Drying Shrinkage has much complexity as it has relations with both internal elements of concrete and external factors. Therefore, experiments on Concrete Drying Shrinkage are carried out in this study under simplified circumstances applying temperature & Humidity test chamber which enables constant temperature and humidify. Comparative analyses have been made respectively according to the consequences aiming at modelling for prediction of Concrete Drying Shrinkage and making out measures to reduce it. As a result Strain Rate of Drying Shrinkage of concrete was measured to increase by average $10{\times}10^{-5}$ in proportion to additional 4% increase in fine aggregate ratio, when water/cement ratio constant. Strain Rate of Drying Shrinkage in pit sand concrete increased 20% higher than measured when in river sand under the condition of 90-day material age. 6. Strain Rate of Drying Shrinkage in sea sand concrete increased $10%{\sim}15%$ higher than measured when in river sand. The results of prediction of Rate of Drying Shrinkage by Response Surface Analysis are as fellows. The coefficient of correlation of Drying Shrinkage in concrete was over 90%.
From sludge of S municipal wastewater treatment plant in Busan, Korea, we isolated the denitrifier DN-9 which showed the ability of denitrification under aerobic conditionby the color change and gas formation in liquid culture with Giltay medium. The isolated strain was identified as Pseudomonas sp. DN-9 on the basis of the morphological, physiological, biochemical and nucleotide sequence analysis of l6S rRNA. The isolated strain, Pseudomonas sp. DN-9, has cytochrome $cd_1$, nirS of nitrite reductase. By the co-existance of additional ammonium and nitrate ion, the strain was not affected largely on growth in SL series broth. It seemed the result of denitrification. Although Pseudomonas sp. DN-9 has a good nitrate reduction activity under aerobic condition, the activity is less than Pseudomonas stutzeri in same cultivation condition. However, Escherichia coli had little the activity of aerobic denitrification and Pseudomonas putida showed lower activity of aerobic denitrification than Pseudomonas sp. DN-9 and Pseudomonas stutzeri in this study.
FK506 hyper-yielding mutant, called the TCM8594 strain, was made from Streptomyces tsukubaensis NRRL 18488 by mutagenesis using N-methyl-N'-nitro-N-nitrosoguanidine, ultraviolet irradiation, and FK506 sequential resistance selection. FK506 production by the TCM8594 strain improved 45.1-fold ($505.4{\mu}g/mL$) compared to that of S. tsukubaensis NRRL 18488 ($11.2{\mu}g/mL$). Among the five substrates, wheat bran was selected as the best solid substrate to produce optimum quantities of FK506 ($382.7{\mu}g/g$ substrate) under solid-state fermentation, and the process parameters affecting FK506 production were optimized. Maximum FK506 yield ($897.4{\mu}g/g$ substrate) was achieved by optimizing process parameters, such as wheat bran with 5 % (w/w) dextrin and yeast extract as additional nutrients, 70 % (v/w) initial solid substrate moisture content, initial medium pH of 7.2, $30^{\circ}C$ incubation temperature, inoculum level that was 10 % (v/w) of the cell mass equivalent, and a 10 day incubation. The results showed an overall 234 % increase in FK506 production after optimizing the process parameters.
This paper has presented a finite element analysis of structural behaviour of box culvert during and after fires. The fire tests were carried out in a furnace on RC slabs using the ISO 834 standard fire curve. The load capacity after cooling of the RC slab that was not loaded during the fire tests was evaluated by means of additional 3 points bending tests. In the past, stress-strain models of concrete under fire loading have been proposed by several researchers. Comparisons are made with the load-displacement relations of RC slabs after fire loading using the existing stress-strain models with temperature, such as Schneider, EUROCODE 2, Lie, Shi and Nan model. By comparing the load-displacement relations, Lie model was found to result in a maximum load about 2.0% higher than that of test. Based on the fire test results of RC slabs, this paper presents an extensive analytical study on the fire response of box culvert during and after fires.
In the present study the behavior of fire and the residual strength of fire-ignited RC slabs are investigated by experimental tests and numerical simulations. The fire tests of RC slabs were carried out in a furnace using the ISO 834 standard fire. The load capacity of the cooled RC slabs that were not loaded during the fire tests was evaluated by additional 3 point bending tests. The influence of the proportion of PP (polypropylene) fibers in the RC slabs on the structural behavior of the RC slabs after the fire loading was investigated. The results of the fire tests showed that the maximum temperature of concrete with PP fiber was lower than that of concrete without PP fiber. As the concrete was heated, the ultimate compressive strength decreased and the ultimate strain increased. The load-deflection relations of RC slabs after fire loading were compared by using existing stress-strain-temperature models. The comparison between the numerical analysis and the experimental tests showed that some numerical analyses were reliable and therefore, can be applied to evaluate the ultimate load of RC slabs after fire loading. The ultimate load capacity after cooling down the RC slabs without PP fiber showed a considerable reduction from that of the RC slabs with PP fiber.
Finite element analyses are carried out to understand the piezoelectric behaviors of ZnO nanowires. Three different types of ZnO nanowires, with aspect ratios of 1:2. 1:31, and 1:57, are analyzed for uniaxial compression, pure bending, and buckling. Under the uniaxial compression with a strain of $1.0{\times}10^{-4}$ as the reference state, it is predicted that all three types of nanowires develop the same magnitude of the piezoelectric fields, which suggests that longer nanowires exhibit higher piezoelectric potential. However, this prediction is not in agreement with the experimental results previously reported in the literature. Such discrepancy is understood when the piezoelectric behaviors under bending and buckling are considered. When only the strain field due to bending is present in bending or buckling, the antisymmetric nature of the through-thickness stain distribution indicates that two piezoelectric fields, the same in magnitude and opposite in sign, develop along the thickness direction, which cancels each other out, resulting in a zero net piezoelectric field. Once additional strain contribution due to axial deformation is superposed on the bending, such field cancelling is compensated for due to the axial component of the piezoelectric field. Such numerical predictions seem to explain the reported experimental results while providing a guideline for the design of nanowire-based piezoelectric devices.
