Research Trends on Compounds that Promote Melanin Production Related to Hair Graying (모발 백발화와 관련된 melanin 생성을 촉진시키는 화합물의 연구동향)

  • Hair graying is the result of a malfunction in the signaling pathways that control melanogenesis, and it is activated by UV light, melanocyte-stimulating hormone (MSH), stem cell factor (SCF), Wnt, and endothelin-1 (ET-1). To prevent hair graying, synthetic and natural compounds can be used to stimulate melanogenesis effectively under the control of tyrosinase, tyrosine hydroxylase, tyrosinase-related protein-1 (TRP-1), TRP-2, and microphthalmia-associated transcription factor (MITF). This article describes a crucial strategy to resolve the problem of hair graying, as well as recent advances in the signaling pathway related to melanogenesis and hair graying. In particular, the article reviews potentially effective therapeutic agents that promote melanogenesis, such as antioxidants that modulate catalase, methionine sulfoxide reductase, and sirtuin 1 (SIRT1) activators including resveratrol, fisetin, quercetin, and ginsenoside. It also discusses vitiligo inhibitors, such as corticosteroids, calcineurin inhibitors, and palmitic acid methyl ester, as well as activators of telomerase expression and activity, including estrogen, androgen, progesterone, and dihydrotestosterone. Furthermore, it explores compounds that can inhibit hair graying, such as latanoprost, erlotinib, imatinib, tamoxifen, and levodopa. In conclusion, this article focuses on recent research trends on compounds that promote melanin production related to hair graying.

Anti-aging Effects of Marine Natural Extracts against UVB-induced Damages in Human Skin Cells (UVB로 손상이 유도된 피부세포에 해양소재 추출물의 항노화 효능)

  • The skin is continuously exposed to environmental stresses. One of the most important stress factor is UV radiation. UV radiation causes a variety of biological effects on the skin, including inflammation, pigmentation, photoaging and cancer. Therefore in this study, we tried to search for skin-protective antioxidant materials from marine natural products (Porphyra Thalli, Laminariae japonicae thallus, Ostreae Concha, Sargassum Thallus, Undaria thallus, Haliotidis Concha, Agar, Codium thalli, Hizikia fusiforme thalli; HFE, Thalli) which exhibit protective activities against UVB-induced cytotoxicity and oxidative cell death and antiaging effects. As a results, UVB-induced cytotoxicity and cell death were effectively suppressed by treatment of Sargassum Thallus, Agar, Haliotidis Concha, Codium thalli, Thalli ethanol extracts. UVB-induced cell death was mediated by intracellular accumulation or ROS, which was significantly inhibited by treatment with marine natural products extracts. Also, The protective effect of these marine natural products seemed to be mediated by increased expression of type I collagen and Type I procollagen. These results suggest that marine natural products may have anti-aging effects new functional materials against oxidative stress-mediated skin damages.

A Comparison of Upper Body Sizes and Body Types of Women in Their 20s and 30s-40s - Identifying Problems Generated by Preferences of Women in Their 30s and 40s for Young Casual Brands - (20대와 30~40대 여성의 상반신 신체치수 및 체형 비교 연구 - 30, 40대 여성의 영캐주얼 브랜드 선호 현상에 따른 문제점 파악을 중심으로 -)

  • Women in their 30s and 40s, who are at the center of "ageless" and "down-aging" consumer trends, are likely to encounter problems with fitting and size conformity when they wear casual clothes targeting women in their 20s. Hence, differences in upper body sizes and body types between women in their 20s and women in their 30s and 40s were analyzed. The data for this study was from the 6th Size Korea survey of body measurements of 1,675 female adults in their 20s-40s. SPSS 21.0 for Windows was used for analysis of the collected data. To examine differences in average upper body size between women in their 20s and women in their 30s and 40s, descriptive statistics and independent sample t-tests were conducted. Factor and cluster analyses were used to classify body types by age groups. Comparing direct measurement items showed that women in their 20s tend to have higher average values for most height-related items-including body height-and lower average values for circumference, thickness, and width than women in their 30s and 40s. Factors in determining the upper body shapes of women in their 20s to 40s were narrowed to five; through a cluster analysis, upper body shapes of women were classified into three body types as follows. Type 1 women are shorter and thinner with small frames; Type 2 women have the highest vertical values for their upper bodies and average values for obesity-related categories of circumference, thickness, and width. Type 3 women are the shortest and has the highest body mass index (BMI), verifiable as obese. By analyzing differences in body type distribution according to age groups, it was found that more than 90% of women in their 20s belong to Types 1 and 2. On the other hand, most women in their 30s and 40s are identified as Type 3.

