this study, we established the polymerase chain reaction(PCR) which was a fast, specific and sensitive method for identification of Lawsonia intracellularis (L intracellularis). We designed and synthesized primer on the 16S rDNA and p78 gene encoding L intracellularis. Specificity of the method was confirmed by comparison of the PCR results using other enteric bacteria and the study has shown that PCR method was sensitive to detect 1ng of genomic DNA as a template. Identity of the PCR products was confirmed by comparison of pattern of restriction endonuclease analysis with restriction enzyme Hae III and Pst I. Also, the PCR method was applicable to the naturally affected pigs with PPE. Based on the results from this study, the PCR method could be used as a fast and specific diagnostic tool for PPE.

Changes in Wet Pressing Response of OCC stock by the Beating Time and the Addition of Polymer Aids (골판고지 지료의 고해처리 및 고분자첨가제에 의한 압착탈수 특성변화)

  • The changes in the wet pressing response of old corrugated container(OCC) stock depending on the beating treatment and the addition of polymer aids were investigated with laboratory roll press instrument. Two types of polymer aids, such as high molecular weight polymer with low charge density and low molecular weight polymer with high charge density, were applied in this study. The more beaten OCC stock showed the lower dryness after wet pressing. The addition of polymer aids had great influences on the wet pressing efficiency and paper properties. The dryness after wet pressing was increased by the addition of polymer aids, but the pattern of changes in dryness were different depending on the type of polymer and the properties of stock. The higher molecular weight polymer aids showed the greater increase in the dryness. The properties of paper such as air permeability, bulk, formation, tensile strength were also greatly affected by the addition of polymer aids.

  • Electrophoretic isozyme patterns from mycelia, primordia, cap and stem of Pleurotus spp. collected in Korea were compared. Primordia, cap and stem of fruitbody showed very similar isozyme patterns but mycelial isozyme patterns were different from those of fruitbody. Isozyme patterns of malate dehydrogenase, acid phosphatase in Pleurotus ostreatus collected from different regions in Korea were similar but those of esterase, peroxidase, leucine amino peptidase and superoxide dismutase were different. Interspecific comparison of esterase isozyme patterns among Pleurotus ostreatus P. cornucopiae and, P. florida was very different and may be valuable subsidiary tool to conventional taxonomic techniques for identifying species of Pleurotus. A dendrogram by similarities of isozyme band pattern was presented.

Development of a Molecular Marker for Fruiting Body Pattern in Auricularia auricula-judae

  • The fruiting body pattern is an important agronomic trait of the edible fungus Auricularia auricula-judae, and an important breeding target. There are two types of fruiting body pattern: the cluster type and the chrysanthemum type. We identified the fruiting body pattern of 26 test strains, and then constructed two different near-isogenic pools. Then, we developed sequence characterized amplified region (SCAR) molecular markers associated with the fruiting body pattern based on sequence-related amplified polymorphism (SRAP) markers. Ten different bands (189-522 bp) were amplified using 153 pairs of SRAP primers. The SCAR marker "SCL-18" consisted of a single 522-bp band amplified from the cluster-type strains, but not the chrysanthemum strains. This SCAR marker was closely associated with the cluster-type fruiting body trait of A. auricula-judae. These results lay the foundation for further research to locate and clone genes controlling the fruiting body pattern of A. auricula-judae.

Characteristics of Feeding Behaviors of Myzus persicae (Hemiptera: Aphididae) Depending on Inflow Concentrations of Dinotefuran (작물체내로의 dinotefuran 유입농도에 따른 복숭아흑진딧물 (Myzus persicae) (Hemiptera: Aphididae)의 섭식행동)

  • The changes of feeding pattern and the amount of insecticide penetrated into the leaf were monitored for 420 min after treatment of three concentrations of dinotefuran. At about 30 min after treatment of insecticide, the overall feeding pattern in EPG (Electrical Penetration Graph) monitoring was started to change, and there was significant difference in proportions of NP (non-penetration), PA (pathway activity), and phloem-feeding patterns among the different concentrations of dinotefuran treatment. Especially, as the amount of insecticide penetrated into a leaf increased, the reactive behaviors against this insecticide such as withdrawal of proboscis and movements of stylet of aphid were more quickly exhibited. And also, total time at which the proboscis of the aphids did not penetrate the plant was getting longer. The amount of dinotefuran penetrated into a leaf was monitored with HPLC. When the recommended concentration (100 mg/L) of the insecticide was treated, 5.24${\sim}$7.24 mg/L of the insecticide was detected from the leaf, and the proboscis of the aphids was apparently withdrawn from the leaf at approximately 60 minutes after treatment of this insecticide.

Chemical and Morphological Change and Discoloration of Cedar Wood Stored Indoor (실내 보관 삼나무 목재의 재색 및 화학적·현미경적 변화 특성)

  • The modification of wood color occurs rapidly during the service period at indoor. It is crucial to investigate the characteristics of color change, chemical and microscopical modification of wood at indoor. Wood products made of Japanese cedar at different years were used for this work. The tests were performed in order to evaluate the degree of color change of wood surface, breakpoint of brightness from surface to inside of wood, chemical analysis with FT-IR, and microscopical characteristics using the LM and TEM. Surface color of cedar wood stored indoor were rapidly changed at early stage, particularly ${\Delta}a$ (yellow), and ${\Delta}b$ (red) values were steeply decreased for one year old indoor wood, ${\Delta}L$ (white) value was dropped until 5 years old indoor wood compared with control sample. Decrease of peaks related to polysaccharide and lignin was noticed, especially, lignin was severely degraded. Although degradation of cell wall limited only to surface layers of indoor wood, degradation pattern of indoor wood showed similar degradation pattern to natural weathering of wood during outdoor weathering or wood behavior under artificial UV irradiation.

Evidence of genome duplication revealed by sequence analysis of multi-loci expressed sequence tagesimple sequence repeat bands in Panax ginseng Meyer

  • Background: Panax ginseng, the most famous medicinal herb, has a highly duplicated genome structure. However, the genome duplication of P. ginseng has not been characterized at the sequence level. Multiple band patterns have been consistently observed during the development of DNA markers using unique sequences in P. ginseng. Methods: We compared the sequences of multiple bands derived from unique expressed sequence tagsimple sequence repeat (EST-SSR) markers to investigate the sequence level genome duplication. Results: Reamplification and sequencing of the individual bands revealed that, for each marker, two bands around the expected size were genuine amplicons derived from two paralogous loci. In each case, one of the two bands was polymorphic, showing different allelic forms among nine ginseng cultivars, whereas the other band was usually monomorphic. Sequences derived from the two loci showed a high similarity, including the same primer-binding site, but each locus could be distinguished based on SSR number variations and additional single nucleotide polymorphisms (SNPs) or InDels. A locus-specific marker designed from the SNP site between the paralogous loci produced a single band that also showed clear polymorphism among ginseng cultivars. Conclusion: Our data imply that the recent genome duplication has resulted in two highly similar paralogous regions in the ginseng genome. The two paralogous sequences could be differentiated by large SSR number variations and one or two additional SNPs or InDels in every 100 bp of genic region, which can serve as a reliable identifier for each locus.
