  • A bacterial strain capable of synthesizing polyhydroxyalkanoates (PHAs) with an unusual pattern of monomer units was isolated from activated sludge using the enrichment culture technique. The organism, identified as Pseudomonas aeruginosa P-5, produced polyesters consisting of 3-hydroxyvalerate and medium-chain-length (MCL) 3-hydroxyalkanoate monomer units when $C_{-odd}$ alkanoic acids such as nonanoic acid and heptanoic acid were fed as the sole carbon source. Solvent fractionation experiments using chloroform and hexane revealed that the 3-hydroxyalkanoate monomer units in these polyesters were copolymerized. The molar concentration of 3-hydroxyvalerate in the polyesters produced were significantly elevated up to 26 mol% by adding 1.0 g/L valeric acid as the cosubstrate. These copolyesters were sticky with low degrees of crystallinity. The PHA synthase genes were cloned, and the deduced amino acid sequences were determined. P. aeruginosa P-5 possessed genes encoding MCL-PHA synthases (PhaC1 and PhaC2) but lacked the short-chain-length PHA synthase gene, suggesting that the MCL-PHA synthases from P. aeruginosa P-5 are uniquely active for polymerizing (R)-3-hydroxyvaleryl-CoA as well as MCL (R)-3-hydroxyacyl-CoAs.

  • Natural products are attractive as the source of new drug development. Especially, numerous unknown marine bioresources are an object of attention because the ocean occupies three fourth of the earth. Survival of marine bioresources in extreme environment may induce the production of biological active compounds. As previous study, we examined over 40 specimens of marine sponges collected from Micronesia and screened their anti-proliferative activities in various cancer cell lines. Among them, we investigated Coscinoderma sp.'s activity and mechanism in human colon carcinoma HCT116 and RKO cells. Furthermore, we also used the p53-knockout of HCT116 cells and the p53 loss of RKO cells for elucidating the role of p53. Coscinoderma sp. inhibited cellular viability independently of the p53 status. Therefore, we compared the expression level of cell death-related proteins by Coscinoderma sp. in HCT16 and in HCT116 p53KO cells. Coscinoderma sp. increased p53 level and NOXA levels and induced apoptosis under the condition of p53 existence. On the other hand, Coscinoderma sp. increased p21 and mTOR levels in HCT116 p53KO cells. These results suggest that Coscinoderma sp. induced anti-proliferation effect through different pathway depending on p53 status.

  • The fructus of Crataegus pinnatifida Bunge (CBF) has been used as medicinal and food source in worldwide. In this study, antimicrobial activity of the methanol extract and its sequential organic solvent fractions of CBF against different pathogenic bacteria and fungi, including multidrug resistant Pseudomonas aeruginosa and Candida sp., were investigated. The methanol extract of CBF was active against various gram-positive and gram-negative bacteria, and the ethylacetate and butanol fractions of CBF showed strong antibacterial activity against Listeria monocytogenes, Staphylococcus epidermidis, Staphylococcus aureus, Bacillus subtilis, Salmonella typhimurium, Proteus vulgaris, Escherichia coli and various multidrug resistant Pseudomonas aeruginosa with minimal inhibitory concentration of 1.0~7.5 mg/mL. Also the fractions showed anti-Candida activity against C. albicans, C. kruseis and C. geochares. The methanol extract of CBF and its solvent fractions, except n-hexane fraction, did not show any hemolytic activity against human red blood cell up to $500\;{\mu}g/mL$, respectively. The hemolysis in n-hexane fraction at $500\;{\mu}g/mL$ was less than 9.9%. Our results suggest that the CBF could be developed as a potent antibacterial agent, especially for multidrug resistant Pseudomonas aeruginosa.

  • Sphene U-Pb ages were determined for the granodiorites from Gimcheon, Seongju and Anui areas of the middle Yeongnam massif. The determined ages were in the narrow range of 195.7±2.4∼200.8±1.9(2σ) Ma that are approximately coincident with the boundary between Triassic and Jurassic. Even though the studed plutons are aerially separated, they reveal quite similar major element compositions and almost identical ages, suggesting that they were generated from the similar source materials under the identical tectonic environment and thus they can be considered to form a single suite. Considering the age and spatial distribution of the Triassic to Lower Jurassic plutons of the Yeongnam Massif and Okcheon Belt, it seems that there were episodic changes in tectonic environment in both areas with relatively short intervals. In general, the compressive environment of active continental margin was prevailed. However, the tensional environment of within-plate was also appeared several times intermittently. In conclusion, Yeongnam Massif and Okcheon Belt experienced distinct tectonic environments during Triassic to Lower Jurassic, providing important clue to reveal the crustal evolution of the Korean Peninsula.