Whitening Effect and Skin Regeneration Effect of Red Sea Cucumber Extract (홍해삼 추출물의 멜라닌 형성 억제를 통한 미백효과 및 피부 재생효과에 관한 연구)

  • Recently, several researchers have been developing cosmetics from natural ingredients for skin whitening and anti-aging products. The red sea cucumber (RSC), Apostichopus japonicas, is a species of sea cucumber in the family stichopodiae, which is widely distributed in China, Japan, and Korea. To use Red Sea Cucumber as a cosmetic ingredient, its inhibitory effects on melanogenesis and the anti-aging effects of RSC extracts were investigated. First, a tyrosinase activity assay was performed, which showed that RSC inhibited tyrosinase activity at a concentration of $200{\mu}g/ml$. An MTT assay was carried out to evaluate cell toxicity, and the results showed that RSC extract has no cytotoxicity in HaCaT cells. Furthermore, the mRNA expression levels of tyrosinase, tyrosinase related protein 1 (TRP-1), tyrosinase related protein 2 (TRP-2), microphthalmia-associated transcription factor (MITF), and matrix metalloproteinase (MMPs) genes treated with RSC extract in B16F10 and HaCaT cells decreased. Moreover, a wound-healing assay was performed to identify the cell regeneration effect of RSC extracts. Also, a skin turnover effect was confirmed by creating a three-dimensional cell culture with HaCaT and human fibroblasts. Altogether, the results suggested that Red Sea Cucumber may possess a high ability to induce whitening and anti-wrinkle effects as a cosmeceutical ingredient.

Correlations of Plasma Taurine, Homocysteine and Serum Lipid Levels of Elderly Women (노년기 여성들의 혈장 타우린의 농도와 혈장 호모시스테인 및 혈청 지질농도와의 상관관계)

  • This study was conducted to assess blood components caused by metabolic syndrome increasing in postmenopausal elderly women. The blood samples of these subjects were analyzed to investigate the correlation of plasma taurine levels and plasma homocysteine levels, and serum lipid profiles. The subjects were 33 elderly women($72.8{\pm}4.4$ years). Their mean height, weight and BMI were $150.5{\pm}5.7\;cm$, $57.5{\pm}6.3\;kg$ and $25.4{\pm}2.5\;kg/m^2$. 16 women of this study subjects have been chronic diseases such as hypertension or diabetes. Their fasting blood glucose was $98.2{\pm}24.0\;mg/d{\ell}$, and their plasma total cholesterol (TC), HDL-C, LDL-C, triglyceride(TG) were $216.5{\pm}29.9$, $52.1{\pm}10.7$, $145.7{\pm}27.9$ and $141.2{\pm}59.6\;mg/d{\ell}$, respectively. Their blood lipid profiles were higher than the standard levels of metabolic syndrome, thus these levels of lipid profiles may play a role as risk factors on the elderly person. Plasma taurine level of the subjects was $278.5{\pm}48.1\;{\mu}mol/{\ell}$, and their plasma homocysteine level was $12.8{\pm}2.9\;{\mu}mol/{\ell}$. The concentration of plasma vitamin $B_{12}$ was significantly decreased by aging(p<0.05). The correlation of plasma homocysteine and plasma folate showed significantly negative(p<0.05). Thus, the decreased levels of plasma vitamin $B_{12}$ and folate by aging might affect on the plasma homocyteine concentration acting as a risk factor of cardiovascular diseases for elderly person. The correlation of plasma taurine and hemoglobin, and their platelet showed significantly positive(p<0.05). In conclusion, the diet on the elderly person is one of the important factors to prevent their health from chronic diseases. This study recommends that well balanced diets are needed for elderly person to keep their health and prevent from metabolic syndrome.

