High-Cell-Density Fed-Batch Culture of Saccharomyces cerevisiae KV-25 Using Molasses and Corn Steep Liquor

  • High-cell-density cultivation of yeast was investigated using the agricultural waste products corn steep liquor (CSL) and molasses. The Saccharomyces cerevisiae KV-25 cell mass was significantly dependent on the ratio between C and N sources. The concentrations of molasses and CSL in the culture medium were statistically optimized at 10.25% (v/v) and 16.87% (v/v), respectively, by response surface methodology (RSM). Batch culture in a 5-l stirred tank reactor using the optimized medium resulted in a cell mass production of 36.5 g/l. In the fed-batch culture, the feed phase was preceded by a batch phase using the optimized medium, and a very high dried-cell-mass yield of 187.63 g/l was successfully attained by feeding a mixture of 20% (v/v) molasses and 80% (v/v) CSL at a rate of 22 ml/h. In this system, the production of cell mass depended mainly on the agitation speed, the composition of the feed medium, and the glucose level in the medium, but only slightly on the aeration rate.

Sludge Returned CMAS에 의한 전기부속품제조공장 폐수처리

  • Sludge Returned CMAS process was applied to treat the wastewater from electric accessory manufacturing company while this type of wastewater was usually treated by chemical process. This result show that the removal rate of TCOD was about 70-80% regardless of hydraulic retention time, On the contrary, the removal rate of BOD was abtained in a range of 77-92% depending on hydraulic retention time. In order to remove more than 80% of organic materials with the proposed process, the F/M ratio should be maintained below 0.17. In this case, the calculated value of organic removal rate, Km, was calculated to be 1.26 hr$^{-1}$, and the ratio of cell synthesis/total energy was 0.32 and 0.26 for COD and BOD base, respectively. The yield coefficient was calculated to be 0.242 and the half velocity coefficient was 0.3 hr$^{-1}$. The value of endogenous respiration coefficient was 0.02 hr$^{-1}$. The measured effluent BOD concentration, MLSS concentration in aeration tank, oxygen uptake rate, and sludge production were matched relatively well with the calculated values using above coefficients, In order to optimize the dewatering of sludge, the hydraulic retention time was recommended to be 15. 6 hrs. These results indicate that the wastewater from an eletric accessory manufacturing company can be treated safely with a biological process.

  • Removal rate of swine wastewater containing organic matter was 99 percent in case of high loading rate. The studies of swine wastewater treatment aim to development of process using soil microorganism. Microorganism was devoted to improve the treatment efficiency of the process. Biochemical oxygen demand (BOD) concentration of treated swine wastewater was under 150 mg/L in anaerobic reactor. Also, value of km and Y were $0.73\;hr^{-1}$ and 0.433 g/VSS/g BODrm/d at microorganism of post aeration tank in anaerobic reactor.

A Pilot Study on Air Flotation Processes for Retrofitting of Conventional Wastewater Treatment Facilities (하수처리시설의 Retrofitting을 위한 파일럿 규모 공기부상공정 연구)

  • The pilot study was conducted to evaluate the applicability of air flotation(AF) processes combined with biological nutrient removal(BNR) for the retrofitting of conventional wastewater treatment facilities. The BNR system was operated in pre-denitrification and intermittent aeration; developed ceramic membrane diffusers were installed to separate the solid-liquid of activated sludge at the bottom of a flotation tank. Before performing a pilot scale study, the size distribution of microbubbles generated by silica or alumina-based ceramic membrane diffusers was tested to identify the ability of solid-liquid separation. According to the experimental results, the separation and thickening efficiency of the alumina-based ceramic membrane diffuser was higher than the silica-based ceramic membrane diffuser. In a $100m^3/d$ pilot plant, thickened and return sludge concentration was measured to be higher than 15,000mg SS/L, therefore, the MLSS in the bioreactor was maintained at over 3,000mg SS/L. The effluent quality of the AF-BNR process was 4.2mg/L, 3.7mg/L, 10.6mg/L and 1.6mg/L for $BOD_5$, SS, T-N and T-P, respectively. Lastly, it was revealed that the unit treatment cost by flotation process is lower than about $1won/m^3$ compared to a gravity sedimentation process.

Three Dimensional Simulation Model of Fuel Delivery Jet Pump (연료 송출용 제트펌프 3차원 전산해석 모델)

  • Jet pump in automotive fuel tank module is used to deliver fuel to fuel pump so that the pump is operated without aeration in suction side. In this study, three dimensional simulation model of jet pump is developed to understand performance variation over design parameters. Performance of jet pump is also investigated experimentally in terms of operating pressures. The experimental data is used to verify the three dimensional simulation model of jet pump. Verification results show that the three dimensional simulation model of jet pump is about 1% error with experiment. The simulations are conducted in terms of throat ratio and primary flow induction angle. As the throat ratio is increased, the flux ratio is trade-off at 3 times of throat diameter. On the other hand, as primary flow induction angle is increased, vapor pressure inside the nozzle is decreased. In summary, the results show that liquid jet pump has to be optimized over design parameters. Additionally, high velocity of induced flow is able to evolve cavitation phenomena inside the jet pump.