Field monitoring techniques offer an attractive approach for understanding bridge behavior under in-service loads. However, the investigations on bridge behavior under high-speed train load using field monitoring data are limited. The focus of this study is to explore the structural behavior of an in-service long-span steel truss arch bridge based on field monitoring data. First, the natural frequencies of the structure, as well as the train driving frequencies, are extracted. Then, the train-induced bearing displacement and structural strain are explored to identify the effects of train loads and bearings. Subsequently, a sensitivity analysis is performed for the impact factor of strain responses with respect to the train speed, train weight, and temperature to identify the fundamental issues affecting these responses. Additionally, a similar sensitivity analysis is conducted for the peak acceleration. The results indicate that the friction force in bearings provides residual deformations when two consecutive trains are in opposite directions. In addition, the impact factor and peak acceleration are primarily affected by train speed, particularly near train speeds that result in the resonance of the bridge response. The results can provide additional insight into the behavior of the long-span steel truss bridges under in-service high-speed train loads.
본 연구에서는 지중배전선로를 건설함에 있어 전력케이블용 보호관으로 사용되고 있는 PE파형관의 매설깊이별 작용압력-변형률 특성을 분석하기 위해 전산해석 및 실내 모형토조실험을 수행하였으며, 더불어 고전이론에 의한 해석과 비교 분석하여 지중연성관이 어느 매설깊이에서 안전하고 그 때의 거동특성이 어떤 양상을 보이는지 규명하였다. 실내 모형토조실험에 사용된 토조는 현장조건을 정확히 모사하기 위해 대규모로 제작(350$\times$220$\times$300cm)하였으며, 토조의 크기는 전산해석값을 이용하여 벽체에 의한 응력의 반발이 없도록 치수를 결정하였다. 가압장치로는 최대 25ton까지 압력을 가할 수 있는 Actuator시스템을 이용하였다. 각 해석방법에 적용된 매설형태는 A-1급 도로포장 아래 에 PE파형관이 묻히고, 임계하중으로 DB-24차량이 매설관 상단 중앙에 정차했을 때 관이 받는 작용압력-변형률 특성을 분석하였다. 해석결과, KS-C8455에 명시되어 있는 지중연성관의 허용변형률 기준인 3.5%에 만족하는 매설깊이는 약 40cm 이상인 것으로 판명되었고, 실내 모형토조실험 결과값에 대한 회귀분석을 통해 우리나라 지중배전선로의 현장조건에 잘 적용될 수 있는 지중연성관의 매설깊이에 따른 변형률 상관공식을 제안하였다.

Recent human gut microbiome studies have supported that the genus Bifidobacterium is one of the most beneficial bacteria for human intestinal health. To develop a new probiotic strain for functional food applications, fourteen fecal samples were collected from healthy Koreans and the strain BCBR-583 was newly selected and isolated from a 25-year-old Korean woman's fecal sample using the selective medium for Bifidobacterium. Subsequent fructose-6-phosphate phosphoketolase (F6PPK) test and 16S rRNA gene sequencing analysis of the strain BCBR-583 confirmed that it belongs to B. longum subsp. longum. The stress resistance tests showed that it has oxygen and heat tolerance activities (5- and 3.9-fold increase for 24 h at 60 and 120 rpm, respectively; $78.61{\pm}6.67%$ survival rate at $45^{\circ}C$ for 24 h). In addition, gut environment adaptation tests revealed that this strain may be well-adapted in the gut habitat, with gastric acid/bile salt resistance ($85.79{\pm}1.53%$, survival rate under 6 h treatments of gastric acid and bile salt) and mucin adhesion ($73.72{\pm}7.36%$). Furthermore, additional tests including cholesterol lowering assay showed that it can reduce $86.31{\pm}1.85%$ of cholesterol. Based on these results, B. longum BCBR-583 has various stress resistance for survival during food processing and environmental adaptation activities for dominant survival in the gut, suggesting that it could be a good candidate for fermented food applications as a new probiotic strain.

Candida albicans is an opportunistic fungus possessing multiple virulence factors controlling pathogenicity. Cell wall proteins are the most important among these factors, being the first elements contacting the host. Ddr48 is a cell wall protein consisting of 212 amino acids. A DDR48 haploinsufficient mutant strain was previously found necessary for proper oxidative stress response and drug resistance. In this study, we aimed to further elucidate the role of Ddr48 by performing additional phenotypic characterization assays. A combinatory proteomic and bioinformatics approach was also undertaken to determine differentially expressed cell wall proteins. Results showed that the mutant strain exhibited a 10% decrease in adhesion mirrored by a 20% decrease in biofilm formation, and slight sensitivity to menadione, diamide, and SDS. Both strains showed similar hyphae formation, virulence, temperature tolerance, and calcofluor white and Congo red sensitivities. Furthermore, a total of 8 and 10 proteins were identified exclusively in the wild-type strain grown under filamentous and non-filamentous conditions respectively. Results included proteins responsible for superoxide stress resistance (Sod4 and Sod6), adhesion (Als3, Hyr4, Pmt1, and Utr2), biofilm formation (Hsp90, Ece1, Rim9, Ipp1, and Pra1) and cell wall integrity (Utr2 and Pga4). The lack of detection of these proteins in the mutant strain correlates with the observed phenotypes.