Aquatide Activation of SIRT1 Reduces Cellular Senescence through a SIRT1-FOXO1-Autophagy Axis

  • Ultraviolet (UV) irradiation is a relevant environment factor to induce cellular senescence and photoaging. Both autophagy- and silent information regulator T1 (SIRT1)-dependent pathways are critical cellular processes of not only maintaining normal cellular functions, but also protecting cellular senescence in skin exposed to UV irradiation. In the present studies, we investigated whether modulation of autophagy induction using a novel synthetic SIRT1 activator, heptasodium hexacarboxymethyl dipeptide-12 (named as Aquatide), suppresses the UVB irradiation-induced skin aging. Treatment with Aquatide directly activates SIRT1 and stimulates autophagy induction in cultured human dermal fibroblasts. Next, we found that Aquatide-mediated activation of SIRT1 increases autophagy induction via deacetylation of forkhead box class O (FOXO) 1. Finally, UVB irradiation-induced cellular senescence measured by $SA-{\beta}-gal$ staining was significantly decreased in cells treated with Aquatide in parallel to occurring SIRT1 activation-dependent autophagy. Together, Aquatide modulates autophagy through SIRT1 activation, contributing to suppression of skin aging caused by UV irradiation.

Nutritional Status of Antioxidant Vitamins in the Elderly Living in Ulsan Metropolitan City

  • Body antioxidant status is an important factor for the prevention of many chronic diseases in the elderly. This study was done to evaluate the nutritional status of antioxidant vitamins of the elderly by determining their intakes and plasma levels. It was also aimed to compare daily intakes and plasma levels of antioxidant vitamins by sex of age. Subjects were 225 elderly persons aged over 60 years old (63 males, 162 females) living in Ulsan area. Subjects were divided by groups according to age(< 65, 65-74, 75) and sex. Dietary Intakes were assessed by semi-quantitative food frequency questionnaires(FFQ). Plasma Vitamin C level was measured by 2,4-dinitrophenylhydrazine method and plasma levels of vitamin E, A and ${\beta}$-carotene were measured by HFLC. The average intakes of vitamin C were 104.9g(150% of RDA) and 104.4g(149% of RDA) in the elder1y males and females, respectively. Vitamin C intake of the elderly was significantly decreased by aging but not different by sex. The average intakes of vitamin A were 678$\mu\textrm{g}$RE(96.9% of RDA) and 604$\mu\textrm{g}$:RE(86.2% of RDA) in elderly males and females, respectively. The average levels of Plasma vitamin C were 6.22mg/L and 11.45mg/L in the elderly males and females, respectively. Those levels are within normal range(6-20mg/L). However the percentage of the elderly males with deficiency(< 2mg/L) and marginal level(2-4mg/L) of vitamin C were 27.4% and 16.1%, respectively. Plasma retinaol levels were 0.39mg/L for the elderly males and 0.37mg/L for the elderly females, which were within normal range. But the percentage of subjects with marginal level were 28% ill both males and females. Plasma ${\alpha}$-tocopherol levels of the elderly were lower than normal range(5-12mg/L). Plasma levels of vitamin C, E and ${\beta}$-carotene, except retinol, were significantly higher in the elderly females compared to males and showed significant decrease by aging.