A Study on Characteristic Factors of Tubular Membranes in a Membrane coupled Activated Sludge Process (Tubular형 막을 이용한 활성슬러지법에 있어서 여과특성에 미치는 인자에 관한 연구)

  • We elucidated factors such as membrane pore size, activity of the activated sludge and MLSS. affecting filtering characteristics or filtering resistance in an activated sludge bio-process using tubular type membranes. We used a commercial membrane module with $0.5m^2$ of effective membrane area and a bundle of 7 tubes(13mm of ID and 2m long) in the experiments. As a result, the flux was maintained in the range of $35{\sim}105l/m^2$ hr at 5,000~20,000mg/l of MLSS concentration, and there seemed to be a correlation between the flux and the MLSS. Also the greater the pores were, the higher the flux was in some comparison experiments using UF and MF membranes, but the water permeated through both of the membranes showed almost the same quality. However, the MF membrane showed a great reduction in filtering resistance initially, but went back to a stable stage afterwards. The activity of the activated sludge in the aeration tank was also found to be considered as a factor showing permeation characteristics of the membrane.

Higher Production of Biolipids from Botryococcus braunii using Pre-treated Solvent Extraction Methods (해양생물 Botryococcus braunii에서 유래한 바이오연료의 고급생산기술: 전처리 용매추출법)

  • A lipid-enriched strain of Botryococcus braunii (UTEX 572) was cultivated in a semi-batch aeration tank to enhance biomass as well as to develop intracellular lipids and fatty acids. A 30 day period of incubation produced 1.39 g/L of biomass and 0.31 g/L of total lipids in the biomass. The grown biomass was pre-treated using several methods to extract the total lipid content efficiently: ultrasonication was found to yield the highest percentage of lipids-namely 19.8% per biomass. Direct heating of biomass in an autoclave also showed better performance than when using only conventional solvent extraction. To enhance the biomass harvest and lipid extraction efficiency, coagulation and flocculation steps were added to the extraction process. It is noteworthy that not only the solvent type but also the solvent/biomass ratio greatly affected efficiency. In addition, the moisture content of the harvested(wet) biomass affected the efficiency significantly. This study elucidated the need for future research on optimizing this extraction process.

The Bacterial Community Structure in Biofilms of the RABC Process for Swine Butchery Wastewater Treatment (돼지 도축폐수 처리를 위한 RABC 공정의 생물막 세균군집 구조)

  • Culture-independent microscopic observations and 16S rDNA analyses were applied to describe the bacterial community inherent to the biofilm structure of the RABC (Rotating Activated Bacillus Contactors) process for swine butchery wastewater treatment. The ratios of Gram-positive bacterial counts to total bacterial counts of the RABC process were significantly increased in the last aeration tank as well as returned sludge, while those of the existing A2O (Anaerobic-Anoxic-Oxic) process maintained constant from aeration tanks to returned sludge. Totally nine phyla were recovered by 16S rDNA analysis, two of which were major groups: the Proteobacteria (64.1%) and the Actinobacteria (18.4%). The third major group was the endospore-forming Firmicutes (5.4%). The remaining six minor groups are the Bacteroidetes (3.3%), the Chlorobi (2.2%), the Nitrospirae (1.1%), the Chlorofleix (1.1%), the Acidobacteria (1.1%), and the Fusobacteria (1.1%). The ratio of endospore-forming bacteria was 19.4%, which was composed of the members of the Firmicutes phylum (5.4%) and the Intrasporangiaceae family (14.0%) of the Actinobacteria phylum. Nitrifying and denitrifying related- and phosphorus accumulating related-sequences were composed of 6.5% and 5.4% of total community, respectively, these could mean the high capacity of the RABC process to remove odor compounds and reduce eutrophication by efficient removing inorganic nutrients.

A Study on Adjustment of Operational Factor in A2O process (A2O공정 운전인자 조정에 대한 연구)

  • An alternative was investigated to maximize the treatment efficiency of wastewater treatment plant without large scale expansion. 79% of detention time was required for enough nitrogen control. As aeration time was extended, nitrification was processed, but opposite trend was seen in phosphorus. High concentration of $NO_3-N$ interrupted PAOs activity not to absorb phosphorus. An alternative was devised for selective use of anaerobic or anoxic zone. Trisection was suggested for alternative use of center room. The result was relatively successful. The concentration of phosphorus was reduced with reduction of nitrogen. Extended anaerobic condition seemed to stimulate denitrification. Valve connection of internal return from aeration tank will make it possible to use middle room alternatively. This method will be a good alternative for seasonal variation of water temperature.

Estimation of Acidic Wastewater Toxicity on the Activated Sludge (활성슬러지에 미치는 산폐수의 독성도 예측)

  • Respiration rate should be a reasonable state variable for the activated sludge and could be used to simulate the performance of the activated sludge process. Toxic materials are classified into three groups, competitive, noncompetitive and uncompetitive. They increase/decrease the half saturation coefficient or specific growth rate. that means decreasing of the substrate removal capacity. In this research, a pilot-scale activated sludge process was operated under extended aeration method, and a representative noncompetitive inhibitor, acidic wastewater was applied to establish a respirometry-based toxicity model. Using this model. the correlation coefficient between measured and calculated respiration rate was 0.96 when acidic wastewater(pH 3.9~5.5) was introduced continuously to the aeration tank. Even though respiration rate was decreased by toxic effect of acidic wastewater, effluent substrate concentration represented to COD was deteriorated just a little bit. It might be caused by the low ratio of readily biodegradable substrate in the input substrate. Reduction of respiration rate by decreasing of input substrate concentration was much lower than that by acidic wastewater, and hence it was estimated that the possibility of false toxic alarm caused by decreasing of substrate concentration should be low.