  • In this review we discussed the effective ways to catalytically derive value-added chemicals from propane which has been utilized only as an energy source so far. Among various propane-derived products, the most valuable chemicals such as propylene and acrylonitrile were mainly focused herein. Propylene could be manufactured through oxidative dehydrogenation of propane using $O_2,\;CO_2$, etc. as an oxidant for the purpose of overcoming thermodynamic limitations of propane dehydrogenation. On the other hand, propane ammoxidation would be an alternative to propylene ammoxidation for producing acrylonitrile since propane is much cheaper than propylene as a starting material. Although effective $MoVTeNbO_x$ catalysts have been developed fur propane ammoxidation in recent years, more detailed studies should be thoroughly performed. In carrying out both oxidative dehydrogenation and ammoxidation of propane fur a long period, the most critical issue is definitely considered to find out the most active and selective catalysts, which makes it possible to commercialize both reactions into economically viable processes.

  • Chrysanthemum zawadskii, a herbaceous perennial plant belonging to the Compositae, grows wild in Asian countries, including Japan, China, and Korea. The biological, antioxidative, anti-inflammatory, and antibacterial activities of C. zawadskii have been reported, its antiobesity activity has not been elucidated. In the present study, the effect of C. zawadskii methanol extract (CZME) on pancreatic lipase enzyme activity, adipocyte differentiation, and adipogenesis was investigated using an in vitro assay and a cell model system. CZME effectively suppressed lipase enzyme activity in a dose-dependent manner. CZME also inhibited insulin, dexamethasone, 3-isobutyl-1-methylxanthine (MDI)-induced adipocyte differentiation, lipid accumulation, and the level of triglyceride in 3T3-L1 preadipocytes in a dose-dependent manner, without cytotoxicity. The antiobesity effect of CZME might be modulated by gene and protein expression of cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins (C/EBP) α, C/EBPβ, and the peroxisome proliferator-activated receptor γ (PPAR γ). CZME also triggered lipolysis in a dose-dependent manner in MDI-induced 3T3-L1 preadipocytes. Taken together, these results provide important new insights into the antiobesity activities of C. zawadskii, showing that they involve pancreatic lipase inhibition, as well as antiadipogenic and lipolysis effects. CZME might be a promising source in the field of nutraceuticals. However, the active compounds that confer the antiobesity activities of CZME need to be identified.

  • In this study, the anti-obesity activity of Smilax china methanol extract (SCME) was evaluated using a pancreatic lipase enzyme inhibition assay, and a cell culture model system. Results indicated that, SCME effectively inhibited pancreatic lipase enzyme activity in a dose-dependent manner. Furthermore, SCME significantly suppressed insulin, dexamethasone, 3-isobutyl-1-methylxanthine-induced adipocyte differentiation, lipid accumulation, and triglyceride contents on 3T3-L1 preadipocytes, in a dose-dependent manner. The anti-adipogenic effect was modulated by cytidine-cytidine-adenosine-adenosine-thymidine (CCAAT)/enhancer binding proteins (C/EBP) ${\alpha}$, $C/EBP{\beta}$, and the peroxisome proliferator-activated receptor ${\gamma}$ gene and protein expressions. Moreover, SCME triggered lipolysis effects dose-dependently on adipocyte. Taken together, these results provide an important new insight into SCME, indicating that it possesses anti-obesity activity through pancreatic lipase inhibition, anti-adipogenic and lipolysis effects. SCME may therefore be utilized as a promising source in the field of nutraceuticals. The identification of active compounds that confer the anti-obesity activities of SCME may be a logical next step.

  • In recent years, many institutions predict that cloud services and big data will be popular IT trends in the near future. A number of leading IT vendors are focusing on practical solutions and services for cloud and big data. In addition, cloud has the advantage of unrestricted in selecting resources for business model based on a variety of internet-based technologies which is the reason that provisioning and virtualization technologies for active resource expansion has been attracting attention as a leading technology above all the other technologies. Big data took data prediction model to another level by providing the base for the analysis of unstructured data that could not have been analyzed in the past. Since what cloud services and big data have in common is the services and analysis based on mass amount of data, efficient operation and designing of mass data has become a critical issue from the early stage of development. Thus, in this paper, I would like to establish data processing architecture based on technological requirements of mass data for cloud and big data services. Particularly, I would like to introduce requirements that must be met in order for distributed file system to engage in cloud computing, and efficient compression technology requirements of mass data for big data and cloud computing in terms of cost-saving, as well as technological requirements of open-source-based system such as Hadoop eco system distributed file system and memory database that are available in cloud computing.
