Transcriptional Regulation of Escherichia coli serC-aroA Operon : Further Support for cAMP-Dependent Expression

  • The Escherichia coli mixed-function serC-aroA operon encodes biosynthethic enzymes for unrelated pathways leading to the syntheses of serine and aromatic amino acids. It has been proposed that the operon is expressed in a cAMP-dependent manner. In this work experiments were performed to investigate the cAMP-dependent expression of the operon. Exogenous cAMP increased ${\beta}$-galactosidase synthesis in the $cya^+$ and cya strains harboring the serC-aroA-lac fusion plasmid. This enhancement was more dramatic in the $cya^-$ strain grown in a minimal medium. In a dot blot assay the serC-aroA mRNA content increased in a concentration-dependent pattern after the addition of exogenous cAMP. The activity of phosphoserine aminotransferase, encoded by the serC gene, apparently increased in E. coli cells after the addition of cAMP. All results obtained confirmed that the expression of the E. coli serC-aroA operon is positively regulated by cAMP at the level of transcription.

  • The purpose of research is to reveal the basic factors which innovation behaviors is motivated in the small-and-medium sized company for strengthening the competitiveness. This study empirically investigates and I was able to obtain participations of 114 questionnaire, and derived statistics by means of SPSS/PC version 18.0. In this study I found that strategic approach of organization and educational supports are significant to effective shop-floor process control. Also, results of process control are significant to customer satisfaction and business performance.

Survey of Maternity wear Purchasing Status in Pregnant Women (임산부의 임부복 구매 실태)

  • The purpose of this study is to most appropriately develop maternity wear according to the preferences of specific age groups of pregnant women. The specific age groups were women in their twenties, early thirties, and late thirties. This survey was conducted in the metropolitan area around Seoul, South Korea through a questionnaire given to 150 pregnant women. The study found that pregnant women purchased significantly cheaper clothes after their pregnancy than before. The participants responded that they mostly bought clothes using the Internet, but they also reported many instances of returning the clothes due to fit and sizing. It was shown that women in their twenties tended to consider the price, design, and activity more than women in their thirties when purchasing maternity wear. The study showed that pregnant women tended to wear maternity wear from early in the pregnancy-preferring leggings, one-piece dresses, pants, shirts, briefs, and bras with the girdle on the outside. More women in their late thirties purchased maternity support belts than younger age groups. In any case, in purchasing maternity wear, clothing features that the respondents focused on were comfort, functionality, fit, the covering of their body changes, and fashion. Still, many respondents 35 years and older reported more interest in safety rather than fashion in comparison to the responses of respondents under the age of 35.

Peroxiredoxin 6 Promotes Lung Cancer Cell Invasion by Inducing Urokinase-Type Plasminogen Activator via p38 Kinase, Phosphoinositide 3-Kinase, and Akt

  • The peroxiredoxin family of peroxidase has six mammalian members (Prx 1-6). Considering their frequent up-regulation in cancer cells, Prxs may contribute to cancer cells' survival in face of oxidative stress. Here, we show that Prx 6 promotes the invasiveness of lung cancer cells, accompanied by an increase in the activity of phosphoinositide 3-kinase (PI3K), the phosphorylation of p38 kinase and Akt, and the protein levels of uPA. Functional studies reveal that these components support Prx 6-induced invasion in the sequence p38 kinase/PI3K, Akt, and uPA. The findings provide a new understanding of the action of Prx 6 in cancer.

A Study of Development and Evaluation of Tutorial Management Strategy for Web-based Nursing Education (웹 기반 간호 교육을 위한 튜터의 운영 전략 개발 및 효과 검증 연구)

  • Purpose: This study was attempted and completed in order to settle down and qualitatively improve web-based nursing education by evaluating effect and managing strategy of tutor. Method: The development of tutor's managing strategy was based on "The Self-regulated Learning" and "The supportive Learning", then it was applied to 79 learners in one of the cyber-learning centers. After applying the tutor's managing strategy, self-regulated learning scale, attitude for school, preference for computer and academic achievement were evaluated. The development of tutor's managing strategy for web-based nursing education are consisted of participation promotion, psychological support and motivation, recognition and promotion strategy of learning activity, management strategy of evaluating stage. Result: The levels of learner's self-regulated learning, recognition, behavior, attitude on the school and learning achievement were meaningfully increased in statistics after applying for the managing strategy of tutor. The motivation level and learning participation kept high scores from the beginning with no significant statistical changes. Conclusion: It is required to develop an educational program for cultivating well-educated tutors in order to help the effective learning process of nurses based on understanding characteristics of learners.