  • Along with economic growth in the 1970s, the nation's economy, railroads, and airports have soared, but the facilities have been aging for 30 years. The aging SOC lowers the national competitiveness and threatens the safety of the people. SOC is a key asset of the national life and economic activity, and is a key factor for evaluating the national competitiveness. Therefore, it is necessary to systematically manage and operate the SOC related to people's lives. In the USA, the SOC report card is issued regularly. Those reports are used as a basis for securing budgets. In Korea, facilities management institutions are different. Therefore, there is no integrated SOC monitoring system. Hence, this study developed a 'SOC information system' that can collect reports scattered throughout the country and support report cards. The demonstration system was implemented through the web and the feasibility of implementing the evaluation support system was confirmed. The collection distributed SOC data should be supported by national policy. Although it takes a long time, it is necessary to improve national competitiveness and public safety.

Genome-wide hepatic DNA methylation changes in high-fat diet-induced obese mice

  • BACKGROUND/OBJECTIVES: A high-fat diet (HFD) induces obesity, which is a major risk factor for cardiovascular disease and cancer, while a calorie-restricted diet can extend life span by reducing the risk of these diseases. It is known that health effects of diet are partially conveyed through epigenetic mechanism including DNA methylation. In this study, we investigated the genome-wide hepatic DNA methylation to identify the epigenetic effects of HFD-induced obesity. MATERIALS AND METHODS: Seven-week-old male C57BL/6 mice were fed control diet (CD), calorie-restricted control diet (CRCD), or HFD for 16 weeks (after one week of acclimation to the control diet). Food intake, body weight, and liver weight were measured. Hepatic triacylglycerol and cholesterol levels were determined using enzymatic colorimetric methods. Changes in genome-wide DNA methylation were determined by a DNA methylation microarray method combined with methylated DNA immunoprecipitation. The level of transcription of individual genes was measured by real-time PCR. RESULTS: The DNA methylation statuses of genes in biological networks related to lipid metabolism and hepatic steatosis were influenced by HFD-induced obesity. In HFD group, a proinflammatory Casp1 (Caspase 1) gene had hypomethylated CpG sites at the 1.5-kb upstream region of its transcription start site (TSS), and its mRNA level was higher compared with that in CD group. Additionally, an energy metabolism-associated gene Ndufb9 (NADH dehydrogenase 1 beta subcomplex 9) in HFD group had hypermethylated CpG sites at the 2.6-kb downstream region of its TSS, and its mRNA level was lower compared with that in CRCD group. CONCLUSIONS: HFD alters DNA methylation profiles in genes associated with liver lipid metabolism and hepatic steatosis. The methylation statuses of Casp1 and Ndufb9 were particularly influenced by the HFD. The expression of these genes in HFD differed significantly compared with CD and CRCD, respectively, suggesting that the expressions of Casp1 and Ndufb9 in liver were regulated by their methylation statuses.

Anti Photoaging Effects of Hyaluronic Acid following Oral Administration in SKH-1 Hairless Mice (SKH-1 Hairless Mice에서의 히알루론산 경구 투여를 통한 피부 광노화 개선 효과)

  • Photoaging is the main extrinsic aging factor that is induced due to UVB. Many studies have revealed that application of hyaluronic acid to the skin is effective in healing photoaging. However, the effect of hyaluronic acid through oral administration is unclear. The aim of this research was to investigate the effect on skin photoaging after oral administration of hyaluronic acid. During a ten week study, hyaluronic acid was fed to hairless mice, which were illuminated by UV radiation. After ten weeks, wrinkle indicator and histological changes were determined. Compared with the control group, the sample group had a decrease in wrinkle depth, thickness, and number. Especially, the HA 160 mg/kg group had a similar value of wrinkle depth ($19.44{\pm}0.75{\mu}m$), number ($654.00{\pm}98.34$), and thickness ($1.35{\pm}0.08mm$), when compared with the RA (retinoic acid) group ($19.28{\pm}0.95{\mu}m$, $653.57{\pm}83.54$, and $1.34{\pm}0.07mm$ respectively). Also, the treated group showed improved elastosis and decreased collagen degradation resulting from UV irradiation. Thus, we can conclude that hyaluronic acid has a positive effect in improving skin photoaging induced by UV radiation. Hyaluronic acid therefore has the potential to be an ingredient in skin health function foods.