A Study on Factor Related to Fatigue in Cancer Patients Receiving Radiotherapy (방사선 치료를 받는 암환자의 피로 관련 요인간의 상관관계 연구)

  • Purpose: The purpose of this study is to investigate fatigue and its related factors in cancer patients receiving radiotherapy. Method: The subjects of this study consisted of 98 patients receiving radiotherapy. Subjects were recruited from C University Hospital radiation oncology unit located in Gwangju from March to May, 2001. Questionnaire and medical records were used for data collection. The obtained data was analyzed using SAS program that included descriptive statistics, t-test, ANOVA, Post-hoc test(Fisher's LSD) and Pearson's correlation coefficients. Result: The fatigue perceived by the subjects was middle level ($5.59{\pm}1.59$) and 72.4% of them reported greater than 5 points. The subjects in no religion, low income, and spouse caregiver groups experienced the higher fatigue than another groups, respectively. The subjects in nasopharyngeal cancer, head & neck radiation site, and analgesics medication groups did, experience fatigue as well. The fatigue not only positively correlated with symptom distress, disruption of usual activity, sleep dissatisfaction, and mood state, but also negatively with less family support. Conclusion: Cancer patients receiving radiotherapy experience the middle level of fatigue and it correlates with the multi-dimensional factors. However, further research is needed to identify the changes in fatigue over the radiotherapy period through longitudinal design and to develop nursing intervention for fatigue decrease.

Inhibitory Aromatase Effects of Flavonoids from Ginkgo Biloba Extracts on Estrogen Biosynthesis

  • Ginkgo biloba extract (GBE) is a popular phytomedicine and has been used for disorders of the central nervous system, cardiovascular, renal, respiratory, and circulatory diseases. Although GBE is a complex mixture of over 300 compounds, its major components are 24% flavonoids and 6% terpene lactones. In this study, we tested the inhibitory effects of the three major flavonoids (kaempferol, quercetin, and isorhamnetin) from GBE, independently and as mixtures, on aromatase activity using JEG-3 cells (human placental cells) and recombinant proteins (human placental microsome). In both systems, kaempferol showed the strongest inhibitory effects among the three flavonoids; the flavanoid mixtures exerted increased inhibitory effects. The results of exon I.1-driven luciferase reporter gene assays supported the increased inhibitory effects of flavonoid mixtures, accompanied by suppression of estrogen biosynthesis. In the RT-PCR analysis, decreased patterns of aromatase promoter I.1 mRNA expressions were observed, which were similar to the aromatase inhibition patterns of flavonoids and their mixtures. The present study demonstrated that three flavonoids synergistically inhibit estrogen biosynthesis through aromatase inhibition, decrease CYP19 mRNA, and induce transcriptional suppression. Our results support the usefulness of flavonoids in adjuvant therapy for breast cancer by reducing estrogen levels with reduced adverse effects due to estrogen depletion.

  • The user interface design needs to support the complex interactions between human and computers. It also requires comprehensive knowledges many areas to collect customer's requirements and negotiate with them. The user interface designer needs to be a graphic expert, requirement analyst, system designer, programmer, technical expert, social activity scientist, and so on. Therefore, it is necessary to research on an designing methodology of user interface for satisfying various expertise areas. In the paper, We propose the 4 business event's abstract object visualizing phases such as fold abstract object modeling, task abstract object modeling, transaction abstract object modeling, and form abstract object modeling. As a result, this modeling method allows us to enhance visual cohesion of UI, and help unskilled designer to can develope the higy-qualified user interface.

Keumsa Linteusan Suppresses Invasion of Cancer Cells through the Inhibition of Cellular Adhesion and MMP-9 Expression

  • Extracts derived from various medical mushrooms have been reported to have antitumor and immuno-modulatory properties. In order to investigate the antitumor activity of keumsa Linteusan, the water extract of Phellinus Iimteus, HT1080 cells, a human fibrosarcoma cell line, were treated with it and changes in cellular migration potential was tested in vitro. At a concentration range below 1,000 $\mu$g/mL, Linteusan blocked, in a dose dependent manner, the migration of cells through Matrigel as well as Boyden chamber without affecting the viability of the cells. Prolonged treatment of HT1080 cells with Linteusan suppressed TNFa induced production of matrix metalloproteinase (MMP)-9 as well as basal level expression of MMP-2. Linteusan also affected the adhesion of the cells to fibronectin-coated surfaces. The effect of Linteusan on cell signaling pathways was also tested. Linteusan specifically affected TNF-$\alpha$ induced phosphorylation of AKT in a dose-dependent manner, while phosphorylation levels of ERK remained unaffected. These data indicate that Linteusan blocks the migration of HT1080 cells by affecting various processes associated with cell migration such as the expression of matrix degradingenzymes,cell adhesion, and AKT-medicated cellular signaling pathways.

The geophysical survey and rock classification suitable for the design of a tunnel in urban area passing underneath the Gyeongbu Line(Railload) (경부선 직하부를 나란히 통과하는 도심지 터널에 있어서 지표물리탐사 및 암반등급 평가 사례 연구)

  • Urban conditions such as underground facilities and ambient noises due to cultural activity restrict the application of conventional geophysical techniques in general. We used the linear array microtremor technique which uses these noises as strong energy source. The result parameter of the survey is shear wave velocity profile which had been applied as an fundamental information for the determination of the rock support type in tunnel design. This study was the first case in Korea which utilized a surface geophysical technique yielding successful result in urban area especially under the existing rail ways. The quantitative relation between the shear wave velocity from this method and the rock mass rating(RMR) determined from the inspection of the cores recovered from the same boreholes showed high statistical relationship. These correlations were then used to relate the shear-wave velocity to RMR along the entire profile.
